The formation of reaction and barrier zones was studied in the xylem of Eucalyptus globulus and Eucalyptus nitens tree stems after wounding and artificial inoculation with two white rot fungi. The study had two objectives: to describe host responses in Eucalyptus spp. by light microscopy and to determine whether they would differ in a fungal treatment (wounding and inoculation by one of two fungal isolates) when compared to a control treatment (wounding only). Eucalyptus globulus and E. nitens developed similar reaction and barrier zones. The E. globulus barrier zone was characterized by kino vein formation. In both hosts, the reaction zone was primarily influenced by content and distribution of living tracheids and parenchyma cells within the sapwood. By contrast, the anatomy of the barrier zone showed similarities to the basic xylem structure of each host, except for some cell types that were newly formed (sclereids, kino veins) or increased in number (parenchyma cells, tracheids). Other cell types were reduced in number or completely absent. Host response in terms of barrier zone width appeared to be greater in the fungal than control treatment. Both wood decay fungi appeared to induce a wider barrier zone in both species than that associated with non‐specific damage caused exclusively by wounding. However, the small number of replicates available for this study was possibly insufficient to provide statistical evidence for different barrier zone width between fungal and control treatments.  相似文献

Relationships between cell and pulp properties were investigated by examining the within-tree property variations in Eucalyptus camaldulensis and Eucalyptus globulus. Properties investigated included proportions of ray and axial parenchyma, thickness of cell walls and cell wall percentages. The characteristics of the ray and axial parenchyma (their proportions and wall thickness) were found to have a significant influence on all measured pulp properties, including paper strength properties. Multiple regression of pulp properties in relation to cell properties revealed that nearly all measured pulp properties were explained by cell properties at the 1% significance level. It was concluded, therefore, that all cell types are important for predicting pulp properties, and it is strongly recommended that tree breeding programs for Eucalyptus include the measurement of all cell types. Eucalyptus globulus sapwood and heartwood were delignified at 130°, 150°, or 170°C by kraft pulping. Pulp yields of heartwood were lower than those of sapwood (46.5% vs. 50.4% at 170°C). Delignification was modeled using consecutive and simultaneous kinetic models. The modeling was similar for heartwood and sapwood, and either approach could be used, with both yielding good correlations between experimental and model data. The consecutive model identified two delignification phases with similar reaction rates and activation energies for heartwood and sapwood at 150° and 170°C. At 130°C only one phase was identified. Three reactive types of lignin fractions were identified using the simultaneous model, without differences between heartwood and sapwood. Their reaction rates were 0.152, 0.138, and 0.003 min−1 at 170°C, and the activation energies were 132, 119, and 102 kJ.mol−1. The presence of heartwood did not influence the kinetic development of delignification. The negative impact of heartwood in pulping is related to the higher content of extractives (9.8% vs. 3.9% in heartwood and sapwood) and to their influence on the process, namely in the heating-totemperature phase when a substantial mass loss occurs (30% vs. 20% for heartwood and sapwood).  相似文献

