Tissues were evaluated from emerald tree boas, Corallus caninus, from a collection in which chlamydiosis was diagnosed. To determine the strain of chlamydia infecting these snakes, tissue samples from 5 frozen snakes were tested by a quantitative TaqMan polymerase chain reaction (PCR) test and a PCR sequence analysis test. Of the 22 samples tested, 9 were categorized as either positive or weakly positive with the TaqMan test, and 6 yielded an amplicon using a serial PCR test that amplified a portion of the 23S ribosomal RNA gene. A PCR product suitable for sequencing was obtained from the heart of one of the snakes. Sequence analysis showed that the snake had been infected with Chlamydophila pneumoniae. These findings show that C. pneumoniae can infect emerald tree boas, broadening the range of reptiles known to be infected by this primarily human pathogen.  相似文献

Dantrolene is a skeletal muscle relaxant used commonly in performance horses to prevent exertional rhabdomyolysis. The goal of the study reported here was to begin to characterize cytochrome P450-mediated metabolism of dantrolene in the horse and describe the pharmacokinetics of the compound, formulated as a capsule or a compounded paste formulation, following oral administration. Dantrolene is rapidly metabolized to 5-hydroxydantrolene both in vivo and in vitro. Preliminary work with equine liver microsomes suggest that two enzymes are responsible for the metabolism of dantrolene, as evidenced by two distinct K(m) values, one at high and one at low substrate concentrations. For the pharmacokinetic portion of the study, a randomized, balanced 2-way crossover design was employed wherein eight healthy horses received a single oral dose of either capsules or paste followed by a 4 week washout period prior to administration of the second formulation to the same horse. Blood samples were collected at time 0 (prior to drug administration) and at various times up to 96 h postdrug administration. Plasma samples were analyzed using liquid chromatography-mass spectrometry and data analyzed using both noncompartmental and compartmental analysis. Peak plasma concentrations were 28.9 ± 21.6 and 37.8 ± 12.8 ng/mL for capsules and paste, respectively and occurred at 3.8 h for both formulations. Dantrolene and its major metabolite were both below the limit of detection in both plasma and urine by 168 h postadministration.  相似文献

Malignant catarrhal fever was diagnosed in a newly imported swamp buffalo calf. This disease is prevalent in deer and cattle in New Zealand and causes sporadic losses in these species. Sixteen ruminally cannulated, English-crossbred heifers (378 ± 28.4 kg) grazing small-grain pasture (SGP) were used in a completely randomized design to evaluate effects of supplementing different amounts of corn dried distillers grains with solubles (DDGS; 0, 0.2, 0.4, and 0.6% of BW; as-fed basis) on forage intake, digestibility, and rumen fermentation characteristics. The experiment was conducted from April 6 through April 20, 2007. Heifers grazed in a single SGP with supplements offered individually, once daily at 0700 h. Forage and total OM, CP, and NDF intake were not affected (P ≥ 0.21) by DDGS amount. Digestibility of NDF and ether extract (EE) increased linearly (P < 0.001) when heifers consumed more DDGS. Intake of DM (kg/d and g/kg of BW), ruminal volume (L), fluid dilution rate (%/h), fluid flow rate (L/h) turnover time (h), and particle dilution rate (SGP and DDGS) were not affected (P ≥ 0.32) by increasing DDGS supplementation amount. In situ DDGS CP kinetic parameters were not affected (P ≥ 0.25) by increasing DDGS supplementation amount. Forage masticate in situ soluble CP fraction and CP effective degradability increased quadratically (P = 0.01) with increasing DDGS supplementation amount. However, amount of DDGS did not affect forage masticate CP slowly degradable fraction (%; P = 0.39) or degradation rate (%/h; P = 0.63). Rate of in situ disappearance (%/h) for DDGS DM (P = 0.94), forage masticate DM (P = 0.89), and NDF (P = 0.89) were not affected by DDGS supplementation amount, nor was rumen undegradable intake protein (% of CP) for DDGS (P = 0.28) and forage masticate samples (P = 0.93). Ruminal concentration of VFA and ammonia and ruminal pH were not affected (P ≥ 0.21) by increasing DDGS amount. Results indicated that DDGS can be used in SGP supplements without negatively affecting forage intake, digestibility, or ruminal fermentation.  相似文献

OBJECTIVE: To develop a flow-limited, physiologic-based pharmacokinetic model for use in estimating concentrations of sulfamethazine after IV administration to swine. SAMPLE POPULATION: 4 published studies provided physiologic values for organ weights, blood flows, clearance, and tissue-to-blood partition coefficients, and 3 published studies provided data on plasma and other tissue compartments for model validation. PROCEDURE: For the parent compound, the model included compartments for blood, adipose, muscle, liver, and kidney tissue with an extra compartment representing the remaining carcass. Compartments for the N-acetyl metabolite included the liver and the remaining body. The model was created and optimized by use of computer software. Sensitivity analysis was completed to evaluate the importance of each constant on the whole model. The model was validated and used to estimate a withhold interval after an IV injection at a dose of 50 mg/kg. The withhold interval was compared to the interval estimated by the Food Animal Residue Avoidance Databank (FARAD). RESULTS: Specific tissue correlations for plasma, adipose, muscle, kidney, and liver tissue compartments were 0.93, 0.86, 0.99, 0.94, and 0.98, respectively. The model typically overpredicted concentrations at early time points but had excellent accuracy at later time points. The withhold interval estimated by use of the model was 120 hours, compared with 100 hours estimated by FARAD. CONCLUSIONS AND CLINICAL RELEVANCE: Use of this model enabled accurate prediction of sulfamethazine pharmacokinetics in swine and has applications for food safety and prediction of drug residues in edible tissues.  相似文献

