Following alarming statements (French Senate, Académie des Sciences) on the present situation concerning entomology and systematics in France, the Conseil Général Vétérinaire designated one of us (D.C.) to carry out a survey on the status of medical and veterinary entomology (MVE) with respect to research orientations and university curricula. Around 100 participants, including scientists, teachers and several directors of research and educational bodies, were interviewed and filled in questionnaires for this survey. On the basis of the results, it was concluded that the deterioration of MVE in France is associated with: (1) the hasty reorganisation of training and research in the life sciences, leading to the disappearance of several disciplines. Hence, the postgraduate DEA degree in entomology was eliminated, and even the name 'entomology' no longer appears in teaching programmes or on research contracts; (2) France's withdrawal from action research programmes in developing countries. Although these programmes were efficient in controlling outbreaks of major endemic diseases, integrated pest and vector management programmes have been replaced by basic health care ('Health for everyone in 2000') and vaccination programmes; (3) the general shift from field to laboratory research, focused mainly on molecular mechanisms. The survey results confirmed generally acknowledged trends concerning many points and highlighted several specific problems, such as the disappearance of systematics experts. Conventional magnetic resonance methods that provide interior temperature profiles, which find use in clinical applications such as hyperthermic therapy, can develop inaccuracies caused by the inherently inhomogeneous magnetic field within tissues or by probe dynamics, and work poorly in important applications such as fatty tissues. We present a magnetic resonance method that is suitable for imaging temperature in a wide range of environments. It uses the inherently sharp resonances of intermolecular zero-quantum coherences, in this case flipping up a water spin while flipping down a nearby fat spin. We show that this method can rapidly and accurately assign temperatures in vivo on an absolute scale.  相似文献

This study aimed to investigate both the pharmacokinetic behavior and tolerance of methotrexate (MTX) in horses to design a specific dosing regimen as a new immunomodulatory drug for long-term treatment. To determine the primary plasma pharmacokinetic variables after single intravenous, subcutaneous or oral administration, six horses were administered 0.3 mg/kg MTX in a crossover design study. After a 10-week washout, MTX was administered subcutaneously to three of the six previously treated horses at a dose of 0.3 mg/kg once per week for 3 months. In both studies, MTX and metabolite concentrations were measured using LC-MS/MS. The absolute bioavailability of MTX was 73% following subcutaneous administration but less than 1% following oral administration. The plasma clearance was 1.54 ml min−1 kg−1 (extraction ratio = 2%). After 24 hr, plasma concentrations were below the LOQ. No adverse effects were noted except for a moderate reversible elevation in liver enzymes (GLDH). With regards to the main metabolites of MTX, very low concentrations of 7-hydroxy-MTX were found, whereas polyglutamated forms (mainly short chains) were found in red blood cells. A subcutaneous dose of 0.2 mg kg−1 week−1 may be safe and relevant in horses, although this has yet to be clinically confirmed.  相似文献

Reasons for performing the study: During riding, horses are frequently exposed to pressure from the rider, e.g. through the bit and reins, but few studies have investigated at which point rein tension becomes uncomfortable for the horse. Objectives: To investigate how much rein tension young inexperienced horses are willing to accept in order to obtain a food reward; whether the tension acceptance changes during 3 consecutive test days; and whether rein tension correlates with the expression of conflict behaviour and heart rate. Hypotheses: Pressure‐naïve horses will apply only little rein tension in the first voluntary trial, but their acceptance will gradually increase. High levels of rein tension will lead to expression of conflict behaviour and increases in heart rate. Methods: Fifteen 2‐year‐old, bridle‐naïve mares were encouraged to stretch their head forward (across a 0.95 m high metal bar) to obtain a food reward in a voluntary test situation. On each test day, each horse was exposed to 2 control sessions (loose reins), an intermediate and a short rein session (1 min/session). Rein tension, heart rate and behaviour were recorded. Results: The horses applied significantly more tension on the first day (mean rein tension: 10.2 N), compared to the second and third test day (Day 2: 6.0 and Day 3: 5.7 N). The horses showed significantly more conflict behaviour in the short rein treatment. There was no treatment effect on heart rate. Conclusions and potential relevance: The horses applied the highest rein tension on the first day, and apparently learned to avoid the tension, rather than habituate to it. Rein tension correlated with expression of conflict behaviour, indicating that the horses found the tension aversive. Further studies should focus on the correlation between rein tension and conflict behaviour in ridden horses.  相似文献

Andromonoecy is a widespread sexual system in angiosperms characterized by plants carrying both male and bisexual flowers. In melon, this sexual form is controlled by the identity of the alleles at the andromonoecious (a) locus. Cloning of the a gene reveals that andromonoecy results from a mutation in the active site of 1-aminocyclopropane-1-carboxylic acid synthase. Expression of the active enzyme inhibits the development of the male organs and is not required for carpel development. A causal single-nucleotide polymorphism associated with andromonoecy was identified, which suggests that the a allele has been under recent positive selection and may be linked to the evolution of this sexual system.  相似文献