Summary The kinetics of HCl-catalysed delignification of Eucalyptus globulus wood by 70% (w/w) acetic acid solution were satisfactorily explained by a model in which 5% of the lignin fraction could not be eliminated and the remaining 95% was eliminated by a single process which, in view of the activation energy calculated from the data was attributed to the hydrolysis of -aryl ether bonds. The selectivity of the process was effectively independent of HCl concentration and operating temperature for pulp yields >50%. At high temperature (160 °C) and catalyst concentration (0.027 MHCl), lignin condensation and precipitation became significant and the data were better fitted by a model comprising two consecutive processes: lignin solubilization followed by lignin condensation. The authors are grateful to the CICYT for financial support of this work (Project AGF93-0605) and also to the DGICYT for a research grant awarded to J. Non-isothermal autohydrolysis treatments of Eucalyptus globulus wood were carried out in batch reactors in order to assess the kinetics of hemicellulose degradation, as well as the effects caused on cellulose and lignin. Experiments were carried out at a liquor to wood ratio of 8 g/g, and two heating profiles were explored. The maximum temperature considered was 224 °C, and the longest reaction time was 0.94 h. The experimental variables considered were solid residue yield, solid residue composition and composition of liquors. Up to 90.4% of the initial xylan was removed in treatments, which also caused some delignification (up to 13.8% of the initial lignin was removed), whereas cellulose was almost quantitatively retained in solid phase. The conversion of xylan into xylooligomers, with further hydrolysis to xylose and decomposition of this sugar into furfural was assessed by means of a kinetic model based on sequential, first order reactions involving kinetic coefficients with Arrhenius-type dependence on temperature. Previous research has shown that carbon isotope signatures (Δ13C) may correlate well with water status in 1–2 years old, plantation grown Eucalyptus globulus. Here we investigate this relationship for mid-rotation aged (5 years) trees and whether a simple field determination of phloem sugar concentrations is a reliable surrogate for Δ13C. We sampled six plantations covering much of the climatic range of E. globulus plantations in Australia (average annual rainfall 618–1094 mm). We found significant correlations between phloem Δ13C and phloem sugar concentrations across and within plantations. Both parameters were also correlated with a climate wetness index. We thus conclude that phloem sugar concentration is a useful surrogate for Δ13C and hence water status of mid-rotation E. globulus. We also observed strong correlations of phloem Δ13C and phloem sugar concentrations with growth across all plantations, providing some confirmation that water limits growth of E. globulus at a larger scale. This relationship was much weaker or absent within plantations, suggesting that factors other than water supply contribute significantly to growth limitations at smaller (within-plantation) scales.  相似文献

Mycosphaerella leaf disease (MLD) can cause significant discoloration, necrosis and defoliation in commercial plantations of young Eucalyptus globulus. This study investigated the spectral reflectance changes that result in E. globulus foliage from Mycosphaerella infection. Spectral reflectance measurements were made on healthy and infected E. globulus foliage in the visible and near‐infrared wavelengths (400–1000 nm). Reflectance (R) at 760 nm produced the largest difference between the extremes of disease severity classes. The wavelength most sensitive to infection severity was R678 nm. Other sensitive wavelengths were located between 480 and 500 nm. The wavelengths near 708 and 550 nm appeared relatively insensitive to infection severity. The reflectance index best correlated to leaf infection was R678/R550 (r = 0.841, p < 0.0001). This index was used in a linear regression model for successfully predicting leaf infection levels from an independent data set (r = 0.95, p < 0.0001).  相似文献

The genus Mycosphaerella Johanson contains many pathogens capable of causing a severe impact on the growth of susceptible eucalypt species. The lack of knowledge about which species are present in Tasmania and their potential risk to the plantation industry prompted this study into the Mycosphaerella species occurring on Eucalyptus globulus and Eucalyptus nitens plantations in Tasmania. A total of 36 plantation and five road verge sites of E. globulus and E. nitens were sampled. Five Mycosphaerella species and three species from associated anamorph genera were isolated and identified in Tasmania; Mycosphaerella nubilosa, Mycosphaerella cryptica, Mycosphaerella tasmaniensis, Mycosphaerella grandis, Mycosphaerella vespa, Coniothyrium ovatum, Sonderhenia eucalypticola and Sonderhenia eucalyptorum. The most frequently isolated species with the highest incidence and severity of infection were M. cryptica and M. nubilosa. These two species appear to have the greatest potential to damage juvenile eucalypt plantations in Tasmania. A link between Mycosphaerella vespa and Coniothyrium ovatum is described for the first time.  相似文献

Resistance of Eucalyptus globulus juvenile foliage to Teratosphaeria leaf disease (TLD) damage has been shown to be under genetic control. Constitutive traits of juvenile leaves such as stomatal density (counted with wax on and with wax removed), total phenolics, total leaf wax, leaf mass per area (LMA) and leaf anatomical features were assessed to determine differences between pairs of resistant and susceptible families. Anatomic features assessed included cuticle, epidermis, palisade spongy mesophyll and total leaf thickness, as well as palisade cell size, shape and intercellular airspace. One interprovenance, one intraprovenance and two within‐family contrasts were used to compare resistant and susceptible pairs of families of Australian origin. The more resistant families had a significantly higher LMA and smaller percentage airspace compared with susceptible families. It is argued that one of the mechanisms by which E. globulus resists TLD involves the closely packed palisade mesophyll cells that increases LMA and reduces airspace, thereby restricting pathogen spread once infection has occurred.  相似文献

