Feline renal diseases are increasingly noted in veterinary practice. It is important to diagnose and identify the pathological basis of renal dysfunction accurately at an early stage, but there are only a few reports on this area in clinical veterinary medicine. We investigated the efficacy of measurement of urinary albumin (u-Alb) and urinary transferrin (u-Tf) for early diagnosis using 5-µl urine samples collected noninvasively by catheterization from normal (IRIS stage I) cats and cats with stage I chronic kidney disease (CKD). The u-Alb levels in normal and stage I CKD cats were 6.0 ± 4.5 and 11.2 ± 8.4 mg/dl, respectively, and the u-Tf levels were 0.09 ± 0.42 and 0.52 ± 0.79 mg/dl, respectively. Based on ROC curve analysis, the sensitivity and specificity of u-Alb and u-Tf were higher than those of the currently used biomarker, the plasma creatinine level. The sensitivity of u-Alb was higher than that of u-Tf, whereas
the specificity of u-Tf was higher than that of u-Alb. The validity of the threshold albumin level (20 mg/dl) was confirmed by measurements using SDS-PAGE. Since leakage of u-Tf in urine precedes leakage of u-Alb, inclusion of u-Tf in biochemistry tests may be appropriate for IRIS staging as a diagnostic marker of early diagnosis of renal disorder in cats. 相似文献

Most anesthetics have an immuno-suppressive effect on cellular and neurohumoral immunity,
and research shows that total intravenous anesthesia (TIVA) with propofol has a greater
immuno-protective effect than inhalational anesthesia in human medicine. However, in
veterinary clinics, these effects remain ambiguous. To clarify the details, we focused on
propofol and isoflurane, investigating clinical blood hematology and immunological
profiles drawn from healthy dogs under and after two anesthesia techniques. Twelve healthy
adult beagles were included in this study, randomly assigned to the propofol anesthesia
group (group P: n=6) or the isoflurane anesthesia group (group I: n=6). In both groups,
the number of lymphocytes in peripheral blood decreased after 2 hr of anesthesia (2 hr),
but group P showed significantly less decrease than group I. For T-lymphocyte subsets
examined by flowcytometry, the ratio of CD3+, CD4+ and CD8+ lymphocytes in the peripheral
blood mononuclear cell (PBMC) of group P at 2 hr also exhibited a high level compared to
group I. Moreover, for mRNA expression of cytokines measured by real-time PCR, the IL2
(pro-inflammatory cytokine) of group P showed no decrease like group I. The IL10
(anti-inflammatory cytokine) of group P also showed no increase like group I, while both
cytokines maintained nearly the same level until 2 hr. These results suggest that,
compared to propofol, isoflurane had more strongly immuno-suppression caused by
anesthesia, and propofol itself might have some immuno-protective effects. Thus, TIVA with

Tumor-promoting effects of ethyl tertiary-butyl ether (ETBE) were investigated in a 2-stage carcinogenesis bioassay with regard to hepatic and renal carcinogenesis in rats. Male 6-week-old Wistar rats were given drinking water containing N-ethyl-N-(2-hydroxyethyl)nitrosamine (EHEN), as an initiator, at a dose of 500 ppm for 2 weeks. Starting one week thereafter, the animals were administered ETBE at dose levels of 0 (control), 100, 300, 500 or 1,000 mg/kg/day by gavage for 19 weeks from week 4 to 22. Necropsy of all rats was performed at week 23, and livers and kidneys were examined histopathologically. Incidences of hepatocellular adenomas, and those of combined hepatocellular adenomas and carcinomas were significantly elevated in rats given 1,000 mg/kg/day ETBE, but not 100‒500 mg/kg/day ETBE, and there was a significant increase in the average numbers of lesions. No significant differences in incidences and average numbers of renal tubule neoplasms were found in rats administered 100‒1,000 mg/kg/day ETBE. However, the average numbers of atypical tubule hyperplasias, considered to be preneoplastic lesions, were significantly increased in rats given ETBE at 1,000 mg/kg/day, but not in rats given 500 mg/kg/day or lower doses. Thus, these results imply that ETBE has hepatic and renal tumor-promoting activities that affect EHEN-induced carcinogenesis in male rats, and the no-observed-effect level is 500 mg/kg/day under the present experimental conditions. 相似文献

