What we know about the life history of fungi that cause disease in plants is commonly based on studies of the pathogen’s interaction with a susceptible host: how and when infection occurs, growth and reproduction within the host, and survival during the interval when a growing host is not available. This focus is appropriate, given the need for information that will facilitate management of disease affecting an economically important crop, but it can limit recognition of the full range of resources that may be utilized by fungi that we classify as plant pathogens. This was certainly the case for Fusarium circinatum, which causes a destructive disease of pines known as pitch canker. Although F. circinatum was initially known only as a necrotrophic, wound-infecting pathogen of coniferous trees, recent research has revealed that an isolate of this fungus that will kill shoot tissue when inoculated into a wound can also have a biotrophic relationship with roots of pine seedlings, infect and grow within grasses without causing symptoms, and cause ear rot of corn. Thus, although F. circinatum became known to science because it induced visible symptoms on pines, it has the capacity for a much broader range of ecological activities than is captured by its designation as a necrotrophic pathogen. The physiological plasticity manifested by F. circinatum illustrates the challenge of obtaining a comprehensive understanding of the life history of a plant pathogenic fungus.  相似文献

Pitch canker of pines is caused by the fungus Fusarium circinatum. In South Africa, this pathogen has mostly been a nursery problem. From 2005, however, outbreaks of pitch canker have been reported from established Pinus radiata and P. greggii in the Western and Eastern Cape Provinces. Most recently, pitch canker-like symptoms were observed on 10-year-old P. greggii trees in a plantation in the midlands of the KwaZulu-Natal (KZN) Province. The aim of this study was to: (i) identify the causal agent of the observed symptoms, (ii) determine the genetic diversity, and (iii) the mode of reproduction of this fungal population. Furthermore, the aggressiveness of isolates from these trees was compared with that of isolates obtained previously from P. patula in South Africa. Isolates from the P. greggii trees in KZN were confirmed as F. circinatum based on both morphology and DNA sequence analyses. Microsatellite marker analyses revealed the presence of five genotypes of F. circinatum, not previously reported from other plantations in South Africa, with one of these genotypes being dominant. These genotypes were all pathogenic to P. patula and P. elliottii. No evidence of sexual reproduction was detected in the KZN population of the fungus. This was consistent with the fact that isolates from P. greggii were all of the MAT-2 mating type, in contrast to previously collected isolates from across South Africa that included both mating types. The results suggest that the outbreak of pitch canker on P. greggii in KZN represents a separate introduction of F. circinatum into the region with important implications for managing the disease.  相似文献

The oomycete Phytophthora infestans is the causal agent of potato late blight, one of the most destructive and historically significant pathogens in agricultural production. A virus-induced gene silencing-based screening of the solanaceous model plant N. benthamiana resulted in revealing a wide range of resistance mechanisms of solanaceous plants against this pathogen. In this article, we present an overview of the various pathways involved in the N. benthamianaP. infestans pathosystem, including some of the follow-up work that was triggered by these findings. The purpose of this review is to assemble these findings and integrate them into our current understanding of plant pathogen defense mechanisms and discuss their potential application for the development of potato resistance to P. infestans.  相似文献

Fungal and oomycete communities in symptomatic roots, crowns and lower stem tissues of field pea plants from sites across the Canadian prairies were characterized using plate culture and sequence-based identification. Symptomatic plants were sampled at the mid-flowering stage from a total of 21 fields in 2014 and 2015. Fungi and oomycete species were isolated on agar medium and grouped into operational taxonomic groups (OTU) based on their morphology. From the OTUs, 40 fungal and oomycete species were identified according to translation elongation factor-1 alpha or ITS sequences. Fusarium spp. accounted for more than 60 % of total isolations. The fungal communities were similar in roots and crowns, but differed from communities in lower stems. In 2014, the most prevalent fungi isolated from roots were F. avenaceum, F. solani and F. redolens, but F. solani was dominant in 2015. In crown tissues, F. avenaceum was most prevalent in 2014 but F. solani was more prevalent in 2015. Stem tissues were primarily colonized by Peyronellaea spp. (= Ascochyta / Phoma spp.) and Alternaria spp. in 2014, but there was a drastic decrease in Peyronellaea spp. in 2015. This study indicated that climate, primarily precipitation levels, and local edaphic characteristics may play a profound role in the structure of phytopathogen communities associated with the root and foot rot diseases complex of field pea on the Canadian prairies.  相似文献

