Reasons for performing study: An improvement in sperm quality after single layer centrifugation (SLC) has been seen in previous studies using small sample sizes (for example, n = 10 stallions). There is a need to investigate whether this improvement is repeatable over several breeding seasons with a larger number of stallions (n ≥ 30 stallions). Objective: To make a retrospective analysis of the results of SLC performed on more than 250 sperm samples (176 ejaculates) from 31 stallions in 3 consecutive breeding seasons. Methods: Sperm quality (motility, proportion of morphologically normal spermatozoa and the proportion of spermatozoa with undamaged chromatin) was assessed before and after SLC. Results: All parameters of sperm quality examined were significantly better in sperm samples after SLC than in their unselected counterparts (P<0.001 for each parameter). The yield of spermatozoa obtained after SLC was influenced by the type of extender used and also by the concentration of spermatozoa in the original ejaculate, with fewer spermatozoa being recovered when the loading dose contained a high concentration of spermatozoa. The optimal concentration was approximately 100 × 106/ml. Sperm concentration in the samples loaded on to the colloid influenced the sperm yield while the type of semen extender affected sperm quality and survival. Furthermore, the scaled‐up SLC method was found to be suitable for use with a range of ejaculates, with similar sperm kinematics being observed for standard and scaled‐up preparations. Conclusions: SLC consistently improved the quality of stallion sperm samples from a large number of ejaculates. The method could be scaled‐up, allowing larger volumes of ejaculate to be processed easily from a wide range of stallions. 相似文献

The Less Favoured Area (LFA) scheme is a major element of the EU Rural Development Policy, aimed at supporting farming in areas with natural handicaps or low soil productivity. It has been in place since 1975 and accounts for 14% of total Community funding. In 2003, the European Court of Auditors recommended that the socio‐economic criteria on which the current scheme is based be replaced by biophysical criteria. Reviews of the proposals suggest that in Atlantic climates of Northwest Europe, the new criteria do not delineate adequately areas where agricultural productivity is constrained by the biophysical environment and that such areas are instead demarcated by the occurrence of excess soil moisture conditions. In this paper, we review the impact of excess soil moisture conditions on the sustainability of farming systems and their role in constraining strategic and tactical farm management practices. In particular, we review the scientific evidence on the impact of excess soil moisture conditions on herbage growth, herbage utilization, farm operations and environmental sustainability. On the basis of this, we propose an additional biophysical criterion for the new delineation of LFAs, namely the length of time that soil water is in excess of field capacity (‘field capacity days’). While there is no clear threshold for field capacity days above which agricultural sustainability is acutely constrained, the evidence reviewed in this paper suggests that the sustainability of intensive livestock farming and tillage systems is particularly challenging in scenarios where the 80 percentile of field capacity days exceeds 220–230 days. 相似文献

Abstract White crappie, Pomoxis annularis Rafinesque, provides important sport fisheries across North America but sometimes exhibits slow growth and fails to reach sizes acceptable to anglers. Three dissimilar white crappie populations were studied to assess whether differences in growth were associated with size and age at maturity and subsequent larval production. Similar to other species, female white crappie matured at younger ages and smaller sizes and contained larger ovaries in the population with the slowest growth; this population also had the lowest annual mortality. Larval production (number m?3) in this population was intermediate to that of the other populations. Attempts to improve growth of white crappie in the slow‐growing population will probably be hindered by these individuals’ greater reproductive investment, which reduces the amount of energy available for somatic growth. 相似文献

