In this study, we investigated the effects of lanthanum (La), one of the rare earth elements (REEs), on microbial biomass C as well as the decomposition of 14C-labelled glucose in a fluvo-aquic soil in 28 days. The soil was collected from the field plots under maize/wheat rotation in Fengqiu Ecological Experimental Station of Chinese Academy of Sciences, Henan Province, China. Application of La decreased soil microbial biomass C during the experimental period, and there was a negative correlation (P < 0.01) between microbial biomass and application rate of La. La increased microbial biomass 14C after 14C glucose addition, and the increase was significant (P < 0.05) at the rates of more than 160 mg kg−1 soil. La slightly increased 14CO2 evolution at lower rates of application but decreased it at higher rates 1 day after 14C glucose addition, while there was no significant effect from days 2 to 28. For the cumulative 14CO2 evolution during the incubation of 28 days, La slightly increased it at the rates of less than 120 mg kg−1 soil, while significantly decreased (P < 0.05) it at the rate of 200 mg kg−1 soil. The results indicated that agricultural use of REEs such as La in soil could decrease the amount of soil microbial biomass and change the pattern of microbial utilization on glucose C source in a short period.  相似文献

The most frequently used models simulating soil organic matter (SOM) dynamics are based on first-order kinetics. These models fail to describe and predict such interactions as priming effects (PEs), which are short-term changes in SOM decomposition induced by easily available C or N sources. We hypothesized that if decomposition rate depends not only on size of the SOM pool, but also on microbial biomass and its activity, then PE can be simulated. A simple model that included these interactions and that consisted of three C pools - SOM, microbial biomass, and easily available C - was developed. The model was parameterized and evaluated using results of 12C-CO2 and 14C-CO2 efflux after adding 14C-labeled glucose to a loamy Haplic Luvisol. Experimentally measured PE, i.e., changes in SOM decomposition induced by glucose, was compared with simulated PE. The best agreement between measured and simulated CO2 efflux was achieved by considering both the total amount of microbial biomass and its activity. Because it separately described microbial turnover and SOM decomposition, the model successfully simulated apparent and real PE.The proposed PE model was compared with three alternative approaches with similar complexity but lacking interactions between the pools and neglecting the activity of microbial biomass. The comparison showed that proposed new model best described typical PE dynamics in which the first peak of apparent PE lasted for 1 day and the subsequent real PE gradually increased during 60 days. This sequential decomposition scheme of the new model, with immediate microbial consumption only of soluble substrate, was superior to the parallel decomposition scheme with simultaneous microbial consumption of two substrates with different decomposability. Incorporating microbial activity function in the model improved the fit of simulation results with experimental data, by providing the flexibility necessary to properly describe PE dynamics. We conclude that microbial biomass should be considered in models of C and N dynamics in soil not only as a pool but also as an active driver of C and N turnover.  相似文献

Summary The effects of trifluralin and 12 of its soil-formed metabolites on the decomposition of radio-labelled glucose, protein and cellulose were determined, using 14CO2 evolution from soil as a measure of decomposition. Trifluralin increased 14C-glucose mineralization rates, but these increases could be eliminated by adding N. Trifluralin had no inhibitory effect on the mineralization of protein or cellulose, but five of the metabolites inhibited glucose mineralization. None of the trifluralin metabolites affected protein mineralization. Seven trifluralin metabolites increased the rate of cellulose mineralization when applied at rates exceeding those that would be expected in soil. After considering the rate of metabolite application and the magnitude of the responses observed these compounds are expected to have no major effects on the microbial decomposition processes in soil.  相似文献

Summary Degradation of the herbicide phosphinothricin (L-homoalanine-4-yl-(methyl)-phosphinic acid) in a phaeozem was investigated by monitoring the 14CO2 release from [1-14C] and [3,4-14C]phosphinothricin. The degradation was largely due to microbial activity, since the rate decreased by more than 95% when the soil was sterilized by -radiation. Data obtained with both labels suggested that decarboxylation of phosphinothricin preceded oxidation of its C-atoms 3 and 4, since a metabolite, probably 3-methylphosphinico-propanoic acid, was only labelled when [3,4-14C]phosphinothricin was used as the substrate. Maximum rates of 14CO2 production from both the 1- and 3,4-label positions occurred without a lag phase during the breakdown of phosphinothricin as monitored for a total of 30 days at 5-day intervals. This result indicated that a phosphinothricin-degrading microbial community was already present in the soil. With low concentrations of [1-14C]phosphinothricin (10.7 mg kg-1 soil), complete decarboxylation at 25°C was observed within 30 days of incubation, compared to 15.9% 14CO2 release from [3,4-14C]phosphinothricin. Increasing the quantity of the herbicide in the soil (10.7–1372 mg kg-1) resulted in increased degradation rates, irrespective of whether the herbicide was labelled in the positions 1 or 3 and 4. Addition of glucose and other carbohydrates stimulated 14CO2 release while addition of a yeast extract had a negative effect. Glucose stimulation was reversed by ammonium nitrate, suggesting that the microorganisms were using the herbicide as a source of N.  相似文献

