Genetic improvement in reproductive efficiency through selection is difficult because many reproductive traits are binomial and have low heritabilities. Before genetic markers can be generated for fertility in cows, greater characterization of reproductive phenotypes is needed to understand the components of the trait. The current study tested the hypotheses that: 1) breeds vary in postpartum interval to estrus (PPIE) and estrous cycle length, 2) a longer estrous cycle immediately before breeding increased pregnancy rates, and 3) a greater number of cycles before breeding increased conception rates. The postpartum interval to estrus, estrous cycle length, and number of cycles before breeding were examined in F1 cows (n = 519) obtained from mating Hereford, Angus, and MARC III cows to Hereford, Angus, Simmental, Limousin, Charolais, Gelbvieh, and Red Angus sires. Cows were classified as having 0, 1, 2, or 3 observed estrous cycles before breeding. All traits analyzed were adjusted to constant BCS. Sire breed of the cow influenced length of the PPIE and number of cycles before the start of breeding (P <0.001). Simmental-sired cows had the shortest PPIE and greatest number of cycles before breeding, whereas Limousin-sired cows had the longest PPIE and least number of cycles before breeding. Cows with a greater number of cycles before breeding did not have greater conception rates than cows that had not exhibited standing estrus before breeding (P = 0.87). In cows that cycled before breeding, the length of the estrous cycle immediately before breeding was influenced by dam breed and BCS (P <0.01). Cows out of Hereford dams had shorter estrous cycles than cows out of MARC III or Angus dams, and estrous cycle length increased as BCS increased. Conception rate decreased as length of the estrous cycle immediately before breeding increased (P = 0.05, -2.2% per d of cycle length). Therefore, previously anestrous cows were just as likely to conceive as cows that had cycled before breeding, and an increased number of observed estrous cycles before breeding did not increase conception rates. There may be an influence of the length of estrous cycle immediately before breeding on conception rates, possibly because a longer estrous cycle results in a persistent follicle with greater potential for a lower quality oocyte. Breed differences in PPIE and estrous cycle length suggest that there are genetic components to these traits.  相似文献

The objectives of this study were to 1) estimate the allelic frequencies in US beef cattle of 6 DNA markers reported to be associated with variation in dairy production traits; and 2) evaluate the association of these markers with beef production traits. Several genetic markers have been associated with milk yield or composition, including polymorphisms in secreted phosphoprotein 1 (SPP1; also called osteopontin), growth hormone receptor (GHR), casein S1 (CSN1S1), diacylglycerol O-acyltransferase 1 (DGAT1), peroxisome proliferator-activated receptor gamma co-activator-1alpha (PPARGC1A), and ATP-binding cassette subfamily G (white) member 2 (ABCG2). Allelic frequencies for these 6 markers, and their association with 21 phenotypes, were evaluated in 2 crossbred beef cattle populations that sample influential industry sires. Five of 6 markers were segregating in beef cattle populations; the exception was ABCG2. The SPP1 marker was associated with yearling weight (P = 0.025), live weight at slaughter (P = 0.016), postweaning ADG (P = 0.007), and HCW (P = 0.007) in a large, multisire population representing the 7 most populous beef breeds in the United States. Postweaning growth trait associations were confirmed in an independent population of similar construction, including sires from tropically adapted breeds. The SPP1 marker was associated with yearling weight (P = 0.034), live weight at slaughter (P = 0.011), and postweaning ADG (P = 0.015) and showed a trend toward association with HCW (P = 0.083) in this population. Whereas DGAT1, GHR, and CSN1S1 polymorphisms showed association with some traits in individual populations, the lack of consistent predictive merit between populations indicates they may not be suited for beef cattle selection. No significant associations were observed for the PPARGC1A marker and any of 21 recorded traits, indicating this marker had no apparent value in selection for the beef cattle traits tested in these populations. The SPP1 marker had consistent associations and effect sizes (10.5 to 11.5 kg of live weight at slaughter) in both populations, providing strong evidence for utility of the SPP1 marker for postweaning growth in beef cattle.  相似文献

