Conservation of forest-dwelling caribou (Rangifer tarandus caribou) is of great concern across most of its range. Anthropogenic disturbances, primarily logging activities, have been identified as the most important cause of caribou decline, although the mechanisms underlying this decline are not fully understood. Caribou commonly display fidelity to calving sites or seasonal ranges, but the potential role of this life-history trait has been largely overlooked in research and conservation planning. This is surprising because sites and ranges with high inter-annual use should have high conservation value. We investigated the relationship between habitat disturbances and home-range fidelity of forest-dwelling caribou across three study sites in Québec, Canada, using a broad range of natural and anthropogenic disturbances. Between 2004 and 2007, we tracked 47 adult female caribou using GPS collars. Home-range fidelity varied between seasons, being higher during calving and summer, and lower during winter. Caribou reduced fidelity following natural and anthropogenic disturbances, the latter having a stronger negative influence. Anthropogenic disturbances had a strong negative impact on home-range fidelity during annual, summer and winter periods, whereas natural disturbance was the dominant factor during calving. Despite this negative influence on fidelity, caribou tended to demonstrate range fidelity even in study sites most impacted by human activities. Habitat disturbances could produce two possible outcomes for caribou conservation: (1) a trend for females to reduce home-range fidelity which could translate into lower calf and female caribou survival through reduced familiarity with food distribution, escape cover and predation risk and (2) a global tendency to maintain range fidelity even in a drastically modified landscape which could turn into an ecological trap, particularly for calves when predation risk increases due to increased black bear density in early successional forests. Taking range fidelity behavior into consideration during forest management planning could direct conservation efforts toward the best available sites and therefore facilitate caribou persistence in managed landscapes.  相似文献

Natural structural units of a luvisol under maize crop were studied to assess if soil structure directed sampling could improve the understanding of arrangements of bacteria in spatially constraint location. Three habitats were defined: (i) soil around fine lateral roots (rhizo-aggregates), (ii) soil close to basal roots (core clods) and (iii) unplanted soil between rows (bare soil clods). These habitats were also investigated with maize plants resulting from Azospirillum lipoferum CRT1 inoculated seeds as a model of enhanced fine root system. Rhizo-aggregates were clearly separated from each other (disconnected habitat) in contrast to micro-samples (fragments) from clods, which belong to cohesive macro-structures. Genetic fingerprints on metagenomic extracts were used to characterize the structure of bacterial communities on 95 micro-samples from the three habitats. For eubacteria, automated RISA (Ribosomal Intergenic Spacer Analysis) of ITS (Internal Transcribed Spacer) profiles were performed. PCR-RFLP on nifH gene were used to describe the N-fixer guilds. Exploratory multivariate analyses (PCA and MDS) revealed bacterial community patterns in the sampled habitats. On the basis of ITS profiles, rhizo-aggregates harboured closely related communities, distant from those of the unplanted soil, and each sampled rhizo-aggregate could therefore be considered as a sub-unit of the whole macro-habitat, comprising all the fine roots. The observed low dissimilarity of disconnected rhizo-aggregates is likely to result from the direct influence of maize root tips on the recruitment of rhizosphere bacteria. Molecular fingerprints of nifH from basal root clods (core) were more similar to bare soil than to rhizo-aggregates, indicating similar ecological conditions without, or with, at least, poor maize exudating root influence. Although our study was performed on a limited number of situations, the distribution of bacteria was revealed to be patterned by soil structure units, which is a first step to improve the modelling of microbial ecology in soils.  相似文献

Hypersensitivity dermatitides (HD) are commonly seen in cats, and they are usually caused by environmental, food and/or flea allergens. Affected cats normally present with one of the following clinical reaction patterns: head and neck excoriations, usually symmetrical self-induced alopecia, eosinophilic skin lesions or miliary dermatitis. Importantly, none of these clinical presentations is considered to be pathognomonic for HD skin diseases, and the diagnosis of HD is usually based on the exclusion of other pruritic diseases and on a positive response to therapy. The objectives of this study were to propose sets of criteria for the diagnosis of nonflea-induced HD (NFHD). We recruited 501 cats with pruritus and skin lesions and compared clinical parameters between cats with NFHD (encompassing those with nonflea, nonfood HD and those with food HD), flea HD and other pruritic conditions. Using simulated annealing techniques, we established two sets of proposed criteria for the following two different clinical situations: (i) the diagnosis of NFHD in a population of pruritic cats; and (ii) the diagnosis of NFHD after exclusion of cats with flea HD. These criteria sets were associated with good sensitivity and specificity and may be useful for homogeneity of enrolment in clinical trials and to evaluate the probability of diagnosis of NFHD in clinical practice. Finally, these criteria were not useful to differentiate cats with NFHD from those with food HD.  相似文献

