Surface hydrophobicity (SH) of milk proteins treated physicochemically (by heating and Maillard reaction) or modified enzymatically (by transglutaminase, lactoperoxidase, laccase, and glucose oxidase) was assessed in relation to their techno-functional properties. Heat-treatment increased SH of whey protein isolate and decreased SH of sodium caseinate and bovine serum albumin. Maillard reaction of milk proteins caused time-depended decreases of SH. Only for total milk protein reacting with glucose and lactose elevated SH-values were detected. Protein modification with transglutaminase, laccase, and lactoperoxidase strongly increased the SH of whey protein isolate and total milk protein. Incubation with glucose oxidase elevated SH values of sodium caseinate, whey protein isolate, and total milk protein. When correlating SH with techno-functional properties, a positive correlation was observed between SH and foam formation, and a negative correlation was observed between SH and foam stability as well as emulsion stability. No clear correlation was detected between SH and emulsifying activity, surface tension, viscosity, and heat stability of enzymatically and physicochemically treated milk proteins.  相似文献

Procyanidin dimers and trimers, needed as reference compounds for biological studies, have been synthesized from various natural sources using a semisynthetic approach and purified by high-speed countercurrent chromatography (HSCCC). In the past, it has been difficult to elucidate the structure of these compounds, especially the determination of the interflavanoid bond. Here, the structure of two B-type procyanidin dimers, with (+)-catechin ((+)-C) in the upper unit, and eight C-type procyanidin trimers, with (-)-epicatechin ((-)-EC) in the upper unit, have been elucidated using low-temperature (1)H NMR spectroscopy, as well as circular dichroism (CD) spectroscopy. This is the first time NOE interactions have been used to characterize the interflavanoid linkage in underivatized procyanidin trimers. Complete analyses of procyanidin C1 (-)-EC-4β→8-(-)-EC-4β→8-(-)-EC, (-)-EC-4β→8-(-)-EC-4β→8-(+)-C, (-)-EC-4β→6-(-)-EC-4β→8-(-)-EC, (-)-EC-4β→6-(-)-EC-4β→8-(+)-C, (-)-EC-4β→8-(-)-EC-4β→6-(-)-EC, (-)-EC-4β→8-(-)-EC-4β→6-(+)-C, (-)-EC-4β→8-(+)-C-4α→8-(-)-EC, procyanidin C4 (-)-EC-4β→8-(+)-C-4α→8-(+)-C, and procyanidin dimers B6 (+)-C-4α→6-(+)-C and B8 (+)-C-4α→6-(-)-EC are presented.  相似文献

Dissolved organic nitrogen and carbon (DOC) are significant in the C and N cycle in terrestrial ecosystems. Little is known about their dynamics in the field and the factors regulating their concentrations and fluxes. We followed the fluxes and concentrations of the two in a Norway spruce (Picea abies (L.) Karst.) forest ecosystem in Germany from 1995 to 1997 by sampling at fortnightly intervals. Bulk precipitation, throughfall, forest floor percolates from different horizons and soil solutions from different depths were analysed for major ions, dissolved organic N and DOC. The largest fluxes and concentrations were observed in percolates of the Oi layer, which contain amino N and amino sugar N as the major components. The average ratio of dissolved organic C to N in forest floor percolates corresponded to the C/N ratio of the solid phase. Concentrations and fluxes were highly dynamic with time and decreased with depth. The largest fluxes in forest floor percolates occurred when the snow melted. The concentrations and fluxes of dissolved organic N were significantly correlated with DOC, but the correlation was weak, indicating different mechanisms of release and consumption. The dynamics of dissolved organic N and DOC in forest floor percolates were not explained by pH and ionic strength of the soil solution nor by the water flux, despite large variations in these. Furthermore, the release of these fractions from the forest floor was not related to the quality and amount of throughfall. Concentrations of dissolved organic N in forest floor percolates increased with soil temperature, while temperature effects on DOC were less pronounced, but their fluxes from the forest floor were not correlated with temperature. In the growing season concentrations of both dissolved organic N and C in forest floor percolates decreased with increasing intensity of throughfall. Thus, the average throughfall intensity was more important than the amount of percolate in regulating their concentrations in forest floor percolates. Our data emphasize the role of dissolved organic N and DOC in the N and C cycle of forest ecosystems.  相似文献

