Changes in the contents and composition of polyphenolics and resulting antioxidant activities of S. verbenaca by-products were investigated at three phenological stages (flowering, early fruiting and late fruiting stages). The highest accumulation of total phenolics was detected at the flowering stage (58.36 mg GAE/g DW). HPLC analysis of methanolic extracts showed the prevalence of methyl carnosate (821.45–919.82 μg/g DW) and rosmarinic acid (544.51–649.26 μg/g DW). Phenolic diterpenes (1056.90–1148.42 μg/g DW) was the most represented class of compounds. Three complementary tests namely, DPPH? (IC50 value, 49.22 μg/mL) and ABTS?+-radical scavenging assays (146.86 μM TE/mg) and FRAP reducing power test (188.93 mM Fe(II)/mg) were used to evaluate the antioxidant capacity and showed the best performance at the early fruiting period. The current study evidenced the significant effect of phenophase on antioxidants and contributed to valorize S. verbenaca extracts as a source of functional phenolic compounds.

Among extracts of 33 plant species screened against Mycosphaerella eumusae, the causal agent of Eumusae leaf spot disease of banana, water extract of Cassia senna, Zimmu (Interspecific hybrid between Allium cepa × Allium sativum) and Rhincanthus nasutus provided 100% inhibition of spore germination and 1.7–2.0 cm zone of inhibition of mycelial growth under in vitro conditions. The maximum efficacy of mycelial inhibition was observed with Zimmu leaf extract. When Zimmu leaf extract was tested at different concentrations (5, 10, 25, 50 and 100% w/v), all tested concentrations provided complete inhibition of mycelial growth of the pathogen. The field evaluation of Zimmu leaf extract at different concentrations in cv. Grand Naine showed that the application of the water extract of Zimmu leaf at 50% concentration (w/v) provided 55% reduction of disease severity compared to the unsprayed control. Besides, the application of Zimmu leaf extract increased the value of youngest leaf spotted-0 (up to 60.5%) as well as increased the yield of banana (up to 46.8%) as compared to control. The effect of Zimmu in increasing the value of YLS-0 and the bunch yield was comparable with the chemical fungicide Propiconazole 25% EC (0.1%). Thin layer chromatography (TLC) analysis showed that among different major compounds, two lipid compounds (LP-B1 and LP-B2) extracted using methanol had the ability to inhibit M. eumusae growth (0.7–1.5 cm zone of inhibition). The gas chromatography–mass spectrometry (GC–MS) analysis of lipid bands revealed the presence of six different lipid compounds, which may be responsible for the growth inhibition of the pathogen. Since the application of water extract of Zimmu was found to be not only effective in controlling the leaf spot disease severity but also increased the number of green leaves and yield of banana fruits, the Zimmu extract can be used effectively in integrated disease management of Eumusae leaf spot disease for enhancing banana production in an ecologically sustainable manner.  相似文献

The aim of this investigation was to study the antioxidant and anticancer activities of young sprouts of some traditional Korean salad plants. Total phenolics, antioxidant and anticancer activities of the methanol extracts from young sprouts of 11 salad plants were determined. The highest amount of phenolics was found in methanol extracts of Euonymus alatus (235.7 mg kg−1), followed by Hypericum ascyron (197.1 mg kg−1), Zanthoxylum piperitum (194.1 mg kg−1) and Zanthoxylum schinifolium (142.5 mg kg−1). Methanol extracts of E. alatus, H. ascyron, and Z. piperitum at 63 mg kg−1 exhibited the highest dose-depend DPPH radical scavenging activity by 91.2, 91.2 and 83.9%, respectively. According to the MTT results, the methanol extracts from Stellaria aquatica, Eleutherococcus sessilifolrus and Z. schinifolium showed the highest anticancer activities against Calu-6 (IC50 < 25.0 μgml−1) and from S. aquatica—the highest anticancer activities against SNU-601 (153.3 μgml−1), following by E. sessilifolrus (196.7 μg ml−1) and Amaranthus mangostanus (303.1 μgml−1). Total phenolics were highly correlated with the DPPH, suggesting that they contribute to the antioxidant properties of the studied plants. In conclusion: young sprouts of Korean salad possess antioxidant and anticancer properties and could be used as a supplement to proper drugs.  相似文献

