Soybean is a temperate photosensitive crop but has adapted to sub-tropical and tropical countries of lower latitudes also. Photoperiodic and maturity genes confer latitudinal adaptation in this crop. Genotyping of accessions of higher latitudes have shown the role of photoinsensitivity, conferred by recessive photoperiodic alleles (e1/e2/e3/e4), in adaptation of the crop to high latitudes but information is not available for lower latitudinal countries like India. We genotyped and calculated the photosensitivity of 101 cultivated Indian soybean varieties and found that majority of the varieties (86) were photosensitive and had the dominant alleles at these loci. Four genotypic classes (e1-as/E2/E3/E4, E1/e2/e3/E4, E1/e2/E3/E4 and E1/E2/e3/E4) were observed for varieties with recessive alleles. Photoinsensitive alleles at E1 and E2 loci significantly reduced the days to flower, maturity and photosensitivity percentage. Adaptive role of photoperiodic alleles was inferred from breeder seed requirement of these varieties for 35 years. Although the photosensitive class contributed 81% to the total seed requirement the weighted mean contribution of this class (380 Q/year) was far less than that of photoinsensitive class (648 Q/year). Photoinsensitivity is essential for perpetuation of crop in higher latitudes. Present report highlights the novel role of photoinsensitive alleles in adaptation of soybean to rainfed, short growing and sub-tropical conditions of lower latitudes by conferring earliness.

Muscle samples from 890 slaughtered pigs were examined for the presence of sarcocysts. A high prevalence rate of 67.98% was observed. Two types of microsarcocysts were recorded. The sarcocyst wall of one type had redial striations and the other possessed hair-like villar protrusions. The species were identified as Sarcocystis miescheriana and Sarcocystis suihominis; there was a slightly higher incidence of the latter species (47.11%) than of the former (43.14%). S. suihominis has been identified for the first time from pigs in India.  相似文献

Eschericia coli bacteriophages were isolated from the intestines of chickens. These phages had different lytic patterns, and were propagated in nutrient broth containing 0.4 gm calcium chloride/litre. The agar layer technique was used to determine the routine test dilution (RTD) and plaque morphology. The phages differed in their 1) morphology, 2) RTD values, and their ability to lyse E. coli strains from various animals. All phages isolated lysed human K12 E. coli strains, whereas only two phages lysed the chicken E. coli strain. Phages isolated lysed E. coli from chicken, bovine, ovine, equine, and human, but not from porcine, canine and other avian species.  相似文献

The aim of this study was to develop a competitive inhibition ELISA (CI-ELISA) for detection of antibodies to bovine viral diarrhea virus (BVDV) using the helicase domain of NS3 (non-structural) protein and monoclonal antibody (MAb) against it and to estimate its sensitivity and specificity using two commercial ELISA kits as independent references. The 45-kDa helicase domain of NS3 protein of BVDV was expressed in Escherichia coli and 18MAbs were developed against it. MAb-11G8 was selected for use in CI-ELISA on the basis of maximum inhibition (90%) obtained with BVDV type 1 infected calf serum. Based on the distribution of percent inhibition of known negative sera (n=166), a cut-off value was set at 40% inhibition. In testing 914 field serum samples of cattle (810) and buffaloes (104), the CI-ELISA showed a relative specificity of 95.75% and 97.38% and sensitivity of 96% and 94.43% with Ingenesa kit and Institut Pourquier kit, respectively. This study proved that the use of helicase domain of NS3 (45-kDa) is equally good as the whole NS3 protein (80-kDa) used in commercial kits for detection of BVDV antibodies in cattle and buffaloes.  相似文献

To assess the scope for enhancing productivity of soybean (Glycine max L. Merr.), the CROPGRO-Soybean model was calibrated and validated for the diverse soybean-growing environments of central and peninsular India. The validated model was used to estimate potential yields (water non-limiting and water limiting) and yield gaps of soybean for 21 locations representing major soybean regions of India. The average water non-limiting potential yield of soybean for the locations was 3020 kg ha−1, while the water limiting potential was 2170 kg ha−1 indicating a 28% reduction in yield due to adverse soil moisture conditions. As against this, the actual yields of locations averaged 1000 kg ha−1, which was 2020 and 1170 kg ha−1 less than the water non-limiting potential and water limiting potential yields, respectively. Across locations the water non-limiting potential yields were less variable than water limited potential and actual yields, and strongly correlated with solar radiation during the season (R2 = 0.83, p ≤ 0.01). Both simulated water limiting potential yield (R2 = 0.59, p ≤ 0.01) and actual yield (R2 = 0.33, p ≤ 0.05) had significant but positive and curvilinear relationships with crop season rainfall across locations. The gap between water non-limiting and water limiting potential yields was very large at locations with low crop season rainfall and narrowed down at locations with increasing quantity of crop season rainfall. On the other hand, the gap between water limiting potential yield and actual farmers yield was narrow at locations with low crop season rainfall and increased considerably at locations with increasing amounts of rainfall. This yield gap, which reflects the actual yield gap in rainfed environment, is essentially due to non-adoption of improved crop management practices and could be reduced if proper interventions are made. The simulation study suggested that conservation of rainfall and drought resistant varieties in low rainfall regimes; and alleviation of water-logging and use of water-logging tolerant varieties in high rainfall regimes will be the essential components of improved technologies aimed at reducing the yield gaps of soybean. Harvesting of excess rainfall during the season and its subsequent use as supplemental irrigation would further help in increasing crop yields at most locations.  相似文献

A post-transfer investigation was carried out to analyze the process of gene introgression in a conventional backcross-breeding program taking low erucic acid as target trait in Brassica juncea. FAE1 locus is involved in the elongation of oleic acid (C18:1) to erucic acid (C22:1). A high concentration of erucic acid in the seeds of Brassica species has been reported to be nutritionally undesirable. Molecular markers like AFLP markers, microsatellite markers, and gene-based SNP markers were used to determine the size of the donor parent chromosomal segments retained around the FAE1 genes in the individuals selected from different backcross generations. The genotype of the individuals was inferred from the genotype of the markers and the graphical genotypes were constructed using GGT software. Molecular marker analysis led to the identification of rare recombinants near the target locus with reduced size of introgressed segment from the donor parent. Based on the present study, we propose that marker-assisted backcross breeding in B. juncea could prove to be a promising tool for the transfer of many quality traits from unadapted East European germplasm to Indian cultivars.  相似文献