Low molecular weight (LMW) organic compounds in soil solution are easily biodegradable and could fuel respiration by soil microorganisms. Our main aim was to study the mineralization kinetics of monosaccharides using 14C-radiolabelled glucose. Based on these data and the soil solution concentrations of monosaccharides, we evaluated the contribution of monosaccharides to basal respiration for a variety of tropical forest soils. Further, the factors controlling the mineralization kinetics of monosaccharides were examined by comparing tropical and temperate forest soils. Monosaccharides comprised on average 5.2 to 47.7% of dissolved organic carbon in soil solution. Their kinetic parameters (V max and KM ), which were described by a single Michaelis-Menten equation, varied widely from 11 to 152?nmol?g?1?h?1 and 198 to 1294?µmol?L?1 for tropical soils, and from 182 to 400?nmol?g?1?h?1 and 1277 to 3150?µmol?L?1 for temperate soils, respectively. The values of V max increased with increasing microbial biomass-C in tropical and temperate soils, while the KM values had no correlations with soil biological or physicochemical properties. The positive correlation between V max values and microbial biomass-C indicates that microbial biomass-C is an essential factor to regulate the V max values in tropical and temperate forest soils. The biodegradation kinetics of monosaccharides indicate that the microbial capacity of monosaccharide mineralization far exceeds its rate at soil solution concentration. Monosaccharides in soil solution are rapidly mineralized, and their mean residence times in this study were very short (0.4–1.9?h) in tropical forests. The rates of monosaccharide mineralization at actual soil solution concentrations made up 22–118% of basal respiration. Probably because of the rapid and continuous production and consumption of monosaccharides, monosaccharide mineralization is shown to be a dominant fraction of basal respiration in tropical forest soils, as well as in temperate and boreal forest soils.  相似文献

Priming effect (PE) on soil organic carbon (SOC) decomposition caused by the addition of organic carbon (C) is an important ecological process in regulating soil C cycle. Additionally, most priming studies are confined to laboratory trials, while the assessment of soil PE under field conditions with variable weather conditions is scarce. This study assessed the direct effects of glucose addition and in-situ warming on the extent of PE under field conditions in a temperate forest. We evaluated soil PE using 13C-glucose labelling, a simple and novel technique, based on the Keeling plot method. Glucose addition significantly enhanced native SOC decomposition and induced strong PE. However, the effect of in-situ warming on the extent of PE was not significant. This study confirms the importance of PE in regulating SOC turnover under field conditions.  相似文献

Tannins are purported to be an important factor controlling nitrogen cycling in forest ecosystems, and the ability of tannins to bind proteins in protein-tannin complexes is thought to be the primary mechanism responsible for these effects. In this study, we examined the influence of well-characterized tannins purified from five different plant species on C and N dynamics of a forest soil A horizon. Tannic acid, a commonly used and commercially available hydrolyzable tannin (HT), and cellulose were also included for comparison. With the exception of tannins from huckleberry (Vaccinium ovatum), the amendments increased respiration 1.4-4.0 fold, indicating that they were acting as a microbial C source. Tannic acid was significantly more labile than the five purified tannins examined in this study. All treatments decreased net N mineralization substantially, through greater N immobilization and decreased mineralization. The six tannins inhibited gross ammonification rates significantly more than cellulose. This suggests that added tannins had effects in addition to serving as an alternative C source. Tannins purified from Bishop pine (Pinus muricata) were the only tannins that significantly inhibited potential gross nitrification rates, however, rates were low even in the control soil making it difficult to detect any inhibition. Differences in tannin structure such as condensed versus HTs and the hydroxylation pattern of the condensed tannin B-ring likely explain differences observed among the tannin treatments. Contrary to other studies, we did not find that condensed tannins were more labile and less inhibitory than HTs, nor that shorter chained tannins were more labile than longer chained tannins. In addition to supporting the hypothesis that reduced N availability in the presence of tannins is caused by complexation reactions, our data suggests tannins act as a labile C source leading to increased N immobilization.  相似文献