The plant pathogenic oomycete, Phytophthora infestans, is the causal agent of late blight disease in tomato and potato. For characterizing Egyptian P. infestans isolates by DNA marker analysis, 40 isolates of P. infestans were collected from different locations in Egypt during two growing seasons (2012/2013 and 2013/2014). The 40 isolates were grouped into seven genotypes, in which 24 alleles were detected. The identified genotypes were not completely associated with geographic location and sample collection years. These results provide genetic and geographical information for developing a program to manage late blight disease.  相似文献

In this investigation, leaf extracts of Ocimum basilicum and Mangifira indica were used as reducing agents for biosynthesis of silver nanoparticles (AgNPs). The biosynthesized AgNPs were authorized by UV-vis spectrophotometry and X?ray diffraction (XRD) analysis. AgNPs were obtained after 5?min of reaction at 80oC. The formation of AgNPs was confirmed by the presence of absorption peaks at 439?nm using extract from O. basilicum and at 442.5?nm from M. indica. X?ray spectra showed strong peaks for the crystalline Ag. Shape and size of the biosynthesized AgNPs were studied using high resolution transmission electron microscope (HR-TEM). Size of the produced AgNPs was found to be 9–35?nm. Effect of the synthesized nanosilver was then investigated on some biochemical attributes of wheat plant (Triticum aestivum cultivar saka 92). The growth parameters such as shoot lengths, fresh and dry weight of shoot, chlorophyll, carbohydrate and protein contents in shoot of wheat plant were investigated. Application of AgNPs synthesized from Ocimum basilicum and from Mangifira indica at the concentrations of 20,40?ppm showed an increase in shoot length, fresh and dry weight of shoot, chlorophyll, total carbohydrate and protein content in shoot of wheat plants, beyond these concentrations an inhibitory effects were shown.  相似文献

Wood-staining fungi, moulds and decay fungi colonize freshly cut wood. Of these, only ophiostomatoid fungi are considered major agents of sapstain on logs and freshly sawn lumber because they can cause permanent staining and discoloration. Sapstain in pine pallets significantly reduces their market value and use in the food industry. The aim of this study was to identify the sapstaining fungi that colonize Scots pine wood used in pallet production. In addition, we evaluated the growth and stain intensity of six isolates of ophiostomatoid fungi on freshly cut Scots pine billets. Fungi were isolated from samples of Scots pine and identified based on morphology and DNA sequence comparisons for three gene regions (ITS, β-tubulin, TEF-1α). A total of 1259 isolates representing 31 fungal species were obtained from the pine samples in Poland during July and September. The isolates represented different ecological and taxonomical groups and belonged to the categories of staining fungi, decay fungi and surface moulds. The most frequently isolated fungi were ophiostomatoid fungi (14 species including an unknown Leptographium sp. and Ophiostoma sp.) and moulds (mainly Trichoderma and Mucor spp.). Of the ophiostomatoid species, Ophiostoma floccosum, O. piceae, O. piliferum and Endoconidiophora pinicola were the predominant species. The results of pine billet inoculation showed that among the six fungal species tested, E. pinicola exhibited the fastest growth in all three directions on the billets. Ophiostoma minus and O. piliferum displayed moderate growth rates in pine billets, while O. floccosum, O. piceae, and Leptographium lundbergii grew very slowly, especially in the tangential and radial directions. The information provided in this paper will help develop more effective control strategies for sapstain prevention in Scots pine.  相似文献