The trend in wildlife management over the last two decades has been to develop locally based approaches for responsiveness to local conditions, but some state wildlife agencies are finding the amount of staff time required to service this approach prohibitive. Although local engagement strategies have been lauded as assuring that public trust obligations of state government to citizens are met, we can expect that states with a local focus as their operational level of stakeholder engagement may opt to change their approach to reflect their resource limitations. We argue for comprehensive regional level effort to understand stakeholders augmented with local engagement processes where needed to deal with special circumstances in smaller areas within a region. Such an approach can be anticipated to have implications for stakeholder engagement and human dimensions research needs, which we discuss in the context of public trust resource administration and good governance of wildlife resources.  相似文献

Variance components (VC) were estimated for the semen production trait ejaculate volume, sperm concentration and sperm motility in the Swiss cattle breeds Brown Swiss (BS), Original Braunvieh (OB), Holstein (HO), Red‐Factor‐Carrier (RF), Red Holstein (RH), Swiss Fleckvieh (SF) and Simmental (SI). For this purpose, semen production traits from 2,617 bulls with 124,492 records were used. The data were collected in the years 2000–2012. The model for genetic parameter estimation across all breeds included the fixed effects age of bull at collection, year of collection, month of collection, number of collection per bull and day, interval between consecutive collections, semen collector, bull breed as well as a random additive genetic component and a permanent environmental effect. The same model without a fixed breed effect was used to estimate VC and repeatabilities separately for each of the breeds BS, HO, RH, SF and SI. Estimated heritabilities across all breeds were 0.42, 0.25 and 0.09 for ejaculate volume, sperm concentration and sperm motility, respectively. Different heritabilities were estimated for ejaculate volume (0.42; 0.45; 0.49; 0.40; 0.10), sperm concentration (0.34; 0.30; 0.20; 0.07; 0.23) and number of semen portions (0.18; 0.30; 0.04; 0.14; 0.04) in BS, HO, RH, SF and SI breed, respectively. The phenotypic and genetic correlations across all breeds between ejaculate volume and sperm concentration were negative (?0.28; ?0.56). The other correlations across all breeds were positive. The phenotypic and genetic correlations were 0.01 and 0.19 between sperm motility and ejaculate volume, respectively. Between sperm motility and sperm concentration, the phenotypic and genetic correlations were 0.20 and 0.36, respectively. The results are consistent with other analyses and show that genetic improvement through selection is possible in bull semen production traits.  相似文献

Selenium deficient calves when compared to selenium supplemented calves had increased plasma thyroxine concentrations and decreased plasma tri-iodothyronine concentrations. These changes in the selenium deficient calves were accompanied by significant increases in plasma urea and creatinine concentrations and decreased plasma alkaline phosphatase activity. The demonstration that low selenium status can cause imbalances in thyroid hormone metabolism may provide an explanation for some of the effects of the deficiency.  相似文献

REASONS FOR PERFORMING STUDY: Many racing jurisdictions monitor pre-race serum concentration of total carbon dioxide (TCO2) among racing horses. To our knowledge, factors influencing concentration of TCO2 among horses participating in racing have not been systematically evaluated and reported. OBJECTIVES: To determine if characteristics of horses and racing conditions routinely recorded were significantly associated with pre-race concentration of TCO2, while accounting for and estimating effects of trainer and horse. METHODS: Pre-race serum TCO2 concentrations from 5028 starts made by 2,349 horses trained by 287 trainers at 2 racetracks in California during 2005 were examined. Data regarding characteristics of starters and race conditions obtained from a commercial database were recorded for each start. Data were analysed using mixed-effects, with TCO2 concentration as the dependent variable, and trainer and horse nested within trainer as random effects. RESULTS: Sex, class and distance of race, frusemide administration and cloudy weather conditions were significantly (P<0.001) associated with pre-race TCO2 concentration. Horses that finished in the top 3 positions had values that were slightly (0.2 mmol/) but significantly (P<0.001) greater than horses not finishing in the top 3. There were significant effects of trainer on pre-race TCO2 concentration. CONCLUSIONS: A variety of factors may influence pre-race TCO2 concentration in horses. Horses with better performance tend to have higher pre-race TCO2 concentrations. POTENTIAL RELEVANCE: TCO2 concentration is associated with improved performance although the magnitude of effect was quite small. Regulatory programmes based on monitoring should consider the influence of other factors on TCO2 concentration.  相似文献
