Abstract

Potato (Solanum tuberosum) showed a well-established mutualistic association with arbuscular mycorrhizal fungi. In the present study, earthen pots containing autoclaved soil were taken in which surface sterilized potato seeds were sown. The seed sowed earthen pots were inoculated by sterilized spores of arbuscular mycorrhizal fungi along with sterilized inoculated maize-root fragments, while the rest half of the seed filled pots, without any inoculation depicting control but were provided with non-inoculated maize root fragments. The inoculation was performed twice; first inoculation was done 3 days prior to sowing of potato seeds and second at the time of seedling emergence. Sampling of the inoculated as well as of control plants was performed at 20-day intervals till 80?days after tuber initiation. An increment in the level of metabolites as well as mineral nutrient was found in mycorrhizal inoculated potato tubers in contrast to non-inoculated. Our work demonstrates that inoculation has a great potential in enriching storage metabolites and nutrients in potato plant in low yielding soils.

Mycorrhizae are fungal symbionts forming mutualistic relationships with plant roots. This study was undertaken to evaluate the overall influence of arbuscular mycorrhizal fungi (AMF) on the overall growth and development of carrot plant. Surface-sterilized seeds of carrot were sown in earthen pots filled with sterile soil. Half of the pots were inoculated with AMF spores; the other half without any AMF inoculation represented control. After germination inoculated plants, along with the controls, were sampled at 20, 40, 60 and 80 days of growth after seedling emergence. There is a progressive net increase in growth with each 20-day interval after seedling emergence. Storage roots of carrot had a higher level of metabolites and nutrients like nitrogen, phosphorus and potassium in mycorrhiza colonized plants than the control. High performance liquid chromatography analysis indicated an increase in the carbohydrate fractions in AMF-inoculated roots. Thus, mycorrhiza has been found increasing the growth, metabolites and nutrition of carrot plant.

The effect of inoculation with arbuscular mycorrhizal fungi (AMF) on crop productivity under small-scale farming conditions and the persistence of the fungal inoculum in the field were investigated over 2 years. Sweet potato plants were inoculated with various combinations of AMF and grown under traditional Chinese farming procedures. Plantlets from germinated tubers were inoculated in a soil/sand mixture at the time of hand transplanting into the field. A technique for long-term preservation of the root samples and a fast, reliable DNA extraction method were developed to track and evaluate the persistence of the selected AMF in two field trials in China. The AMF rDNA was specifically amplified by nested PCR from colonized sweet potato roots collected from the field trials, and polymorphism of the 5′-end of the large ribosomal subunit was used to monitor fungi at the species level. AMF varied in their ability to establish after inoculation, and in their effect on yield and quality of sweet potato tubers.

The mycorrhizal enhancement of plant growth is generally attributed to increased nutrients uptake. A greenhouse experiment was conducted to investigate the effect of arbuscular mycorrhizal fungi (AMF) inoculation on the growth and nutrient uptake of directly seeded wetland rice. Seeds were germinated and inoculated with arbuscular mycorrhizal fungi or left uninoculated. The plants were grown at 60% of ‐0.03 MPa to establish the mycorrhizas. After 5 weeks, half of the pots were harvested and the rest were flooded with deionized water to maintain 3–5 cm of standing water until harvesting (122 days after sowing). Mycorrhizal fungal colonization of rice roots was 36.2% at harvest. Mycorrhizal fungi inoculated rice seedlings grew better compared to uninoculated seedlings and had increased grain yield (10%) at the harvesting stage. Shoot and root growth were effectively increased by AMF inoculation at the harvesting stage. The nitrogen (N) and phosphorus (P) acquisition of direct seeding wetland rice were significantly increased by AMF inoculation. The AMF enhanced N and P translocation through the hyphae from soils to roots/shoots to grains effectively.