The antifungal activity of fifteen wild plant species grown in the Al-Qassim region, Saudi Arabia, was investigated against the fungi species Fusarium solani, Botrytis cinerea, Alternaria alternata and Stemphylium botryosum at concentrations of 10, 20, 40 and 80?g/L. The inhibitory effect of test extracts varied among examined fungi, and Fusarium solani exhibited the least sensitivity compared to other fungi species. Antifungal activity of the tested extracts was proportional to the applied dose. Extracts of Lactuca virosa, Neurada procumbens, Ochradenus baccatus and Cyperus conglomerates showed relatively low inhibition effects on the fungi species. The most effective plants were Pulicaria undulata, Artemisia monosperma, Prosopis juliflora, Withania somnifera and Rumex vesicarius. At 80?g/L, extracts of these plant species reduced mycelial growth of Botrytis cinerea by 16.5–32.2%, of Fusarium solani by 11.1–27.9%, of Alternaria alternaae by 26.9–63.5% and of Stemphylium botryosum by 22–40%. The methanolic extract of the most effective plant species was further fractioned with hexane, methylene chloride and ethyl acetate. The obtained fractions varied in their effects on mycelial growth of the four tested fungi. Using the same fraction resulted in different inhibition effects on mycelial growth of all tested fungi. The antifungal activity of each crude extract tended to be distributed among its three fractions, probably because the bioactive components were also distributed among the fractions.  相似文献

In September 2014, Phytophthora rot on wasabi plants [Wasabia japonica (Miq.) Matsum.] was found for the first time in the city of Okutama, Tokyo, Japan. A Phytophthora sp. strain was constantly isolated from brown stem bases and rhizomes of infected plants. The same symptoms as those observed in the field were produced in vitro through inoculation of test plants with the isolated Phytophthora sp. The fungus was identified as Phytophthora drechsleri based on morphological and DNA sequence comparison. Phytophthora rot, “eki-byo” in Japanese, is proposed for this disease common name.  相似文献

RNA silencing pathways in filamentous fungi are composed of multiple component proteins and known to be involved in vegetative growth, virulence or sexual reproduction. We found that the tomato wilt fungus, Fusarium oxysporum f. sp. lycopersici (Fol), carries four homologues genes of Qde-2, an argonaute protein gene and one of the main component protein genes in Neurospora crassa. Gene targeting revealed that FoQde-2, one of the Qde-2 homologues in Fol, is involved in virulence to tomato but not in vegetative growth.  相似文献

In previous research, concentrated metabolites produced by bacteria of the genera Xenorhabdus and Photorhabdus (which are symbionts of entomopathogenic nematodes) were reported to be highly suppressive to fungal and oomycete plant pathogens. Conceivably, application of non-concentrated bacterial filtrates would be more economically feasible compared to using concentrated metabolites. We evaluated the potency of 10 % v/v cell-free supernatants of the bacteria X. bovienii, X. nematophila, X. cabanillasii, X. szentirmaii, P. temperata, P. luminescens (VS) and P. luminescens (K22) against Fusicladium carpophilum (peach scab), F. effusum (pecan scab), Monilinia fructicola (brown rot), Glomerella cingulata (anthracnose) and Armillaria tabescens (root rot). A bioactive compound derived from Photorhabdus bacteria, trans-cinnamic acid (TCA), was also compared with the bacterial filtrates. Fungal colony size based on manual measurements was compared for accuracy to measurements taken by image analysis. Supernatants of Xenorhabdus spp. exhibited stronger suppressive effects on spore germination and vegetative growth when compared with Photorhabdus spp. Overall, TCA was the most effective treatment; vegetative growth was completely inhibited by TCA (1.27 mg/ml). TCA treatments also suppressed spore germination of F. carpophylium and F. effussum by approximately 90 %. The efficacy of supernatants varied among Xenorhabdus species depending on the species tested, but X. szentirmaii filtrates tended to cause greater inhibition relative to the other bacteria supernatants. Manual measurement of colony diameter required at least two replicate estimates of the colony to avoid a type II error. Area measurements were slightly overestimated based on ruler measurements, but did not affect the outcome of the analysis. Supernatants of Xenorhabdus spp., Photorhabdus spp., or TCA, did not cause any phytotoxic effects when applied to various plant species in the greenhouse. Our results indicate the potential of using TCA or Xenorhabdus cell free supernatants as bio-fungicides. Such a product, based on bacterial culture supernatants, would be economically viable, marketable and easily applicable by the end-users in many situations.  相似文献