In a previous study, a reduced efficiency of ileal digestible threonine (THR) use for body protein deposition was observed in growing pigs when pectin was included in the diet. This response was not due to increased physical endogenous ileal THR loss. Our aim was to explore the contribution of diet-induced increases in protein synthesis in the colon, especially mucins, to dietary THR requirements. Twelve barrows (21 kg mean BW) were fed either a cornstarch–soybean meal-based diet (Control) or Control with 12% pectin (Pectin). Pigs were given intravenously 1.5 mmol/kg BW of L-1-13C valine (40 mol%) to measure fractional and absolute synthesis rates (FSR, ASR, respectively) of mucosal and whole intestinal protein in the jejunum and colon. Dietary pectin inclusion increased plasma levels of glucose, isoleucine and glutamine (P < 0.05) but had no effect on insulin or urea nitrogen (P > 0.10). There were no differences in FSR and ASR of whole intestinal protein in jejunum and colon (P > 0.10). The FSR of mucosal proteins in colon, not in jejunum, was increased with dietary pectin supplementation (P < 0.05). Assuming mucosal protein mass is constant, these results imply that the higher protein synthesis in colon mucosa contributes to the reduced THR efficiency observed in pectin-supplemented diet.  相似文献

The effects of dietary level of tryptophan (TRP) and CP content and composition on voluntary feed intake, growth performance, and carcass characteristics in finishing pigs were studied in two experiments, with an equal number of females and castrated males. In Exp. 1, involving 120 Large White pigs from 44 to 99 kg BW with ad libitum access to feed, six treatments were compared according to a 2 x 3 factorial arrangement: 1) two levels of TRP (.09 and .13%), suboptimal and optimal for growth, respectively, 2) three types of CP supply (a 12.5% CP diet based on corn-soybean meal, and adequately balanced for essential amino acids [EAA] other than TRP; 15.7% CP diet with additional protein from corn gluten meal; 16.2% CP diet with additional nonessential amino acids [NEAA, in the form of L-glutamic acid.HCl and glycine], and the same levels of EAA as in the 12.5% CP diet. In Exp. 2, including four of the six previous factorial combinations (.09 and .13% TRP, 12.3 and 15.8% CP with additional protein), 32 pigs of 50-kg initial BW were used during 21 d, and further observations on meat quality characteristics, plasma free amino acid levels, and serotonin concentrations in the posterior hypothalamus were made. The major observed effects were interactions of different magnitude according to sex between TRP level and the amount and the composition of additional CP. At the suboptimal level of .09% TRP, the increase in protein content severely decreased daily feed intake and growth compared with the .13% level, especially in females. Conversely, the addition of NEAA at both TRP levels had little effect on daily feed intake and growth. Deficiency of TRP exerted a significant increase of pH in adductor femoris and semimembranosus muscles measured 45 min and 24 h postmortem, but only in females. Voluntary feed intake, as affected by dietary TRP and CP levels, was linearly related with concomitant changes in TRP to large neutral amino acids (TRP:LNAA) ratio both in feed and in plasma, which in turn was directly associated to hypothalamic serotonin concentration. It was concluded that an overly low concentration of serotonin in the hypothalamus, especially in females, as a result of TRP:LNAA imbalance, could be involved in the reduction of voluntary feed intake.  相似文献

