BackgroundCanine herpesvirus type 1 (CaHV-1) infects dogs and is associated with neonatal deaths and reproductive, ocular, neurological, and respiratory problems. In Brazil, reports of CaHV-1 have been restricted to the southeast and south regions, particularly in municipalities in the state of Rio Grande do Sul.ObjectivesTo assess the presence and variability of CaHV-1 in canine populations in the state of Pará, North Brazil.MethodsBiological samples from 159 dogs from 4 municipalities in the State of Pará were evaluated using polymerase chain reaction and phylogenetic analyses, with the target being the viral enzyme, thymidine kinase.ResultsCaHV-1 was detected in 13 dogs (8.2%), with 2 animals being from the municipality of Santa Bárbara do Pará, 8 from Algodoal Island, 2 from Salinópolis, and one from Capanema. The study sequences revealed 100% identity among themselves and 64% to 100% identity with the other nucleotide sequences from Australia, Brazil, United Kingdom, and United States, including 100% identity with the 2002 isolate from Australia. The 1996 isolate from France was grouped in a branch that was different from the sequence of this study.ConclusionsThis study presents the first molecular detection of CaHV-1 in dogs from the Amazon region in northern Brazil. The nucleotide identity between the strains and cytosine insertion in the sequences isolated in this study suggests at least 2 strains of CaHV-1 circulating in Brazil (Pará and BTU-1).  相似文献

Chlorophyll fluorescence spectral analysis permits detection, monitoring, and evaluation of abiotic stresses upon healthy plants using illumination of a light source in the UV?CVIS spectral range. This technique indirectly assesses the amount of physiological stress caused by photosynthetic damage, specifically damage to photosystem II, in plants. The objective of this study was to detect the toxicity of cadmium in maize plants via spectral analysis of chlorophyll fluorescence. The analysis is noninvasive and nondestructive and is used to follow the temporal evolution of changes in the chlorophyll content and physiological state of Zea mays L. seedlings under cadmium stress. Conventional techniques were also used to evaluate the dry matter production and Cd accumulation in plant leaves. Plants exhibited a notable reduction in dry matter production and chlorophyll levels with the administration of increasing doses of Cd in the nutrient solution. The fluorescence analysis was sensitive to changes caused by Cd in maize plants, detecting damage caused by different treatments before visual symptoms were observed. This technique has a practical application and produces rapid results that can be used in the evaluation of Cd-induced stress in plants and the detection of areas contaminated by this element.  相似文献

Nutrient solution composition plays an important role in root uptake rate due to interactions among nutrients and internal regulation. Studies to determine the optimum nutrient solution concentration are focused on individual ions, ignoring the adaptation mechanisms triggered by plants when growing in a varying external nutrient concentration. The objective of the present study was to determine the response in growth and tissue ion concentration of lilium cv. ‘Navona’ to nutrient mixtures of varying proportions of nitrogen (N), potassium (K+), and calcium (Ca2+) in solution using mixture experiments methodology in order to determine the optimum concentration. Bulbs of lilium were transplanted in plastic crates and drip-irrigated with the treatment solutions, which consisted of a mixture of N, K+, and Ca2+ whose total concentration was 340 mg L?1 and minimum concentrations of each ion was 34 mg L?1. Chlorophyll concentration (SPAD), shoot fresh weight (FW), leaf FW, and leaf area were measured 60 days after transplanting and ion analysis was performed on shoot tissues from selected treatments. Lilium exhibited a moderate demand for N and K+ (136–170 mg L?1 N and 116–136 mg L?1 K+) and a very low demand for Ca2+ (34–88 mg · L?1). This low demand may be due to the remobilization of the nutrients stored in the bulbs. Integrating the predictions of the models estimated to produce >90% of maximum growth, the optimum nutrient solution should contain Ca2+ at a concentration between 34 and 126 mg · L?1, K+ between 119 and 211 mg · L?1, and N between 92 mg · L?1 and 211 mg · L?1. Increasing external N concentration affected internal N concentration but not internal K+ or Ca2+ concentrations, despite that the increase in external N was associated with a decrease in external K+ and Ca2+. Similar trends were observed for external K+ and Ca2+ concentration. In conclusion, lilium was able to maintain a relatively constant K+ and Ca2+ concentration regardless of the lower concentration in the nutrient solution when N was increased (similar response was observed for K+ and Ca2+) and it has a low Ca2+ demand and moderate N and K+ supply.  相似文献

