The uptake and distribution of intramuscularly (IM) administered tritium-labeled polysulfated glycosaminoglycan (3H-PSGAG) in serum, synovial fluid, and articular cartilage of eight horses was quantitated, and hyaluronic acid (HA) concentration of the middle carpal joint was evaluated in a pharmacokinetic study. A full-thickness articular cartilage defect, created on the distal articular surface of the left radial carpal bone of each horse served as an osteochondral defect model. 3H-PSGAG (500 mg) was injected IM, between 14 and 35 days after creation of the defects. Scintillation analysis of serum and synovial fluid, collected from both middle carpal joints at specific predetermined times up to 96 hours post-injection, revealed mean 3H-PSGAG concentrations peaked at 2 hours post-injection. 3H-PSGAG was detected in cartilage and subchondral bone 96 hours post-injection in samples from all eight horses. There were no statistically significant differences in 3H-PSGAG concentration of synovial fluid or cartilage between cartilage defect and control (right middle carpal) joints.

HA assay of synovial fluid revealed concentrations significantly increased at 24, 48, and 96 hours post-injection in both joints. The concentration nearly doubled 48 hours post-injection. However, no statistically significant differences were found between synovial concentrations of HA in cartilage defect and control joints.

3H-PSGAG administered IM to horses, was distributed in the blood, synovial fluid, and articular cartilage. HA concentrations in synovial fluid increased after IM administration of polysulfated glycosaminoglycan.  相似文献

Three trickle irrigation schedules, two of which were scheduled according to soil water potential ( soil) (tensiometer method) and daily stem contraction (DSC) (dendrometer method) respectively and the other one was a schedule of restricted water supply, were applied to a mature peach orchard.The annual water application based on soil was greater than that based on DSC. However, tree growth, fruit size and leaf water potential (leaf) on the trees in the dendrometer scheduling plot did not differ from those in the tensiometer scheduling plot while the premature fruit drop and fruit bud initiation were greatly different. The restricted water supply treatment limited significantly both tree and fruit growth. In addition, the lower leaf was observed on the trees in this plot.Further study shows that use of the dendrometer method for scheduling irrigation satisfies the water needs of the plant and that the tensiometer method is less accurate.Abbreviations leaf leaf water potential - soil soil water potential - DSC daily stem contraction - LVDT linear variable displacement transducer - PET potential evapotranspiration  相似文献

通过基因枪转基因方法和双质粒共转化体系将Xa21基因转入优良恢复系明恢63,得到转基因系M12和M22,并进一步做田间试验、新品种选育和食用安全性评价。结果显示:M12和M22对中国的所有白叶枯病小种都表现出高度抗性,但与原受体品种明恢63相比,农艺性状上有许多变异,主要表现在:结实率、千粒重等农艺性状变差,与珍汕97A的配合力显著降低;但与温敏不育系(X07S、056S)配组的F1有较好的田间表现。通过多代回交和分子标记辅助选择,成功地将Xa21基因从M12株系转到保持系80-4B和不育系80-4A中,得到抗白叶枯病的皖21B和皖21A。并利用皖21A不育系选育出优良杂交组合抗优97(皖21A×R-18),该组合在两年区域试验和一年生产试验中产量表现突出,米质优良。对不同世代和不同遗传背景的转基因品系进行白叶枯病鉴定和Southern分析表明:Xa21基因都能稳定遗传和正常表达,连续16代的种植并没使其白叶枯病的抗性减弱或丧失,而且不论Xa21基因是纯合的还是杂合的都有相同的抗性表现。对大鼠和小鼠的饲喂试验表明:转基因大米实质上等同于非转基因大米,是安全无害的。  相似文献