Four separate controlled and blinded studies were conducted to confirm the dose of spinosad and milbemycin oxime (MO) administered orally in combination to dogs for the treatment and control of naturally acquired infections of adult whipworm (Trichuris vulpis), hookworm (Ancylostoma caninum) and ascarids (Toxocara canis, Toxascaris leonina). Dogs were allocated randomly based on pre-treatment quantitative nematode egg counts of each species of interest to one of two treatment groups of 10 or 11 animals each. In each study, spinosad and MO in combination, was given orally to dogs using the lower half (30-45 mg/kg spinosad; 0.5-0.75 mg/kg MO) of the US commercial dose band (30-60 mg/kg spinosad; 0.5-1.0mg/kg MO) of each active ingredient on Day 0 using a tablet formulation. A corresponding vehicle control group was treated similarly in each individual study. Dogs were necropsied post-treatment on Day 7/8. All nematodes in the intestinal tract collected at necropsy were identified and counted by species and stage. The spinosad and MO combination group demonstrated significantly different adult intestinal nematode efficacy in each individual study as compared to the vehicle control group. Efficacy values for whipworm, hookworm, T. canis and T. leonina were 100%, 99.8%, 100%, 93.3%, respectively. Minor non-serious adverse events were observed in a small number of control and treated dogs that were attributed primarily to the natural nematode infections. In summary, flavored spinosad and MO combination tablets administered orally to dogs were both safe and highly efficacious delivering >93% up to 100% adult intestinal nematode control in naturally infected dogs.  相似文献

Nitrogen (N) is an important plant nutrient and is crucial for the plant growth and grain yield formation of field crops such as wheat (Triticum aestivum L. ssp. aestivum). However, little is known about the influence of N on secondary metabolites in wheat grains which are supposed to be beneficial for human health due to their antioxidant potentials. Therefore, we investigated the influence of N fertilization on plant growth and yield performance of winter wheat, as well as on total phenolic concentration, antioxidant capacity, and the accumulation of (in)soluble phenolic acids in wheat grains during the grain‐filling phase. It was found that ferulic acid was the predominant phenolic acid in wheat grains. As expected, higher amounts of N fertilizer led to increasing grain yields, whereas the concentration of soluble ferulic acid decreased. In contrast, insoluble bound ferulic acid, total phenolic content, and antioxidant capacity were not affected by the N treatment. Insoluble phenolic compounds seemed to be less susceptible to variations in N supply.  相似文献

Fragaria x ananassa enone oxidoreductase (FaEO), earlier putatively assigned as quinone oxidoreductase, is a ripening-induced, negatively auxin-regulated enzyme that catalyzes the formation of 4-hydroxy-2,5-dimethyl-3(2H)-furanone (HDMF), the key flavor compound in strawberry fruit by the reduction of the alpha,beta-unsaturated bond of the highly reactive precursor 4-hydroxy-5-methyl-2-methylene-3(2H)-furanone (HMMF). Here we show that recombinant FaEO does not reduce the double bond of straight-chain 2-alkenals or 2-alkenones but rather hydrogenates previously unknown HMMF derivatives substituted at the methylene functional group. The furanones were prepared from 4-hydroxy-5-methyl-3(2H)-furanone with a number of aldehydes and a ketone. The kinetic data for the newly synthesized aroma-active substrates and products are similar to the values obtained for an enone oxidoreductase from Arabidopsis thaliana catalyzing the alpha,beta-hydrogenation of 2-alkenals. HMMF, the substrate of FaEO that is formed during strawberry fruit ripening, was also detected in tomato and pineapple fruit by HPLC-ESI-MSn and became 13C-labeled when d-[6-13C]-glucose was applied to the fruits, which suggested that a similar HDMF biosynthetic pathway occurs in the different plant species. With a database search (http://ted.bti.cornell.edu/ and http://genet.imb.uq.edu.au/Pineapple/), we identified a tomato and pineapple expressed sequence tag that shows significant homology to FaEO. Solanum lycopersicon EO (SlEO) was cloned from cDNA, and the protein was expressed in Escherichia coli and purified. Biochemical studies confirmed the involvement of SlEO in the biosynthesis of HDMF in tomato fruit.  相似文献