The amino acid composition and the physicochemical and functional properties of quinoa protein isolates were evaluated. Protein isolates were prepared from quinoa seed by alkaline solubilization (at pH 9, called Q9, and at pH 11, called Q11) followed by isoelectric precipitation and spray drying. Q9 and Q11 had high levels of essential amino acids, with high levels of lysine. Both isolates showed similar patterns in native/SDS-PAGE and SEM. The pH effect on fluorescence measurements showed decreasing fluorescence intensity and a shift in the maximum of emission of both isolates. Q9 showed an endotherm with a denaturation temperature of 98.1 degrees C and a denaturation enthalpy of 12.7 J/g, while Q11 showed no endotherm. The protein solubility of Q11 was lower than that of Q9 at pH above 5.0 but similar at the pH range 3.0-4.0. The water holding capacity (WHC) was similar in both isolates and was not affected by pH. The water imbibing capacity (WIC) was double for Q11 (3.5 mL of water/g isolate). Analysis of DSC, fluorescence, and solubility data suggests that there is apparently denaturation due to pH. Some differences were found that could be attributed to the extreme pH treatments in protein isolates and the nature of quinoa proteins. Q9 and Q11 can be used as a valuable source of nutrition for infants and children. Q9 may be used as an ingredient in nutritive beverages, and Q11 may be used as an ingredient in sauces, sausages, and soups.  相似文献

Summary Aluminum toxicity due to the cation Al+3 is a major factor limiting yields in acid soils. Wide genetic variability to aluminum tolerance is found in oat genotypes. The objectives of this study were to determine the number of genes controlling aluminum tolerance in oats and to verify if any detrimental effects were present of the aluminum tolerance genes on grain yield and grain quality in Al+3free soils. Aluminum tolerance was estimated as the average regrowth of the main root after exposure to toxic levels of Al+3 in a hydroponic solution under controlled conditions. The number of genes controlling that trait was estimated from the distribution of the average root regrowth frequencies in a population of 333 recombinant inbred lines (RIL's) in generations F5:6 and F5:7. The effects on grain yield and grain quality were assessed in a subpopulation of 162 RIL's chosen based on their aluminum tolerance response. Aluminum tolerance in the evaluated population was controlled by one dominant major gene with the tolerant genotypes carying Al a Al a and the sensitive ones al a al a alleles. The use of saline waters is an option for the irrigation of salt tolerant ornamentals as competition for high quality water increases. However, despite the importance of ornamental shrubs in Mediterranean areas, salt tolerance of such species has received little attention. The aims of our investigation were to quantify the growth response and any injury symptom of 12 widely cultivated ornamental shrubs to irrigation with saline water and to investigate any possible relation with the concentration of Na+ and Cl in the plants. Species were irrigated with different salinities (10, 40, and 70 mM NaCl) for a 120-day period. At the end of salt treatment, plants were sampled and dry biomass recorded; the relative growth rate (RGR) was also calculated. Root and leaf samples from each species were used to evaluate Na+, K+ and Cl concentrations. Growth rates were significantly reduced in Cotoneaster lacteus, Grevillea juniperina and Pyracantha ‘Harlequin’, which also showed the highest percentage of necrotic leaves. The increasing external NaCl lead to an increase of Na+ and Cl in roots and leaves of the different species, although less Na+ was accumulated than Cl: growth reduction well correlated with the concentration of Cl and/or Na+ in the leaves. The most sensitive species (i.e. C. lacteus, G. juniperina and Pyracantha ‘Harlequin’) had high concentrations of Na+ and/or Cl in their leaves and also showed a decrease in their leaf K+/Na+ ratios. Even though other species (i.e. Bougainvillea glabra, Ceanothus thyrsiflorus, Leptospermum scoparium, Leucophyllum frutescens and Ruttya fruticosa) demonstrated a high ion concentration in their leaves, they could be considered relatively salt tolerant as there was little growth reduction and few symptoms of injury in the leaves. In some other cases (i.e. Cestrum fasciculatum, Escallonia rubra and Viburnum lucidum) the observed tolerance was related to higher ion concentration in the roots compared to the leaves, probably indicative of a limited transport to the shoots. Only in Eugenia myrtifolia was the absence of symptoms associated with a limited Na+ and Cl uptake from the rhizosphere.  相似文献