Bottom soil samples were collected from 35 ponds in the vicinity of Samutprakarn, Thailand. Ponds ranged in age from 3 to 39 years and had been used continuously for production of tilapia. Liming materials had been applied in large amounts, and bottom soils of all ponds had pH above 7, low exchange acidity, and free carbonate. Pond soils often contained between 1% and 2% total sulphur, suggesting that they were potential acid–sulphate soils. However, acidity from sulphide oxidation was not expressed because carbonate in the soil neutralized it. Concentrations of total carbon seldom exceeded 4% and the average for organic carbon was 1.90%. The correlations between pond age and both total carbon and organic carbon concentration were weak (r=0.34 and 0.36 respectively). Concentrations of nitrogen in bottom soils did not differ with pond age and ranged from 0.1% to 0.3% with an average of 0.19%. The average ratio of concentrations of carbon and nitrogen was 11. Acid‐extractable phosphorus concentrations averaged 217 mg kg?1, but the phosphorus adsorption capacity averaged 768 mg kg?1 suggesting that soils still have considerable reserve capacity to adsorb phosphorus. Ponds can be used annually for semi‐intensive production of tilapia, and presumably other species, for many years without serious deterioration of bottom soil quality.  相似文献

The combination of (1)H NMR fingerprinting of lipids from gilthead sea bream (Sparus aurata) with nonsupervised and supervised multivariate analysis was applied to differentiate wild and farmed fish and to classify farmed specimen according to their areas of production belonging to the Mediterranean basin. Principal component analysis (PCA) applied on processed (1)H NMR profiles made a clear distinction between wild and farmed samples. Linear discriminant analysis (LDA) allowed classification of samples according to the geographic origin, as well as for the wild and farmed status using both PCA scores and NMR data as variables. Variable selection for LDA was achieved with forward selection (stepwise) with a predefined 5% error level. The methods allowed the classification of 100% of the samples according to their wild and farmed status and 85-97% to geographic origin. Probabilistic neural network (PNN) analyses provided complementary means for the successful discrimination among classes investigated.  相似文献

Hypersensitivity dermatitides (HD) are commonly seen in cats, and they are usually caused by environmental, food and/or flea allergens. Affected cats normally present with one of the following clinical reaction patterns: head and neck excoriations, usually symmetrical self-induced alopecia, eosinophilic skin lesions or miliary dermatitis. Importantly, none of these clinical presentations is considered to be pathognomonic for HD skin diseases, and the diagnosis of HD is usually based on the exclusion of other pruritic diseases and on a positive response to therapy. The objectives of this study were to propose sets of criteria for the diagnosis of nonflea-induced HD (NFHD). We recruited 501 cats with pruritus and skin lesions and compared clinical parameters between cats with NFHD (encompassing those with nonflea, nonfood HD and those with food HD), flea HD and other pruritic conditions. Using simulated annealing techniques, we established two sets of proposed criteria for the following two different clinical situations: (i) the diagnosis of NFHD in a population of pruritic cats; and (ii) the diagnosis of NFHD after exclusion of cats with flea HD. These criteria sets were associated with good sensitivity and specificity and may be useful for homogeneity of enrolment in clinical trials and to evaluate the probability of diagnosis of NFHD in clinical practice. Finally, these criteria were not useful to differentiate cats with NFHD from those with food HD.  相似文献

This study was conducted to compare water quality and channel catfish production in earthen ponds located in two dissimilar physiographic regions of the southeastern United States and supplied with water of disparate quality. Ponds at Auburn, Alabama are on acidic Piedmont soils and filled with poorly mineralized runoff water; ponds at Stoneville, Mississippi are on slightly alkaline alluvial clays and filled with groundwater of high total alkalinity and hardness. Channel catfish were stocked at 8,750 fish/ha, fed daily, and provided nightly aeration in 0.04-ha ponds at both sites. Ponds were managed as similarly as possible. Minimum daily water temperatures and pH were higher at Stoneville than at Auburn, and there were greater concentrations of suspended clay turbidity, dissolved inorganic phosphorus, total ammonia-nitrogen, and nitrite-nitrogen at Auburn than at Stoneville. The taxonomic composition of the phytoplankton community was broadly different between the two sites. Taste tests revealed off-flavor in fish at both sites, but there were no significant differences (P > 0.05) in flavor scores between sites. The quality of flavor was somewhat different between sites, and these differences in quality were thought to result from observed differences in the taxonomic composition of phytoplankton communities. All differences in water quality seemed to be directly or indirectly related to the dissimilarity in the quality of the water supply and soils at the two locations. Although some water quality variables differed between sites and changed over time at both sites, environmental conditions never deteriorated enough at either site to cause serious stress or mortality in fish. There were no significant differences (P > 0.05) in average net fish production, survival, weight of individual fish at harvest, or feed conversion ratios. Average net fish production and feed conversion ratios, respectively, were 4,905 kg/ha and 1.27 at Auburn and 5,286 kg/ha and 1.27 at Stoneville. The results of this study demonstrate the need for site-specific investigations when conducting certain types of aquaculture research.  相似文献