Semiconductor quantum dots (QDs) are nanometer-sized fluorescent probes suitable for advanced biological imaging. We used QDs to track individual glycine receptors (GlyRs) and analyze their lateral dynamics in the neuronal membrane of living cells for periods ranging from milliseconds to minutes. We characterized multiple diffusion domains in relation to the synaptic, perisynaptic, or extrasynaptic GlyR localization. The entry of GlyRs into the synapse by diffusion was observed and further confirmed by electron microscopy imaging of QD-tagged receptors.  相似文献

Wheat bran tissue fractionation using biochemical markers   总被引:1,自引:0,他引:1  
Phenolic acid analysis of hand-isolated outer grain layers and endosperm led to the identification of markers of pericarp and aleurone layers, respectively. A new dehydrotrimer of ferulic acid (DHT) was found to be concentrated in the outer pericarp of wheat bran whereas p-coumaric (p-CA) acid was mainly in the aleurone layer. Phytates were also used as a marker of aleurone layer and starch as a marker of starchy endosperm. Biochemical markers constitute an original method for determining the histological composition of any technological bran fractions. A pin milling process was applied to coarse bran produced by a conventional milling process. Three different fractions (B1, B2 and B3) were obtained by sieving the bran products and then the smallest bran particle fraction (B3) was air-classified to obtain two particle size fractions (B3a and B3b with a D50 of 83 and 7 μm, respectively). The biochemical composition of these fractions was used to calculate the distribution of tissues according to the sieving process. The dissociation behavior of individual bran tissues upon mechanical fractionation was investigated in relation to particle size and discussed according to their mechanical properties.  相似文献

The classical debate about the roles of endogenous factors and external drivers in regulating populations continues to be a very active area of scientific research, and these roles are even more complex in populations located in geographically marginal areas. We tested three hypotheses on two separate brown trout populations inhabiting the southernmost basins of the Iberian Peninsula, with the aims of determining the relationships among (a) rainfall and air temperature for each season preceding fish sampling and the observed age group densities; (b) environmental synchrony and density synchrony for all possible pairs of sites; and (c) physical habitat similarity and the similarity of the density response to climatic drivers among pairs of sites. For this purpose, demographic series (98 surveys at 14 sampling sites), climatic conditions (rainfall and air temperature) and site habitat characteristics (physiogeographic and quality index values) were analysed between 2006 and 2014. The results show how the synchrony in the study populations is the result of a Moran effect directed mainly by winter precipitation and conditioned by the habitat similarity (highlighting the altitude, the distance to the upper limit and IHF index, along with the Euclidian distance among pairs of sites of the whole set of habitat variables). Preserving the heterogeneity of these habitat characteristics will increase the resilience of trout populations that inhabit this rear edge of distribution. The current context of global climate warming predicts extremely drastic changes in the dynamics of external drivers that regulate the trout populations in the study region.  相似文献

Enzootic dermatophytosis in a shelter with approximately 140 cats was treated according to a protocol combining identification, isolation and treatment of subclinical carrier and affected animals in accordance with a three‐area system: healthy animals (no lesions and negative cultures), subclinical carrier animals (no lesions but with positive cultures) and clinically affected animals (lesions and positive cultures). The cats were examined and inspected under a Wood’s lamp and had samples taken for fungal culture every 2 weeks. Thirty‐three per cent of the cats had a positive fungal culture at the start of the study. Clinically affected animals and carriers were treated with a 0.2% enilconazole lotion (Imaverol®) twice a week and given itraconazole (Itrafungol®) 5 mg/kg SID orally every other week. The environment was treated once a day with a 1% bleach solution and once a week with a 0.6% enilconazole (Clinafarm®) solution. Treated animals were considered cured after two consecutive negative fungal cultures. All cats were cured within 56 days. Prophylactic measures against dermatophytosis were implemented for new arrivals consisting of individual quarantine and the systematic taking of fungal cultures. No relapses were observed based on the fungal cultures taken from the animals and the environment over the first 10 months.  相似文献

  • ? The objective of this review was to summarize scientific data on the symbiotic status of exotic tree species useful in tropical Africa, and to update reports about their growth improvement through microbial inoculations, especially ectomycorrhizal symbionts.
  • ? The studies reviewed microbial symbionts associated to exotic tree species belonging to Myrtaceae, Pinaceae, Casuarinaceae and Leguminosae. In their native areas, these trees are associated either with ectomycorrhizal (ECM) fungi (e.g. Pinaceae) or both ECM and arbuscular mycorrhizal (AM) fungi (e.g. Eucalyptus), or sometimes as in Casuarina and Acacia with three coexisting symbionts (nitrogen fixing bacteria, ECM and AM fungi). In their new habitats, using highly efficient mycorrhizal fungi, controlled mycorrhization experiments have pointed out the importance of root symbionts in establishment and growth of exotic tree species such exotic pines or Australian acacias.

Since the challenge in sylviculture and reforestation was to determine the best compromise between symbiotic compatibility and efficiency of both partners under local soil constraints, further researches have to be encouraged to elucidate the complexity of these tree symbioses in terms of diversity, interaction and effectiveness of their symbiotic partners for their better exploitation in reforestation programmes.
