1. Several economically important traits in two Leghorn populations (over 9000 birds) were examined for additive and non‐additive components of genetic variance and sex‐linked effects. Data were analysed by two different statistical models based on least‐squares procedures.

2. Six different covariances were first calculated between relatives; i.e., full‐sibs, 3/4‐sisters, half‐sisters, dam‐daughters, grandam‐granddaughters and aunt‐nieces.

3. From the covariances, weighted least‐squares equations were used to obtain estimates of variance components for additive genetic, dominance, maternal and sex‐linkage effects.

4. The estimates of non‐additive components were highly variable but generally small compared with the additive genetic estimates.

5. In general this study suggests that for most traits, with the possible exception of rate of egg production, there is relatively little non‐additive genetic variation.

6. The consequences of possible negative correlations between additive effects and maternal effects are considered as they might apply to egg production in poultry.  相似文献

The permeable nature of frog skin presents an alternative route for the delivery of therapeutic chemicals to treat disease in frogs. However, although therapeutic chemicals are often topically applied to the skin of frogs, their pharmacokinetics have rarely been reported. To provide evidence to guide both candidate drug and formulation selection, we highlight factors expected to influence percutaneous absorption through frog skin, including the anatomy and physiology of the skin and the physicochemical properties of applied therapeutic chemicals. Importantly, we also highlight the effects of the formulation on percutaneous absorption, especially the inclusion of potential penetration enhancers as excipients. Finally, we collate empirical data on the topical application of various therapeutic chemicals in postmetamorphic frogs and show that, in contrast to mammalian species, even large chemicals (i.e. >500 Da) and those with a wide range of log P values (?4 through +6) are likely to be absorbed percutaneously. Topical application in frogs thus promises a convenient and effective method for delivering systemic treatments of a diverse range of chemicals; however, further experimental quantification is required to ensure optimal outcomes.  相似文献

OBJECTIVE: To study the effect of unilateral synovitis in the distal intertarsal and tarsometatarsal joints on locomotion, including the compensating effects within and between limbs. ANIMALS: 4 clinically normal horses. PROCEDURE: Gait analyses including kinematics, force plate, and inverse dynamic analysis were performed at the trot before lameness, after which synovitis was induced by injecting endotoxin into the right distal intertarsal and tarsometatarsal joints. Gait analyses were repeated 24 to 30 hours later during lameness. Differences between the stride variables during the 2 conditions (lame and sound) were identified. RESULTS: Tarsal joint range of motion, peak vertical force, and vertical impulse were decreased during lameness. Mechanical deficits included a decrease in negative work performed by the tarsal extensors during the early stance phase and a decrease in positive work by the tarsal extensors during push off. No compensatory changes in work were performed by other joints within the lame hind limb during the stance phase. Vertical impulse in the diagonal forelimb decreased, but there were no significant changes in forces or impulses in the ipsilateral forelimb or contralateral hind limb. CONCLUSION AND CLINICAL RELEVANCE: Results indicate that horses are able to manage mild, unilateral hind limb lameness by reducing the airborne phase of the stride rather than by increased loading of the compensating limbs.  相似文献

Zusammenfassung Um eine schonende Keimhemmung bei Kartoffeln der SorteBona zu erzielen, wurden Versuche mit sehr weichen R?ntgenstrahlen von 12 keV Energie durchgeführt. Sogar mit einer Bestrahlungsdosis von 10.000 bis 15.000 r wird — bei der hier angewandten weichen R?ntgenstrahlung — eine über 22 Wochen andauernde Keimhemmung erreicht ohne Verschlechterung des ?usseren Ansehens und Geschmacks. Der Schwund der Knollen wurde einzeln bis 5.000 r gemessen und mit anwachsender Dosis bis zu 15.000 r eine starke Verminderung des Schwundes erzielt. Au?erdem wurde nachgewiesen, da? durch langstündiges, vorsichtiges Rollen in der beschriebenen Versuchsanordung ebenfalls eine Keimhemmung zu erreichen ist. Es wird versucht, diese Erscheinung biologisch und rechnerisch zu deuten. Aus praktischen Gründen w?re es wünschenswert, den neuen, erw?hnten Effekt weiter zu untersuchen.
Summary In order to bring about sparing sprout inhibition of theBona variety of potatoes experiments were undertaken with very weak X-rays of an energy of 12 keV. A radiation dose of only 10.000 to 15.000 r (with the weak X-radiation employed in this instance) was sufficient to inhibit sprouting for over 22 weeks without adversely affecting the appearance and flavour. The tuber shrinkage was individually measured up to 5.000 r and a marked reduction of shrinkage obtained with an increasing dose up to 15.000 r. It was also shown that sprouting may be inhibited by protracted, careful rolling in the described order of experiments. An attempt is made to explain this phenomenon biologically and analytically. From practical considerations it would be desirable to make a further study of the novel effect referred to.

