The objective of this study was to quantify the genetic variability and determine the inheritance of Al tolerance in lucerne (or alfalfa) using a 4-parent diallel mating design. Regrowth root length (RRL), along with root length (RL) and total root length (TRL), was measured to indicate relative Al stress response using a hydroponic system in a 3 μM Al solution at pH 4.5. A diallel analysis indicated the significance of general combining ability (gca) variance for RRL but not the specific combining ability (sca) variance; the same result was obtained for TRL but not for RL. For both RRL and TRL, genetic variance appeared to be more important than the environmental variance. For RRL, a strong but non-significant correlation was indicated between parental performance and their gca effects; while mid-parent heterosis and/or over-dominance were detected, as associated with the expressed sca effects in several combinations. These results suggest the complex genetic nature and expression of Al tolerance in the 4-parent diallel crossing system tested. Al-tolerant parent, GAAT‘S’, was the most promising parent, conferring the highest gca effects for RRL as well as for other two characters. The existence of significant gca variance in RRL may also suggest the feasibility of improving Al tolerance through enhanced root regrowth using phenotypic recurrent mass selection to pyramid desirable Al-tolerant genes, focussing on parental lines and/or elite individual plants expressing long regrowth roots.  相似文献

The plant species composition of seminatural woods in Northeast Ireland was recorded. Three main types of woodland were identified in habitats where combinations of steep slope, poor drainage and accumulations of large boulders have, in the past, made them unsuitable for agriculture. Despite these characteristics, clearance for agriculture is proceeding rapidly, but mainly in woods from poorly drained sites. Badly drained woods also have a relatively higher proportion of regionally rare plant species and there is, therefore, an argument for some priority for their conservation.

The population size-structures of trees were used to predict future changes in canopy composition, possibly originating from the introduction of myxomatosis to Ireland. Such changes may give rise to management problems associated with canopy closure and a subsequent reduction of species diversity.  相似文献

An obscure sesquiterpene, rotundone, has been identified as a hitherto unrecognized important aroma impact compound with a strong spicy, peppercorn aroma. Excellent correlations were observed between the concentration of rotundone and the mean 'black pepper' aroma intensity rated by sensory panels for both grape and wine samples, indicating that rotundone is a major contributor to peppery characters in Shiraz grapes and wine (and to a lesser extent in wine of other varieties). Approximately 80% of a sensory panel were very sensitive to the aroma of rotundone (aroma detection threshold levels of 16 ng/L in red wine and 8 ng/L in water). Above these concentrations, these panelists described the spiked samples as more 'peppery' and 'spicy'. However, approximately 20% of panelists could not detect this compound at the highest concentration tested (4000 ng/L), even in water. Thus, the sensory experiences of two consumers enjoying the same glass of Shiraz wine might be very different. Rotundone was found in much higher amounts in other common herbs and spices, especially black and white peppercorns, where it was present at approximately 10000 times the level found in very 'peppery' wine. Rotundone is the first compound found in black or white peppercorns that has a distinctive peppery aroma. Rotundone has an odor activity value in pepper on the order of 50000-250000 and is, on this criterion, by far the most powerful aroma compound yet found in that most important spice.  相似文献

The disruption of casein micelles at alkaline pH was investigated using turbidity measurements. The rate and extent of disruption of casein micelles at alkaline pH (8.0-11.0) increased with pH. Furthermore, the extent of alkaline disruption increased with increasing temperature (5-40 degrees C). Preheating milk for 10 min at 90 degrees C did not influence the extent of alkaline disruption of casein micelles, suggesting that whey proteins do not influence the alkaline disruption process. Levels of ionic calcium and serum calcium and phosphate decreased in a logarithmic fashion with increasing pH, indicating precipitation of calcium phosphate onto the casein micelles. A mechanism for alkaline disruption of casein micelles is proposed, in which increasing the milk pH improves the solvent quality for the caseins, thereby leading to the disruption of casein micelles into their constituent nanoclusters; increases in the net-negative charge on the caseins on increasing pH may contribute to micellar dissociation.  相似文献