Microsatellites or Simple Sequence Repeats(SSRs) are informative molecular genetic markers in many crop species. SSRs are PCR-based, highly polymorphic, abundant, widely distributed throughout the genome and inherited in a co-dominant manner in most cases. Here we describe the presence of SSRs in cDNAs of cotton. Thirty one SSR primer pairs of 220 (∼14%) tested led to PCR amplification of discrete fragments using cotton leaf cDNA as template. Sequence analysis showed 25% of 24randomly selected cDNA clones amplified with different SSR primer pairs contained repeat motifs. We further showed that sequences from the SSR-containing cDNAs were conserved across G. barbadense and G. hirsutum, revealing the importance of the SSR markers for comparative mapping of transcribed genes. Data mining for plant SSR-ESTs from the publicly available databases identified SSRs motifs in many plant species,including cotton, in a range of 1.1 to4.8% of the submitted ESTs for a given species. Purpose

Suspended particulate matter (SPM) plays an important role in the transport and fate of contaminants in the environment. To better understand the relationships between contaminants and SPM, SPM properties, and their variations with flow regime, river size, land use, and season should be considered.

Materials and methods

The grain size distribution, elemental composition, and mineralogy of SPM from different stations along the Moselle River (Lorraine, France) were investigated at the particle scale during different flow regimes. The resulting data were compared with the elemental composition of the dissolved compartment to understand the role of particles in element transport.

Results and discussion

The grain size distribution, elemental composition, and mineralogy of SPM along the Moselle River and during different flow regimes showed only slight variations, except for the Fensch and Orne tributaries, two rivers that were impacted by inherited steel-making industrialization and different land use. In the Moselle River, SPM mainly consisted of clay minerals, while in Fensch and Orne Rivers, SPM mainly consisted of multiple types of anthropogenic particles. The diffuse urbanization gradient was hardly recognized based on the Trace Metal Element (TMEs) content in the river SPM, while the rivers impacted by the steel industries had greater TME contents. Finally, the TME content in the Moselle SPM was more strongly influenced by water flow than by the position of sampling on the linear reach of the Moselle River. The partitioning of TMEs in the particles and water at the main Moselle station (Frouard) revealed that SPM predominantly contributed to TMEs transport.

Conclusions

This study confirmed that catchment geology greatly contributed to the SPM composition in the mean-sized rivers. In addition, the high anthropogenic pressure could be deciphered for small tributaries. Furthermore, this study allowed us to observe the high contribution of particles to TMEs and Rare Earth Element (REEs) transportation.

Summary Bradyrhizobial strain BTAi 1 nodulates both stems and roots of Aeschynomene spp. Previous work has shown that it contains bacteriochlorophyll a and forms photosynthetic reaction centers, and has provided indirect evidence of photosynthesis by bacteroids within stem nodules. Here we report physiological and biochemical characteristics of BTAi 1 ex planta, which also suggest the presence of photosynthetic activity. Light-stimulated uptake of 14CO2 by BTAi 1 was detected at all stages of growth. Inhibitors of photosynthesis, 1,10-orthophenanthroline and 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), and the uncoupler NH4Cl, immediately suppressed light-driven 14CO2 uptake and increased O2 uptake. BTAi 1 is strictly aerobic and was unable to grow without organic C even in the light; also, it was unable to grow chemoautotrophically in an atmosphere enriched with H2 and CO2. In micro-aerobic conditions, strain BTAi 1 expressed acetylene reducing activity ex planta in an N-free medium. The highest rates of light-stimulated 14CO2 uptake and acetylene-reducing activity occurred during the exponential and early stationary phases of growth. Acetylene-reducing rates at a low glucose concentration were increased following a light-dark cycle in comparison with continuous dark conditions.  相似文献