While it is well known that soil moisture directly affects microbial activity and soil organic matter (SOM) decomposition, it is unclear if the presence of plants alters these effects through rhizosphere processes. We studied soil moisture effects on SOM decomposition with and without sunflower and soybean. Plants were grown in two different soil types with soil moisture contents of 45% and 85% of field capacity in a greenhouse experiment. We continuously labeled plants with depleted 13C, which allowed us to separate plant-derived CO2-C from original soil-derived CO2-C in soil respiration measurements. We observed an overall increase in soil-derived CO2-C efflux in the presence of plants (priming effect) in both soils. On average a greater priming effect was found in the high soil moisture treatment (up to 76% increase in soil-derived CO2-C compared to control) than in the low soil moisture treatment (up to 52% increase). Greater plant-derived CO2-C and plant biomass in the high soil moisture treatment contributed to greater priming effects, but priming effects remained significantly higher in the high moisture treatment than in the low moisture treatment after correcting for the effects of plant-derived CO2-C and plant biomass. The response to soil moisture particularly occurred in the sandy loam soil by the end of the experiment. Possibly, production of root exudates increased with increased soil moisture content. Root exudation of labile C may also have become more effective in stimulating microbial decomposition in the higher soil moisture treatment and sandy loam soil. Our results indicate that moisture conditions significantly modulate rhizosphere effects on SOM decomposition.

The rates of sulphur (S) released to and removed from the soil inorganic pool were estimated using the isotopic dilution technique. In an initial study fresh soil was mixed with combinations of two inorganic S levels (0 and 10 μg S g−1 soil) and three plant residues (wheat straw, perennial ryegrass and oilseed rape) and followed over 32 days of incubation. As 35S recovery was inadequate prior to day 2 and re-mineralisation of immobilised 35S occurred after day 8 thereby invalidating the method, estimates of gross S transformation rates should be based on data sampled between days 2 and 8. In the main experiment 16 plant residues with ranges in S contents of 0.08-0.81%, C/S ratios of 50-604 and lignin content of 0.9-10.8 were mixed with soil and carrier-free 35S label. Net turnover rates varied from 58% of S in Persian clover being immobilised to 76% of S in winter cress being mineralised within 5 days of incubation. Gross S mineralisation varied from 0.9-14.9 μg S g−1 soil d−1, whereas gross immobilisation only varied from 0.5 to 3.1 μg S g−1 d−1. Gross S immobilisation was strongly correlated to the C/S ratio of the plant material (P<0.001), whereas gross S mineralisation showed a weaker, but still significant, correlation with lignin content (P<0.05). The results indicate that immobilisation may predominantly have been a biological process in response to carbon addition while early mineralisation may have been dominated by the biochemical hydrolysis of organic sulphates in the residues. If attention is paid to the various constraints and limitations, isotopic pool dilution using 35S offers a tool that may prove valuable in understanding and modelling soil S turnover.  相似文献

Soil was amended with 14C-labelled unripe straw only (C:N ratio ca. 20), with 14C-labelled unripe straw plus unlabelled ripe straw (C:N ratio ca. 100) or with 14C-labelled unripe straw plus glucose. Half the samples with 14C-labelled straw and half the samples with 14C-labelled plus unlabelled straw were cropped with rape plants. A decreased rate of mineralization of the 14C-labelled straw was found in the planted soil compared with the unplanted soil. The reduction was most profound in the soil amended with both labelled and unlabelled straw, indicating that at least part of the reduction was due to competition between plants and microorganisms for mineral N. No other explanations for the decrease in mineralization in the presence of plants were found. The soil amended with glucose which simulated the effect of root exudates showed an increased rate of mineralization. Therefore, the reduction in the presence of plants was probably not due to microbial use of the rhizodeposition in favour of the labelled straw. Only a minor part of the reduction was apparently due to uptake of labelled C by the plant, as only small amounts were found in the roots and shoots at harvest. The difference in 14C mineralization between treatments was not reflected in the number of bacteria in the soil at harvest. The number of bacteria, which was determined by plate counts and direct microscopy, was the same in all the soils, rhizosphere soils as well as bulk soils.  相似文献

