Contagious bovine pleuropneumonia (CBPP), caused by Mycoplasma mycoides subsp. mycoides (Mmm) is a severe cattle disease, present in many countries in sub-Saharan Africa. The development of improved diagnostic tests and vaccines for CBPP control remains a research priority. Polyacrylamide gel electrophoresis and mass spectrometry were used to characterize the Triton X-114 soluble proteome of nine Mmm strains isolated from Europe or Africa. Of a total of 250 proteins detected, 67 were present in all strains investigated. Of these, 44 were predicted to be lipoproteins or cytoplasmic membrane-associated proteins and are thus likely to be members of the core in vitro surface membrane-associated proteome of Mmm. Moreover, the presence of all identified proteins in other ruminant Mycoplasma pathogens were investigated. Two proteins of the core proteome were identified only in other cattle pathogens of the genus Mycoplasma pointing towards a role in host–pathogen interactions. The data generated will facilitate the identification and prioritization of candidate Mycoplasma antigens for improved control measures, as it is likely that surface-exposed membrane proteins will include those that are involved in host–pathogen interactions.  相似文献

Contagious bovine pleuropneumonia (CBPP) caused by Mycoplasma mycoides subsp. mycoides small colony type (MmmSC) has been eradicated in the developed world, but it is still present in many countries of sub-Saharan Africa. After initially successful control measures in the 1960s it has been spreading due to a lack of money, fragmentation of veterinary services, uncontrolled cattle movement, insufficient vaccine efficacy and sensitivity of current diagnostic tests.In this study we used two-dimensional polyacrylamide gel electrophoresis followed by immunoblot with sera from MmmSC-infected animals and MALDI-ToF mass spectrometry to identify novel immunogenic proteins as candidate molecules for improved diagnostics and vaccines. We identified 24 immunogens recognized by pooled sera from experimentally infected cattle. Furthermore, a serum from an animal with acute clinical disease as well as severe pathomorphological lesions recognized 13 additional immunogens indicating variation in the antibody responses to CBPP amongst cattle. Most immunogens showed compelling similarity to protein/gene sequences in the two ruminant pathogens M. capricolum subsp. capricolum and M. mycoides subsp. mycoides large colony type both belonging to the mycoides cluster. Three of these proteins, namely glycerol-3-phosphate oxidase, adenylosuccinate synthase, and glyceraldehyde-3-phosphate dehydrogenase, had no compelling homologue in the other distantly related bovine pathogen M. agalactiae. In addition, translation elongation factor Tu, heat shock protein 70, pyruvate dehydrogenase, and FKBP-type peptidyl-prolyl isomerase, which have been found to mediate adhesion to host tissue in other mycoplasmas were shown to be expressed and recognized by sera. These proteins have potential for the development of improved diagnostic tests and possibly vaccines.  相似文献

Contagious bovine pleuropneumonia (CBPP) is a disease of economic importance that is widely distributed in sub-Saharan African and contributes significantly to cattle morbidity and mortality. Control of CBPP offers a number of challenges as a result many developing countries in Africa are still struggling with this disease. In this study, we look at the challenges encountered in CBPP control in sub-Saharan Africa from the Zambian perspective. In conducting this study, we reviewed scientific literature and reports from the Ministry of Agriculture and Livestock and related animal institutions, and also made interviews with experts and key government officials involved in CBPP control in Zambia. Among the challenges identified for the successful control of CBPP were as follows: failure in the delivery of veterinary services, lack of a cattle identification system, natural phenomenon, livestock husbandry systems in the traditional sector, human movements, traditional practices among cattle farmers and cattle marketing systems. It was seen that the epidemiology of CBPP in Zambia is influenced by both ecological and anthological factors. Therefore, design and implementation of any control or eradication programme should be area/regional-dependent taking into account the different factors influencing disease transmission and maintenance.  相似文献

