采用蜡磨菌与VA 菌根真菌分别对蓝桉和尾叶桉进行单接种或混合接种,研究结果表明,两种菌根类型的真菌均能在桉树苗木根系上成功地定殖,分别合成了外生菌根、VA 菌根和混合菌根,证实了桉树不仅是菌根营养型树种,而且能形成多种菌根类型。VA 菌根感染率在接种初期较高,但随时间的推移有降低的趋势;而外生菌根菌初期合成菌根的速度较慢,但单位长度根段内菌根根尖数目有明显增加的趋势。与单接种VA菌根真菌苗木相比,外生菌根真菌抑制了VA 菌根菌在根系上的进一步侵染,体现在较低的VA 菌根感染率;而外生菌根菌显示出了较强的竞争能力,并能在接种后9 周时开始形成子实体。在混合菌根中,外生菌根有逐步替代VA 菌根的发展趋势,两种类型的菌根真菌间存在一定的竞争性作用关系。基质磷(P)素水平对菌根的形成也有一定的影响  相似文献

Eucalypts are very popular for revegetation in many parts of south China because of their capacity to tolerate degraded sites and unfertile soils,and their fast growth potential to coppice. This paper reviews a decade of field trials in china, undertaken as part of several bilateral researchprograms in plantation forestry, concerning the use of fertilizers, harvest residue management and inoculation with ectomycorrhizal fungi. The major vernalisation genes of VRN1 are well understood at the molecular level. However, their quantitative contributions to flowering time and grain yield related traits are not clear. In this study, we used a double haploid population (225 lines) of Westonia × Kauz in which the Vrn-A1a (Westonia), Vrn-B1a (Westonia) and Vrn-D1a (Kauz) were segregating, and a high resolution genetic map of 1,159 loci, to determine the quantitative contributions of Vrn-A1a, Vrn-B1a and Vrn-D1a for the days to anthesis and grain yield related traits in diverse environments. The major quantitative trait loci (QTL) of spikelet number per spike and days to anthesis were contributed by the winter alleles of VRN1. The QTL of the time of grain filling were contributed by the spring alleles of VRN1. The wild genotype (vrn-A1vrn-B1vrn-D1) showed the latest flowering, the highest spikelet number per spike, lowest peduncle proportion and thousand grain weight in three environmental analyses, and the largest spikelet number per spike, which resulted in high kernel number per spike (KN) and grain weight (GW) in well-watered environments. One QTL of KN was located on 5B, contributed by winter allele of vrn-B1 in three environmental analyses, and one GW QTL was detected on 5A, contributed by the spring allele of Vrn-A1a in a drought environment. The results indicated that the genotype Vrn-A1avrn-B1Vrn-D1a would shorten the time to anthesis and give high GW and KN in drought environments. The early anthesis associated phenotype, peduncle proportion would provide an indicator in breeding programs.  相似文献

Bird species richness in 22 reserves in the Western Australian wheatbelt was shown to be related not to isolation from adjacent uncleared land, either spatially or with time since clearing of land in their vicinity, but to area of reserve and certain reserve habitat variables. The nature of these relationships was examined with multiple regression analysis. Reserve area was the most important variable, except in some passerine groups, where numbers of plant species, vegetation associations, and plant life form and density classes in each vegetation stratum were, separately, more important than area. Eighty-two percent of the variation in the number of bird species was explained by area of reserve and number of plant species, indicating the importance of floristics to the total bird assemblage within reserves. The number of formations present (the broadest vegetation structural grouping used) did not explain any of the variation in species numbers in any of the bird groupings.There were more resident passerine species present in wheatbelt reserves than there were breeding passerine species on similar-sized southwestern Australian late-Pleistocene/Holocene islands, although the species versus area relationship of the resident habitat-specific passerines (P5 group) was very similar to that of the island land-bird faunas. The P5 species group contained many species which probably responded to wheatbelt reserves as islands; some of these species may fail to persist in the wheatbelt in the long term. Twenty-three species usually resident in reserves and 15 ‘non-residents’ were identified as being ‘vulnerable’ or of uncertain conservation status in the wheatbelt.Some vegetation formations within reserves were more important to birds than others; woodlands were most important both to resident and transient species. Most species do not appear to distinguish between shrublands and heaths as major habitats; Species richness versus formation area relationship suggest that insular patchiness for birds in the wheatbelt was manifest at the level of vegetation formations. Reserves as small as 80 ha were important sanctuaries for birds in the wheatbelt, although 1500 ha was considered a minimum area of reserve to conserve a local avifauna. Reserves of the order of 30,000–94,000 ha were required to contain most of the avifauna of the wheatbelt.  相似文献