The responses of juvenile leaves of two Eucalyptus species, with contrasting susceptibility to infection by Mycosphaerella leaf disease, were compared. The anatomical changes, accumulation of phenolics, suberin, lignin and anthocyanin and the retention of chlorophyll were studied in leaf lesions of varying developmental stages caused by species of Mycosphaerella. Enhanced resistance of Eucalyptus nitens in southern Australia was attributed to the formation of an effective lignified and suberized necrophylactic periderm, to restrict pathogen spread. Leaves of E. nitens contained isobilateral palisade which resulted in both abaxial and adaxial cell division and the initiation of a strong reinforced cellular zone from an early lesion stage. Eucalyptus globulus formed a slower, distorted necrophylactic periderm through hypertrophic changes to existing mesophyll and limited cell divisions of the single adaxial palisade layer. Deposits of lignin and suberin did not occur until later in lesion development, which were not effective in preventing further disease development. From this study it is hypothesized that tolerance of eucalypts to Mycosphaerella pathogens may be associated with constitutive mesophyll density.  相似文献

Eucalyptus globulus is the main eucalypt species grown in Australian plantations. The focus on seedling deployment systems, coupled with exploitation of large, open-pollinated base populations for breeding purposes over the last two decades, has required a detailed understanding of the reproductive biology of this species. We review our research on the reproductive biology of E. globulus, with a focus on its breeding system and advances made in seed production systems. While most improved seed is still obtained from open-pollinated seedling or grafted seed orchards, the development of the one-stop/single-visit pollination procedure has revolutionised the breeding and deployment of this species. The reduced costs of controlled pollination has meant full pedigree-control can now be maintained in large advanced-generation breeding populations and E. globulus is one of the few eucalypt species where large-scale production of manually pollinated seed for family forestry is being undertaken.  相似文献

Eucalyptus globulus wood was subjected first to HCl–catalysed delignification with 70% acetic acid under conditions realizing an incomplete 3 × 3 × 3 factorial design (HCl concentration 0, 0.025 or 0.05%; temperature 120, 140 or 160 °C; reaction time 1, 2.5 or 4 h), and then to enzymatic hydrolysis. The hydrolysis kinetics conformed to both Ghose's empirical model and a biexponential equation. The biexponential fit implies the presence of both readily and reluctantly hydrolysed cellulose fractions, and the fitted coefficients show hydrolysis yield to depend largely on the digestibility of the latter. Multiple regression of performance variables on pulping conditions showed that neither the rate nor the extent of hydrolysis is greatest for pulps with minimum lignin or xylose contents; we attribute this circumstance to the condensation and precipitation of lignin under severe pulping conditions, which protects the cellulose of the pulp from enzymatic attack. There is a need to calibrate models for carbon accounting in forest systems if they are to be applied for carbon trading and off-set schemes. One such model, Full Carbon Accounting Model (FullCAM), calculates stem mass by taking annual inputs of tree growth in stem volume and multiplying these by basic stem wood density. Stem mass is then multiplied by user-entered coefficients to determine the mass of other tree components. Using datasets of Pinus radiata and Eucalyptus globulus that comprised of between 73 and 187 observations, we determined empirical relationships that can be used in FullCAM to relate basic stem wood density to stand age, and masses of bark, foliage or branches to mass of stem wood for these two species. All fitted relationships were highly significant (p < 0.001), explaining between 35 and 89% of the variance. These calibrations were then tested using three case studies where data on volume yield curves and repeated measures of biomass of stand components were available: one of P. radiata and two of E. globulus. Although accumulation of biomass in foliage and branches were not well predicted by the model, sensitivity analysis showed that this was relatively unimportant to total carbon storage because of the dominance of the stem, particularly once the stand is older than 5 years. Indeed, FullCAM accounted for 99% of the variance in measured above-ground biomass at all three sites because calibrations for the mass of stem was reasonably well constrained. Uncertainty analysis showed that despite the standard errors of parameters used in relationships for basic density and biomass partitioning, and for estimates of carbon contents of tree components, we can be 95% confident that sequestration of carbon in trees and debris of Pinus radiata and Eucalyptus globulus plantations are, on average, within 13% of that predicted by FullCAM. Teratosphaeria gauchensis and T. zuluensis both cause a stem canker of eucalypts that leads to serious damage in various parts of the world. Until recently, this disease was unknown in Portugal. Nevertheless, severe damage to Eucalyptus globulus has been observed in Portugal since 2006 when symptoms appeared as necrotic spots on young green stems and leaves, twig lesions, and dark oval‐shaped lesions on stems and trunks. The isolates from affected E. globulus tree plantations were identified using sequence data of the ITS1‐5.8S‐ITS2 and EF‐1α clusters, together with morphological characteristics. Based on these results, the causal organism was identified as T. gauchensis, which represents the first report of this pathogen from Portugal.  相似文献