Twenty strains of arbuscular mycorrhizal fungi (AMF), native to West Africa, and three commercial AMF, were evaluated for their protective effect against root-knot nematodes, Meloidogyne spp., in pots and field experiments in Benin. In pots, these strains were assessed in sterilized soil following inoculation of nematodes and in non-sterilized soil naturally infested with nematodes using tomato. The four strains showing greatest potential in suppressing nematode development were further assessed in the field with a relatively high natural infestation level of nematodes (155 per 100 cm3 soil) over a tomato–carrot double cropping. In the pot experiments, most native strains provided significant suppression of nematode multiplication and root galling, but in most cases the level of nematode control depends on either sterilized or non-sterilized soils. In the field experiments, application of AMF mostly resulted in significant suppression of nematode multiplication and root galling damage on both crops indicating that the AMF persists and remains protective against root-knot nematodes over two crop cycles. Field application of AMF increased tomato yields by 26% and carrot yields by over 300% compared with the non-AMF control treatments. This study demonstrates for the first time, the protective effect of indigenous West African AMF against root-knot nematodes on vegetables. The potential benefits of developing non-pesticide AMF-based pest management options for the intensive urban vegetable systems are evident.

ABSTRACT

This work was conducted to study phosphorus (P) efficiency of two maize genotypes (Zea mays, L.) in calcareous soil grown in potted soil with two levels of P in soil by adding 40 and 270 mg P/kg soil. Half of the pots were inoculated with arbuscular mycorrhizal fungi (AMF) (Rhizoglomus irregulare). The maize genotypes were harvested two times at 35 and 50 days after transplanting. The plant dry matter, root length and Plant P uptake of maize genotype Hagen 1 without mycorrhizal fungi (AMF) increased significantly compared with Hagen 9 at a low P level. In contrast, there was no significant difference between two maize genotypes inoculated with AMF under the same P level. The predicted value increased rapidly with increasing P levels from about 70% up to 97% in both maize genotypes with and without mycorrhizal fungi. At a low P level, the mycorrhizae hyphae contributed by about 31.6% and 30.2% of the predicted total P uptake in maize genotype Hagen 1 and Hagen 9, respectively. The results of this study suggested that the P-inefficient genotype Hagen 9 improved with inoculation with mycorrhizal fungi under a low P level at the same conditions of this experiment. Also, root growth system and mycorrhizal hyphae length would be a suitable plant parameter for studying P efficient maize genotypes, especially under limited P supply. The current study clearly pointed out that the mechanistic simulation model (NST 3.0) provides useful tools for studying the role of AMF in P uptake of plant.

Aims : The aim of this study was to explore interactive effects between quality (types) and quantity (application rates) of biochar as well as of arbuscular mycorrhiza (AM) symbiosis on the growth of potato plants. Methods : A low P sandy loam soil was amended with 0%, 1.5%, or 2.5% (w/w) of either of 4 types of biochar, which were produced from wheat straw pellets (WSP) or miscanthus straw pellets (MSP) pyrolyzed at temperatures of either 550°C or 700°C. Potato plants grown in pots containing the soils or soil biochar mixture were inoculated with or without AM fungus (AMF), Rhizophagus irregularis. The experiment was carried out under fully irrigated semi‐field conditions and plants were harvested 101 days after planting. Results : Application of high temperature biochar decreased growth, biomass and tuber yield of potato plants, while the low temperature biochar had a similar effect on yield as plants grown without biochar amendment. Total biomass of potato plants were decreased with the increasing rate of biochar. Arbuscular mycorrhizal fungus inoculation stimulated the growth of potato plants in all organs, increased tuber biomass significantly in 1.5% MSP700 amended plants, and to a lesser degree for WSP700, MSP550, and WSP550. In addition, plant biomass gain was linearly related to N, P, and K uptake, the ratio of P to N in the leaf of plants indicated that all treatments were mainly P‐limited. A multiple linear regression using P uptake and biochar rate as independent variables explained 91% of the variation in total biomass. The single effect of AMF inoculation, type and rate of biochar affected plant N, P and K uptake similarly. While AMF inoculation significantly increased P uptake in potato plants grown in soil with WSP700 or MSP700 despite of the rate of biochar. In general, application of biochar significantly increased AMF root colonization of potato plants. Conclusions : The application of MSP550 at 1.5% combined with AMF stimulated growth of potato the most. Furthermore, the results indicated that the interactive effect of AMF inoculation, biochar type and application rate on potato growth to a large extent could be explained by effects on plant nutrient uptake.