The leaves of Rosmarinus officinalis harvested from three different locations of Turkey were extracted by both methanolic and supercritical CO2 extraction. Subsequently, six extracts and the active compounds, carnosic acid, and rosmarinic acid were applied to various human cancer cell lines including NCI-H82 (human, small cell lung, carcinoma), DU-145 (human, prostate, carcinoma), Hep-3B (human, black, liver, carcinoma, hepatocellular), K-562 (human chronic myeloid leukemia), MCF-7 (human, breast, adenocarcinoma), PC-3 (human, prostate, adenocarcinoma) and MDA-MB-231 (human, breast, adenocarcinoma) by MTT assay. Supercritical CO2 extracts had superior antiproliferative effect compared to the soxhlet extracts. Although the extracts exhibited various cytotoxic effects against different cell lines, comparatively low IC50 values ranging between 12.50 and 47.55 μg/ml were attained against K-562, being the most sensitive cell line. Moreover, carnosic acid caused the lowest cell viability with values ranging from 13 to 30 % at a concentration of 19 μM after 48 h of treatments, resulting in superior antiproliferative effect. Rosemary extract is a potential candidate to be included in the anti-cancer diet with pre-determined doses avoiding toxicity.  相似文献

The objectives of this study were to examine the free radical scavenging activity and the protective effects against macromolecular oxidation as well as the cytotoxic activity of Aphanes arvensis aqueous and methanolic extracts. Free radical scavenging activity was determined by DPPH method. The methanolic extract showed a scavenging activity nearly equivalent to Trolox and Vitamin C and has an IC50 value of 4.54 μg/mL. Total antioxidant capacity was determined by CUPRAC method. The antioxidant capacity of aqueous and methanolic extract was 0.792 and 1.550 mmol TE/g DWE, respectively. The protective effect of A. arvensis extracts against lipid peroxidation was evaluated using a liposome oxidation system. The methanolic extract was more active than the aqueous extract. The aqueous extract possessed protective effect against protein oxidation in a dose dependent manner. Both extracts showed inhibitory effect on DNA oxidation as measured by plasmid relaxation assay. Results presented here indicate that A. arvensis possess strong antioxidant activity and protective effects with very little cytotoxic effect, and they can therefore be used as a natural additive in food, cosmetic and pharmaceutical industries.  相似文献

Antioxidant activity of fresh Allium sativum L. (garlic) is well known and is mainly due to unstable and irritating organosulphur compounds. Fresh garlic extracted over a prolonged period (up to 20 months) produces odourless aged garlic extract (AGE) containing stable and water soluble organosulphur compounds that prevent oxidative damage by scavenging free radicals. The aim of this study was to investigate the in vitro antioxidant activity of aged (up to 20 months) 15% hydroethanolic extracts of different parts (bulbs, bulblets, flower bulblets, flowers, and leaves) of three Allium spontaneous species which are endemic for Italian flora: Allium neapolitanum Cyr., Allium subhirsutum L., Allium roseum L. and to compare it with the in vitro antioxidant activity of aged 15% hydroethanolic extracts of bulbs and leaves of garlic. The antioxidant potential of aged extracts of all species has been evaluated using two different spectrophotometric assays: 2,2-diphenylpicrylhydrazyl (DPPH) test and the ferric reducing/antioxidant power (FRAP) assay. Furthermore the polyphenol content was determined. The aged extracts obtained from the leaves showed the best antioxidant activity, followed by flowers and then by bulbs in both used tests, while flower bulblets and bulblets exhibited lower results or no activity. The polyphenol content was generally directly correlated with antioxidant/antiradical activity. This study confirms the data obtained in previous researches, the wild-type species of Allium and in particular organs other than bulbs are more active and efficacious than garlic bulb. Surely leaves of these Allium spp. deserve special attention.  相似文献

Onion (Allium cepa L.) is an important vegetable crop in Korea, but its production is severely affected by fungal pathogens during plant growth and bulb storage. We investigated the occurrence of fungi on market onion bulbs; identified the predominant fungal species based on the internal transcribed spacer region, β-tubulin region, and elongation factor 1-α gene sequences; and tested the pathogenicity of each predominant fungal species in onion bulbs. The genera Aspergillus (63.9%), Penicillium (15.5%), Fusarium (6.4%), Rhizopus (5.2%), and others (9.0%) were detected in the samples. Among these genera, Aspergillus awamori, Fusarium oxysporum, Penicillium brasilianum, and Rhizopus oryzae were identified as the predominant species. All of the fungi tested could infect both the inner layers and outer surfaces of onion bulbs and be re-isolated from the infected tissues. To our knowledge, this is the first report that P. brasilianum is a fungal pathogen of onion bulbs.  相似文献

