New clonal selections with increased vine vigor and stress resistance have been identified for the potato cultivar ‘Russet Norkotah’. However, the importance of clonal variation in nitrogen (N) uptake and root morphological properties is not well known. The objective of this study was to determine the effect of N fertilization on dry matter and N accumulation and root morphological parameters of two clonal selections of ‘Russet Norkotah’. A field experiment was conducted in 2002 using the standard ‘Russet Norkotah’ clone (SRC) and Texas selection 112 (TX112) of ‘Russet Norkotah’, grown at 0 and 150 kg N ha? 1. Whole plants were excavated at 54, 76, and 96 days after planting; partitioned into tubers, vines, roots, stolons, and fruits; and their dry matter and N accumulation were determined. Soil cores were obtained from 10 spatial locations relative to the plant, and used for determination of root length (RL), root length density (RLD), root average diameter (RAD), and root dry weight (RDW). Soil inorganic N content was also measured. Nitrogen fertilization increased tuber yield and dry matter and N accumulation. Fertilizer N application did not affect RL, RLD, or RDW, but resulted in a larger proportion of roots close to the top of the potato hill. Tuber yield and dry matter and N accumulation were similar for the two clonal selections. The TX112 clone, however, partitioned more dry matter and N to vines and less dry matter and N to tubers compared with the SRC clone. Soil nitrate concentration was significantly higher for SCR than for the TX112 clone in the fertilized treatment at 54 DAP, and was low and similar between clones thereafter. Root length and RLD were significantly higher for the TX112 clone compared with SRC, and both clones had a similar spatial distribution of roots. Under the conditions of this study where moisture and disease stress were limited and under a short growing season, the larger root system and increased vine vigor of the TX112 clone did not provide any advantage in terms of plant production as either dry-matter accumulation or tuber yield.  相似文献

With the problem of parasitic nematode drug resistance increasing, vaccine development offers an alternative sustainable control approach. For some parasitic nematodes, native extracts enriched for specific proteins are highly protective. However, recombinant forms of these proteins have failed to replicate this protection. This is thought to be due to differences in glycosylation and/or conformation between native and recombinant proteins. We have exploited the free-living nematode Caenorhabditis elegans to examine its suitability as an alternative system for recombinant expression of parasitic nematode vaccine candidates. We focussed on Haemonchus contortus aminopeptidase H11 glycoprotein, which is enriched in a gut membrane fraction capable of inducing significant protection against this important ovine gastrointestinal nematode. We show that H. contortus H11 expressed in C. elegans is enzymatically active and MALDI mass spectrometry identifies similar di- and tri-fucosylated structures to those on native H11, with fucose at the 3- and/or 6-positions of the proximal GlcNAc. Some glycan structural differences were observed, such as lack of LDNF. Serum antibody to native H11 binds to C. elegans recombinant H11 and most of the antibody to rH11 or native H11 is directed to glycan moieties. Despite these similarities, no reduction in worm burden or faecal egg count was observed following immunisation of sheep with C. elegans-expressed recombinant H11 protein. The findings suggest that the di- and tri-fucosylated N-glycans expressed on rH11 do not contribute to the protective effect of H11 and that additional components present in native H11-enriched extract are likely required for enhancing the antibody response necessary for protection.  相似文献

Background

Fishing activities can influence foraging behaviour of many seabird species worldwide. Seabirds are attracted by fishing vessels which can facilitate access to demersal fish as a novel food resource that otherwise would be unavailable. On the other hand, intense fishing activities cause depletion of fish stocks with a reduction of natural prey available for seabirds. Moreover, fisheries discards can have lower nutritional value than natural prey. However, the importance of fisheries discard for seabirds and the possible implications on their foraging ecology is still poorly understood. In this study, we analysed the interactions of Scopoli’s shearwaters (Calonectris diomedea) during their foraging trips with fishing vessels. We combined the GPS and accelerometer data of shearwaters with the GPS data gathered during the same period from fishing vessels. Accelerometers allowed us to identify the main behaviours of birds.

Results

The presence of fishing vessels significantly affected the individual behaviour of Scopoli’s shearwaters. Birds increased the time spent sitting on the water within 1.28?±?0.13 km of fishing vessels likely feeding or waiting for discards. Approaches towards vessels within the interaction distance were therefore classified as an interaction and were recorded in about 40% of individuals. Birds interacting with fisheries had longer flight time during their foraging trips and covered longer distances to reach more distant foraging areas compared with individuals not approaching vessels.