Résumé Pour réaliser de fa?on délicate une inhibition de la germination des pommes de terre de la variétéBona, nous avons effectué des essais de traitement par les rayons X très mous d'une énergie de 12 keV. Même l'irradition à une dose de 10.000 à 15.000 r sufit déjà — dans le cas des rayons très mous utilisés dans ces essais — pour inhiber la germination pendant 22 semaines au moins sans nuire à l'aspect ni au go?t des tubercules. La perte de poids des tubercules fut mesurée isolément jusqu'à 5000 r. Les doses croissantes jusqu'à 15.000 r réduisaient fortement la perte de poids. Il a également été constaté que dans les conditions de l'essai, un roulement très lent et doux des tubercules permet aussi de retarder la germination. Nous avons tenté d'expliquer ce phénomène au point de vue biologique et statistique. Pour des raisons pratiques, il serait désirable que l'étude de cet effet nouvellement observé soit approfondie.

Constitutive Hedgehog (Hh) pathway activity is associated with initiation of neoplasia, but its role in the continued growth of established tumors is unclear. Here, we investigate the therapeutic efficacy of the Hh pathway antagonist cyclopamine in preclinical models of medulloblastoma, the most common malignant brain tumor in children. Cyclopamine treatment of murine medulloblastoma cells blocked proliferation in vitro and induced changes in gene expression consistent with initiation of neuronal differentiation and loss of neuronal stem cell-like character. This compound also caused regression of murine tumor allografts in vivo and induced rapid death of cells from freshly resected human medulloblastomas, but not from other brain tumors, thus establishing a specific role for Hh pathway activity in medulloblastoma growth.  相似文献

Pesticide contamination of soil and groundwater at agricultural chemical distribution sites is a widespread problem in the USA. Alternatives to land-farming or solid waste disposal include biostimulation and phytoremediation. This research investigated the ability of compost, corn stalks, corn fermentation byproduct, peat, manure, and sawdust at rates of 0.5% and 5% (w/w) to stimulate biodegradation of atrazine [6-chloro-N-ethyl-N'-(1-methyethyl)-1,3,5-triazine-2,4-diamine], metolachlor [2-chloro-N-(2-ethyl-6-methylphenyl)-N-(2-methoxy-1-methylethyl)acetamide], and trifluralin [2,6-dinitro-N,N-dipropyl-4-(trifluoromethyl)benzenamine] added as a mixture to soil. Initial concentrations were 175ᆾ mg atrazine kg-1 soil, 182ᆭ mg metolachlor kg-1 soil, and 165ᆫ mg trifluralin kg-1 soil. After amendment addition, 30% of the atrazine, 33% of the metolachlor, and 44% of the trifluralin was degraded over 245 days, which included 63 days' aging prior to amendment additions. Atrazine degradation was enhanced by 0.5% manure, 5% peat, and 5% cornstalk amendments compared to nonamended soils. Metolachlor degradation was enhanced by all amendments at the 5% level, except for compost and peat. Amendments had no effect on trifluralin degradation. The 5% addition of compost, manure, and cornstalks resulted in significant increases in bacterial populations and dehydrogenase activity. A second experiment compared the persistence of atrazine, metolachlor, and trifluralin applied in a mixture to their persistence in soil individually. A combined average of 123 mg atrazine kg-1 remained in soil treated with the three-herbicide mixture compared to 31 mg atrazine kg-1 remaining in soil treated with atrazine only. Atrazine mineralization and atrazine-degrading microorganisms were suppressed by high concentrations of metolachlor, but not by trifluralin.  相似文献