Interactions with food components may alter the resistance of food proteins to digestion, a property thought to play an important role in determining allergenic properties. The kinetics of breakdown of the bovine milk allergen alpha-lactalbumin during in vitro gastrointestinal digestion was found to be altered by interactions with physiologically relevant levels of phosphatidylcholine (PC), a surfactant that is abundant both in milk and is actively secreted by the stomach. Breakdown during gastric digestion was slowed in the presence of PC and accompanied by small alterations in the profile of resulting peptides, with little effect being observed during subsequent duodenal digestion. alpha-Lactalbumin was found to unfold at gastric (acid) pH, giving a CD spectrum similar to that obtained for the partially folded state it is known to adopt at pH values below its isoelectric point. Fluorescence polarization studies performed at low pH indicated that this partially unfolded form of the protein was able to penetrate into the PC vesicles. These interactions are probably responsible for the slowing of gastric digestion by reducing the accessibility of the protein to pepsin. These findings show that interactions with other food components, such as lipids, may alter the rate of breakdown of food proteins in the gastrointestinal tract. It underlines the importance of the food matrix in affecting patterns of food allergen digestion and hence presentation to the immune system and that in vitro digestion systems used for assessing digestibility of allergens must take account of surfactants.  相似文献

A quantitative study of the amount of air transported between the boundary layer and the free atmosphere is important for understanding air quality and upper tropospheric ozone, which is a greenhouse gas. Frontal systems are known to be an effective mechanism for the vertical transport of pollutants. Numerical experiments have been performed with a simple two-dimensional front model to simulate vertical transport of trace gases within developing cold fronts. Three different trace gases experiments have been done numerically according to the different initial fields of trace gases such as aerosol, ozone and H2O2. Trace gas field tilts to the east while the front tilts to the west. Aerosol simulation shows that pollutants can be transported out of the boundary to altitudes of about 10 km. The stratospheric ozone is brought downwards in a tropopause fold behind of the frontal surface. The meridional gradient in trace gas (H2O2) can cause a complicate structure in the trace field by the meridional advection.  相似文献

Long-term diversity-disturbance responses of soil bacterial communities to copper were determined from field-soils (Spalding; South Australia) exposed to Cu in doses ranging from 0 through to 4012 mg Cu kg−1 soil. Nearly 6 years after application of Cu, the structure of the total bacterial community showed change over the Cu gradient (PCR-DGGE profiling). 16S rRNA clone libraries, generated from unexposed and exposed (1003 mg Cu added kg−1 soil) treatments, had significantly different taxa composition. In particular, Acidobacteria were abundant in unexposed soil but were nearly absent from the Cu-exposed sample (P<0.05), which was dominated by Firmicute bacteria (P<0.05). Analysis of community profiles of Acidobacteria, Bacillus, Pseudomonas and Sphingomonas showed significant changes in structural composition with increasing soil Cu. The diversity (Simpsons index) of the Acidobacteria community was more sensitive to increasing concentrations of CaCl-extractable soil Cu (CuExt) than other groups, with decline in diversity occurring at 0.13 CuExt mg kg−1 soil. In contrast, diversity in the Bacillus community increased until 10.4 CuExt mg kg−1 soil, showing that this group was 2 orders of magnitude more resistant to Cu than Acidobacteria. Sphingomonas was the most resistant to Cu; however, this group along with Pseudomonas represented only a small percentage of total soil bacteria. Changes in bacterial community structure, but not diversity, were concomitant with a decrease in catabolic function (BioLog). Reduction in function followed a dose-response pattern with CuExt levels (R2=0.86). The EC50 for functional loss was 0.21 CuExt mg kg−1 soil, which coincided with loss of Acidobacteria diversity. The microbial responses were confirmed as being due to Cu and not shifts in soil pH (from use of CuSO4) as parallel Zn-based field plots (ZnSO4) were dissimilar. Changes in the diversity of most bacterial groups with soil Cu followed a unimodal response - i.e. diversity initially increased with Cu addition until a critical value was reached, whereupon it sharply decreased. These responses are indicative of the intermediate-disturbance-hypothesis, a macroecological theory that has not been widely tested in environmental microbial ecosystems.  相似文献

A Spatio-Temporal Downscaler for Output From Numerical Models   总被引:2,自引:0,他引:2  
Often, in environmental data collection, data arise from two sources: numerical models and monitoring networks. The first source provides predictions at the level of grid cells, while the second source gives measurements at points. The first is characterized by full spatial coverage of the region of interest, high temporal resolution, no missing data but consequential calibration concerns. The second tends to be sparsely collected in space with coarser temporal resolution, often with missing data but, where recorded, provides, essentially, the true value. Accommodating the spatial misalignment between the two types of data is of fundamental importance for both improved predictions of exposure as well as for evaluation and calibration of the numerical model. In this article we propose a simple, fully model-based strategy to downscale the output from numerical models to point level. The static spatial model, specified within a Bayesian framework, regresses the observed data on the numerical model output using spatially-varying coefficients which are specified through a correlated spatial Gaussian process.  相似文献
