Genetic modification of nitrogen metabolism via bacterial NADPH- dependent glutamate dehydrogenase (GDH; E.C.4.1.2.1) favorably
alters growth and metabolism of C3 plants. The aim of this study was to examine the effect of expression of GDH in the cytoplasmic
compartment of Zea mays cells. The gdhA gene from Escherichia coli , that encoded a NADPH-GDH, was ligated to the ubiquitin promoter that incorporated the first intron enhancer and used to
transform Z. mays cv. ‘H99’ embryo cultures by biolistics. R0–R3 generations included selfed inbreds, back-crossed inbreds, and hybrids with
B73 derivatives. The lines with the highest GDH specific activity produced infertile R0 plants. The highest specific activity
of GDH from the fertile Z. mays plants was sufficient to alter phenotypes. Plant damage caused by the phosphinothricin in gluphosinate-type herbicides, glutamine
synthetase (GS; EC 6.1.3.2) inhibitors, was less pronounced in Z. mays plants with gdhA pat than in gusA pat plants. Germination and grain biomass production were increased in gdhA transgenic plants in the field during seasons with significant water deficits but not over all locations. Water deficit

Forest management practices often generate clear-cut patches, which may be colonized by ants not present in the same densities in mature forests. In addition to the associated changes in abiotic conditions ants can initiate processes, which do not occur in old-growth stands. Here, we analyse the effects of ants and aphid honeydew on litter solution of Norway spruce, microbial enzyme activities, and needle decomposition in a field and greenhouse experiment during summer 2003. In the field, low ant densities had relatively little effects on litter solution 30 cm away from a tree trunk, but significantly increased organic carbon concentrations and decreased inorganic nitrogen concentrations next to a trunk where ants tend to build their nests. In a greenhouse experiment, the addition of ants to lysimeters containing spruce litter significantly increased dissolved organic carbon (DOC), dissolved organic nitrogen (DON), NH4-N, NO3-N and K concentrations in litter solutions compared to the control treatment, while the simulation of aphid infestation (addition of honeydew) significantly increased DOC as a direct result of honeydew leaching, and decreased inorganic N concentrations in leachates. The presence of ants resulted in a changed composition of dissolved organic matter (DOM) with more aromatic and complex compounds, and microbial enzyme activity was significantly higher in litter extracts from the ant treatment compared to the honeydew and control treatment. However, mass loss, litter %C and %N were not affected by ants or honeydew. Our results suggest that ants have a distinct and immediate effect on solution composition and microbial activity in the litter layer indicating accelerated litter decay whereas the effect of honeydew was insignificant. 相似文献

Using the 160,000-km2 drainage basin of the Madre de Dios and Orthon rivers in the southwest Amazon as a test case, we piloted an approach for large-scale conservation planning for freshwater systems characterized by a near-complete lack of biological and physical data. We used newly available spatial and remote sensing datasets, including spaceborne optical and radar observations, and new techniques of spatial data analysis to generate subbasin (?100 km2), stream, and floodplain and wetland habitat classifications. We then generated a preliminary plan for a network of conservation areas to protect the most intact examples of representative habitat types while maximizing longitudinal and lateral connectivity. Proposed additions for freshwater conservation complement existing reserves and build on earlier conservation planning efforts for terrestrial ecosystems. In the resulting integrated plan, at least 20% of the area of each major freshwater habitat type is represented and two continuous corridors exist from the mouth of the Madre de Dios to its headwaters in the Andes. In total, we highlighted 84 currently unprotected subbasins to fulfill our representation and connectivity goals. About two-thirds of these subbasins were considered relatively undisturbed and are identified as Level I (critical management zones) or Level II (indigenous territories), whereas one-third are potentially degraded and thus were designated as Level III (threat mitigation zones). This exercise provides an example of how newly available remote-sensing datasets and analytical tools may be used to advance freshwater conservation planning, particularly in data-poor regions. 相似文献

The effect of soil fauna-mediated leaf litter (faecal pellets) versus mechanically fragmented (finely ground) leaf litter on biomass production of rice (Oryza sativa, var. Primavera) was assessed in pot tests. Rice seedlings were either grown in soil samples amended with faecal pellets of diplopods and isopods fed on leaf litter of a legume cover crop (Pueraria phaseoloides (Roxb.) Benth) and a peach palm (Bactris gasipaes) or in soil amended with finely ground leaf litter. The addition of faecal pellets caused a significant and dose-related increase in plant biomass compared to pure soil. Ground leaf litter induced a significantly smaller positive effect on plant biomass development with Pueraria litter > Bactris litter > mixed primary forest litter. In contrast, soil microbial biomass development during the 4 weeks plant test was higher in the soil amended with ground litter as compared to soil amended with feacal pellets. The results show a clear positive effect of the soil fauna on soil fertility and indicate differences in the availability of nutrients from the organic substrates to higher plants and soil microorganisms. 相似文献

Inflammation, trauma, or nerve injury may cause enduring hyperalgesia, an enhanced sensitivity to painful stimuli. Neurons in lamina I of the spinal dorsal horn that express the neurokinin 1 receptor for substance P mediate this abnormal pain sensitivity by an unknown cellular mechanism. We report that in these, but not in other nociceptive lamina I cells, neurokinin 1 receptor-activated signal transduction pathways and activation of low-threshold (T-type) voltage-gated calcium channels synergistically facilitate activity- and calcium-dependent long-term potentiation at synapses from nociceptive nerve fibers.