Cowpea is an important crop that serves as a legume and vegetable source to many smallholder farmers in sub-Saharan Africa. Soil fertility is a significant limitation to its production thus; inoculation with beneficial soil biota such as arbuscular mycorrhizal fungi (AMF) could improve its performance. However, plant–AMF interaction could vary based on crop cultivar hence affecting overall crop production. The present study aimed at determining the effect of AMF inoculation and soil sterilization on root colonization and growth of a wild-type and three modern cowpea cultivars grown by smallholder farmers in Kenya. Potted cowpea plants were inoculated with a commercial AMF inoculum comprising of Rhizophagus irregularis, Funneliformis mosseae, Glomus aggregatum and Glomus etunicatum and maintained in a greenhouse for 40 days. After harvesting, mycorrhizal colonization, nodule number and dry weight, root and shoot dry weights, nitrogen (N,) phosphorus (P) and potassium (K) content were determined. Interestingly, the modern cultivars showed significantly (p < 0.001) higher root colonization, nodulation, shoot P and N compared to the wild-type cultivar. Moreover, a strong positive correlation between AMF root colonization and shoot P (r2 = 0.73, 0.90, p < 0.001), AMF root colonization and shoot N (r2 = 0.78; 0.89, p < 0.001) was observed in both sterilized and non-sterilized soil, respectively. Soil sterilization affected root colonization and growth parameters with plants grown in non-sterilized soil performing better than those grown in sterilized soil. This study provides major evidence that modern cowpea cultivars are still responsive to mycorrhizal inoculation suggesting that modern breeding programs are not deleterious AMF symbiosis.

Abstract

In general, according to previous studies, pioneer species do not require arbuscular mycorrhizal fungi (AMF) to increase their growth and survival in tropical systems. The aim of this study was to determine the dependence response to AMF of Heliocarpus appendiculatus, a pioneer species, at different phosphorus (P) levels. In a greenhouse experiment, H. appendiculatus seedlings were grown in pots with a sterile vermiculite-sand mixture (1:1). Two sets of pots were set up: One set was inoculated (150 spores per pot) with indigenous AMF from a tropical rain forest at “Los Tuxtlas” (Veracruz, Mexico); the other set was not inoculated. To each set, 0, 0.02, 0.2, and 2 g L?1P was added. All pots were watered with 250 mL of nutrient solution. Mycorrhizal plants showed a higher total dry weight and relative growth rate in 0.02 g L?1P concentration, while nonmycorrhizal plants responded positively at 0.2 g L?1P; a decrease in plant responses at higher P levels was observed in both treatments. H. appendiculatus showed to have higher relative dependence at lower P concentration (≈50%). As levels of P increased, mycorrhizal colonization decreased. Successful growth of pioneer species during succession process may be improved if there is AMF content in soils, prior to disturbance.

Arbuscular mycorrhizal fungi (AMF) have the capability to improve crop yields by increasing plant nutrient supply. A pot experiment was conducted under natural conditions to determine the response of AMF inoculation on the growth of maize (Zea mays L.), sorghum (Sorghum bicolor L.), millet (Pennisetum glaucum L.), mash bean (Vigna mungo L.), and mung bean (Vigna radiata. L.) crops during 2008. The experiment was conducted as a completely randomized design in three replications using phosphorus (P)–deficient soil. Three plants were grown in 10 kg soil up to the stage of maximum growth for 70 days. Spores of AMF were isolated from rhizosphere of freshly growing wheat and berseem crops and mixed with sterilized soil with fine particles. Crops were inoculated in the presence of indigenous mycorrhiza with the inoculum containing 20 g sterilized soil mixed with 40–50 AMF spores. Inoculation with AMF improved yield and nutrient uptake by different crops significantly over uninoculated crops. Inoculated millet crop showed 20% increase in shoot dry matter and 21% in root dry matter when compared with other inoculated crops. Increases of 67% in plant nitrogen (N) and iron (Fe) were observed in millet, 166% in plant P uptake was observed in mash beans, 186% in zinc (Zn) was measured in maize, and 208% in copper (Cu) and 48% in manganese (Mn) were noted in sorghum crops. Maximum root infection intensity of 35% by AMF and their soil spore density were observed in millet crop followed by 32% in mash beans. Results suggest that inoculation of AMF may play a role in improving crop production and the varied response of different crops to fungi signifies the importance of evaluating the compatibility of the fungi and plant host species.