Betalains are water-soluble plant pigments that are widely used as food colorants, and have a wide range of desirable biological activities, including antioxidant, anti-inflammatory, hepatoprotective, anti-cancer properties. They can be produced from various plants, notably beetroot, but betalain products obtained in this way also have some undesirable properties and are difficult to standardize. A potentially attractive alternative is to use hairy root cultures. In the study reported here, we found that betalain extracts obtained from hairy root cultures of the red beetroot B. vulgaris cv. Detroit Dark Red also had higher antioxidant activity than extracts obtained from mature beetroots: six-fold higher 2,2-dyphenyl-1-picrylhydrazyl radical scavenging ability (90.7% inhibition, EC50 = 0.11 mg, vs 14.2% inhibition, EC50 = 0.70 mg) and 3.28-fold higher oxygen radical absorbance capacity (4,100 μM TE/g dry extract, vs 1,250 μM TE/g dry extract). The high antioxidant activity of the hairy root extracts was associated with increased concentrations (more than 20-fold) of total phenolic concomitant compounds, which may have synergistic effects with betalains. The presence of 4-hydroxybenzoic acid, caffeic acid, catechin hydrate, and epicatechin were detected in both types of extract, but at different concentrations. Rutin was only present at high concentration (1.096 mg.g−1 dry extract) in betalain extracts from the hairy root cultures, whereas chlorogenic acid was only detected at measurable concentrations in extracts from intact plants.  相似文献

The effect of the drying temperature on the volatile components and sensory acceptance of the Roselle (Hibiscus sabdariffa) extract in powder was investigated. The Roselle extraction was carried out by maceration with 7 L of 30% ethanol (v/v), 560 g of fresh Roselle calyces for 168 h. The Roselle extracts were spray dried at different temperatures 150, 160, 170, 180, 190, 200 and 210 °C, giving different outlet values about yield and final moisture. The volatile compounds in Roselle extract and dried samples were performed using needles of solid phase microextraction (SPME) and gas chromatography/mass spectrometry (GC/MS HP-5890). Twenty volatile compounds were identified in the extracts among them terpenoids, esters, hydrocarbons and aldehydes. Fourteen volatile compounds were identified in the powder sample, but only ten were present in the Roselle extract. This indicates that some compounds were lost and some others were generated due to a degradation process. An acceptability sensory analysis showed that the best powder sample was the Roselle extract dehydrated using temperature between 190 °C and 200 °C (p < 0.05). There was not statistically significant difference in the pH of Roselle extracts ranging from 3.4 to 3.9. It was concluded that the spray drying temperature of the Roselle extracts has an effect on the volatile compounds losses.  相似文献

Background:To study the anticancer activity of Plantago major, we assessed the effect of ethanolic, methanolic and acetonic extracts of this plant on HCT-116, SW-480, and HEK-293 cell lines as control. Methods:The cytotoxic activity, biocompatibility, and toxicity were evaluated by MTT assay, hemolysis, and Artemia salina-LD50 (on mice) tests, respectively. The analysis of the extracts was performed by GC-MS analysis. Results:The results showed that all the extracts had the most antiproliferative properties on the HCT-116 cell line. The P. major root extract was more effective than the aerial parts, and IC50 values for ethanolic, methanolic and acetonic root extracts were 405.59, 470.16, and 82.26 µg/mL, respectively on HCT-116 cell line at 72 h. Hemolysis degree of the ethanolic extract of aerial and root parts were approximately 1% at 400 μg/mL.. Using the ethanolic extracts, the Artemia survived every concentration, and no toxicity was observed. One week after the oral administration of different parts of P. major extracts, none of the mice died, even those were administered 2000 mg/kg. The results of GC/MS analysis showed that P. major extracts contain potential anticancer compounds, such as stearic acid (8.61%) in aerial parts of methanolic extract and 1,2- Benzenedicarboxylic acid, mono(2-ethylhexyl)ester (88.07% and 40.63%) in aerial and root parts of acetonic extract of P. major. Conclusions:Our findings suggest that the P. major is a source of potential compounds with antiproliferative properties. To contribute to the development of new methods to reduce the contamination of coffee beans with Aspergillus ochraceus and ochratoxin A, a toxin produced by this fungus, the present work initially aimed to select plant extracts active against A. ochraceus. Among the extracts from 43 plant species, the most active was the one obtained from leaves of Heteropterys byrsonimifolia. This extract was subjected to a fractionation process that resulted in the purification of four flavonoids, among which the most active against the fungus was rutin, which presented a minimal inhibitory concentration (MIC, 32.5 μg/mL) close to the value obtained for the commercial fungicide benzalkonium chloride (7.84 μg/mL). In vitro studies with the isolated flavonoids and other four from commercial sources showed that the MIC for the flavonoid chrysin is equal to that observed for rutin, and results from an in silico study suggested that these flavonoids act against A. ochraceus by binding to a protein kinase produced by this fungus. Consequently, this enzyme and the studied flavonoids are potentially useful for the development of new products to control A. ochraceus in coffee beans.  相似文献