Orchard and vineyard producers conduct preplant site evaluations to help prevent planting permanent tree and vine crops on lands where the crop will not perform to its highest potential or attain its full life expectancy. Physical soil characteristics within specific soil profiles and spatially throughout an orchard influence decisions on land preparation, irrigation system selection, horticultural choices, and nutrient management. Producers depend on soil surveys to help them understand the soil characteristics of the land and may be interested in technology that provides additional information. Electromagnetic induction (EM38) and four-probe soil resistance sensors (VERIS) are being used in combination with global positioning systems to map spatial variability of soils using apparent soil electrical conductivity (ECa). The hypothesis evaluated in this study is whether rapid, in situ, and relatively low-cost methods of measuring ECa (EM38 and VERIS) can effectively identify and map physical soil variability in non-saline soils. The supposition is that in non-saline soils, ECa levels will relate well to soil texture and water-holding capacity and can be used to map physical soil variability. In turn, the information can be used to guide decisions on preplant tillage, irrigation system design, water and nutritional management, and other horticultural considerations. Two sites in the Sacramento Valley were mapped each with EM38 and VERIS methods. Site-specific management zones were identified by each provider on ECa maps for each site, and then soil samples were collected by University of California researchers to verify these zones. Results showed that on non-saline soils, ECa measured with both EM38 and VERIS correlate with physical soil properties such as gravel, sand, silt, and clay content but the relationship between conductivity and these physical soil properties varied from moderately strong to weak. The strength of the correlation may be affected by several factors including how dominant soil texture is on conductivity relative to other soil properties and on methods of equipment operation, data analysis and interpretation. Overall, the commercial providers of ECa surveys in this study delivered reasonable levels of accuracy that were consistent with results reported in previous studies. At one site, an ECa map developed with VERIS provided more detail on physical soil variability to supplement published soil surveys and aided in the planning and development of a walnut orchard. At a second site, almond yield appeared to correlate well with distinctly different soil zones identified with EM38 mapping.  相似文献

The adult Drosophila midgut contains multipotent intestinal stem cells (ISCs) scattered along its basement membrane that have been shown by lineage analysis to generate both enterocytes and enteroendocrine cells. ISCs containing high levels of cytoplasmic Delta-rich vesicles activate the canonical Notch pathway and down-regulate Delta within their daughters, a process that programs these daughters to become enterocytes. ISCs that express little vesiculate Delta, or are genetically impaired in Notch signaling, specify their daughters to become enteroendocrine cells. Thus, ISCs control daughter cell fate by modulating Notch signaling over time. Our studies suggest that ISCs actively coordinate cell production with local tissue requirements by this mechanism.  相似文献

A new HTLV-III/LAV protein encoded by a gene found in cytopathic retroviruses   总被引:25,自引:0,他引:25  
The DNA of the HTLV-III/LAV group of retroviruses contains certain additional open reading frames that are not found in typical avian or mammalian retroviruses. The role of these sequences in encoding for gene products that may be related to pathogenesis remains to be resolved. An open reading frame whose 5' end overlaps with the pol gene, but is unrelated to the env gene, has been observed in HTLV-III/LAV and visna virus, both cytopathic mammalian retroviruses. Evidence presented here shows that this open reading frame is a bona fide coding sequence of HTLV-III/LAV and that its product, a protein with a molecular weight of 23,000, induces antibody production in the natural course of infection.  相似文献

Antibodies from the serum of patients with the acquired immune deficiency syndrome (AIDS) or with the AIDS-related complex and from the serum of seropositive healthy homosexuals, recognize two major glycoproteins in cells infected with human T-cell lymphotropic virus type III (HTLV III). These glycoproteins, gp160 and gp120, are encoded by the 2.5-kilobase open reading frame located in the 3' end of the HTLV-III genome, as determined by amino terminus sequence analysis of the radiolabeled forms of these proteins. It is hypothesized that gp160 and gp120 represent the major species of virus-encoded envelope gene products for HTLV-III.  相似文献
