To evaluate the exposure of Koreans to ochratoxin A, we conducted a survey in 2003 for ochratoxin A in various domestic food commodities: 60 polished rices, 22 barleys, 35 wheat flours, 46 beers, and 14 unstrained rice wine (makkolli) samples. They were analyzed for ochratoxin A using immunoaffinity column and high-performance liquid chromatography (HPLC)-fluorescence detection, and the positive samples were confirmed using HPLC-tandem mass spectrometry. By combining results from different surveys on the levels of ochratoxin A in selected foods and the consumption patterns, we obtained the Korean probable daily intakes (PDI) of ochratoxin A. The polished rice commodity had the highest mean levels of ochratoxin A, which ranged from 0.2 (not detected, i.e., ND = 0) to 1.0 ng/g (ND = limit of detection, i.e., LOD). The estimated PDI for all Koreans fell into the range of 0.8-4.1 ng/kg bw/day, while for heavy consumers the estimates ranged from 1.7 to 9.1 ng/kg bw/day, which did not exceed the PTDI value (14 ng/kg bw/day). Staple rice is the major contributor (>90%) to the Korean dietary intake of ochratoxin A. On the basis of these estimates, it may be concluded that there is at present no considerable risk of ochratoxin A exposure for the average Korean consumer.  相似文献

This study presents concentrations of perfluorinated compounds in food and the dietary intake of perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA) in The Netherlands. The concentrations of perfluorinated compounds in food were analyzed in pooled samples of foodstuffs randomly purchased in several Dutch retail store chains with nation-wide coverage. The concentrations analyzed for PFOS and PFOA were used to assess the exposure to these compounds in The Netherlands. As concentrations in drinking water in The Netherlands were missing for these compounds, conservative default concentrations of 7 pg/g for PFOS and 9 pg/g for PFOA, as reported by European Food Safety Authority, were used in the exposure assessment. In food, 6 out of 14 analyzed perfluorinated compounds could be quantified in the majority of the food categories (perfluoroheptanoic acid (PFHpA), PFOA, perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluoro-1-hexanesulfonate (PFHxS), and PFOS). The highest concentration of the sum of these six compounds was found in crustaceans (825 pg/g product, PFOS: 582 pg/g product) and in lean fish (481 pg/g product, PFOS: 308 pg/g product). Lower concentrations were found in beef, fatty fish, flour, butter, eggs, and cheese (concentrations between 20 and 100 pg/g product; PFOS, 29-82 pg/g product) and milk, pork, bakery products, chicken, vegetable, and industrial oils (concentration lower than 10 pg/g product; PFOS not detected). The median long-term intake for PFOS was 0.3 ng/kg bw/day and for PFOA 0.2 ng/kg bw/day. The corresponding high level intakes (99th percentile) were 0.6 and 0.5 ng/kg bw/day, respectively. These intakes were well below the tolerable daily intake values of both compounds (PFOS, 150 ng/kg bw/day; PFOA, 1500 ng/kg bw/day). The intake calculations quantified the contribution of drinking water to the PFOS and PFOA intake in The Netherlands. Important contributors of PFOA intake were vegetables/fruit and flour. Milk, beef, and lean fish were important contributors of PFOS intake.  相似文献

Despite the growing public interest in perfluorinated compounds (PFCs), very few studies have reported the sources and pathways of human exposure to these compounds in China. In this study, concentrations of 10 PFCs were measured in human blood, water (tap water and surface water), freshwater fish, and seafood samples collected from China. On the basis of the data, we calculated daily intakes of PFCs, regional differences in human exposures, and potential risks associated with ingestion of PFCs from diet, drinking water, and indoor dust for the Chinese population. Perfluorooctane sulfonate (PFOS) was the most predominant PFC found with a mean concentration of 12.5 ng/mL in human blood from Tianjin and 0.92 ng/g wet wt in freshwater fish and seafood; perfluorooctanoic acid (PFOA) was the major PFC found in drinking water at a concentration range of 0.10 to 0.92 ng/L. The estimated daily intake of PFOS and PFOA via fish and seafood consumption (EDI(fish&seafood)) ranged from 0.10 to 2.51 and 0.13 to 0.38 ng/kg bw/day, respectively, for different age groups (i.e., toddlers, adolescents and children, and adults) from selected locations (i.e., Tianjin, Nanchang, Wuhan, and Shenyang). The EDI(fish&seafood) of PFCs decreased (p < 0.05) with age. The estimated daily intake of PFOS and PFOA via drinking water consumption (EDI(drinking water)) ranged from 0.006 to 0.014 and 0.010 to 0.159 ng/kg bw/day, respectively. Comparison of EDI(fish&seafood) and EDI(drinking water) values with those of the modeled total dietary intake (TDI) of PFCs by adults from Tianjin, Nanchang, Wuhan, and Shenyang showed that contributions of fish and seafood to TDI of PFOS varied depending on the location. Fish and seafood accounted for 7%, 24%, 80%, and 84% of PFOS intake in Nanchang, Shenyang, Wuhan, and Tianjin, respectively, suggesting regional differences in human exposure to PFOS. Drinking water was a minor source of PFOS (<1%) exposure in adults from all the study locations.  相似文献