Application of readily-oxidizable organic substrate to laboratory soil-water systems and fish ponds caused anaerobic conditions in bottom soil and water, and concentrations of soluble reactive phosphorus (SRP) increased. Aeration of ponds increased total phosphorus (TP) concentrations by suspending soil particles in the water, but SRP concentrations declined because of increased oxy- genation of bottom water and soil, Alum [Al2(SO4)3·14H2O] treatment of ponds reduced SRP and TP concentrations in ponds, but the low concentration of alum used, 20 mg/L, had little residual effect on phosphorus concentration. Application of agricultural limestone at 0.2 kg/m2 to ponds with soil pH of 5.5 and Ca2+ concentration of 5 mg/L did not affect SRP and TP concentration. Unless pond soils were anaerobic at their surfaces, a condition not acceptable in thermally-unstratified fish ponds, soils released little phosphorus to the water. Strong adsorption of phosphorus by soils in intensive ponds with feeding is beneficial, because removal of phosphorus by aerobic soils is a control on excessive phytoplankton growth. In fertilized ponds, phosphorus must be applied at frequent intervals to replace phosphorus removed from the water by soils.  相似文献

Balanced plant nutrition is essential to achieve high yields of canola (Brassica napus L.) and get the best economic return from applied fertilizers. A field study was conducted at nine site‐years across eastern Canada to investigate the effects of nitrogen (N), sulfur (S) and boron (B) fertilization on canola nutrient uptake, nutrient balance, and their relationship to canola yields. The factorial experiment consisted of four N rates of 0 (N0), 50 (N50), 100 (N100), and 150 (N150) kg ha?1, two S rates of 0 (S0) and 20 (S20) kg ha?1, and three B treatments of 0 (B0), 2 kg ha?1 at preplant (B2.0P), and 0.5 kg B ha?1 foliar‐applied at early flowering stage (B0.5F). Each site‐year used the same experimental design and assigned treatments in a randomized complete block design with four replications. Fertilizer S application greatly improved seed yields at six out of nine site‐years, and the highest N use efficiency was in the N150+S20 treatment. Sulfur application generally increased seed S concentration, seed S removal, and plant total S uptake, while B fertilization mainly elevated straw B concentration and content, with minimal effect on seed yields. At the early flowering stage, plant tissue S ranged from 2.2 to 6.6 mg S g?1, but the N : S ratio was over or close to the critical value of 12 in the N150+S0 combination at five site‐years. On average across nine site‐years, canola reached a plateau yield of 3580 kg ha?1 when plants contained 197 kg N ha?1, 33 kg S ha?1 and 200 g B ha?1, with a seed B content of 60 g B ha?1. The critical N, S, and B values identified in this work and their potential for a posteriori nutrient diagnosis of canola should be useful to validate fertilizer requirements for canola production in eastern Canada.  相似文献

Total alkalinity and total hardness are familiar variables in aquatic animal production. Aquaculturists—both scientists and practitioners alike—have some understanding of the two variables and of methods for adjusting their concentrations. The chemistry and the biological effects of alkalinity and hardness, however, are more complex than generally realized or depicted in the aquaculture literature. Moreover, the discussions of alkalinity and hardness—alkalinity in particular—found in water chemistry texts are presented in a rigorous manner and without explanation of how the two variables relate to aquaculture. This review provides a thorough but less rigorous discussion of alkalinity and hardness specifically oriented toward aquaculture. Alkalinity and hardness are defined, their sources identified, and analytical methods explained. This is followed by a discussion of the roles of the two variables in aquaculture, including their relationships with carbon dioxide, pH, atmospheric pollution, ammonia, and other inorganic nitrogen compounds, phytoplankton communities, trace metals, animal physiology, and clay turbidity. Liming and other practices to manage alkalinity and hardness are explained. Changes in alkalinity and hardness concentrations that occur over time in aquaculture systems are discussed. Emphasis is placed on interactions among alkalinity, hardness, water quality, and aquacultural production.  相似文献
