The development of three separate rapid ELISAs for detecting antibodies in host serum to three different viruses is described. These include: 1. A direct antigen assay using enzyme labelled anti-canine Ig for detecting antibodies to canine parvovirus, 2. A competitive ELISA using a feline infectious peritonitis virus-specific monoclonal antibody labelled with enzyme, and 3. The stable carbon isotope technique has been widely used to infer the dietary ecology of a range of animal species; however calibration of the technique with animals fed known diets is essential for accurate back-calculation of dietary preferences. The aim of this study was to identify suitable samples and back-calculation methods to predict short-term (2 to 3 week) dietary selection by sheep among plants with C3 and C4 photosynthetic pathways. Variation in integration time of dietary carbon into plasma and faeces; diet-tissue discrimination of carbon isotopes (fractionation) and the importance of accounting for the digestible or indigestible components of the diet was investigated. The results indicate that faecal and rumen samples provided the most accurate prediction of short term dietary changes in sheep selecting between C3 and C4 plants. The most accurate back-calculation method for these samples used δ13C of the C3 and C4 plants and accounted for both diet-tissue discrimination and differences in the indigestibility between the C3 and C4 forage. For faecal samples, the organic matter content of the diet originating from C4 plants could be predicted with a mean error as low as 2.7%. Wool and plasma samples were not conducive to predicting proportion of C4 forage in the diet within 18 days after a change in diet; however plasma could be used to discriminate between animals fed 100% C3 and C4 diets after 3 days. The δ13C technique provides a valuable tool for researchers when designing pastures for dual environmental and production purposes. In 7 horses, 4 anastomoses were done in the small intestine in each, using the combinations of synthetic absorbable monofilament and multifilament suture materials with continuous- and interrupted-suture patterns in the serosubmucosal layer of a 2-layer inverting-suture technique. Horses were evaluated 30 days after the operation for adhesion formation, lumen diameter, evidence of chronic obstruction, and suture tract inflammation at the anastomosis. Postoperative obstruction occurred in 5 of the 7 horses, and 6 horses survived. One horse was euthanatized on postoperative day 6 after 48 hours of ileus and obstruction; necropsy revealed a partial intussusception involving the anastomosis done with continuous multifilament-suture material. Two other horses that became obstructed between postoperative days 3 and 5 had protracted ileus and gastric reflux up to 48 hours' duration, but survived. Horses that had obstruction after the 6th postoperative day recovered within 4 hours of onset. The continuous inverting-suture pattern in the serosubmucosal layer resulted in significantly (P less than 0.05) fewer adhesions than did the interrupted pattern, and suture material had no effect on adhesion formation. There was no significant difference in the percentage of reduction of lumen diameter between the variations of the 2-layer technique, and there was no evidence of chronic obstruction related to any of the anastomoses. Suture tract inflammation was moderate in the anastomoses, using continuous-multifilament suture, with neutrophils being the predominant cell type. The anastomoses with continuous-monofilament suture had mild inflammation with focal accumulations of neutrophils. Concerns exist that some uses of antimicrobials in cattle may lead to the emergence, proliferation, dissemination and persistence of resistant pathogenic bacteria in animal agriculture, which in turn can infect humans via the food supply. The degree of perceived risk varies with the clinical indication for which the antimicrobial in question is used. In this study, four uses of antimicrobials are considered, including in acutely sick, chronically sick, at-risk, and high-risk cattle, contrasting the degree of risk among these uses. Using a random sample of 103 feedlot cattle veterinarians and variables drawn from the theory of planned behavior, we predict differences in risk perception by clinical indication with differences in perceived efficacy of antimicrobials, social pressures to use antimicrobials, and moral obligations to use antimicrobials. In most models, veterinarians who perceived that others in the feedlot industry (i.e., other feedlot veterinarians, nutritionists, feedlot clients, and retained owners of cattle) were more likely to expect them to use antimicrobials in one situation versus another, the less likely those veterinarians perceived the risk of antimicrobial risk to be greater in the former versus the latter situation. Only two of these contrasts contained influences outside the immediate feedlot relationships. This exception involves the 'downstream' public: meat packers, retailers, and consumers. Veterinarians who believe that using antimicrobials for acutely sick cattle is more beneficial than using antimicrobials for chronically sick cattle were more likely to believe that antimicrobial resistance was a less probable outcome in acutely sick cattle than in chronically sick cattle. 相似文献