A comparative study on the natural occurrence of aflatoxins and Fusarium toxins was conducted with corn samples from high- and low-incidence areas for human primary hepatocellular carcinoma (PHC) in Guangxi, China. In samples from the high-risk area, aflatoxin B(1) was the predominant toxin detected in terms of quantity and frequency, with its concentration ranging between 9 and 2496 microg/kg and an 85% incidence of contamination. Among the samples, 13 (76%) exceeded the Chinese regulation of 20 microg/kg for aflatoxin B(1) in corn and corn-based products intended for human consumption. Significant differences in aflatoxin B(1), B(2), and G(1) and total aflatoxin concentrations in corn between the areas were found (P < 0.05). The average daily intake of aflatoxin B(1) from corn in the high-risk area was 184.1 microg, and the probable daily intake is estimated to be 3.68 microg/kg of body weight/day, 3.20 times the TD(50) in rats. Corn samples from both areas were simultaneously contaminated with fumonisins B(1), B(2), and B(3). Aflatoxin B(1) may play an important role in the development of PHC in Guangxi.  相似文献

Inorganic arsenic (iAs) is considered to be a human carcinogen. In this paper, total (As) and iAs contents of 215 food products and drinks (i.e., seafood, fruits and vegetables, meat products, oils and fats, rice and rice products, seasonings, and alcoholic drinks) marketed in Catalonia (Spain) were quantified by inductively coupled plasma-mass spectrometry. The analytical method described was used for different food products, obtaining feasible results without the need to couple LC-ICP-MS for iAs. Daily As and iAs intakes for the average adult Catalan consumer were estimated at 354 and 6.1 μg/day/person, respectively, using consumption data from the Catalan Nutrition Survey (ENCAT). The highest As content was found in seafood, contributing 96% of dietary As intake, whereas rice presented the highest iAs values, corresponding to 67% of dietary iAs intake. As cooking process may affect iAs content, boiled rice was evaluated, showing an iAs reduction (up to 86%) when using higher water volumes (30:1 water/rice ratio) than those used in previous studies. This iAs exposure was slightly below the exposure risk range stated by the European Food Safety Authority (0.3-8 μg/kg of body weight/day), although the possibility of a risk to the population with high rice consumption cannot be excluded.  相似文献

Aflatoxicol (AFL) and aflatoxins B1 and M1 were found in tissues (kidney, liver, and muscle) of feeder pigs given an estimated LD50 oral dose of B1 (1.0 mg/kg body weight) provided as a rice culture of Aspergillus flavus and of market-weight pigs fed a naturally contaminated feed, containing aflatoxin B1 at a level of 400 ng/g from corn, for 14 days. The residues in all tissues decreased with time after treatment in both groups, with no detectable residues (approximate detection limits, ng/g, B1 0.03, M1 0.05, AFL 0.01) in pig tissues from the feeding experiment 24 h after withdrawal of aflatoxin-contaminated feed. B1 and M1, when found in the feeding experiment, were at about the same levels in all tissues except the kidney, in which M1 was the dominant aflatoxin. The level of AFL, when detected, was about 10% of the B1 level.  相似文献