A cross sectional study was undertaken from October 2010 to March 2011 to determine the seroprevalence of contagious bovine pleuropneumonia (CBPP) and its related risk factors in export quarantine centers. A total of 3,111 cattle sera were collected from different export quarantine farms located in and around Adama, namely, Bekero, Jogo, Kedir, and Dera farms, and tested for the presence of Mycoplasma mycoides subsp. mycoides small colonies antibody using competitive enzyme-linked immunosorbant assay. Of the total 3,111 cattle sera examined, 124 (4 %) were found positive for CBPP. Among the potential predisposing factors assessed, origin, transportation condition, confinement level, and stay time of the animals in quarantine center were not found significantly (P?>?0.05) associated with the occurrence of the disease. Whereas age was found significantly (P?<?0.05) associated with the occurrence of the disease in which a high seroprevalence was recorded in aged (9.5 %) animals than young (3 %). Generally, this study showed that CBPP is a threat for Ethiopian livestock export market and a well established disease in Borana and Bale areas, where the animals originated.  相似文献

L-α-glycerophosphate oxidase (GlpO) plays a central role in virulence of Mycoplasma mycoides subsp. mycoides SC, a severe bacterial pathogen causing contagious bovine pleuropneumonia (CBPP). It is involved in production and translocation of toxic H2O2 into the host cell, causing inflammation and cell death. The binding site on GlpO for the cofactor flavin adenine dinucleotide (FAD) has been identified as Gly 12−Gly13Gly 14−Ile15−Ile16Gly 17. Recombinant GlpO lacking these six amino acids (GlpOΔFAD) was unable to bind FAD and was also devoid of glycerophosphate oxidase activity, in contrast to non-modified recombinant GlpO that binds FAD and is enzymatically active. Polyclonal monospecific antibodies directed against GlpOΔFAD, similarly to anti-GlpO antibodies, neutralised H2O2 production of M. mycoides subsp. mycoides SC grown in the presence of glycerol, as well as cytotoxicity towards embryonic calf nasal epithelial (ECaNEp) cells. The FAD-binding site of GlpO is therefore suggested as a valuable target site for the future construction of deletion mutants to yield attenuated live vaccines of M. mycoides subsp. mycoides SC necessary to efficiently combat CBPP.  相似文献

Contagious bovine pleuropneumonia (CBPP), caused by Mycoplasma mycoides subsp. mycoides SC (MmmSC), is one of the most significant cattle disease in Africa. The control measures, which led to eradication from numerous countries are not feasible in Africa where the only prophylaxis relies on vaccination. However, the attenuated vaccines, used up to now in Africa, are of low efficiency. The development of an improved vaccine is, therefore, a necessity. The purpose of this study was to compare some immunological parameters in MmmSC-infected cattle (endobronchial versus natural in-contact infection) and assess the response in correlation with the clinical outcome (death versus recovery). Characterization of the immune parameters elicited in recovered animals, known to be refractory to new infection, will be an important step towards development of new vaccines against CBPP. A significant outcome of this study was the demonstration that all MmmSC-infected cattle developed a MmmSC-specific cell-mediated immune response. A kinetic analysis of the MmmSC responsiveness showed that the main difference between endobronchially- and in-contact infected animals was the delay before the onset of the MmmSC-specific immune response. The first MmmSC-responding PBMC sample was selected from each animal for cell phenotyping. The phenotypic analysis of this early MmmSC-induced response revealed the predominant contribution of the CD4 T-cells in all animals whereas IFNgamma was only constantly produced in recovered animals. Evolution of this early MmmSC-specific immune response was then followed by a kinetic analysis of the MmmSC-induced CD4 T-cell response and IFNgamma released. The results demonstrated that in recovered animals, the MmmSC-specific CD4 Th1-like T-cell response was maintained until slaughtering whereas in animals with acute disease, progression of CBPP was associated with a decreased ability of the PBMC to produce IFNgamma. The results led to the identification of immune parameters, which correlate with protection against CBPP and to a relevant strategy for the development of improved vaccines against this disease.  相似文献