Streptococcus uberis is the most common environmental mastitis pathogen causing udder inflammations of different severities in dairy cows. The aim of the study was to investigate if the different clinical outcome of mastitis induced by different strains of S. uberis can be reflected in the mammary immune response. Mammary epithelial cells and somatic milk cells were treated with heat inactivated and living S. uberis of strain A and strain B in vitro. Strain A was repeatedly isolated from a chronically infected quarter during 8 months, and persisted in the quarter despite antibiotic treatment. Strain B caused an acute clinical mastitis and was not further isolated after a single antibiotic treatment. Treatment with Strain B induced a more pronounced increase of mRNA-expression of various immune factors (interleukin-8, interleukin-1beta, RANTES, and lactoferrin) in mammary epithelial cells than strain A. In contrast to mammary epithelial cells the response of removed somatic milk cells showed no differences between the stimulation with two S. uberis strains. Tumor necrosis factor-alpha mRNA expression was not differently induced by the two strains. In conclusion, the characteristics of different severities of mastitis that are induced by different S. uberis strains in vivo can also be reflected at the level of the immune response of the mammary gland in vitro.  相似文献

A fully computer-controlled apparatus was designed. It combines a glass reactor with a temperature-controlled hood, in which headspace volatiles are captured. Flavored liquids can be introduced into the reactor and exposed to conditions of temperature, air flow, shear rate, and saliva flow as they occur in the mouth. As the reactor is completely filled before measurements are started, creation of headspace just before sampling start prevents untimely flavor release resulting in real time data. In the first 30 s of flavor release the concentrations of the volatiles can be measured up to four times by on-line sampling of the dynamic headspace, followed by off-line trapping of the samples on corresponding Tenax traps and analysis using GC-TDS-FID. Flavor compounds from different chemical classes were dissolved in water to achieve concentrations typically present in food (micrograms to milligrams per liter). Most of the compounds showed constant release rates, and the summed quantities of each volatile of three 10 s time intervals correlated linearly with time. The entire method of measurement including sample preparation, release, sampling, trapping, thermodesorption, and GC analysis showed good sensitivity [nanograms (10 s)(-1)] and reproducibility (mean coefficient of variation = 7.2%).  相似文献

We hypothesized that maternal nutrition during the first 50 d of gestation would influence the abundance of hexose transporters, SLC2A1, SLC2A3, and SLC2A5, and cationic amino acid transporters, SLC7A1 and SLC7A2, in heifer uteroplacental tissues. Angus-cross heifers (n = 43) were estrus synchronized, bred via artificial insemination, and assigned at breeding to 1 of 2 dietary intake groups (CON = 100% of requirements to achieve 0.45 kg/d of BW gain or RES = 60% of CON intake) and ovariohysterectomized on day 16, 34, or 50 of gestation (n = 6 to 9/d) in a completely randomized design with a 2 × 3 factorial arrangement of treatments. Uterine cross-sections were collected from the horn ipsilateral to the corpus luteum, fixed in 10% neutral buffered formalin, sectioned at 5 µm, and stained via immunofluorescence for transporters. For each image, areas of fetal membrane (FM; chorioallantois), luminal epithelium (ENDO), superficial glands (SG), deep glands (DG), and myometrium (MYO) were analyzed separately for relative intensity of fluorescence as an indicator of transporter abundance. Analysis of FM was only conducted for days 34 and 50. No transporters in target areas were influenced by a day × treatment interaction (P ≥ 0.06). In ENDO, all transporters were differentially abundant from days 16 to 50 of gestation (P ≤ 0.04), and SLC7A2 was greater (P = 0.05) for RES vs. CON. In SG, SLC7A1 and SLC7A2 were greater (P ≤ 0.04) at day 34 vs. day 16. In DG, SLC2A3 and SLC7A1 were greater (P ≤ 0.05) for CON vs. RES heifers; furthermore, SLC7A1 was greater (P < 0.01) at day 50 vs. days 16 and 34 of gestation. In MYO, SLC7A1 was greater (P < 0.01) for CON vs. RES and was greater (P = 0.02) at days 34 and 50 vs. day 16. There were no differences in FM (P ≥ 0.06). Analysis of all uterine tissues at day 16 determined that SLC2A1, SLC2A3, and SLC7A2 were all differentially abundant across uterine tissue type (P < 0.01), and SLC7A1 was greater (P = 0.02) for CON vs. RES. Analysis of all uteroplacental tissues at days 34 and 50 demonstrated that all transporters differed (P < 0.01) across uteroplacental tissues, and SLC7A1 was greater (P < 0.01) for CON vs. RES. These data are interpreted to imply that transporters are differentially affected by day of gestation, and that hexose and cationic amino acid transporters are differentially abundant across utero-placental tissue types, and that SLC7A1 is responsive to maternal nutritional treatment.  相似文献
