Transplantation of ovarian tissue has high potential for female gamete conservation. However, optimal timing of oocyte recovery for in vitro maturation and fertilization is still critical. Therefore the aim of the present study was to use high-resolution transcutaneous ultrasonography to monitor follicular development within xenografted ovarian tissue. Ovarian cortex fragments (n=44) from domestic cats were transplanted into athymic nude rats (n=12). Graft development in the animals was assessed weekly by high frequency ultrasound (10-22 MHz) under two different FSH regimes. Blood collection for serum estradiol determination and vaginal smears were performed simultaneously. The xenografts were removed at different time points according to the ultrasound findings. The survival rate of the transplants 4 weeks after surgery was 54.5% and antral follicular growth was observed within 10 grafts from 5 different hosts (8.6 +/- 6.43 follicles per graft). Early follicle antrums could be detected from 0.4 mm onwards. The growth rate of the antral cavity was calculated from weekly measurements (0.56 +/- 0.44 mm per week). Although vaginal cells and estradiol levels followed a cyclic pattern, no correlation was found between follicular diameter, estradiol and keratinized vaginal cells. We recovered 5, 1 and 4 cumulus oocyte complexes from three different individuals during weeks 19, 21, and 23 respectively. Extrusion of a polar body (1 oocyte) and germinal vesicle break down (7 oocytes) indicated progression of maturation after in vitro culture. We conclude that ultrasonography und provided a reliable method to examine xenograft survival and follicular development within the grafts. Furthermore, this technique is suitable for assessment of the efficiency of hormonal treatment and narrowing of the optimal time frame for oocyte retrieval. According to current scientific opinion the risk of human infection with H5N1 via preparation and consumption of poultry meat is negligible.This opinion has not yet been challenged by a formal risk assessment, due to the lack of empirical data. We have developed a scenario pathway model as a conceptual framework for a formal assessment of the H5N1 risk to humans through consumption of poultry meat and parameterise the model using information derived from expert opinions. The aim of this study was to investigate whether the notion of an overall negligible risk via the oral infection route is consistent with ad hoc data and expert opinions on the relevant parameters of the model.The model is mainly based on expert opinion. A stochastic Monte-Carlo simulation was conducted which took into consideration (amongst others) the exposure and infection of chicken (broiler and layer), turkeys, ducks and geese, the probabilities of detection prior to slaughter, virus survival and contamination during slaughter, as well as during the cutting and preparation of meat in commercial plants and in private households, respectively. The empirical consumption pattern for poultry meat in Germany was taken into account in the simulation.The results show that the risk for the individual consumer is practically zero whereas up to 23 cases per year in Germany might occur if the upper (more pessimistic) ranges of the expert opinions apply.The finding of a low but non-negligible risk to the population is discussed in relation to the epidemiological information available from recent outbreaks in South East Asia. 相似文献

Pharmacokinetics of xylazine after 2-, 4-, and 6-hr durations of continuous rate infusions in horses

Intravenous (i.v.) bolus administration of xylazine (XYL) (0.5 mg/kg) immediately followed by a continuous rate infusion (CRI) of 1 mg kg−1 hr−1 for 2, 4, and 6 hr produced immediate sedation, which lasted throughout the duration of the CRI. Heart rate decreased and blood pressure increased significantly (p > .05) in all horses during the first 15 min of infusion, both returned to and then remained at baseline during the duration of the infusion. Compartmental models were used to investigate the pharmacokinetics of XYL administration. Plasma concentration–time curves following bolus and CRI were best described by a one-compartment model. No differences were found between pharmacokinetic estimates of the CRIs for the fractional elimination rate constant (Ke), half-life (t1/2e), volume of distribution (Vd), and clearance (Cl). Median and range were 0.42 (0.15–0.97)/hr, 1.68 (0.87–4.52) hr, 5.85 (2.10–19.34) L/kg, and 28.7 (19.6–39.5) ml min−1 kg−1, respectively. Significant differences were seen for area under the curve ( ) (p < .0002) and maximum concentration (Cmax) (p < .04). This indicates that with increasing duration of infusion, XYL may not accumulate in a clinically relevant way and hence no adjustments are required in a longer XYL CRI to maintain a constant level of sedation and a rapid recovery. 相似文献

BACKGROUND: Sepsis is still a major cause of death in both human and veterinary medicine. Early diagnosis is essential for appropriate treatment. Identification of patients at risk for developing sepsis is already possible in human medicine through the measurement of plasma interleukin-6 (IL-6) levels. In veterinary medicine, however, this has been investigated only in canine experimental models. OBJECTIVES: The purpose of this study was to measure IL-6 plasma levels in dogs with naturally occurring systemic inflammatory response syndrome (SIRS) and sepsis and to analyze the value of IL-6 as a predictive parameter for severity and mortality. METHODS: Included in the study were 79 dogs that had been admitted to the small animal clinics of Munich and Berlin from July 2004 to July 2005 and that satisfied the diagnostic criteria for SIRS and sepsis as defined using established parameters. Measurement of plasma IL-6 levels on days 0, 1, and 2 was performed by the use of a colorimetric bioassay based on IL-6-dependent cell growth. RESULTS: Septic foci were identified in 43 patients (septic group), and 36 patients were enrolled in the SIRS group. The frequency of positive blood cultures was 11%. The overall mortality rate was 48%. Higher plasma IL-6 levels on the day of admission were significantly correlated with a more severe degree of disease, increased mortality rate, and earlier fatality. CONCLUSIONS: Plasma IL-6 concentration is predictive of outcome in canine SIRS and sepsis and may be a valuable laboratory parameter for assessing critically ill dogs. 相似文献