The genetic diversity of 60 field strains of Mycoplasma mycoides ssp. mycoides, small colony type (M. mycoides SC), comprising 56 isolates from cattle in Tanzania, one from Kenya, two from Botswana and one from Portugal, as well as the type (PG1T) and vaccine (T1‐SR49) strains, was investigated. The strains were analysed for variations in the EcoRI and Csp6I restriction sites in the genomic DNA using the amplified fragment length polymorphism (AFLP) technique, and variations in the BamHI restriction sites using pulsed‐field gel electrophoresis (PFGE). Six AFLP types were detected among the analysed strains. The AFLP profiles of the type and vaccine strains were indistinguishable from each other. Indistinguishable AFLP profiles were found for 55 Tanzanian field strains, one of them isolated in 1990 and the other 54 isolated in 1998/1999, although one strain isolated in 1999 showed a different profile. Strains from different countries revealed different AFLP profiles. Six PFGE types were detected among the analysed strains, with all the 56 Tanzanian field strains displaying indistinguishable PFGE profiles. Strains from different countries revealed different PFGE profiles, and so did the type and vaccine strains. The strong genomic homogeneity among M. mycoides SC strains associated with outbreaks of contagious bovine pleuropneumonia in different regions of Tanzania suggests that the outbreaks of the disease in the 1990–99 period might have been caused by a single epidemic clone. Moreover, this study has demonstrated that AFLP and PFGE are potential tools for molecular epidemiological studies of M. mycoides SC infections.  相似文献

Contagious bovine pleuropneumonia (CBPP), a highly infectious and fatal disease of cattle present in many countries in sub-Saharan Africa, is usually controlled by mass vaccinations. However, vaccination against CBPP is known to cause site reactions in a percentage of cattle especially in primary vaccinations. In Zambia, a record of site reactions was kept for seven consecutive years from 2005 to 2011 to establish the level of the problem. In some areas, after 3 years of consecutive vaccination campaigns, immunization could not be implemented for a period of 2 years because of logistical difficulties or owner resistance. Whereas in the three preceding years when animals were vaccinated annually, site reactions were in the range of 6.2 %; on resumption of vaccination in the herds that had not been immunized for 2 years, site reactions averaged 21.3 %. This data shows that the T1/44 vaccine may cause severe local reactions in cattle if there is any break in annual vaccinations. It is therefore important for authorities to ensure that the cattle at risk of contracting CBPP are regularly vaccinated to avoid discouraging farmers from presenting their animals.  相似文献

Contagious bovine pleuropneumonia (CBPP) is a respiratory disease of cattle; CBPP is caused by Mycoplasma mycoides subsp. mycoides small colony. CBPP is a major cause for concern for African countries (because of mortality, animal-production losses and cost of control). The clinical form of the disease is the more infectious (contagion occurs essentially through coughing). However, chronic lung lesions with viable mycoplasmas can persist in recovering cattle. Animals presenting these lesions might have a time-delimitated infectious phase. Such carriers are suspected to generate field outbreaks (although this hypothesis remains debated). We investigated the potential quantitative effects of these chronic carriers on the within-herd CBPP spread. Data were collected during a longitudinal field herd survey in a mixed crop-livestock system in the Ethiopian highlands. Two stochastic Markov-chain models' outputs (seroconversion dynamics, basic reproduction ratio R0, cumulative clinical incidence and risk of herd infection) were compared given different hypotheses on the carrier infectiousness. The late seroconversions observed in the field data were fitted correctly only for the highest carrier infectiousness we considered (mean chronic duration of 1 year and carriers 50-times less infectious than clinical cases). Although sensitivities (in terms of disease impact in the herd) were in general negligible when the carrier infectiousness was low (e.g. when carriers were assumed to be 1000-times less infectious than clinical cases), they rapidly became important when the infectiousness increased.  相似文献