A sensitive, efficient, and reproducible method, based on solid phase extraction and derivatization with acetic anhydride followed by gas chromatography-mass spectrometry in selected-ion monitoring mode, was developed for the determination of bisphenol A (BPA) in liquid infant formula. The method quantification limit was 0.5 ng g(-1). Extraction recoveries were 85-94% over the concentration range of 2.5-20 ng g(-1). Good reproducibility of the method was observed at levels of 0.54 and 10.4 ng g(-1) with relative standard deviations of 5.0 and 2.8%, respectively. The method was used to analyze samples of 21 canned liquid infant formula products for BPA. BPA was detected in all samples at levels ranging from as low as 2.27 ng g(-1) to as high as 10.2 ng g(-1). The probable daily intakes of BPA due to consumption of canned liquid infant formula were estimated for infants from premature to 12-18 months of age. The maximum probable daily intake of BPA was 1.35 microg kg(-1) of body weight day(-1) for 0-1-month-old infants with the maximum formula intake, which is below the provisional tolerable daily intake for BPA established by Health Canada, 25 microg kg(-1) of body weight day(-1).  相似文献

The production and consumption of home-brewed Xhosa maize beer is a widespread traditional practice in the former Transkei region of South Africa. HPLC determination of fumonisins B1 (FB1), B2 (FB2), and B3 (FB3) in maize beer samples collected in two magisterial areas, Centane and Bizana, showed a wide range of levels. All samples were positive for FB(1), with a mean level of 281 +/- 262 ng/mL and a range from 38 to 1066 ng/mL. Total fumonisins (FB1 + FB2 + FB3) ranged from 43 to 1329 ng/mL, with a mean of 369 +/- 345 ng/mL. Data on the consumption of home-brewed beer are not available. On the basis of published data for the consumption of commercial beer in South Africa, the fumonisin exposure in these districts among the consumers of maize beer was found to be well above the provisional maximum tolerable daily intake of 2 mug/kg of body weight/day set by the Joint FAO/WHO Expert Committee on Food Additives.  相似文献

The scope of this study was to evaluate the exposure of the Italian population to ochratoxin A (OTA) attributable to wine consumption. With this aim 1166 wine samples (773 red wines, 290 white, 75 rose, and 28 dessert wines), collected in 19 different Italian regions and mostly produced between 1988 and 2004, were analyzed for OTA content. The obtained results are reported by year of harvest, geographical area of production, and type of wine. Red wine showed the highest maximum level of contamination (7.50 ng/mL), even though rose wines were characterized by a higher mean value (0.01 ng/mL). A gradually increasing mean concentration was also observed from the north (0.05 ng/mL) to south of Italy (0.54 ng/mL). Exposure calculations, performed using two different consumption databases, indicate a daily intake for consumer only of 0.59 up to 1.24 ng/(kg of b.w.)/day and of 0.33 up to 0.90 ng/(kg of b.w.)/day for the total population. Even in the worst case, corresponding to the calculation of the intake for consumers only in southern Italy and Islands and considering the mean consumption data increased by 1 standard deviation, a quite low exposure (1.68 ng/(kg of b.w.)/day, accounting for 9.8% of TDI) was obtained. Considering the overall OTA dietary exposure, obtained exposure rates indicate that wine did not pose a risk to the Italian population health.  相似文献

Sequential sampling for aflatoxin testing in pistachios is evaluated using the aflatoxin distribution and Monte Carlo results previously obtained (J. Agric. Food Chem. 1999, 47, 3771-3775). The sequential protocol is modeled on the current EU test protocol by applying a three-step sampling, using 10, 20, and 30 kg sample averages. An acceptance level of 15 ng/g of total aflatoxin, under consideration for U.S. standards, is applied. Optimization leads to indifference regions of 2-30 ng/g for the first two steps. The resulting OC curve approximates that for a single 50 kg sample. The sequential protocol is applied to the results for a set of 1293 lots of the 1998 crop year, each tested with a single 10 kg sample. Ninety-five percent of the lots would have been accepted on the basis of the single test and 1.5% would have been rejected, whereas 3.5% of the lots would have required retesting.  相似文献