The aim of this study was to evaluate resistance to Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) by antixenosis on 57 Lycopersicon esculentum Mill. accessions from the Horticultural Germplasm Bank (HGB) of Universidade Federal de Viçosa and by the three commercial cultivars (Santa Clara, Moneymaker and TOM-601) under greenhouse conditions. A randomized complete block design was used with three replications. Infestations with T. absoluta adults were performed weekly and the following characteristics were evaluated: number of small, large and total mines/leaf and % of leaves mined at days 60, 75 and 90 after planting. Low infestation occurred at days 60 and 75 but at day 90, infestation was sufficient to evaluate insect damage. Based on these data it was concluded that only accessions HGB-674 and HGB-1497 appeared to be the most promising. In addition, to determine possible chemical causes of resistance, hexane extracts were analyzed at day 90 by gas chromatography/mass spectrometry and the major peaks identified by a mass spectral database using similarity index. Nine hydrocarbons, viz., hexadecane, heptadecane, eicosane, tricosane, 2-methyltricosane, tetracosane, hexacosane, octacosane and triacontane were identified in the hexane extracts in many samples. Tricosane, tetracosane and hexacosane presented significant correlations with the leaves mined. Only tricosane presented a negative correlation with the number of small mines (r = −0.28), total number of mines (r = −0.27) and % of leaves mined (r = −0.22). However, tetracosane and hexacosane presented significant positive correlations (r = 0.25 and 0.24, respectively) with the % of leaves mined.  相似文献

The state of Morelos, Mexico has gradually become an important producer of gladiolus. Some preconditioning treatments of corms are empirically done causing uneven emergence and low quality of flowers. In this investigation, before planting, gladiolus corms var. ‘Blanca Borrego’ were dipped in chitosan (chitosan reagent and commercial chitosan Biorend®), in hot water at various temperatures and in treatments combined with Biorend® at 1.5% and hot water. Results indicated that the most influenced variables were corm germination, number of flowers per spike, number of cormlets and vase life. Overall, the commercial product Biorend® at 1.5% accelerated corm emergence in approximately 4 days, the number of flowers increased by 2–7 and the vase life extended for 3 days. The number of cormlets was also duplicated. Corms dipped in the commercial chitosan Biorend® at 1.5% at different intervals of time were not greatly affected except for the emergence and number of cormlets. However, for this experiment there were significant effects on the number of leaves and flowers because of the interactions between chitosan and the immersion time. The temperature of 55 °C affected plant development because emergence was delayed by 6 days; and there were less number of leaves, flowers and cormlets. On the other hand, the incidence of Fusarium oxysporum in naturally infected corms was 0% at temperatures of 55 °C and 50 °C. Immersion times (0, 10, 15 and 20 min) in hot water at 50 °C did not show significant effects on plant development and vase life. Corms dipped in Biorend® at 1.5% and hot water at 50 °C accelerated their emergence for about 1–7 days, the number of flowers increased by two, extended the storage life for 1–3 days and increased the number of cormlets. The integration of these two treatments -Biorend® and hot water- might be a good option for increasing the gladiolus plant quality and vase life.  相似文献

Transferring ecological information across scale often involves spatial aggregation, which alters information content and may bias estimates if the scaling process is nonlinear. Here, a potential solution, the preservation of the information content of fine-scale measurements, is highlighted using modeled net ecosystem exchange (NEE) of an Arctic tundra landscape as an example. The variance of aggregated normalized difference vegetation index (NDVI), measured from an airborne platform, decreased linearly with log(scale), resulting in a linear relationship between log(scale) and the scale-wise modeled NEE estimate. Preserving three units of information, the mean, variance and skewness of fine-scale NDVI observations, resulted in upscaled NEE estimates that deviated less than 4% from the fine-scale estimate. Preserving only the mean and variance resulted in nearly 23% NEE bias, and preserving only the mean resulted in larger error and a change in sign from CO2 sink to source. Compressing NDVI maps by 70–75% using wavelet thresholding with the Haar and Coiflet basis functions resulted in 13% NEE bias across the study domain. Applying unique scale-dependent transfer functions between NDVI and leaf area index (LAI) decreased, but did not remove, bias in modeled flux in a smaller expanse using handheld NDVI observations. Quantifying the parameters of statistical distributions to preserve ecological information reduces bias when upscaling and makes possible spatial data assimilation to further reduce errors in estimates of ecological processes across scale.  相似文献
