Serum vascular endothelial growth factor in dogs with various proliferative diseases

Angiogenesis plays an important role in the proliferation and metastasis mechanisms of malignant tumors. Vascular endothelial growth factor (VEGF), a group of cytokines that contribute to angiogenesis and vasculogenesis. This study aimed to investigate the serum VEGF-A concentrations in dogs with various proliferative diseases. A total of 202 dogs that were histopathologically diagnosed with proliferative diseases were included in the study. Serum VEGF-A concentrations were measured using enzyme-linked immunosorbent assay. Median serum VEGF-A concentrations in dogs were as follows: healthy dogs, 4 pg/ml [0–21 pg/ml]; hepatocellular carcinoma, 30 pg/ml [0–158 pg/ml, P=<0.001]; hepatocellular adenoma, 32 pg/ml [0–49 pg/ml, P=0.003]; hepatic nodular hyperplasia, 18 pg/ml [0–51 pg/ml, P=0.595]; adrenal pheochromocytoma, 32 pg/ml [0–187 pg/ml, P=<0.001]; adrenocortical carcinoma, 32 pg/ml [3–161 pg/ml, P=0.002]; adrenocortical adenoma, 27 pg/ml [0–106 pg/ml, P=0.005]; colorectal adenocarcinoma, 36 pg/ml [0–75 pg/ml, P=0.002]; colorectal adenoma, 43 pg/ml [0–48 pg/ml, P=0.144]; inflammatory colorectal polyps, 37 pg/ml [0–111 pg/ml, P=<0.001]; pulmonary adenocarcinoma, 35 pg/ml [4–107 pg/ml, P=0.002]; pulmonary histiocytic sarcoma, 35 pg/ml [0–131 pg/ml, P=0.016]; and follicular thyroid carcinoma, 35 pg/ml [0–106 pg/ml, P=0.009]. The serum VEGF-A concentrations were significantly higher in dogs with neoplastic lesions compared to healthy dogs, except for colorectal adenoma. High serum VEGF-A concentrations were observed in dogs with proliferative diseases. The present study suggests that angiogenesis-inhibiting therapy, which targets VEGF-A, may be useful for canine neoplastic diseases.     

Effects of synchronization of donor cell cycle on embryonic development and DNA synthesis in porcine nuclear transfer embryos

The relationship between donor cell cycle and the developmental ability of somatic cell nuclear transfer (SCNT) embryos has not fully been elucidated. Donor cells that are usually prepared by serum starvation or confluent-cell culture for SCNT represent a heterogeneous population that includes mainly G0 phase cells, other cells in different phases of the cell cycle and apoptotic cells. In this study, we compared the developmental ability of porcine SCNT embryos reconstructed from G0 phase cells (G0-SCNT embryos) and strictly synchronized-G1 phase cells (G1-SCNT embryos), and examined the developmental rates and timing of first DNA synthesis. The G0 phase cells were synchronized by confluent culture, and the G1 phase cells were prepared from actively dividing M phase cells. The G1-SCNT embryos showed a significantly higher (P<0.05) developmental rate to the blastocyst stage per cleaved embryo (59%) than the G0-SCNT embryos (43%). Moreover, initiation of first DNA synthesis and cleavage occurred significantly earlier in the G1-SCNT embryos than in the G0-SCNT embryos. Delay of initiation of first DNA synthesis in the SCNT embryos by aphidicolin resulted in decreased developmental rates to the blastocyst stage without any effect on cleavage rates. Our data demonstrates that synchronized-G1 phase cells can be used as donor cells for SCNT embryos and that earlier initiation of first DNA synthesis may be important for subsequent development of SCNT embryos. The SCNT system using G1-synchronized cells, in terms of their highly uniform and viable cell states, can be useful for studying the reprogramming processes and embryonic development of SCNT embryos.     

Comparison of behavioral effects of morphine and fentanyl in dogs and cats

Behavioral effects induced by intravenous administration of morphine at 0.3, 0.6, 1.2, and 2.4 μg/kg and fentanyl at 5, 10, 20, and 40 μg/kg were evaluated in dogs and cats. In dogs, fentanyl and morphine depressed activity and level of consciousness in a dose- dependant manner. In cats, higher doses of fentanyl stimulated activity temporarily, but excitement, so-called "opioid mania," was not observed. Morphine induced distinctive behavioral changes characterized by sitting with fixed staring, and "opioid mania" was not observed in cats.     