Rubber wood, one of the most preferred plantation timbers in the tropical regions, is easily prone to various moulds, decay and stain fungi. Lasiodiplodia theobromae, the dominant sapstain fungus of rubber wood, causes high economic loss to the industry through its bluish black sap wood discolouration. In our previous laboratory trials, Bacillus subtilis B1 isolated from market available compost was proved as a potent biocontrol agent against the sapstain fungus L. theobromae. The main aim of the present study was to demonstrate the biocontrol potential of B. subtilis B1 against rubber wood sapstain fungus on fresh rubber wood pieces during pre-monsoon, southwest monsoon and northeast monsoon seasons in the field. The visual ratings provided for the wood fungal growth and internal wood stain exhibited the bio-protectant efficiency of B. subtilis B1 even in the southwest monsoon season, which is considered as the most favourable period for the wood fungal growth. Biocontrol agent, B. subtilis B1 demonstrated significant inhibitions of sapstain fungus on open stacked rubber wood pieces in all the seasons. The frequent rainfalls during monsoon periods provide a conducive environment for fungal growth on felled rubber wood. Successful field evaluations of the biocontrol agent during pre-monsoon, monsoon and post-monsoon periods would guarantee year round protection against rubber wood sapstain. B. subtilis B1 displayed holistic biocontrol efficiency towards other stain (Ceratocystis sp., Fusarium sp.), mould (Aspergillus sp., Penicllium sp.) and decay fungi (Trametes versicolor) as well.  相似文献

Phytophthora nicotianae is one of the most important soil-borne plant pathogens. Sporangia and zoospores of P. nicotianae are responsible for primary infection and disease dissemination. The disease caused by P. nicotianae was difficult to control by fungicide. Boron, an essential plant micronutrient, was found to have a direct effect on other pathogens. In this paper, the effects of B on the growth, antioxidant system and gene differential expression of P. nicotianae were tested. The results showed that 0.1 mM B could dramatically decrease the sporangiogenesis and zoosporogenesis of P. nicotianae. Mycelial growth of P. nicotianae was significantly inhibited when the concentration of B reached 8 mM. A high-quality differential expression sequence csn4 was obtained by gene differential expression analysis. Under the treated of B, csn4 expression was inhibited, activity of superoxide dismutase (SOD) and catalase (CAT) significantly decreased and the malondialdehyde (MDA) content notably increased compared to control. It is suggested that B could serve as a potential fungicide for the control of plant disease caused by P. nicotianae.  相似文献

RNA-based disease control has shown great potential for controlling pest and diseases in crop plants. While delivery of inhibitory noncoding double-stranded (ds)RNA by transgenic expression is a promising concept, it requires the generation of transgenic crop plants, which may cause substantial delay for application strategies depending on the transformability and genetic stability of the crop plant species. Focusing on agronomic important barley - Fusarium spec. pathosystems, we have sought for alternative strategies to apply dsRNAs for fungal control. Recently, we have demonstrated that a spray application of a long noncoding dsRNA termed CYP3RNA, which targets the three fungal Cytochrome P450 lanosterol C-14α-demethylase genes FgCYP51A, FgCYP51B, and FgCYP51C, inhibits Fusarium graminearum (Fg) on barley leaves (Koch et al., PLoS Pathogens, 12, e1005901, 2016). Here we show that another Fusarium species, F. culmorum (Fc), also is sensitive to CYP51-derived dsRNAs. Treating Fc with various dsRNAs targeting the genes FcCYP51A, FcCYP51B and FcCYP51C was destructive to the fungus and resulted in growth retardation in in vitro cultures. We discuss important consequences of this finding on future RNA-based disease control strategies. Given the ease of design, high specificity, and applicability to diverse pathogens, the use of target-specific dsRNA as an anti-fungal agent offers unprecedented potential for novel plant protection strategies.  相似文献

