Brazil has various species of domestic animals, which developed from breeds brought by the Portuguese settlers soon after the discovery of the country. After being submitted to a long process of natural selection, these breeds present characteristics adapted to specific Brazilian environmental conditions, and are known as “criollo”, “local” or naturalized breeds. From the beginning of the 20th century, some exotic breeds, selected in temperate regions, have begun to be imported, and, although more productive, they do not have adaptive traits, such as resistance to disease and parasites found in the naturalized breeds. Even so, they gradually replaced the native breeds, to such an extent that the latter are in danger of extinction. To avoid further loss of this important genetic material, in 1983 the National Research Center for Genetic Resources and Biotechnology (Cenargen) of the Brazilian Agricultural Research Corporation (Embrapa) decided to include conservation of animal genetic resources among its priorities. The conservation activities have been conducted under the Brazilian Genetic Resources Network — Renargen, and is carried out by various research centres of Embrapa, universities and State research institutions, as well as by private farmers, with a single coordinator at national level, Cenargen. The in situ conservation of cattle, horses, buffaloes, donkeys, goats, sheep and pigs is being carried out by Conservation Nuclei, located in the animal's original habitat. Ex situ conservation is centred at the Brazilian Animal Germplasm Bank (AGB), kept at Cenargen. This is responsible for the storage of semen and embryos of various breeds of domestic animals threatened with extinction. Presently the AGB has almost 60,000 doses of semen and more than 250 embryos, as well as over 7000 DNA samples. An important challenge for this program is to increase awareness within the different segments of society in terms of the importance of the conservation of animal genetic resources. The development and evolution of Renargen and its efforts to facilitate the conservation of genetic resources of livestock in Brazil are described in this paper. 相似文献

The increasing incidence of candidiasis has drawn the attention of scientists and clinicians attempting to improve methods of studying Candida yeasts. PCR amplification followed by agarose gel electrophoresis (PCR-AGE) and the manual method (morphological characteristics, biochemical profiles and culturing on CHROMagar-Candida) and VITEK 2 automated method were used to test a total of 30 fungal strains from dog sources. The strains were obtained from cases of dermatitis, otitis externa and from the ears, oral mucosa, vaginal mucosa, prepuce and perianal region of clinically normal dogs. After identification as Candida yeasts by the manual method, the strains were analyzed using both VITEK and PCR-AGE methods. Isolates of C. parapsilosis ATCC 22019, C. krusei ATCC 6258 and C. albicans ATCC 10231 were included as controls. The universal primers ITS1, ITS3 and ITS4 were used in two independent PCR reactions. Of 30 yeast isolates, 3 isolates (Saccharomyces cerevisiae, C. rugosa and C. parapsilosis) that were incompletely identified by the manual method were identified with the PCR-AGE and VITEK methods. The results revealed a 96.7% and 86.7% concurrent identification between the PCR-AGE and VITEK methods versus the manual method, respectively. PCR-AGE showed a greater level of concordance with the manual method, besides being faster and more sensitive than the other methods examined, and is therefore indicated for routine diagnostic testing of Candida spp. strains from veterinary sources. 相似文献
Parasites resembling Neospora caninum or Toxoplasma gondii were detected by cytologic examination of cerebrospinal fluid (CSF) from a dog with neurologic disease. The dog became severely ill and was euthanized. Canine tissue homogenates were used for direct parasite isolation in cell culture, bioassay in 2 mouse lineages, and PCR. T. gondii was isolated in monkey kidney cells, and species identity was confirmed by PCR. Inoculated parasites were highly virulent for mice, which developed clinical signs and were euthanized immediately. PCR-RFLP for T. gondii using the cultured isolate (TgDgBA22) was conducted with 12 genetic markers, and a unique recombinant strain was identified. Detection of T. gondii by CSF cytology, although described in humans, had not been reported previously in dogs, to our knowledge, and was crucial for the diagnosis of toxoplasmosis in the examined dog. 相似文献
The objective was to evaluate the intake, digestibility, milk yield and composition of cows fed mesquite pod meal (MPM). Five crossbred dairy cows, with an average body weight of 450?±?5 kg and producing 10?±?1 kg of milk/day were distributed in a 5?×?5 Latin square arrangement with five periods of 20 days each, comprised of 15 days for adaptation to the experimental diets (T1?=?0%, T2?=?1.5%, T3?=?3.0%, T4?=?4.5% and T5?=?6.0% MPM) and 5 days for data collection. Variables were assessed by analysis of variance and regression at P?<?0.01 and P?<?0.05 of significance, using the Student’s test. Animals were kept on pastures and the concentrates were provided at 6 kg/animal divided in two meals per day. The inclusion of 3.0% mesquite pod meal did not affect the intake, digestibility of nutrients and milk yield, but it changed milk composition. The dietary inclusion of 3.0% mesquite pod meal as a palatability enhancer increases the percentage of milk solids, lactose, fat and protein. Therefore, mesquite pod meal can be included in diets for dairy cows to improve the nutritive value of milk.