N-linked protein glycosylation is the most abundant posttranslation modification of secretory proteins in eukaryotes. A wide range of functions are attributed to glycan structures covalently linked to asparagine residues within the asparagine-X-serine/threonine consensus sequence (Asn-Xaa-Ser/Thr). We found an N-linked glycosylation system in the bacterium Campylobacter jejuni and demonstrate that a functional N-linked glycosylation pathway could be transferred into Escherichia coli. Although the bacterial N-glycan differs structurally from its eukaryotic counterparts, the cloning of a universal N-linked glycosylation cassette in E. coli opens up the possibility of engineering permutations of recombinant glycan structures for research and industrial applications.  相似文献

Blackleg and soft rot diseases, caused by the bacteriumErwinia carotovora, are among the diseases that cause important losses in culture and storage of potato. In this paper, we introduced bacterial resistance into potato, via genes encoding for proteins with antibacterial activity. For this purpose, potato clones were transformed either with the gene encoding the acidic attacin protein fromHyalophora cecropia, or with the gene encoding the cecropin analog peptide SB37. These clones were evaluated for soft rot and blackleg resistance, after inoculation with the bacterial strainErwinia carotovora subsp.atroseptica T7. Results reported in this paper indicate that a considerable percentage of the potato clones (15–22%) showed increased resistance to bacterial infection, revealed by reduced severity of blackleg or soft rot symptoms. Expression of the transgenes was demonstrated in some of the clones by Northern blot analysis. This is the first report indicating that expression of the gene encoding for an attacin protein and for the cecropin SB-37 peptide in transgenic potato confers increased resistance to bacterial infection.  相似文献

Flow cytometry is a useful tool that provides an accurate, objective and rapid evaluation of semen quality. The use of this technique could significantly improve the quality of buffalo semen samples used in artificial insemination. This study was carried out to evaluate, by flow cytometry, frozen–thawed buffalo spermatozoa quality parameters such as sperm viability by SYBR‐14/propidium iodide staining; mitochondrial function by JC‐1 potentiometric probe; sperm chromatin stability (SCSA) by acridine orange; and acrosome reaction (AR) by FITC‐PNA staining. Semen samples from five Italian Mediterranean buffalo bulls were used. Sperm viability was not different between bulls and ranged from 33.4% to 43.6%. A consistent rate (55.1 ± 10.8%) of sperm cells showed high mitochondrial membrane potential (Δψhigh), with no significant differences between subjects. Sperm chromatin structure assay differed significantly between the five buffalo bulls; moreover, data showed high stability within each buffalo. DNA fragmentation indexes (DFI), such as %‐DFI, ‐DFI, SD‐DFI, were 11.2 ± 8.6, 153.3 ± 24.6 and 81.6 ± 21.2, respectively. Regarding AR, the percentage of acrosome‐reacted live (ARL) and acrosome‐reacted dead (ARD) spermatozoa was 0.3 ± 0.2 and 15.3 ± 5.5, respectively. This functional parameter differed significantly between buffalo bulls and showed high stability. Following to Ca2+ ionophore A23187 for 3 h, AR significantly differed between subjects and was characterized by an increase in both ARL (10.8%) and ARD population (22.0%). This study indicates that flow cytometry could be a useful tool for a quick multiparametric evaluation of sperm quality in buffalo. In particular, SCSA and AR resulted in sperm functional parameters sensitive enough for the diagnosis of frozen‐thawed semen fertilizing potential.  相似文献