Yacon (Smallanthus sonchifolius (Poepp.) H. Robinson) leaves is traditionally consumed as herbal tea in many countries including Indonesia. This plant’s antidiabetic properties have been extensively researched, but studies on the responsible active compound identification are scarce. Information on the active compounds is critical for the consistency of Yacon herbal tea quality. The aim of this study was to identify α-glucosidase inhibitors in Indonesian Yacon leaves grown in two different locations using FTIR- and LC–MS/MS-based metabolomics in combination with in silico technique. Yacon leaves ethanol (50 and 95%) and water extracts were tested for α-glucosidase inhibitory activity, with the 95% ethanol extract being the most active. Geographical origins were found to have no major impact on the activity. In parallel, chemical profile of Yacon leaves extract was determined using FTIR and LC–MS/MS. Orthogonal Projection to Latent Structure (OPLS) was used to analyze both sets of data. OPLS analysis of FTIR data showed that compounds associated to α-glucosidase inhibitor activity included those with functional groups –OH, stretched CH, carbonyl, and alkene. It was consistent with the result of OPLS analysis of LC–MS/MS data, which revealed that based on their VIP and Y-related coefficient value, nystose, 1-kestose, luteolin-3′-7-di-O-glucoside, and 1,3-O-dicaffeoilquinic acid isomers, strongly linked to Yacon’s α-glucosidase inhibitor activity. In silico study supported these findings, revealing that the four compounds were potent α-glucosidase inhibitors with docking score in the range of ? 100.216 to ? 115.657 kcal/mol, which are similar to acarbose (? 115.774 kcal/mol) as a reference drug.

Aerial parts (leaves and stem) of Raphanus sativus, which are usually discarded were found to possess potent antioxidant and radical scavenging activity, as measured by standard antioxidant assays. Methanolic and acetone extracts of R. sativus leaves had total polyphenolic content of 86.16 and 78.77 mg/g dry extract, which were comparable to the traditional rich sources such as green tea and black tea. HPLC identification of polyphenolics indicated the presence of catechin, protocatechuic acid, syringic acid, vanillic acid, ferulic acid, sinapic acid, o-coumaric acid, myricetin, and quercetin in leaves and stem. Among the different extraction solvents, methanolic extract of leaves and stem showed potent reductive capacity, significantly inhibited linoleic acid peroxidation and displayed metal chelating activity. Further, they scavenged free radicals effectively with IC50 (half maximal inhibitory concentration) of 31 and 42 μg/ml for DPPH radical, 23 and 52 μg/ml for superoxide radical, 67 and 197 μg/ml for hydrogen peroxide, and 56 and 62 μg/ml for nitric oxide, respectively. Leaves showed most potent antioxidant and radical scavenging activity as compared to stem, which may be accounted for the high polyphenolic content. Leaves and stem of R. sativus, often under-utilized part of this vegetable, thus possessed considerable amount of polyphenolics. Hence, it should be regarded as a potential source of natural antioxidants and could be effectively employed as an ingredient in health or in functional food.  相似文献

This study describes the antioxidant activities and antigenotoxic effects of garlic extracts prepared by different processing methods. Aged-garlic extract (AGE) showed a significantly higher total phenolic content (562.6 ± 1.92 mg/100 g garlic acid equivalents) than those of raw garlic extract (RGE) or heated garlic extract (HGE). The SC50 for DPPH RSA in HGE was significantly the highest at 2.1 mg/ml. The SC50 for SOD-like activity in garlic extracts was, in decreasing order, RGE (7.3 mg/ml) > AGE (8.5 mg/ml) > HGE (9.2 mg /ml). The ED50 of AGE was the highest (19.3 μg/ml) regarding H2O2 induced DNA damage and its inhibition rate was 70.8%. The ED50 of RGE for 4-hydroxynonenal (a lipid peroxidation product) induced DNA damage was 38.6 μg/ml, followed by AGE > HGE. Although the heat treatment of garlic tended to decrease the TPC and SOD-like activity and increased DPPH RSA, garlic, in general, has significant antioxidant activity and protective effects against oxidative DNA damage regardless of processing method.  相似文献