The effect of four extracts from neem seeds (Azadirachta indica) containing 2000, 5000, 9000 and 10,000 ppm of azadirachtin A (AZA), quantified by high-performance liquid chromatography (HPLC) and diluted to 1.25%; 2.5%; 5.0%; 10.0% and 12.8% was verified by in vitro tests with engorged females and larvae of the cattle tick Rhipicephalus microplus. The results from the bioassays with the engorged females showed that the main toxic effect of the extracts was reduction of the reproductive parameters, with a sharp drop in the number of eggs laid and the hatching rate, mainly when the extracts were diluted to 10.0% and 12.8%. The product effectiveness (PE) calculations for all the solutions tested showed that the AZA solution at 10,000 ppm (N10) was the most effective. However, statistical analysis of the PE data obtained for the proportional AZA concentrations in the different diluted extracts showed significance (P<0.05) of the effects included in the model (extract dilution, principle effect (classificatory) of the assay (extract) and the interaction between the two), indicating significant variations due to the dilution, the test and the interaction between the two factors in the tests with engorged females. For solutions N2, N5, and N9, it was not possible to estimate LC(90) values in the dilution range tested. The lowest LC(50) was observed for extract N5, and although extract N10 was the only extract for which the LC(90) could be estimated within the range tested, the LC(50) was higher than for N5 and N9. These results suggest that substances other than AZA present in the extracts influenced the efficacy, especially up to a certain LC range. In the tests with larvae, no mortality was observed, indicating zero effectiveness of all the extracts tested. The results of the tests with engorged females showed that the neem extracts had acaricide activity, inhibiting egg laying and the larval hatching rate. Complementary studies are necessary to develop new methods to isolate and/or identify other substances besides AZA contained in this plant, to enable using products made from it as acaricides. 相似文献
Carbon-isotope values of bulk organic matter from high-altitude lakes on Mount Kenya and Mount Elgon, East Africa, were 10 to 14 per mil higher during glacial times than they are today. Compound-specific isotope analyses of leaf waxes and algal biomarkers show that organisms possessing CO2-concentrating mechanisms, including C4 grasses and freshwater algae, were primarily responsible for this large increase. Carbon limitation due to lower ambient CO2 partial pressures had a significant impact on the distribution of forest on the tropical mountains, in addition to climate. Hence, tree line elevation should not be used to infer palaeotemperatures. 相似文献
Field studies throughout the year focused on the life history of the small rice stink bug, Oebalus poecilus, in southern Brazil. In addition, laboratory experiments examined the bug's life cycle and photoperiodic responses. Adults remain dormant during most of the year. Although activity is restricted to the period between November and March, four generations can develop in each rice-growing season, and populations can reach economically harmful levels. Late-planted rice, however, escapes attack because diapause is induced during early March. The results indicate that delaying and restricting the time of rice planting can be an effective pest management tactic in Rio Grande do Sul, because it results in asynchrony between the pest and the crop. 相似文献