Laboratory incubations were conducted to study the effect of sodium chloride (NaCl) on denitrification and respiratory gases (CO2, O2) from soil treated with ammonium or nitrate and incubated at 20 % moisture. The same samples were assayed for denitrifying enzyme activity (DEA) after incubation at 40 % moisture with glucose and NO3. Under aerobic conditions (20 % water content), a flush of activity was observed at 6 hours after start of incubation and subsided to negligible levels at 12 hours. Sodium chloride significantly depressed N2O and CO2 emissions and O2 consumption. Significantly more loss of N2O occurred from NH4+‐ than NO3‐treated soil at all NaCl levels and was attributed to higher microbial activity. A highly significant positive correlation was obtained between N2O emission and respiratory gases. The respiratory quotient (CO2 evolved/O2) was higher for NH4+‐treated soil and decreased with the amount of NaCl. At 40 % moisture, N2O emissions were higher than at 20 % and peaked at 37 hours followed by a sharp decrease. Short‐term incubations of soil with NH4+ or NO3 did not have an effect on denitrifying enzyme activity (DEA) while NaCl had a positive effect, particularly in previously NO3‐treated soil.  相似文献

Microbial communities in soil A horizons derive their carbon from several potential sources: organic carbon (C) transported down from overlying litter and organic horizons, root-derived C, or soil organic matter. We took advantage of a multi-year experiment that manipulated the 14C isotope signature of surface leaf litter inputs in a temperate forest at the Oak Ridge Reservation, Tennessee, USA, to quantify the contribution of recent leaf litter C to microbial respiration and biomarkers in the underlying mineral soil. We observed no measurable difference (<∼40‰ given our current analytical methods) in the radiocarbon signatures of microbial phospholipid fatty acids (PLFA) isolated from the top 10 cm of mineral soil in plots that experienced 3 years of litterfall that differed in each year by ∼750‰ between high-14C and low-14C treatments. Assuming any difference in 14C between the high- and low-14C plots would reflect C derived from these manipulated litter additions, we estimate that <∼6% of the microbial C after 4 years was derived from the added 1-4-year-old surface litter. Large contributions of C from litter < 1 year (or >4 years) old (which fell after (or prior to) the manipulation and therefore did not differ between plots) are not supported because the 14C signatures of the PLFA compounds (averaging 200-220‰) is much higher that of the 2004-5 leaf litter (115‰) or pre-2000 litter. A mesocosm experiment further demonstrated that C leached from 14C-enriched surface litter or the O horizon was not a detectable C source in underlying mineral soil microbes during the first eight months after litter addition. Instead a decline in the 14C of PLFA over the mesocosm experiment likely reflected the loss of a pre-existing substrate not associated with added leaf litter. Measured PLFA Δ14C signatures were higher than those measured in bulk mineral soil organic matter in our experiments, but fell within the range of 14C values measured in mineral soil roots. Together, our experiments suggest that root-derived C is the major (>60%) source of C for microbes in these temperate deciduous forest soils.  相似文献

We report the first attempt to estimate fungal biomass production in soil by correlating relative fungal growth rates (i.e., acetate incorporation into ergosterol) with fungal biomass increase (i.e., ergosterol) following amendments with dried alfalfa or barley straw in soil. The conversion factor obtained was then used in unamended soil, resulting in fungal biomass productions of 10-12 μg C g−1 soil, yielding fungal turnover times between 130 and 150 days. Using a conversion factor from alfalfa-treated soil only resulted in two times higher estimates for biomass production and consequently lower turnover times. Comparing fungal biomass production with basal respiration indicated that these calculations overestimated the former. Still, the turnover times of fungal biomass in soil were in the same range as turnover times estimated in aquatic systems. The slow turnover of fungal biomass contrasts with the short turnover times found for bacteria. The study thus presents empirical data substantiating the theoretical division of bacteria and fungi into a fast and a slow energy channel, respectively, in the soil food web.  相似文献

The low temperature pyrolysis of organic material produces biochar, a charcoal like substance. Biochar is being promoted as a soil amendment to enhance soil quality, it is also seen as a mechanism of long-term sequestration of carbon. Our experiments tested the hypothesis that biochar is inert in soil. However, we measured an increase in CO2 production from soils after biochar amendment which increased with increasing rates of biochar. The ∂13C signature of the CO2 evolved in the first several days of the incubation was the same as the ∂13C signature of the biochar, confirming that biochar contributed to the CO2 flux. This effect diminished by day 6 of the incubation suggesting that most of the biochar C is slowly decomposing. Thus, aside from this short-term mineralization increasing soil C with young biochar may indeed be a long-term C storage mechanism.  相似文献