Twelve basic taxoids and 22 neutral taxoids were isolated from basic and polar neutral fractions of the extracts of needles
and twigs of Taxus cuspidata. Among them, taxine NA-13, 3,11-cyclotaxinine NN-1, taxinine NN-6, 11(15→1)abeo-taxinine NN-1, taxine NA-8, and taxine NA-4 were isolated first from natural sources by us. The cytotoxic activity of isolated
compounds was evaluated against three human cell lines: normal human fibroblast cells (WI-38), malignant tumor cells induced
from WI-38 (VA-13), and human liver tumor cells (HepG2). 7-Epitaxol, 7-epicephalomannine, taxinine NN-6, taxine NA-2, taxuspine
H, and taxagifine were active toward VA-13 cells and 7-epitaxol, 7-epicephalomannine, taxinine NN-1, 9,10-deacetyltaxinine,
and taxagifine were active toward HepG2 cells. The multidrug-resistant (MDR) cancer reversal activity of isolated compounds
was evaluated on the basis of the amount of vincristine (VCR) accumulated in MDR human ovarian cancer 2780 AD. Taxine NA-8,
taxine NA-2, 5-cinnamoyl-10-acetyltaxicin II, and taxinine NN-1 indicated stronger MDR cancer reversal activity than verapamil.
The result of primary screening based on 39 human cancer cell lines suggests that taxinine NN-1 belongs to a new mechanistic
class and is a new anticancer agents. 7-Epicephalomannine was found to be an effective anticancer agent with tubulin as its
molecular target, which is the same as paclitaxel.

To test whether gonadal development of female eels could be promoted without any exogenous hormone treatments, we observed the effect of water temperature manipulation. After 3–5 months of water temperature treatments, three silver eels showed higher gonadosomatic indices (GSI). In particular, one eel in the 5–15°C fluctuating temperature treatment group (5°C daytime and 15°C night‐time) had the highest GSI of 8.5 with secondary yolk globule stage oocytes of large diameter (OD; 412 μm) after 3 months, which indicated definite gonadal development compared with those in the initial states (mean GSI, 2.4; OD, 226.7 μm). The 5–15°C fluctuating temperature and constant 5°C groups had low oocyte breakdown (atretic) rates. Because daily temperature fluctuations and cold daytime water are experienced by vertically migrating silver eels in the ocean, these temperature conditions may be key maturation process components that could be useful for hormone‐free artificial maturation. 相似文献

The effect of heating on the physicochemical properties of emulsions prepared with soybean soluble polysaccharide (SSPS) was investigated. The emulsions were stable after heating at 90 degrees C for up to 30 min. Heating at different pH values or in the presence of CaCl2 (<10 mM) did not affect the stability; however, at higher concentrations of calcium ions, the emulsion particle size increased. Two fractions, a high molecular weight (HMF) and a low molecular weight (LMF) fraction, were separated from the crude SSPS preparation by gel fitration. Emulsions prepared with SSPS/HMF (MW = 310-420 kDa) showed little change in size with heating, while the protein impurities of the SSPS/LMF fraction formed aggregates by heating at pH 7. Analysis of the heat-induced aggregation of the two fractions of SSPS suggested that the changes in SSPS functionality with heating can be attributed to the protein impurities (LMF) present in the SSPS. 相似文献

The nodulation tendency of indigenous soybean bradyrhizobia on Rj -genotype soybean cultivars was investigated using approximately 260 bradyrhizobia isolated from an Andosol with 13 soybean cultivars of five Rj -genotypes (non- Rj , Rj 2 Rj 3 , Rj 3 , Rj 4 and Rj 2 Rj 3 Rj 4 ). A dendrogram was constructed based on restriction fragment length polymorphism analysis of the polymerase chain reaction products (PCR-RFLP) of the 16S–23S rDNA internal transcribed spacer (ITS) region. Bradyrhizobium USDA strains were used as a reference. The dendrogram indicated nine clusters based on similarities among the reference strains. The ratio of beta diversity to gamma diversity ( H' β / H' γ ), which represents differences in the bradyrhizobial communities by pair-wise comparison between each cultivar, was obtained from Shannon–Wiener diversity indices. The results showed that bradyrhizobial communities among the same Rj -genotype cultivars were similar to each other, whereas bradyrhizobial communities between the Rj 2 -genotype and non- Rj , Rj 3 or Rj 4 -genotype cultivars were significantly different. These results suggest that the Rj 2 -gene might not only affect the nodulation compatibility between Rj -genotype soybeans and bradyrhizobia, but also the nodulation tendency of the bradyrhizobia. 相似文献

对绿茶和儿茶素进行了焙制(品烘焙和炒培)以测定其抗突变效果,结果表明复火茶抑制Trp-P-1诱导突变与1-NP诱导突变的效果分别比原绿茶提高了45.35%-70.24%和45.06%-108.88%;用同样的方法测定儿茶素异构体的抗突变效果,结果表明其抑制Trp-P-1和1-NP的效果都有所增强。 相似文献