The aim of this study was to determine the dietary intake of perfluorinated chemicals (PFCs) by the population of Tarragona County (Catalonia, Spain). PFC levels were determined in 36 composite samples of foodstuffs randomly purchased in various locations. Exposure to PFCs through the diet was estimated for various age/gender groups. Perfluorooctane sulfonate (PFOS), perfluorocarboxylate perfluorooctanoate (PFOA), and perfluoroheptanoic acid (PFHpA) were the only detected PFCs in foodstuffs. On average, for a standard adult man (70 kg of body weight), the dietary intake of PFOS was estimated to be 62.5 or 74.2 ng/day (assuming ND=0 or ND=1/2 LOD, respectively). Fish, followed by dairy products and meats, were the main contributors to PFOS intake. For an adult man, the intake of PFOS (1.07 ng/kg/day) and those of PFOA and PFHpA were lower than that recently reported for Canada (4.0 ng/kg/day), and considerably lower than that previously found in the United Kingdom, the only two countries where, to date, results concerning this issue have been reported. A correlation between dietary intake and blood levels of PFOS is suggested. However, the current results do not justify dietary intake as the main route of exposure governing blood concentrations of other PFCs.  相似文献

Arsenic (As) species were quantified by HPLC-HG-AFS in water and vegetables from a rural area of West Bengal (India). Inorganic species predominated in vegetables (including rice) and drinking water; in fact, inorganic arsenic (i-As) represented more than 80% of the total arsenic (t-As) content. To evaluate i-As intake in an arsenic affected rural village, a food survey was carried out on 129 people (69 men and 60 women). The data from the survey showed that the basic diet, of this rural population, was mainly rice and vegetables, representing more than 50% of their total daily food intake. During the periods when nonvegetarian foods (fish and meat) were scarce, the importance of rice increased, and rice alone represented more than 70% of the total daily food intake. The food analysis and the food questionnaires administrated led us to establish a daily intake of i-As of about 170 microg i-As day (-1), which was above the tolerable daily intake of 150 microg i-As day (-1), generally admitted. Our results clearly demonstrated that food is a very important source of i-As and that this source should never be forgotten in populations depending heavily on vegetables (mainly rice) for their diet.  相似文献

A chemical cleanup procedure for low-level quantitative determination of aflatoxins in major economically important agricultural commodities using HPLC has been developed. Aflatoxins were extracted from a ground sample with MeOH/H2O (80:20, v/v), and after a cleanup step on a minicolumn packed with Florisil, aflatoxins were quantified by HPLC equipped with a C18 column, a photochemical reactor, and a fluorescence detector. Water/MeOH (63:37, v/v) served as the mobile phase. Recoveries of aflatoxins B1, B2, G1, and G2 from peanuts spiked at 5, 1.7, 5, and 1.7 ng/g were 89.5+/-2.2, 94.7+/-2.5, 90.4+/-1.0, and 98.2+/-1.1, respectively (mean+/-SD, %, n=3). Similar recoveries, precision, and accuracy were achieved for corn, brown and white rice, cottonseed, almonds, Brazil nuts, pistachios, walnuts, and hazelnuts. The quantitation limits for aflatoxins in peanuts were 50 pg/g for aflatoxin B1 and 17 pg/g for aflatoxin B2. The minimal cost of the minicolumn allows for substantial savings compared with available commercial aflatoxin cleanup devices.  相似文献