ABSTRACT: Contagious bovine pleuropneumonia (CBPP), caused by Mycoplasma mycoides subsp. mycoides, is an important livestock disease in Africa. The current control measures rely on a vaccine with limited efficacy and occasional severe side effects. Knowledge of the protective arms of immunity involved in this disease will be beneficial for the development of an improved vaccine. In previous studies on cattle infected with M. mycoides subsp. mycoides, a correlation was detected between the levels of mycoplasma-specific IFN-γ-secreting CD4+ T lymphocytes and reduced clinical signs. However, no cause and effect has been established, and the role of such cells and of protective responses acquired during a primary infection is not known.We investigated the role of CD4+ T lymphocytes in CBPP by comparing disease patterns and post mortem findings between CD4+ T cell depleted and non-depleted cattle. The depletion was carried out using several injections of BoCD4 specific murine monoclonal antibody on day 6 after experimental endotracheal infection with the strain Afadé. All cattle were monitored clinically daily and sacrificed 28-30 days post-infection. Statistically significant but small differences were observed in the mortality rate between the depleted and non-depleted animals. However, no differences in clinical parameters (fever, signs of respiratory distress) and pathological lesions were observed, despite elimination of CD4+ T cells for more than a week. The slightly higher mortality in the depleted group suggests a minor role of CD4+ T cells in control of CBPP.  相似文献

The origin of the outbreaks of contagious bovine pleuropneumonia (CBPP) in Italy between 1990 and 1993 were never successfully traced mainly due to the close similarity of the strains of the causative mycoplasma, Mycoplasma mycoides subspecies mycoides small colony (MmmSC) and the limitations of the typing tools available at the time. In this report we examined a selection of strains isolated in the Veneto and Friuli Venezia Giulia regions of Italy by the highly discriminatory variable number tandem repeat (VNTR) procedure. Results were analysed for the first time by a capillary sequencer-based method. It was shown that all the MmmSC strains were genetically very similar and all belonged to the same profiles for both VNTR 4 and 5. This suggests that the outbreaks in Northern Eastern Italy, which eventually spread to other parts of the country, originated from a single source.  相似文献

Contagious bovine pleuropneumonia (CBPP) is a disease of economic importance that is widely distributed in sub-Saharan African and contributes significantly to cattle morbidity and mortality. Lack of resources to implement eradication measures has led to the disease becoming endemic in most areas in sub-Saharan Africa where governments have little resources and the majority of the people are poor. Usually, control and eradication of such diseases as CBPP is treated as a public good by governments and to achieve this, governments are usually assisted by nongovernment organisations, bilateral government programmes and international donors. The private sector, which usually is companies that run businesses to make profit, although not very well established in sub-Saharan Africa could play a big role in the eradication of CBPP in the region. This could play a dual role of promoting investment and also eradicate livestock diseases which have proved a menace in the livestock sector. This paper highlights the role played by the private sector in the control of CBPP in Zambia.  相似文献

Contagious bovine pleuropneumonia (CBPP), caused by Mycoplasma mycoides subsp. mycoides SC (MmmSC), is one of the most important diseases of cattle in Sub-Saharan Africa. The live T1/44 vaccine is normally used for its control but produces only transient protection and gives rise to adverse reactions. The present study evaluated the efficacy of danofloxacin (2.5% Advocintrade mark, Pfizer Ltd.) for the treatment of naturally infected cattle and in the prevention of CBPP transmission to in-contact cattle. Adult cattle, taken from a natural outbreak, were placed into two groups of 10 animals and kept on a research farm in paddocks 50m apart. One group was treated with 2.5mg/kg danofloxacin on days 0, 1 and 2; the other group were saline treated. On day 2, 10 CBPP-free, seronegative cattle were placed in contact with each of the two groups. All cattle were monitored for 3.5 months. No differences were seen in clinical improvement of the CBPP-affected cattle treated with danofloxacin compared with the untreated CBPP-affected cattle with approximately half of each group being withdrawn because of CBPP or showing CBPP lesions at post mortem examination. Clinical scores of the two groups were also similar. However cattle kept in contact with the danofloxacin-treated CBPP-affected animals showed significantly fewer lesions, less mortality and fewer animals were seropositive (P<0.02) and had reduced clinical scores (P<0.001) compared to cattle kept in contact with untreated CBPP-affected cattle. MmmSC was also isolated from fewer contact controls kept with the treated group. These findings could have important implications for the control of CBPP in Africa.  相似文献

