Clarithromycin is a new, safe orally administered macrolide antibiotic active against Mycoplasma sp. in humans. Single-dose and multidose pharmacokinetic parameters were determined for clarithromycin in wild-caught desert tortoises (Gopherus agassizii) seropositive for M. agassizii. Clarithromycin blood levels were measured in three tortoises for up to 72 hr after a single oral dose of 7.5 mg/kg. In a second group of six tortoises, levels were measured after a dose of 15 mg/kg. Noncompartmental iterative two-stage Bayesian and nonparametric expectation maximization pharmacokinetic parameters were determined for each animal assuming first order rate constants. At 15 mg/kg, the maximum concentration was 1.37 microg/ml, the time to maximum concentration was 8.0 hr, and a plasma half-life of 11.69 hr was derived from the latter method. The absorption constant was 0.08/hr, the absorption half-life was 8.47 hr, and the weight-normalized volume of distribution was 5.30 L/kg. Predictions derived by the latter method suggested a dosage of 15 mg/kg p.o. every 24 hr to achieve maximal blood levels of > or =1 microg/ml for multiple dosing. However, results from a preliminary multidose study with three tortoises indicate that the drug is accumulated; therefore, the predicted dose may be closer to 15 mg/kg p.o. every 2-3 days to maintain blood levels of 2-7.5 microg/ml. (For n = 3, 2-point linear regression median estimates for the apparent elimination rate constant (K) and half-life are 0.0227/hr and 30.52 hr, respectively.) This multidose accumulation reflects a slower apparent elimination than that predicted in the eight single-dose tortoises (i.e., K = 0.0593/hr, t1/2 = 11.69 hr). Ferrihydrite occurs in soils undergoing rapid weathering, and in soils containing soluble silicate or organic anions which inhibit the formation of more crystalline iron oxides. Because of its very high specific surface area and adsorptive capacity (analogous to allophane), ferrihydrite can profoundly influence soil properties, even if present in only low concentrations. Ferrihydrite was recognised as a mineral by the International Mineralogical Association in 1975. Its structure and chemical formula, however, are not yet clearly understood. Most evidence to date indicates hexagonal-close-packed layers of O2′, OH, and H2O with Fe(III) occupying octahedral positions and giving a trigonal unit cell (a=0.508 nm; c=0.94 nm). Some samples appear to have only a partially ordered structure and uncertainty exists as to how to name such material. Natural ferrihydrites commonly contain up to about 9% Si and the role and location of silicate are subjects of active research. At concentrations greater than 5-10%, ferrihydrite in soils can usually be identified by X-ray diffraction. At lower concentrations, a combination of methods can be indicative. Acid-oxalate-extractable iron is a convenient and often useful indicator of the presence and quantity of ferrihydrite in a soil, though it cannot be regarded as a means of positive identification. 相似文献

A large number of Bartonella species and genetic variants were compared for their ability to cause bacteremia in different rodent species: the cotton rat (Sigmodon hispidus), white-footed mouse (Peromyscus leucopus), BALB/c mouse and Wistar rat. Experimental data supported field observations that host specificity can occur among certain Bartonella species and rodent species. Bacteremia could only be readily produced in cotton rats or white-footed mice if the strains used for inoculation were originally obtained from the same species or from a phylogenetically close species. A few Bartonella colonies could be observed in the blood of some BALB/c mice by 7 days after inoculation, but no evidence of the persistence of the infection was found. Host specificity suggests the possibility of a long co-speciation of Bartonella species with their rodent hosts. Host–parasite relationships measured by the duration and level of bacteremia and the minimal infectious dose may serve as additional criteria for classification of Bartonella isolates obtained from natural environments. 相似文献

Fifteen middle-aged to older, overweight cats attending a first-opinion clinic were investigated to rule out hyperadrenocorticism as a cause of their weight problem, using two different protocols for the adrenocorticotropic hormone (ACTH) stimulation test. The cats received intravenous synthetic ACTH (tetracosactrin) at an initial dose of 125 microg; a second test was performed between two and three weeks later, using a dose of 250 microg intravenously. The mean basal serum cortisol concentration was 203 nmol/litre (range 81 to 354 nmol/litre). The highest mean serum cortisol concentration occurred at 60 minutes following the 125 microg dose and at 120 minutes following the 250 microg dose. There was, however, no statistically significant difference between these peak cortisol concentrations attained using either dose of tetracosactrin. A significantly higher mean serum cortisol concentration was attained after the higher dose at the 180 minutes time point, indicating a more prolonged response when compared with the lower dose. The cats were followed up for one year after the initial investigations and none were found to develop hyperadrenocorticism during this time. 相似文献