This study analysed the alleviating effect of elevated CO2 on stress‐induced decreases in photosynthesis and changes in carbohydrate metabolism in two wheat cultivars (Triticum aestivum L.) of different origin. The plants were grown in ambient (400 μl l?1) and elevated (800 μl l?1) CO2 with a day/night temperature of 15/10 °C. At the growth stages of tillering, booting and anthesis, the plants were subjected to heat stress of 40 °C for three continuous days. Photosynthetic parameters, maximum quantum efficiency of photosystem II (PSII) photochemistry (Fv/Fm) and contents of pigments and carbohydrates in leaves were analysed before and during the stress treatments as well as after 1 day of recovery. Heat stress reduced PN and Fv/Fm in both wheat cultivars, but plants grown in elevated CO2 maintained higher PN and Fv/Fm in comparison with plants grown in ambient CO2. Heat stress reduced leaf chlorophyll contents and increased leaf sucrose contents in both cultivars grown at ambient and elevated CO2. The content of hexoses in the leaves increased mainly in the tolerant cultivar in response to the combination of elevated CO2 and heat stress. The results show that heat stress tolerance in wheat is related to cultivar origin, the phenological stage of the plants and can be alleviated by elevated CO2. This confirms the complex interrelation between environmental factors and genotypic traits that influence crop performance under various climatic stresses. 相似文献

Quantitative determination of nematode populations in soils frequently necessitates the mixing of representative soil samples to form a homogeneous, compound sample from which the nematodes are extracted. A mixing apparatus was developed and standardized with the aid of a spectrophotometric technique by which the dispersion of a dye through the soil during mixing could be determined. It was found that the mixer is most suitable for use on sandy loam soils. Optimal mixing is obtained at a speed of 15 rpm for 2 minutes. ABSTRACTThis work investigates the impact of municipal solid waste compost (MSW-compost) application (0, 50, and 100 t/ha) on the growth, and on nutrient and trace elements content in lettuce and tomato plants grown in large, 40-L pots. Our findings showed inhibition of plants’ growth with increasing dose of MSW-compost, compared to plants receiving conventional fertilization. Growth inhibition was associated with a sharp decrease in soil NO3–N content. On the other hand, a slower decrease in soil NO3–N content occurred in non-planted pots amended with MSW-compost. These findings provide evidence that N immobilization and/or decreased N mineralization were responsible for inhibited growth by constraining N availability. With regard to the other macro-nutrients, K, P, Mg, Ca, and Fe, their contents in leaves of both crops were maintained at optimum levels. Higher zinc and copper content was measured in leaves of both crops but they did not exceed the optimum range for growth. No accumulation of trace elements was found in the fruits. The content of heavy metals in the tissues of plants grown in MSW-compost amended soil, remained at levels similar to those of the non-amended soil, suggesting that they do not pose a significant risk either for plant growth or public health. The findings of our study suggest that further emphasis should be given on the investigation of the factors regulating N mineralization and availability in order to avoid reductions in crop yield. 相似文献

A field calibration experiment was carried out on salt‐affected clayey soil in Syria, to compare the sensitivity to soil electrical conductivity (ECe), and bulk density (ρb) of two instruments for estimating soil moisture: the neutron probe (NP) and the Diviner 2000 capacitance probe (CP). The results showed that the values of the correlation coefficient of the calibration were decreased when the ECe and ρb values increased; this decrease was more pronounced for the Diviner 2000, indicating that it was more sensitive to ρb and ECe than the NP. When only scaled frequency was used in the fitted equation, the Diviner 2000 in wet soil underestimated soil water content significantly at all depths, but especially in the top layer, by up to 0.09 cm3/cm3 compared with gravimetric determinations. However, in dry soil, the Diviner 2000 overestimated the volumetric water content by up to 0.05 cm3/cm3 in the top 15 cm, and by 0.03 cm3/cm3 at 30‐45 cm depth. The performance of the neutron probe was better overall; using a factory calibration curve no significant differences were observed between NP estimates and the gravimetric values. Including both ρb and ECe in the calibration equations improved the fits, although the regression coefficient (R2) for the Diviner 2000 remained low. 相似文献

A 12-year-old castrated male cocker spaniel dog was presented with a 4-week history of left episcleral injection and pawing at the face. Clinical examination findings included left internal and external ophthalmoplegia, left dorsal strabismus, pain opening the mouth, and intermittent amaurosis. Imaging studies revealed a left orbital apex mass with adjacent sphenoid bone lysis and extension into the cranial cavity. A left exenteration was performed and histopathology confirmed an orbital soft tissue sarcoma.Key clinical message:This report describes an orbital tumor causing orbital apex syndrome. This condition should be differentiated from cavernous sinus syndrome as the latter does not course with optic neuropathy. 相似文献