In the first survey for contagious bovine pleuropneumonia (CBPP) in the southern region of the Sudan (Bahr el Ghazal Province), 8.1% of Dinka-owned, and 9.2% of Fellata-owned, cattle, and 48 and 20% of their respective herds were serologically positive for CBPP. Dinka cattle between 3 months and 3 years of age had significantly higher test reaction rates than those younger or older. There was a positive association between reaction rates and herd size. A similar association was found between crude mortality rates and herd size, though virtually none of the variability among herds in crude mortality was accounted for by presence of CBPP infection. Overall, the study indicated a highly endemic situation for CBPP infection with rare overt disease under conditions of little prior intervention. Under such conditions, it is suggested that a “surveillance and selective actions” rather than a “mass actions” approach might be the most suitable, with vaccination limited by surveillance to those areas at highest risk of outbreaks of clinically important CBPP, namely areas of low herd immunity or those where actual outbreaks of clinical disease have already occurred. Such an approach to CBPP could serve as a “model” for establishing a system of improved disease intelligence overall in the southern Sudan.  相似文献

Mannheimia haemolytica and Histophilus somni are frequently isolated from diseased cattle with bovine respiratory disease (BRD). They compromise animal lung function and the immune responses generated are not sufficient to limit infection. Identification of specific immunogenic antigens for vaccine development represents a great challenge. Immunogenic proteins were identified by immunoproteomic approach with sera from cattle immunized with a commercial cellular vaccine of M. haemolytica and H. somni. Proteins of M. haemolytica were identified as solute ABC transporter, iron-binding protein, and hypothetical protein of capsular biosynthesis. Histophilus somni proteins correspond to porin, amino acid ABC transporter, hypothetical outer membrane protein, cysteine synthase, and outer membrane protein P6. Although these antigens share strong similarities with other proteins from animal pathogens, the ABC system proteins have been associated with virulence and these proteins could be considered as potential vaccine candidates for BRD.  相似文献

Mycoplasma mycoides subsp. mycoides Large Colony (LC) type is a pathogen of goats causing contagious agalactia and respiratory disease, found on all continents where small ruminants are kept. It shares close genetic characteristics with M. mycoides subsp. capri. Substrate oxidation by 22 strains of M. mycoides subsp. mycoides LC from nine countries was compared with that of eight strains of M. mycoides subsp. capri from five countries. There was considerable similarity in the substrates used, but substrate saturation coefficients (Ks) varied for different substrates. Substrate utilization patterns and Ks values did not (1) significantly differentiate the LC strains from each other, (2) show any correlation with geographical origin, or (3) distinguish the LC strains from the capri strains. These results support previous studies justifying the reclassification of these subspecies as a single species.  相似文献

A cross-sectional study was designed to investigate the prevalence of bovine brucellosis and contagious bovine pleuropneumnia (CBPP) in Kongor Rural Council, southern Sudan and to assess associations between the presence of antibodies for these diseases with age, sex and breed. The detection of Brucella antibodies using the Rose-Bengal plate test was significantly associated with increasing age. The difficulty of estimating the true prevalence of brucellosis from the apparent prevalence based on test results is disscussed. Although no linear association between age and CBPP complement fixation test results were found, cattle of 2–4 years of age appeared to be at higher risk. Local factors important in CBPP disease control planning are discussed.  相似文献

African animal trypanosomiasis (AAT) continues to inflict heavy losses on livestock production especially cattle in terms of decreased production and productivity in Uganda. AAT is a disease complex caused by tsetse fly-transmitted Trypanosoma brucei brucei, Trypanosoma brucei rhodesiense, Trypanosoma congolense, and Trypanosoma vivax. The disease is most important in cattle but also known to cause serious losses in pigs, camels, goats, and sheep. Several control measures including live bait technology, mass treatment of cattle with trypanocidal drugs, and deployment of tsetse traps have been used in the past 10 years, but the problem still persists in some areas. This necessitated an exploration of the factors associated with continued trypanosome infections in cattle, which are also known reservoirs for the zoonotic trypanosomiasis. A structured questionnaire was administered to 286 animal owners from 20 villages purposively selected from Lira, Kole, and Alebtong districts of Lango subregion to obtain information on the factors associated with persistence of infection. Over 50% of the respondents reported trypanosomiasis as a major challenge to their livestock. Land ownership (P = 0.029), type of livestock kept (P = 0.000), disease control strategy employed (P = 0.000), source of drugs (P = 0.046), and drug preparation (P = 0.017) were associated with persistent AAT infection. We recommend continued farmer sensitization on the threat of AAT and the available prevention and control options. The use of isometamidium chloride for prophylaxis against trypanosomiasis is highly recommended. There is also a need to foster qualified private veterinary drug supply in the region.