Mucus plays an important role in gut health by favouring colonisation resistance. The aim of this study was to quantify the impact of fibre and protein on mucin recovery in ileal digesta and on goblet cell histochemistry in the proximal colon. A control diet with highly digestible protein and low fibre and three complex diets containing indigestible protein associated with low, soluble or insoluble fibre sources were tested for 14 days in piglets weaned at 28 days of age. Mucin concentration was determined by ethanol precipitation. Goblet cell subtypes in colonic crypts were analysed by histochemistry. The ileal mucin output was higher with the complex diets than with the control diet (31.6 on average vs 21.7 g/kg DM intake). Increases observed with the low and soluble fibre diets were similar (34 g/kg DM intake). This suggests a limited effect of soluble fibre in presence of indigestible protein. Surprisingly, the observed increase was lower with insoluble fibre (27.2 g/kg DM intake). DM and N output and digestibility were not affected by the diets, but a linear relationship was found between DM and mucin output. In proximal colon, crypt depth, goblet cell numbers per crypt and glycosylation subtypes were not affected by the diet suggesting no change in the capacity of mucus to protect the gut. To conclude, introducing highly indigestible protein in the diet increased ileal mucin output, without effects of these factors on crypt goblet cell patterns in the proximal colon. Introducing fibre in a diet containing highly indigestible protein had a marginal effect.  相似文献

Progressive states of maturing lentivirus: maedia visna virus (MVV) and human immunodeficiency virus (HIV), respectively, have been visualized by 2-D electron microscopy and by 3-D electron microscopic tomography. A major fraction of MVV and a low percentage of HIV appear as immature particles 4 to 5 days post virus infection. Upon budding the gag-precursor material is densely packed inside the external envelope. After virus release the major portion of precursors is assembled within an approximately 25 nm thick layer directly attached to the envelope. Structural maturation of the core is different for the two viruses. Pleomorphic cores are observed in mature MVV in contrast to structurally defined cores of HIV. The latter are principally cone-shaped, spanning the entire diameter of the virion with a 40 to 60 nm wide free end and an approximately 20 nm narrow end attached to the envelope with a core-envelope-link.  相似文献

OBJECTIVE: To develop a technique for use in investigation of healing of long-bone defects by creation of a critical-size defect in the left metarsal III and IV bone (metatarsus) of sheep. ANIMALS: 18 healthy adult sheep. PROCEDURE: Sheep were allocated to 4 groups (3, 3, 5, and 7 sheep in groups 1 to 4, respectively). An ostectomy with various segmental length-to-diaphyseal diameter ratios (0.5, 1.0, 2.0, and 2.0 for groups 1 to 4, respectively) was performed on the left metatarsus of each sheep. The defect was left empty in sheep of groups 1, 2, and 3, whereas the defect was filled with a massive corticocancellous bone autograft in sheep of group 4. RESULTS: All sheep tolerated the surgical procedure well and were able to use the affected limb the day after surgery. Radiographic and histologic examinations conducted 16 weeks after surgery revealed nonunion in all sheep of groups 1, 2, and 3, whereas consistent bone healing with abundant bone formation was observed in all sheep of group 4. CONCLUSIONS AND CLINICAL RELEVANCE: Analysis of these findings suggests that the sheep metatarsal model is a critical-size defect model with low morbidity. It should allow the assessment of new technologies for bone regeneration in conditions closely mimicking the clinical setting. IMPACT FOR HUMAN MEDICINE: Use of this technique in sheep should be of benefit for the preclinical study of osteoconductive, osteoinductive, or osteogenic biomaterials for use in humans.  相似文献

The present study describes the occurrence of various gastrointestinal helminths, lungworms and liver flukes in Norwegian sheep and goats as assessed from faecal samples and post mortem examinations performed between 2007 and 2010. Faecal samples for gastrointestinal nematode egg counts were collected from 77 sheep flocks and 30 dairy goat flocks from three geographical regions in Norway. Additionally, thirty-two lambs and 16 adult goats were euthanized for necropsy examination and for identification of adult gastrointestinal nematodes and tapeworms, lungworms and liver flukes. The survey showed that there was a higher mean excretion of trichostrongyle eggs in sheep than in goats at the individual level (392 EPG vs. 154 EPG, p < 0.001). For both host species, the mean prevalence and intensity of excreted trichostrongyle eggs were significantly higher in the southern coastal region compared with the inland and northern regions (p < 0.001). Third stage larvae of Trichostrongylus/Teladorsagia, Haemonchus and Nematodirus type were the most prevalent ones in the coprocultures from sheep, whereas larvae of Trichostrongylus/Teladorsagia and Nematodirus type dominated in goats. The most prevalent gastrointestinal nematode species found at necropsy was Teladorsagia circumcincta (75.0 and 81.2% respectively in sheep and goats), while the largest mean worm burdens were recorded for Haemonchus contortus in sheep (724 ± 623) and T. circumcincta in goats (377 ± 529). Other gastrointestinal nematode species were present at low prevalence or in low numbers.  相似文献