Evening primrose is a valuable oilseed crop with pharmaceutical and nutritional supplementation. Due to presence of valuable and rare gamma linolenic fatty acid, research on this plant has been started since 1970s by German and Britain scientists. Information about evening primrose cultivation is going to rise. Following to pervious studies the goal of this experiment was to clarify the effect of water and temperature stresses on seed yield and seed quality. Results showed that seed yield was significantly influenced by low temperature and water deficit. Plants grown under cold stress during rosette stage produced a lower seed yield than controls. The lowest seed yield was observed in the plants grown under permanent stress. Although the oil content of seeds was not influenced by low temperature stress, water stress strongly influenced the oil content and water stress stimulates oil accumulation. Like oil content, the GLA production seems to be increased under water stress conditions. 相似文献

Pradofloxacin is a new veterinary 8-cyano-fluoroquinolone developed for use against bacterial infections in dogs and cats involving both aerobic and anaerobic bacteria. The minimal bactericidal concentrations have been determined against clinical isolates of Staphylococcus pseudintermedius, Staphylococcus aureus, Escherichia coli, Pasteurella multocida, Streptococcus canis, Proteus spp., Fusobacterium spp., Porphyromonas gingivalis and Prevotella species. A subset of these species was selected, and the in vitro rate of kill by pradofloxacin was determined. For 27 of the 30 tested aerobic strains the pradofloxacin MBC was within two doubling dilutions of the MIC. For the remaining strains, the MIC and MBC were within three to four doubling dilutions. Pradofloxacin also demonstrated bactericidal activity against all anaerobic strains, and the MBC was equal to the MIC for four of the strains, within 1 doubling dilution for three strains, within 2 dilutions for a further 3 strains and within 3 dilutions for the remaining five strains. As pradofloxacin concentration was increased, a faster rate of killing was observed; bactericidal effects were seen in all cases at concentrations ≤ 0.25 μg/mL. The bactericidal activity against the anaerobic strains was marked, of particular relevance was the complete absence of regrowth even at 48 h at concentrations as low as 0.125 μg/mL. In conclusion, pradofloxacin exhibits clear bactericidal activity in terms of MBC and kill kinetics against aerobic and anaerobic clinical isolates from dogs and cats at concentrations that are greatly exceeded within the systemic circulation after administration of the recommended therapeutic doses to the target animals. It is expected that such a rapid rate of kill will play a significant role in clinical efficacy. These data demonstrate the complete and rapid killing of anaerobic bacteria by a veterinary 8-cyano-fluoroquinolone. 相似文献

Precipitation and topsoil samples from a climate transect over the Scandinavian Mountains, Norway, were analyzed for bulk and compound‐specific δ18O values. The natural abundance of 18O in the plant‐derived hemicellulose biomarkers arabinose and xylose correlates positively with δ18O of bulk soil, but not with δ18O of precipitation. This suggests that other factors than δ18Oprec, such as evaporative 18O enrichment of leaf water, exert a strong influence on the natural abundance of 18O in soils. 相似文献

- 1. Numbering no more than 100 individuals and facing many threats, the geographically isolated Eastern Taiwan Strait population of Indo‐Pacific humpback dolphins (Sousa chinensis) is in peril. The estuarine and coastal waters of central‐western Taiwan have historically provided prime habitat for these dolphins, but environmental conditions today bear little resemblance to what they were in the past.
- 2. The humpback dolphins must share their habitat with thousands of fishing vessels and numerous factories built upon thousands of hectares of reclaimed land.
- 3. They are exposed to chemicals and sewage released from adjacent terrestrial activities. Noise and disturbance associated with construction, vessel traffic and military activities are features of everyday life for these animals.
- 4. Measures to slow the pace of habitat deterioration and reduce the many risks to the dolphins are urgently needed. As one practical step in this direction, this paper describes the habitat needs of these small cetaceans so that decision makers will be better equipped to define ‘priority habitat’ and implement much needed protection measures under the terms of local legislation.
- 5. The preferred habitat of these dolphins in Taiwan consists of shallow (<30 m), near‐shore marine waters with regular freshwater inputs.
- 6. For such a small, isolated and threatened population, ‘priority habitat’ should not be limited to areas of particularly intensive dolphin use or high dolphin density, but rather it should encompass the entire area where the animals have been observed (their current ‘habitat’), as well as additional coastal areas with similar bio‐physical features (‘suitable habitat’). Such a precautionary approach is warranted because the loss of only a few individuals could have serious population‐level consequences.
- 7. While conventional socio‐economic analysis might suggest that implementing protection measures over an area stretching ~350 km north–south along Taiwan's west coast and ~3 km out to sea would be too ‘costly’, the loss of this charismatic species from Taiwan's waters would send a troubling message regarding our collective ability to reconcile human activities with environmental sustainability.