Single isolates of four Penicillium species belonging to series Corymbifera (Penicillium allii, Penicillium hirsutum, Penicillium tulipae, Penicillium venetum) plus an isolate of Penicillium polonicum, all from North American sources, were inoculated individually into Crocus sativus (crocus), Allium sativum (garlic), Allium cepa (onion), Allium stipitatum (ornamental onion), Iris hollandica (iris), Ornithogalum umbellatum (grass lily), and varieties of Gladiolus (gladiolus), Tulipa (tulip) and Narcissus (daffodil). All isolates were pathogenic to garlic and onion (but none to ornamental onion), and all but P. polonicum to Tulipa, although virulence varied considerably. Additionally, P. hirsutum was pathogenic to Crocus, Gladiolus and Iris; P. tulipae to Narcissus, Crocus, Ornithogalum and Iris; P. venetum to Gladiolus, Ornithogalum and Iris; and P. polonicum to Iris. Nomenclature and taxonomy of series Corymbifera are summarized, the above results compared to similar investigations in Europe, and management implications provided.  相似文献

The object of the study was to determine the chemical composition of Diospyros lotus L. extract and their antioxidant and antiproliferative properties. Eight compounds were isolated from D. lotus and identified as gallic acid, methylgallate, ellagic acid, kaempferol, quercetin, myricetin, myricetin 3-O-β-glucuronide, and myricetin-3-O-α-rhamnoside. D. lotus extract tested in different in vitro systems (DPPH, ABTS, FRAP, and Fe2+ chelating activity assay) showed significant antioxidant activity. The potential antiproliferative properties of D. lotus extract and isolated compounds against nine human cancer cell lines such as COR-L23, CaCo-2, C32, ACHN, A375, A549, Huh-7D12, MCF-7, and LNCaP were investigated in vitro by SRB assay. D. lotus extract demonstrated the highest inhibitory activity against COR-L23 with an IC50 value of 12.2 μg/ml. Among identified hydrolysable tannins, ellagic acid evidenced strong antiproliferative activity against both C32 and A375 cells with IC50 values of 0.8 and 4.1 μg/ml, respectively. Interesting results were observed, also, with gallic acid that showed the highest cytotoxic activity against CaCo-2 (IC50 2.6 μg/ml). Overall, the results of this study suggest that D. lotus displays a good antioxidant activity and has antiproliferative effects. Both activities are related to identified phenolic compounds.  相似文献

Arbutus unedo, Prunus spinosa, Rosa micrantha and Rosa canina are good sources of phenolic compounds, including anthocyanins. These compounds have potent antioxidant properties, which have been related to anticancer activity. Herein, the in vitro antioxidant and antitumor properties of enriched phenolic extracts (non-anthocyanin phenolic compounds enriched extract- PE and anthocyanins enriched extract- AE) of the mentioned wild fruits were evaluated and compared. PE gave higher bioactive properties than the corresponding AE. It was observed a high capacity of A. unedo phenolic extract to inhibit lipid peroxidation in animal brain homogenates (EC50?=?7.21 μg/mL), as also a high antitumor potential against NCI-H460 human cell line (non-small lung cancer; GI50?=?37.68 μg/mL), which could be related to the presence of galloyl derivatives (exclusively found in this species). The bioactivity of the studied wild fruits proved to be more related to the phenolic compounds profile than to the amounts present in each extract, and could be considered in the design of new formulations of dietary supplements or functional foods.  相似文献

The effects of extracts of different parts of the perennial tropical plant Balanites aegyptiaca (L) Del., including various solvent extracts of roots, methanol extracts from leaves, fruits, flowers and roots, partially purified saponins obtained from its roots and a standard saponin were studied on the life cycle (adult longevity, number of eggs, crawlers, adults, weight of adults and % wax content) of a laboratory-reared parthenogenic line of the mealy bug, Maconellicoccus hirsutus (Homoptera: Pseudococcidae). Extracts derived from various parts of B. aegyptiaca (leaves, fruits, flowers, and roots in methanol) affected the life cycle of M. hirsutus with a methanol root extract being the most effective at a concentration of 500 μg ml−1. Partially purified saponin of B. aegyptiaca and the commercial bark saponin extract (Sigma) from Quillaja saponaria at a concentration of 500 μg ml−1 were effective in reducing the longevity of M. hirsutus. Significant reductions in oviposition by M. hirsutus were found for all the extracts at a concentration of 500 μg ml−1. Extracts also affected the number of emerging crawlers, number of adults as well as the weight and wax content of emerging adults. These studies suggest that B. aegyptiaca plant extracts and saponins can be useful botanical insecticides for the protection of crops from mealy bugs.  相似文献