Use of mathematical modeling and its inverse analysis for precise assessment of pesticide dissipation in a paddy environment

The extrapolability of the lysimeter test as a dissipation simulator in an actual paddy field was evaluated using mathematical models and their inverse analyses for predicting pesticide fate and transport processes in paddy test systems. As a source of experimental data, a four-year comparative experiment in lysimeters and paddy fields was conducted using various paddy pesticides. First, the dissipations for various active ingredients in granule pesticides under submerged applications were statistically compared using simple kinetic modeling. Second, the dissipation pathways, unobserved experimental components, and effect of the experimental setting were evaluated using a higher tier mathematical model with a novel inverse analysis protocol. Finally, owing to experimental constraints, the unobtainable parameters were extracted from the laboratory container test before being transferred to compare the outdoor experimental data under different formulation types.     

Dynamic response of wall-floor joints of wooden light-frame constructions under forced harmonic vibrations

Shaking table tests of the wall-floor joints of wooden light-frame constructions under forced harmonic vibrations are conducted in this study so as to observe the dynamic responsive characteristics. The principal results are as follows: The responsive characteristics of timber constructions under strong earthquakes cannot be directly correlated with their resonant frequencies under free or forced vibrations with low input accelerations, because they behave as continuous bodies when the input accelerations are less than the apparent frictional limits of structural joints. The apparent frictional limits are reduced by periodic fluctuation of the effective vertical loads as a result of the vertical motion of the specimens. The characteristic dynamic responses of wall-floor joints depend clearly upon the frequency and input accelerations of forced vibrations. These dependencies arise from the nonlinear load-slip relationship of the wall-floor joints. The equivalent stiffness in their successive transient phases decreases as joint slip increases, which gradually changes the resonant frequencies of the wall-floor joints. This indicates that the frequency components dominant to ultimate or safety-limit resistance should be distinguished from those dominant to allowable or serviceability-limit resistance.     

Development of a shuttle polymerase chain reaction for the detection of bovine herpesvirus 2

Three different polymerase chain reaction (PCR) protocols were evaluated for their ability to detect bovine herpesvirus 2 (BoHV-2): single-step PCR with 3 reaction stages (denaturation, annealing and extension), 2 reaction stages (denaturation and annealing/extension; shuttle PCR), and semi-nested PCR with 3 reaction stages. All the PCR protocols showed the same sensitivity (detection limit of 0.4 TCID(50)). A non-specific band sometimes appeared in mock cell DNA at annealing temperatures below 64 degrees C. The shuttle PCR was found to be superior to the other protocols under consideration because of the speed of its application. Furthermore, no non-specific band was detected in DNAs of eight other DNA viruses. Thus, the shuttle PCR seems to be an excellent diagnostic tool for BoHV-2 infections.     

Geographic distribution of Waxy gene SNPs and indels in foxtail millet, Setaria italica (L.) P. Beauv.

To elucidate diversity and evolution of the Waxy gene in foxtail millet, Setaria italica, we analyzed sequence polymorphism of Waxy gene in 83 foxtail millet landraces collected from various regions covering the entire geographical distribution of this millet in Europe and Asia. We found a unique geographic distribution pattern at the sequence level of gene haplotypes and also found a large diversity in East Asian landraces. We also found a higher degree of genetic polymorphism in a non-waxy phenotype than in other low amylose types, supporting the hypothesis that low amylose types recently originated from non-waxy type.     

X-ray fluorescence spectrometry of asteroid Itokawa by Hayabusa

X-ray fluorescence spectrometry of asteroid 25143 Itokawa was performed by the x-ray spectrometer onboard Hayabusa during the first touchdown on 19 November 2005. We selected those data observed during relatively enhanced solar activity and determined average elemental mass ratios of Mg/Si = 0.78 +/- 0.09 and Al/Si = 0.07 +/- 0.03. Our preliminary results suggest that Itokawa has a composition consistent with that of ordinary chondrites, but primitive achondrites cannot be ruled out. Among ordinary chondrites, LL- or L-chondrites appear to be more likely than H-chondrites. No substantial regional difference was found on the asteroid surface, indicating its homogeneity in composition.