Summary Effects of zinc (zinc chloride) added to a sandy clay loam soil on its bacterial flora were investigated by the study of cellular fatty acid composition of the isolates. The soil amended with glucose (carbon source) and ammonium sulfate (nitrogen source) was incubated in the presence (1.0 mg/g) or absence of zinc for 96 h. Bacterial strains were isolated on albumin agar from the incubated soils every 24 h. Their cellular fatty acid composition was analyzed and the strains were classified according to the percentage distribution pattern of the fatty acid composition. Two and four dominant patterns of the fatty acid composition were obtained in the isolates from the control soil and the soil with zinc added, respectively. They were different from one another. Most of the dominant strains isolated from the soil with added zinc were zinc-tolerant, and it was suggested that the selection for zinc tolerance readily proceeded.  相似文献

We used a continuous labeling method of naturally 13C-depleted CO2 in a growth chamber to test for rhizosphere effects on soil organic matter (SOM) decomposition. Two C3 plant species, soybean (Glycine max) and sunflower (Helianthus annus), were grown in two previously differently managed soils, an organically farmed soil and a soil from an annual grassland. We maintained a constant atmospheric CO2 concentration at 400±5 ppm and δ13C signature at −24.4‰ by regulating the flow of naturally 13C-depleted CO2 and CO2-free air into the growth chamber, which allowed us to separate new plant-derived CO2-C from original soil-derived CO2-C in soil respiration. Rhizosphere priming effects on SOM decomposition, i.e., differences in soil-derived CO2-C between planted and non-planted treatments, were significantly different between the two soils, but not between the two plant species. Soil-derived CO2-C efflux in the organically farmed soil increased up to 61% compared to the no-plant control, while the annual grassland soil showed a negligible increase (up to 5% increase), despite an overall larger efflux of soil-derived CO2-C and total soil C content. Differences in rhizosphere priming effects on SOM decomposition between the two soils could be largely explained by differences in plant biomass, and in particular leaf biomass, explaining 49% and 74% of the variation in primed soil C among soils and plant species, respectively. Nitrogen uptake rates by soybean and sunflower was relatively high compared to soil C respiration and associated N mineralization, while inorganic N pools were significantly depleted in the organic farm soil by the end of the experiment. Despite relatively large increases in SOM decomposition caused by rhizosphere effects in the organic farm soil, the fast-growing soybean and sunflower plants gained little extra N from the increase in SOM decomposition caused by rhizosphere effects. We conclude that rhizosphere priming effects of annual plants on SOM decomposition are largely driven by plant biomass, especially in soils of high fertility that can sustain high plant productivity.  相似文献

Organic matter dynamics and nutrient availability in saline alkaline soil of the former lake Texcoco will determine the success of a planned reforestation program. Uniformly labelled 14C-maize (MAI-treatment) and glucose (GLU-treatment) with or without 200 mg  kg−1 soil (MAI-N treatment and GLU-N treatment, respectively) were added to soils with electrolytic conductivity (EC) 56 dS m−1 (soil A) and 12 dS m−1 (soil B) to investigate the importance of N availability on decomposition of organic material. Production of CO2 and and inorganic N dynamics were monitored. The amount of 14C-glucose mineralized increased 1.8-times in the soil A, but had no effect in the soil B when 200 mg  kg−1 soil was added, while the amount of 14C-maize mineralized increased 1.7 and 1.3-times when 200  kg−1 soil was added in the soils A and B, respectively. Application of increased priming effect 3.7-times in the MAI-treatment of the soil A and 3.4-times in the GLU-treatment, while in the soil B the increase of priming effect was 4.1-times in the MAI-treatment and 3.7-times in the GLU-treatment. Of the 200 mg  kg−1 added to both soils less than 10 mg NH3-N kg−1 was volatilized within one day, while 22 and 44 mg  kg−1 soil was fixed on the soil matrix in the soil A and the soil B, respectively. Therefore more than 100 mg −N kg−1 was immobilized into the microbial biomass within the first day. Concentration of nitrite increased sharply in all the treatments of soil A at the onset of the incubation followed by a decrease. A similar pattern was observed in the GLU-N and MAI-N treatments of the soil B, but not in the other treatments. A decrease in concentration of was observed in both soils followed by an increase in the MAI-N and GLU-N treatments of the soil B. It was found that application of had a stimulating effect on the decomposition of maize and glucose, and on the priming effect, while assimilatory reduction of resulted in an increase of in the soil A, and nitrification in the soil B.  相似文献

Abstract

The feasibility of using a simple radiochemical method to determine the extent of formylation resulting from interaction of formic acid solutions with soil constituents was investigated. The interaction between formic acid‐l4C solvents and starch was used as a model reaction. Radioassay of 1 cm3 formic acid solution by liquid scintillation counting in 10 cm3 of the scintillator‐solubilizer Insta‐gel was found to be suitable. The method is simple to use and sample preparation is easy.  相似文献
