Reproduction of postweaning multisystemic wasting syndrome in pigs by prenatal porcine circovirus 2 infection and postnatal porcine parvovirus infection or immunostimulation

Postweaning multisystemic wasting syndrome (PMWS) was reproduced in prenatally porcine circovirus 2 (PCV2)-infected pigs by either postnatal infection with porcine parvovirus (PPV) or by immunostimulation. Twenty-four randomly selected piglets from 3 sows, which had been experimentally infected during gestation with PCV2, were randomly divided into 3 groups; group 1 (prenatal PCV2 infection, with postnatal PPV infection), group 2 (prenatal PCV2 infection, with postnatal keyhole limpet hemocyanin, emulsified in incomplete Freund's adjuvant [KLH/ICFA] injection), and group 3 (prenatal PCV2 infection only). Twenty-four randomly selected piglets from 3 uninfected sows were randomly divided into 3 groups; group 4 (no prenatal infection, with postnatal PCV2 and PPV infection), group 5 (no prenatal infection, with postnatal PCV2 infection), and group 6 (negative control pigs). Body weight in negative control pigs (group 6) was increased significantly compared with pigs in groups 1, 2, and 4 at 49, 52, 56, 59, and 63 days of age. The granulomatous inflammatory reaction and lymphoid depletion that are typical lesions in pigs with PMWS were observed in the lymph node of piglets in groups 1, 2, and 4 at 63 days of age. Pigs in group 3 had significantly fewer PCV2-positive cells than those from groups 1, 2, 4, or 5. When the prenatally PCV2-infected pigs were infected with PPV or injected with immunostimulant in the postnatal period, they developed PMWS. Thus, factors that potentiate the progression of prenatal PCV2 infection to PMWS are postnatal infection with PPV or immune stimulation.     

Primary tracheal collapse in a cat

A 7-year-old, neutered male, domestic shorthair cat was presented for severe inspiratory dyspnea of 2 to 3 days' duration. Radiography and tracheobronchoscopy confirmed the diagnosis of primary extrathoracic tracheal collapse. The cat was treated with oxygen, dexamethasone, and terbutaline, but no improvement was seen. Surgical correction was performed using nine prosthetic tracheal ring implants. Clinical signs improved after surgery, and the cat continued to do well 11 months after surgery, despite development of unilateral laryngeal paralysis.     

Stress alters the cellular and proteomic compartments of bovine bronchoalveolar lavage fluid

The stresses of transportation, weaning and commingling are associated with an increased incidence of bacterial and viral pneumonia in cattle. Proteins expressed in the epithelial lining fluid (ELF) of the lungs, in conjunction with resident leukocytes, represent the first line of defence against opportunistic pathogens, and stress-induced alterations in their expression may reveal markers of disease susceptibility. Bronchoalveolar lavage fluid was sampled in weaned and transported calves and ELF protein expression was compared to a control group of calves using two-dimensional electrophoresis (2DE). Serum and pulmonary haptoglobin were increased following stress concurrent with the number of blood neutrophils. Using 2DE, significant changes in expression were observed in spots identified by mass spectrometry as annexin A1 and A5, odorant-binding protein (OBP), isocitrate dehydrogenase, fibrinogen, heme-binding protein, alpha-2-HS-glycoprotein, alpha-1-antichymotrypsin and albumin. Quantification of OBP mRNA by real-time RT-PCR and OBP protein by western blot revealed gender-dependent differences in relative OBP expression in response to stress. These findings reveal stress-associated protein changes in pulmonary ELF and suggest a mechanism through which stress alters respiratory disease susceptibility.     

Intraocular hemorrhage in a goat with thrombocytopenia and metastasis of an adenocarcinoma in the iris

This case report describes the findings in a 10-year-old goat with metastasis of an adenocarcinoma in the iris. Two weeks before admission, the owner had noticed blepharospasm of the left eye. Clinical examination by the referring veterinarian revealed unilateral intraocular hemorrhage. The goat was referred to our clinic for further work-up. The rectal temperature was 40 degrees C. The most important haematological result was severe thrombocytopenia. There was mild corneal oedema of the left eye. Approximately 75 per cent of the anterior chamber was filled with non-coagulated blood. The fluid in the anterior chamber dorsal to the blood was cloudy, and the corpora nigra could not be seen clearly. All other internal parts of the eye could not be seen. Ultrasonography of the left eye confirmed cloudiness of the anterior chamber and revealed moderate thickening of the iris. The right prescapular lymph node was markedly enlarged. Cytological examination of a fine needle aspirate of the lymph node showed a mixed population of neoplastic cells. Based on immunohistochemical evaluation of the cells metastasis of a carcinoma was diagnosed. The goat was subjected to euthanasia, and a postmortem examination was carried out. The anterior chamber of the left eye contained blood, and the iris was thicker than normal and adhered to the posterior surface of the cornea. There were neoplastic alterations in the iris, the oesophagus, the lung lobes, the liver, the kidney and in the prescapular, retropharyngeal, mediastinal and hepatic lymph nodes. Histologically, a diagnosis of carcinoma was confirmed, but the origin of the tumour could not be determined.     

Effect of immunization against somatostatin in the pregnant ewe on growth and endocrine status of the neonatal lamb

Absorption of somatostatin (SRIF) specific antibodies from colostrum of ewes actively immunized against SRIF may improve growth rate of the neonatal lamb by neutralizing the inhibitory effects of SRIF on pituitary and thyroid function. Growth and endocrine parameters in the offspring of SRIF immunized (SI) and control (C) crossbred ewes were examined. Lamb weight was recorded at birth and twice each week to 24 days of age. Blood samples were collected prior to first suckle and twice each week. At 21 to 24 days of age, in separate experiments, lambs were infused with glucose (0.29 g/kg), arginine (0.25 g/kg) or thyrotropin-releasing hormone (TRH; 0.33 microgram/kg). A strong correlation (R = 0.88; P less than .01) was observed between anti-SRIF titre in the ewe at parturition and in the lamb at 3 days of age. No effect on lamb birth weight (SI 4.28 +/- 0.27 kg; C 4.35 +/- 0.23 kg) was observed. At 24 days of age cumulative gain in SI lambs (5.4 +/- 0.32 kg) was greater (P less than .05) than in C lambs (4.5 +/- 0.32 kg). The growth hormone secretory responses to glucose or arginine were not affected by treatment. Plasma IGF-I, plasma thyroxine (T4) and the plasma thyrotropin and T4 responses to TRH were not different between treatments. Plasma triiodothyronine (T3) was higher (P less than .05) in SI (2.46 +/- .10 ng/ml) than in C (2.01 +/- .05 ng/ml) lambs, however, the plasma T3 response to TRH was lower in SI lambs. Plasma glucose (mg/dl) was higher (P less than .05) in SI (118.4 +/- 1.7) than in C (106.0 +/- 4.0) lambs. Plasma insulin was not affected by treatment. Increased plasma T3 and glucose concentrations during SRIF immunoneutralization in the neonate lamb may be important factors contributing to the growth response observed.