Effectiveness of arbuscular mycorrhizal fungi (AMF) is crucial for maximum plant growth and acquisition of mineral nutrients under drought. The objective of this research was to determine effects of varied rates of AMF inoculum on plant growth and acquisition of phosphorus (P), zinc (Zn), copper (Cu), and manganese (Mn) by barley (Hordeum vulgare L. cv. SLB‐6) grown with and without drought stress (WS and nonWS). Plants inoculated with four inoculum rates [control (M0), 120 (M1), 240 (M2), and360 (M3) spores per 100 g dry soil] of Glomus mosseae were grown in a low P silty clay (Typic Xerochrept) soil (pH=8.0) mix in a greenhouse for 45 days. Root AMF colonization increased as inoculum rate increased in plants grown with WS and nonWS. Leaf area and shoot and root dry matter (DM) increased as inoculum rate increased up to M2 regardless of soil moisture. Shoot concentrations of P, Cu, and Mn were generally higher for mycorrhizal (AMF) than for nonmycorrhizal (nonAMF) plants grown with both WS and nonWS. Shoot contents of P, Zn, Cu, and Mn were higher for AMF than for nonAMF plants grown with nonWS, and shoot contents of P were higher for AMF than for nonAMF plants with WS. For plants grown with WS and nonWS, contents of P, Zn, Cu, and Mn were generally higher for plants inoculated with M2 compared to other rates of inoculum. The results of this study indicated that plant responses to root colonization with AMF were dependent on AMF rate and soil moisture. Based on enhancements in plant DM and mineral acquisition traits, M2 inoculum was the most effective rate of inoculation for this AMF isolate.

The capacities of two arbuscular mycorrhizal (AM) fungi, Glomus mosseae and Glomus versderme, tomineralize added organic P were studied in a sterilized calcareous soil. Mycorrhizal (inoculated with either of the AM fungi) and non-mycorrhizal red clover (Trghlium pmtense L.) plants were grown for eight weeksin pots with upper root, central hyphal and lower soil compartments. The hyphal and soil compartmentsreceived either organic P (as Na-phytate) or inorganic P (as KH2PO4) at tbe rate of 50 mg P kg-1. No P wasadded to the root compartments. Control pots received no added P. Yields were higher in mycorrhizal than innon-mycorrhizal clover. Mycorrhizal inoculation doubled shoot P concentration and more than doubled total P uptake of plaflts in P-amended soil, irrespective of the form of applied P. The mycorrhizal contributionto inorganic P uptake was 80% or 76% in plants inoculated with G. mosseae or G. verefforme, respectively. Corresponding values were 74% and 82% when Na-phytate was applied. In the root compartments of the mycorrhizal treatments, the proportion of root length infected, hyphal length density and phosphatase activity were all higher when organic P was applied than when inorganic P was added.

Summary Sweet potatoes were micropropagated and then transplanted from axnic conditions to fumigated soil in pots in the greenhouse. Spores of Glomus clarum were obtained from Brachiaria decumbens or from sweet potatoes grown in soil infected with this fungus and with an enrichment culture of Acetobacter diazotrophicus. Three experiments were carried out to measure the beneficial effects of vesicular-arbuscular mycorrhizal (VAM) fungi-diazotroph interactions on growth, nutrition, and infection of sweet potato by A. diazotrophicus and other diazotrophs obtained from sweet potato roots. In two of these experiments the soils had been mixed with 15N-containing organic matter. The greatest effects of mycorrhizal inoculation were observed with co-inoculation of A. diazotrophicus and/or mixed cultures of diazotrophs containing A. diazotrophicus and Klebsiella sp. The tuber production was dependent on mycorrhization, and total N and P accumulation were increased when diazotrophs and G. clarum were applied together with VAM fungal spores. A. diazotrophicus infected aerial plant parts only when inoculated together with VAM fungi or when present within G. clarum spores. More pronounced effects on root colonization and intraradical sporulation of G. clarum were observed when A. diazotrophicus was co-inoculated. In non-fumigated soil, dual inoculation effects, however, were of lower magnitude. 15N analysis of the aerial parts and roots and tubers at the early growth stage (70 days) showed no statistical differences between treatments except for the VAM+Klebsiella sp. treatment. This indicates that the effects of A. diazotrophicus and other diazotrophs on sweet potato growth were caused by enhanced mycorrhization and, consequently, a more efficient assimilation of nutrients from the soil than by N2 fixation. The possible interactions between these effects are discussed.