Conclusions

Our results suggested that fisheries discard consumption might not be a profitable source of food for Scopoli’s shearwaters. Despite the high density of fishing vessels in the home range of Scopoli’s shearwater, most individuals did not interact with them. Accordingly, scavenging individuals showed a lower foraging efficiency than their conspecifics. Intraspecific competition for foraging areas might play an important role for the foraging decision of birds to consume fisheries discards.

This review critically examines the role of boron (B) in forests in view of recent findings on B nutrition and the continuing occurrence of B deficiency. Many perceptions about the role of B in plants and its uptake and mobility have been altered since the last review on B in forest trees in 1990. Now there is evidence for a fundamental role of B in the formation of the pectic structure in primary cell walls in plants, and further roles in membrane function are being explored. In plants, channel-mediated B uptake, active B uptake and B uptake by mycorrhizas have been shown, B transporters have been identified, and B retranslocation has been shown. We explore these findings and their consequences on forest trees and on ecosystems that they dominate. Particular emphasis is placed on B retranslocation and B in mycorrhizal symbiosis, given their importance in trees.  相似文献

BACKGROUND: McPhail traps, baited with protein food lure, are used worldwide for surveillance of many species of fruit flies. Queensland fruit fly (Qfly) Bactrocera tryoni (Froggatt) (Diptera: Tephritidae) is a native Australian fruit fly and normally monitored using Lynfield traps baited with cuelure. On some occasions, McPhail traps with wet food lures are deployed to detect female flies or to find the incursion epicentre. This paper reviews field results on the merits of Lynfield and McPhail traps for detection of male and female Qfly. RESULTS: Following release of equal numbers of sterile males and females, Lynfield traps baited with cuelure captured more Qfly males than protein autolysate or orange concentrate in McPhail traps. Significantly more male than female Qfly were captured in McPhail traps baited with protein autolysate or orange. There was no significant difference between orange concentrate lure and protein autolysate lure in attracting either males or females. Another Australian native fruit fly, Dacus newmani (Perkins), was attracted to cuelure in Lynfield traps but not to either lure in McPhail traps. CONCLUSIONS: The data obtained run counter to the reputation of McPhail traps baited with protein autolysate or orange concentrate as a specialist lure/trap combination for female Qfly. Twenty-three reserves in the Western Australian wheatbelt were examined in detail to evaluate the adequacy of the reserve system in the region for the conservation of lizards. We recorded most of the lizard species known from the region and conclude that the reserve system is at present adequate for their conservation.Lizard species richness in wheatbelt reserves does not appear to be related to isolation from adjacent uncleared land, either in space or time, but is related to the ecology of the reserve. Lizard species richness-area relationships for wheatbelt reserves are very similar to the most comparable late-Pleistocene/Holocene continental island lizard faunas; the slopes of the regression lines are not significantly different and the intercepts are only marginally significant.Multiple regression analysis shows that 75% of the variation observed in lizard species richness between reserves is accounted for by the algorithm of the number of vegetation associations on these reserves; addition of other reserve variables examined (including area) does not significantly increase this explained variation.Reserves as small as 30 ha are valuable as sanctuaries for lizards although it is suggested that 1500 ha is an optimum size. Some vegetation formations are more important for lizards than others; woodlands are most important.  相似文献

It is known that carbon (C) amendments increase microbial activity in anoxic soil microcosm studies, however the effects on abundance of total and denitrifier bacterial communities is uncertain. Quantitative PCR was used to target the 16S rRNA gene for the total bacterial community, the nosZ functional gene to reflect a broad denitrifier community, and functional genes from narrow denitrifier communities represented by Pseudomonas mandelii and related species (cnorBP) and Bosea/Bradyrhizobium/Ensifer spp. (cnorBB). Repacked soil cores were amended with varying amounts of glucose and red clover plant tissue (0–1000 mg C kg? 1 of soil) and incubated for 96 h. Carbon amendment significantly increased respiration as measured by cumulative CO2 emissions. Inputs of red clover or glucose at 1000 mg C kg? 1 of soil caused increased abundance in the total bacteria under the conditions used. There was about an approximate 2-fold increase in the abundance of bacteria bearing the nosZ gene, but only in treatments receiving 500 or 1000 mg C kg? 1 of soil of glucose or red clover, respectively. Additions of ≥ 500 mg C kg? 1 soil of red clover and ≥ 250 mg C kg? 1 of glucose increased cnorBP-gene bearing denitrifiers. Changes in abundance of the targeted communities were related to C availability in soil, as indicated by soil respiration, regardless of C source. Applications of C amendments at rates that would occur in agricultural soils not only increase microbial activity, but can also induce changes in abundance of total bacterial and denitrifier communities in studies of anoxic soil microcosms.  相似文献