The purpose of this study was to determine if exogenous cholesterol availability influenced Pythiaceae resistance to antibiosis. Characterisation of an isolate of Phytophthora erythroseptica and Pythium ultimum for tolerance to antibacterial compounds found that 0.05 g.l?1 chloramphenicol inhibited mycelial growth by 96.6 % and 23.5 % respectively. However, the addition of cholesterol (0.01 g l?1) to potato dextrose agar (PDA) containing 0.05 g l?1 chloramphenicol was found to increase mycelial growth of P. erythroseptica, indicating a role for cholesterol in tolerance to inhibitory antibacterial compounds. To determine if this property extended to suppressive effects of a potential biocontrol agent, P. erythroseptica and P. ultimum were then tested against a cell-free filtrate of diffusible metabolites produced by a suppressive Trichoderma harzianum isolate in the presence and absence of cholesterol in PDA. In the absence of cholesterol, diffusible metabolites of the T. harzianum isolate were found to inhibit mycelial growth of P. erythroseptica and P. ultimum on PDA by 98 % and 63.6 % respectively (P?<?0.0001). However, the inhibitory effect of the metabolites was mitigated when 0.005 g l?1 of cholesterol was present in PDA, with mycelial growth of P. ultimum and P. erythroseptica reduced by only 60.4 % and 41.8 %, respectively (P?<?0.0001), much less inhibition than was observed in the absence of cholesterol. These results demonstrated that access to exogenous cholesterol can influence the sensitivity of Pythiaceae species to antibiosis by positively influencing mycelial growth.  相似文献

The fungal community on the roots, stem bases, stems and grains of organically grown winter wheat was analysed using terminal restriction fragment length polymorphism (T-RFLP) combined with cloning and sequencing of the ITS region. The changes in the composition of fungi in different plant parts and over time as well as interactions between fungi were also investigated. Among 58 fungal taxa found the most common were Davidiella macrospora, Cladosporium spp., Tetracladium maxilliforme, Didymella exitialis, Microdochium nivale and an unidentified species within Ascomycetes. Several potential wheat pathogens were found: Fusarium spp. including F. poae and G. avenacea (F. avenaceum), Microdochium nivale, Oculimacula yallundae, Parastagonospora nodorum and Zymoseptoria tritici and most of them were present on all plant parts. Plant part affected the most the fungal colonization of wheat as was shown both by multivariate analysis of the whole fungal community as well as the analysis based on the identified species. The composition of fungal communities in different parts changed during the growing season but no pattern common for the whole crop could be observed. The most dynamic and significant changes were found among yeasts. Both positive and negative significant interactions between pairwise combinations of pathogens were observed. Positive significant associations were also found between pathogens and other fungi.  相似文献

The virulence of 29 isolates of Phytophthora infestans collected in potato fields in Hokkaido, Japan, in 2013 and 2014, was tested for race identification. Thirteen different races were identified, each of which had five to eight virulence factors. All of the isolates caused a virulent reaction against plants with R1 and R7, and most of the isolates caused a virulent reaction against plants with R3, R4, R10, and R11. On the other hand, no isolate was virulent against plants with R9. These results demonstrate that the current Japanese P. infestans population is more complex than the population in the 1990s from the viewpoint of race.  相似文献