A pot experiment was conducted to investigate the effect of epigeic earthworm (Eisenia fetida) and arbuscular mycorrhizal (AM) fungi (Glomus intraradices) on soil enzyme activities and nutrient uptake by maize, which was grown on a mixture of sterilized soil and sand. Maize plants were grown in pots inoculated or not inoculated with AMF, treated or not treated with earthworms. Wheat straw was added as a feed source for earthworms. Mycorrhizal colonization of maize was markedly increased in AM fungi inoculated pots and further increased by addition of epigeic earthworms. AM fungi and epigeic earthworms increased maize shoot and root biomass, respectively. Soil acid phosphatase activity was increased by both earthworms and mycorrhiza, while urease and cellulase activities were only affected by earthworms. Inoculation with AM fungi significantly (p?<?0.001) increased the activity of soil acid phosphatase but decreased soil available phosphorus (P) and potassium (K) concentrations at harvest. Addition of earthworms alone significantly (p?<?0.05) increased soil ammonium-N content, but decreased soil available P and K contents. AM fungi increased maize shoot weight and root P content, while earthworms improved N, P, and K contents in shoots. AM fungi and earthworm interactively increased maize shoot and root biomass through their regulation of soil enzyme activities and on the content of available soil N, P, and K.

Nutrient deficiency, especially zinc (Zn) and phosphorus (P), is a common nutritional problem for the production of some crops in Turkey. This problem results in the application of increasing amounts of several fertilizers. Mycorrhizal inoculation or the indigenous potential of mycorrhizae in the soil is a critical factor in crop production under low supply of Zn and P. The effects of selected mycorrhizal inoculation on growth and Zn and P uptake of maize and green pepper were investigated in Zn- and P-deficient calcareous soils from Central Anatolia. Soils were sterilized by autoclaving and plants were grown for 7 weeks in pots under greenhouse conditions with inoculation of two selected arbuscular mycorrhizal (AM) species (Glommus moseea and G. etunicatum) at three rates of P (0, 25, 125 mg P kg?1 soil) and two rates of Zn (0 and 5 mg Zn kg?1soil). Without mycorrhizal inoculation, shoot and root dry matter production were severely affected by P and Zn deficiencies, and supply of adequate amounts of P and Zn significantly enhanced plant growth. When the soil was inoculated with mycorrhizal inoculation, the increasing effects of P and Zn fertilization on plant growth remained less pronounced. In accordance with growth data, mycorrhizae inoculation enhanced P and Zn concentration of plants, especially under low supply of P and Zn. The results obtained indicate that maize and green pepper are highly mycorrizal–dependent (MD) plant species under both low P and Zn supply and mycorrhizae play an essential role in P and Zn nutrition of plants in P and Zn-deficient soils. Although addition of P and Zn increased plant growth and plants are mycorrhizal dependent on P and Zn nutrition however dependence is much more dependent on P nutrition.

The interactive impacts of arbuscular mycorrhizal fungi (AMF, Glomus intraradices) and earthworms (Aporrectodea trapezoides) on maize (Zea mays L.) growth and nutrient uptake were studied under near natural conditions with pots buried in the soil of a maize field. Treatments included maize plants inoculated vs. not inoculated with AMF, treated or not treated with earthworms, at low (25 mg kg−1) or high (175 mg kg−1) P fertilization rate. Wheat straw was added as feed for earthworms. Root colonization, mycorrhiza structure, plant biomass and N and P contents of shoots and roots, soil available P and NO3–N concentrations, and soil microbial biomass C and N were measured at harvest. Results indicated that mycorrhizal colonization increased markedly in maize inoculated with AMF especially at low P rate, which was further enhanced by the addition of earthworms. AMF and earthworms interactively increased maize shoot and root biomass as well as N and P uptake but decreased soil NO3–N and available P concentrations at harvest. Earthworm and AMF interaction also increased soil microbial biomass C, which probably improved root N and P contents and indirectly increased the shoot N and P uptake. At low P rate, soil N mobilization by earthworms might have reduced potential N competition by arbuscular mycorrhizal hyphae, resulting in greater plant shoot and root biomass. Earthworms and AMF interactively enhanced soil N and P availability, leading to greater nutrient uptake and plant growth.