OBJECTIVE: To analyse the association between socio-economic indicators and diet among 2-year-old children, by assessing the independent contribution of parental education and equivalent income to food intake. DESIGN: The analysis was based on data from a prospective birth cohort study. Information on diet was obtained using a semi-quantitative food-frequency questionnaire. Low and high intake of food was defined according to the lowest and the highest quintile of food consumption frequency, respectively. SETTING: Four German cities (Munich, Leipzig, Wesel, Bad Honnef), 1999-2001.Subjects Subjects were 2637 children at the age of 2 years, whose parents completed questionnaires gathering information on lifestyle factors, including parental socio-economic status, household consumption frequencies and children's diet. RESULTS: Both low parental education and low equivalent income were associated with a low intake of fresh fruit, cooked vegetables and olive oil, and a high intake of canned vegetables or fruit, margarine, mayonnaise and processed salad dressing in children. Children with a low intake of milk and cream, and a high intake of hardened vegetable fat, more likely had parents with lower education. Low butter intake was associated with low equivalent income only. CONCLUSIONS: These findings may be helpful for future intervention programmes with more targeted policies aiming at an improvement of children's diets.  相似文献

A 20-day incubation experiment with continuous cereal (CC) versus cereal legume (CL) rotation soils of two semi-arid Sub-Saharan sites (Fada-Kouaré in Burkina Faso, F, and Koukombo in Togo, K) were carried out to investigate the effects of rewetting on soil microbial properties. Site- and system-specific reactions of soil microorganisms were observed on cumulative CO2 production, adenylates (ATP, ADP, and AMP), microbial biomass C and N, ergosterol, muramic acid and glucosamine. Higher values of all parameters were found in the CL rotation soils and in both soils from Fada-Kouaré. While the inorganic N concentration showed only a system-specific response to rewetting, the adenylate energy charge (AEC) showed only a site-specific response. ATP recovered within 6 h after rewetting from ADP and AMP due to rehydration of microorganisms and not due to microbial growth. Consequently, no N seemed to be immobilized by microorganisms and all NO3 in the soil was immediately available to the plants. The fungal cell-membrane component ergosterol was three (CC) and five (CL) times larger at Fada than in the respective soils at Koukombo. The concentrations of the bacterial cell-wall component muramic acid were by 20% and of mainly fungal glucosamine by 10% larger in the CL rotation soils than in the CC soils. Purpose

The transport behavior of human pharmaceuticals in groundwater depends on a multitude of factors such as the physico-chemical conditions in the aquifer and the organic carbon content of the sediment, and, in particular, on the redox conditions in the groundwater. This is of special interest at managed aquifer recharge sites where the occurrence of trace organics is important for drinking water production. The aim of this study was to evaluate the possibility of influencing the redox system of the aquifer in a way that optimizes the potential of managed aquifer recharge systems to reduce the amount of trace organics.

Materials and methods

Column studies were performed using natural and thermally treated sediments from an infiltration basin of the Berlin area, Germany. Special emphasis was placed on thermal treatment of the sediments to influence the total organic carbon (TOC) content in the sediment. In one experiment, the sediment was thermally pretreated at 550 °C, in two experiments the sediment was pretreated at 200 °C, and in one the sediment was untreated. Furthermore, the influence of ozonation, a very common disinfectant used in drinking water production, was studied in the experiments. The retardation and degradation parameters for primidone (PMD), carbamazepine (CBZ), and sulfamethoxazole (SMX) under different redox conditions were evaluated.

Results and discussion

Oxic conditions were obtained in the experiment with low TOC (0.06 wt%) in the sediment pre-treated at 550 °C. Anoxic conditions were predominant in two column experiments with a TOC content of 0.17 wt% in the sediment, irrespective of the mode of treatment (natural or 200 °C). All three pharmaceutical compounds show almost conservative transport behavior with retardation factors between 1.02 and 1.25 for PMD, between 1.06 and 1.37 for CBZ, and between 1.00 and 1.08 for SMX. Differences in the transport behavior were observed depending on the TOC content of the sediment. For CBZ, and to a minor extent for PMD, the higher retardation factors were observed in the sediment with a TOC content of 0.17 wt% under anoxic conditions. The ozonation of the influent water affects the influent concentrations of PMD, CBZ and SMX. However, it has no influence on the oxygen concentration of the column outflow.

Conclusions

CBZ and PMD are retarded in the presence of organic matter in the aquifer. Variations of the TOC content of the sediment have a direct influence on the retardation of CBZ and PMD. The three human pharmaceuticals may be ranked in order of decreasing retardation: CBZ?>?PMD?>?SMX. The microbial activity in the experiments was not studied, although it can be assumed that the thermal pretreatment influences the microbial activity in the sediments. In particular, the microbial activity was severely inhibited at 550 °C, resulting in a shift of the redox conditions.  相似文献