Accurately and non-destructively quantifying the volume, mass or nutrient content of tree components is fundamental for assessing the impact of site, treatment, and climate on biomass, carbon sequestration, and nutrient uptake of a growing plantation. Typically, this has involved the application of allometric equations utilising diameter and height, but for accurate results, these equations are often specific to species, site, and silvicultural treatment. In this study, we assessed the value of incorporating a third piece of information: the height of diameter measurement. We derived a more general volume equation, based on the conical approximation, using a diameter projected to the base of the tree. Common equations were developed which allowed an accurate estimate of stem volume, dry weight and nutrient content across two key plantation grown eucalypt species, Eucalyptus grandis W. Hill ex Maiden and Eucalyptus globulus (Labill.). The conical model was developed with plantation-grown E. grandis trees ranging from 0.28 to 15.85 m in height (1.05 g to 80.3 kg stem wood dry weight), and E. globulus trees ranging from 0.10 to 34.4 m in height (stem wood dry weight from 0.48 g to 652 kg), grown under a range of contrasting cultural treatments, including spacing (E. grandis), site (E. globulus) and fertilization (nitrogen and phosphorus) for both species. With log transformed data the conical function (Vcon) was closely related to stem sectional volume over bark and stem weight (R2 = 0.996 and 0.990, respectively) for both E. grandis and E. globulus, and the same regressions can be applied to both species. Back transformed data compared with the original data yielded modelling efficiencies of 0.99 and 0.97, respectively. Relationships between Vcon and bark dry weight differed for the two species, reflecting differing bark characteristics. Young trees with juvenile foliage had a different form of relationship to older trees with intermediate or adult foliage, the change of slope corresponding to heights about 1.5 m for E. grandis and age 1 year for E. globulus. The Vcon model proved to be robust, and unlike conventional models, does not need additional parameters for estimating biomass under different cultural treatments. More than 99% of the statistical variance of the logarithm of biomass was accounted for in the model. Vcon captures most of the change in stem taper associated with cultural treatments and some of the change in stem form that occurs after the crown base has lifted appreciably. Fertilization increased N and P concentrations in stem wood and bark, and regressions to estimate N and P contents (the products of biomass and concentration) were dependent on treatment. For instance, there was a large growth response to N fertilization in E. globulus corresponding with a change (P < 0.05) in the intercept of the regression to estimate N content.  相似文献

The pathogenicity, physiology and biochemical features of three isolates of Phytophthora. nicotianae var. nicotianae from Eucalyptus delegatensis and E. globulus seedlings were investigated. The acid phosphatase zymograms and total protein profiles confirmed the specific identity of the 3 isolates. Despite significant differences in mycelial growth and zoosporangium production, no differences were noted in pathogenicity. Artificial inoculations revealed a variability in resistance to P. nicotianae within Eucalyptus species.  相似文献

• Introduction

Tree genetic improvement programs usually lack, in general, pedigree information. Since molecular markers can be used to estimate the level of genetic similarity between individuals, we genotyped a sample of a Portuguese Eucalyptus globulus breeding population—a reference population of 125 individuals—with 16 microsatellites (SSR).  相似文献

A little-leaf disease of Eucalyptus tereticornis, E. grandis and E. globulus characterized by stunting of plants and considerable reduction in size of leaves and internodes, was recorded during survey of nurseries and plantations in Kerala State. The survey indicated that though the little leaf disease was widespread, its incidence was quite low. Transmission of the symptoms by sap and graft techniques was unsuccessful. However, positive fluorescence and staining of phloem tissues by Dienes' stain indicates that this disease may be caused by mycoplasmalike organisms (MLO).  相似文献