Background
Dogs are a unique model for examining the effects of exercise on vitamin D status because of their lack of vitamin D synthesis by UV exposure. In addition, the inflammatory response may be associated with hypovitaminosis D.Objectives
To investigate the effects of several days of endurance exercise on plasma vitamin D (25‐(OH)D3, 24,25‐(OH)D3 and 1,25(OH)D3) and serum C‐reactive protein (CRP) concentrations in stage‐stop racing sled dogs.Animals
12 racing sled dogs and 8 control dogs.Methods
Blood was collected before the race and immediately after racing on days 2 and 8. Plasma vitamin D metabolites and serum CRP concentrations were measured.Results
Racing dogs showed a significant increase in 25(OH)D3 on day 2 (P = .027) and day 8 of the race (P < .001), whereas no increases were observed in control dogs. The plasma concentration of 24,25(OH)D3 showed a significant increase by day 8 (P < .001). There were no significant changes in 1,25(OH) D3 concentrations across all time points and groups. Racing dogs had significantly increased CRP concentrations by day 2 (39.3 ± 30.1 μg/mL; P < .001).Conclusions and Clinical Importance
Increases in vitamin D metabolites as well as increases in CRP concentrations were observed in racing sled dogs. This finding was contrary to the hypothesis that decreases in vitamin D status in athletes may be related to the acute phase inflammatory response during exercise. In addition, the increased 24,25(OH)D3 concentrations compared to what is observed in other species suggests metabolic variations in dogs that lead to enhanced disposal of vitamin D. 相似文献28.

Optically pumped nuclear magnetic resonance (OPNMR) measurements were performed in two different electron-doped multiple quantum well samples near the fractional quantum Hall effect ground state nu = 13. Below 0.5 kelvin, the spectra provide evidence that spin-reversed charged excitations of the nu = 13 ground state are localized over the NMR time scale of about 40 microseconds. Furthermore, by varying NMR pulse parameters, the electron spin temperature (as measured by the Knight shift) could be driven above the lattice temperature, which shows that the value of the electron spin-lattice relaxation time tau1s is between 100 microseconds and 500 milliseconds at nu = 13. 相似文献

Halogen atoms from the reactions of sea-salt particles may play a significant role in the marine boundary layer. Reactions of sodium chloride, the major component of sea-salt particles, with nitrogen oxides generate chlorine atom precursors. However, recent studies suggest there is an additional source of chlorine in the marine troposphere. This study shows that molecular chlorine is generated from the photolysis of ozone in the presence of sea-salt particles above their deliquescence point; this process may also occur in the ocean surface layer. Given the global distribution of ozone, this process may provide a global source of chlorine. 相似文献

BACKGROUND: Tribolium castaneum (red flour beetle) is a serious insect pest of stored products around the world. Current control measures for this species have several limitations: loss of registration of insecticides, insecticide resistance and consumer concerns about chemical residues in food. The objective of this study was to determine whether methoprene affects progeny production of T. castaneum. Late‐instar larvae or young adults were exposed to methoprene‐treated wheat, and progeny production was determined. The pairing of male and female adults was performed as untreated × untreated, treated × untreated or treated × treated, to study sex‐based effects. RESULTS: There were three outcomes to late‐instar larvae held on methoprene‐treated wheat kernels (0.001 and 0.0165 ppm): (1) failure to emerge as an adult; (2) emergence as an adult, and almost no offspring produced; (3) emergence as an adult and normal production of offspring. Male larvae were more susceptible to methoprene than female larvae. In contrast, young adults exposed to methoprene (1.67–66.6 ppm) showed no reduction in offspring production. CONCLUSION: Methoprene concentrations will decline with time following its application. However, this research indicates that methoprene can still reduce populations of T. castaneum by reducing their progeny production, even if adults emerge.
