试验选用3种丛枝菌根真菌(AMF)接种苜蓿品种金皇后,研究了5个梯度NaCl(0、50、100、150和200 mmol/L)对金皇后苜蓿抗盐生理特性的影响。结果表明:金皇后苜蓿的脯氨酸含量和可溶性糖含量随着NaCl浓度的增加总体呈先升高后降低的趋势,在100 mmol/L NaCl处理下达到最高,接种Gm真菌的二者含量分别为6.37和1.91μg/g;金皇后丙二醛(MDA)含量随着NaCl浓度的增加呈升高趋势,且在4个NaCl浓度之间差异显著;超氧化物歧化酶(SOD)和过氧化物酶(POD)活性随着NaCl浓度的增加呈先升高后降低的趋势,在150 mmol/L NaCl处理下二者活性下降,接种Gm真菌的2种抗氧化酶活性分别比处理组下降了48.48%和43.71%;同时金皇后在盐环境下光合指标的变化幅度较大,其中Ci随着NaCl浓度的增加而增加,P_n,G_s和T_r随着NaCl浓度的增加而下降;金皇后最大耐盐程度为150 mmol/L NaCl,且接种摩西球囊霉(Gm)效果优于幼套球囊酶(Ge)和根内球囊霉(Gi)。 相似文献

Baitfish such as golden shiners are subjected to stress during harvesting, grading, and transport. Their small size makes
it difficult to measure the stress response with the biological indicator cortisol using conventional assay methods for plasma.
This paper examines the development and validation of methods for whole-body cortisol extraction from individual baitfish.
Three types of extracts were tested: (1) an ethyl ether unaltered extract (UA); (2) an extract reconstituted in phosphate
buffered saline (PBS); (3) an extract that had been increased in volume by the addition of food-grade vegetable oil (VO).
These extracts were evaluated using validation tests with radioimmunoassays (RIA) and enzyme-linked immunosorbent assays (ELISA).
The UA extract produced inadequate volumes of extract for multiple assays and could not be used for the determination of cortisol
in a single fish. The PBS reconstitution method failed the precision recovery of serial dilutions (62.3%), linearity (R
2: 0.7864), and parallelism validation tests. The VO volume-boosting method passed all validation tests [intra-assay coefficent
of variation (%CV): 16.3 for ELISA and 5.9 for RIA; inter-assay %CV: 10.3; spiked recovery: 102.0%; dilution recovery: 93.0%;
linearity R
2: 0.9435; log of serial dilutions was parallel] and provided enough extract for multiple assays from an individual baitfish.
Based on these results, we conclude that the VO volume-boosting method presents a means for determining cortisol from individual

This investigation examined the effects of live prey availability on growth and survival of Sepia officinalis. Two independent experiments, comprising two feed rations each, were performed, using adequate prey size. In the first experiment, cuttlefish hatchlings were fed live mysids, Paramysis nouvelli [(feed ratio I (fr I)], at 15% body weight per day (bw day?1) (fr I15) and 30% bw day?1 (fr I30). In the second experiment, juvenile cuttlefish were fed live Atlantic ditch shrimp, Paleomonetes varians (fr II), under the same experimental design. In both experiments, the final mean weight, feeding rate and instantaneous growth rate were higher when animals were fed feed ratio fr II15 and fr II30 (30% bw day?1). The results indicate that prey availability influenced weight gain, irrespective of the prey used, during the first 2 months of cuttlefish life cycle. This effect seems to be more noticeable when a certain limit of prey is achieved. For cuttlefish fed fr II, the optimal prey density is thought to be under 2.5 g prey L?1 (i.e. 14 shrimp L?1). Results indicate that cuttlefish can withstand prey densities up to 120 mysids L?1 for cuttlefish up to 3 weeks old and 19 shrimps L?1 for cuttlefish up to 6 weeks old. 相似文献