Summary After an absence of about 25 years contagious bovine pleuropneumonia (CBPP) appeared again in 1990 in Tanzania. It was preceded by a spread in Kenya to an area bordering Tanzania. Due to the frequent cattle movements across the border it was soon introduced into Loliondo in northern Tanzania. One month after the first cases, CBPP was suspected in a total of 9 herds comprising 1,500 cattle. However, few animals showed clear clinical signs and frequent antibiotic treatment at an early stage further obscured the clinical picture. In one herd with acute cases, the diagnosis was confirmed by autopsy andMycoplasma mycoides subsp.mycoides, SC type, was isolated. From this herd several serum samples were positive in the complement fixation test and gave high absorbance values in an ELISA withM. mycoides subsp.mycoides antigen. From 5 other herds with suspected cases blood samples were negative by the complement fixation test but in the enzyme-linked immunosorbent assay at least one in each herd was positive.

Resumen Después de un período de 25 años sin ningún brote, la pleuroneumonía contagiosa bovina apareció de nuevo en Tanzania en 1990, después de que la enfermedad se extendiera a una zona de Kenya fronteriza con Tanzania. Debido a los frecuentes movimientos de ganado vacuno a través de la frontera, la enfermedad se declaró pronto en Loliondo, en el norte de Tanzania. Un mes después de los primeros casos, 9 rebaños, que contabilizaban en total 1500 animales, eran sospechosos de estar infectados. Sin embargo, pocos animales mostraron síntomas clínicos claros y et tratamiento precoz con antibióticos contribuyó a disminuir la intensidad de las manifestaciones clinicas. En un rebaño en el que hubo casos agudos el diagnóstico se confirmó mediante necropsia y se aislóMycoplasma mycoides subsp.mycodies del tipo SC. Varias muestras de sangre de este rebaño dieron resultado positivo en el test de fijación del complemento y dieron valores de absorbancia altos en un test de ELISA conM. mycoides subsp.mycoides. Las muestras de sangre provenientes de otros 5 rebaños en los que se sospechó la existencia de la enfermedad dieron resultado negativo en el test de fijación de complemento mientras que al menos un animal de cada rebaño dio resultado positivo en el test ELISA.

Peripneumonie Contagieuse Bovine Dans Le Nord De La Tanzanie, Confirmation Par Culture Et Etudes Serologiques
Résumé Après une absence d'environ 25 ans la péripneumonie contagieuse bovine est de nouveau apparue en 1990 en Tanzanie. Elle a été prècedée d'une progression au Kenya vers une zone bordant la Tanzanie. Par suite de mouvements frontaliers fréquents du bétail, elle a été beintôt introduite dans la région de Loliondo, dans le nord de la Tanzanie. Un mois après les premiers cas, 9 troupeaux totalisant 1500 têtes ont été supectés. Cependant, peu d'animaux ont présenté des signes cliniques nets et, de surcroît, les traitements antibiotiques entrepris en début de maladie obscurissent le tableau clinique. Le diagnostic a été confirmé à l'autopsie dans un troupeau présentant des cas cliniques etMycoplasma mycoides subsp.mycoides, type SC, a été isolé. Plusieurs échantillons de sérum de ce troupeau ont été positifs en fixation du complément et ont donné de grandes valeurs d'absorption dans un test ELISA avec un antigèneM. mycoides subsp.mycoides. Pour 5 autres troupeax avec des cas suspects, les échantillons de sang ont été négatifs en fixation du complément mais un au moins pour chaque troupeaux a été positif dans un test ELISA.