Pome fruits are poor in nitrogenous compounds and the addition of nitrogen can improve colonisation of the fruits by antagonists. Twenty-two nitrogenous compounds were evaluated for their effect on Candida sake (CPA-1) growth in vitro. Ten compounds that induced greater growth were applied with the antagonist to wounded fruits to evaluate their effect on enhancing control of Penicillium expansum. Calcium chloride and 2-deoxy-D-glucose were also tested. L-serine and L-aspartic acid enhanced biocontrol by C. sake against P. expansum on apples. On apples and pears, ammonium molybdate, calcium chloride and 2-deoxy-D-glucose improved the capacity of the antagonist to control P. expansum. The addition of ammonium molybdate at 1 mM allowed C. sake to be used on apples and pears at a lower concentration without diminishing control. Similar results were observed with the addition of calcium chloride to the antagonist. 2-deoxy-D-glucose at 6 and 18 mM enhanced biocontrol on pears by over 81%, but on apples the improvement of biocontrol was observed only at 6 mM. In cold storage, the combination of ammonium molybdate and C. sake completely eliminated the incidence of blue mould on pears, and reduced its severity and incidence by more than 80% on apples. 相似文献

金冠苹果采用38℃,72h或96h热空气处理,可降低0℃贮藏中果实的呼吸强度和乙烯释放量,延缓非溶性果胶的降解,维持较高的硬度、脆度,同时也会加速果皮的褪绿变黄和固酸比的上升,尤其是38℃96h处理的作用效果更为明显,显著提高了货架期后(20℃7d)苹果的质地品质和可接受程度,延缓衰老。苹果分别接种扩展青霉、灰葡萄孢霉,0℃和20℃条件下其病害发生率和腐烂均很严重,采用38℃96h热空气处理可以完全控制病害的发生,避免腐烂。可见,热空气处理一方面能延缓金冠苹果的后熟衰老,另一方面还能控制贮期病害的发生。 相似文献

The influence of 9 rootstocks (M2, M7, M25, M26, MM104, MM106, MM109, MM111 and seedling) on fruit quality at harvest and after storage of ‘Wellspur Delicious’ (WS) and ‘Goldspur’ (GS), and of 3 rootstocks (M7, M26 and MM106) on fruit quality of ‘Red King Delicious’ (RK) and ‘Golden Delicious’ (GD) apple (Malus domestica Borkh.) was evaluated during a 4-year period. Fruits from trees on M26 were larger, developed earlier color and soluble solids (SS), and maintained higher levels of acidity (at harvest and during storage) in comparison with other rootstocks. Fruit from trees on M2 tended to have high SS. Fruit color from trees on MM104, MM106 and MM109 tended to be comparatively poor. Individual growth rates, feeding rates (%BWd?1) and food conversions for cuttlefish (S. officinalis) hatchlings and juveniles were determined during this study. A flow‐through system was used. Water temperature reached 30 °C during the hottest part of the day, gradually decreasing to 25 °C during the night; salinity varied between 37 ± 3 ppt and lights were kept on for 14 h day?1. Hatchlings were placed in separate compartments with a water volume of 1.2 L. Juvenile cuttlefish (from 0.5 to 25 g) were placed in bigger baskets, with a water volume of 5.2 L. Water flow was 120 L h?1. The biggest cuttlefish used in these experiments (> 25 g) were gathered in groups of five and placed in circular tanks (water volume of 250–300 L). Thus, results obtained in this case are means and not individual data. During the first 10, 20, 30 and 40 days, mean growth rates (of all individuals sampled by age group) decreased consistently (11.8 ± 4.1, 9.8 ± 1.8, 8.1 ± 2.2 and 7.3 ± 0.7%BW?1 respectively); in similar fashion, mean feeding rates decreased with age group (33.7 ± 13.5, 22.0 ± 7.9, 17.3 ± 3.9 and 16.7%BWd?1 respectively). Mean food conversions varied between 3.6 and 2.5 between the age groups. When grouping results by weight class, similar patterns occur, as growth and feeding rates decrease consistently as cuttlefish grow bigger. Highest mean growth and feeding rates are obtained by hatchlings (< 0.1 g) with 12.4 ± 4.5 and 35.3 ± 15.1%BWd?1, respectively, while the lowest growth and feeding rates were recorded for the largest animals, between 15 and 25 g (3.4 ± 1.1 and 10.8 ± 4.1%BWd?1 respectively).