The aim of this study was to determine the relationship between the oxidative and reductive metabolic pathways of acrylamide (AA) in the nonsmoking general population. For the first time both the blood protein adducts and the urinary metabolites of AA and glycidamide (GA) were quantified in an especially designed study group with even distribution of age and gender. The hemoglobin adducts N-carbamoylethylvaline (AAVal) and N-( R, S)-2-hydroxy-2-carbamoylethylvaline (GAVal) were detected by GC-MS/MS in all blood samples with median levels of 30 and 34 pmol/g of globin, respectively. Concentrations ranged from 15 to 71 pmol/g of globin for AAVal and from 14 to 66 pmol/g of globin for GAVal. The ratio GAVal/AAVal was 0.4-2.7 (median = 1.1). The urinary metabolites were determined by LC-MS/MS. Of all urine samples examined 99% of N-acetyl- S-(2-carbamoylethyl)- l-cysteine (AAMA) levels and 73% of N-( R/ S)-acetyl- S-(2-carbamoyl-2-hydroxyethyl)- l-cysteine (GAMA) levels were above the LOD (1.5 microg/L). OBJECTIVES: To investigate, measure and assess the food consumption pattern of the adult population living in Beirut, Lebanon, and to identify inadequate or excessive intake of food groups particularly linked to non-communicable diseases. DESIGN: A cross-sectional food consumption survey was conducted in 2001. Dietary habits were assessed by means of a quantitative food-frequency questionnaire. SETTING: Dietary survey of the urban population (Beirut). SUBJECTS: Random sample of 444 adult subjects (aged 25-54 years) in Beirut. RESULTS: The mean consumption of food by the study population was estimated to be 3,030 g day(-1), providing an energy intake of 2,523.57 kcal day(-1). Fat contributed 38.9% to the average daily energy intake, protein 13.4% and carbohydrates 47.2%. Mean consumption of fruits and vegetables was approximately 367 g day(-1) and 45.3% of subjects consumed less than the recommended 400 g daily. Cereals contributed 324.5 g day(-1), providing 35.0% of daily energy intake, with bread being the most highly consumed (146.2 g day(-1)) in this food group. The mean intake of meat and poultry products was 91.7 g day(-1) and provided 8.8% of daily energy intake, with consumption of butchery products especially beef being the highest (47.6 g day(-1)) followed by poultry (36.1 g day(-1)). A low consumption of fish was noted (19.7 g day(-1)), with 73.6% of subjects consuming less than the recommended 2 servings of fish per week. Dairy products contributed 243.1 g day(-1) or 10.9% of daily energy intake, and milk was the least consumed dairy product (56.8% of consumers). The intake of added fats and oils, excluding those in cooked recipes, was 20.4 g day(-1); olive oil was not used in cooking but was added solely at the table and its mean intake was 5 g day(-1). The consumption of butter was low (0.86 g day(-1)) and vegetable oil was the type of fat mostly used in cooking. The average intake of alcoholic beverages was low (33.6 g day(-1)), accounting for 0.7% of total energy intake. Women had significantly higher intakes of milk, dairy products, vegetables and coffee than men (P < 0.05). The percentage of women who reported the use of low-fat items was significantly higher than that of men. Younger people (25-34 years) ate significantly more meat, sugar, alcoholic beverages and soft drinks, and consumed significantly less cooked vegetables and legumes, than older ones (P < 0.05). CONCLUSIONS: The rather high contribution of fat to daily energy intake, the low intake of fish and the relatively high percentage of people consuming less than the recommended amount of fruits and vegetables observed in this study suggest that the adult Lebanese population is at increased risk of cardiovascular diseases, obesity and other non-communicable diseases, which provides the basis for recommending increased intakes of fish, particularly fatty fish, and fruits and vegetables.  相似文献

The cytotoxic and antitumor activity of methanolic extract of rice hulls (MERH) were evaluated by the MTT-dye reduction assay against human colon cancer cells and the colonic aberrant crypt foci (ACF) assay in 1,2-dimethylhydrazine (DMH)-injected F344 male rats, respectively. MERH was found to be highly cytotoxic, with IC50 values of 0.5 microg/mL in vitro. Forty weeks of MERH supplementation (50 mg/kg of body weight/day) reduced colonic pre-neoplastic ACF formation by 35% (p < 0.01). An active compound, momilactone B, was isolated from MERH by silica gel chromatography, Sephadex LH-20 chromatography, and HPLC. The cytotoxic activity of momilactone B was evaluated by the MTT-dye reduction, lactate dehydrogenase (LDH), and colony-forming ability assays in human colon cancer HT-29 and SW620 cells. The results indicated that momilactone B from rice hulls might be a new candidate for chemotherapeutic agent against human colon cancer.  相似文献

A gas chromatography-mass spectrometry (GC-MS) method was used for the quantitative confirmation of phosphine residues in stored products and processed foods. An established extraction technique was utilized for the preparation of headspace samples, which were analyzed by GC-MS and gas chromatography-nitrogen-phosphorus detection (GC-NPD). Wheat, oats, maize, white rice, brown rice, cornflakes, tortilla cornchips, groundnuts, and raisins were validated, showing excellent agreement between detectors when spiked at levels equivalent to 0.001 and 0.01 mg/kg phosphine and for samples containing incurred residues. The GC-MS method was reproducible and accurate when compared to the GC-NPD method and allowed five samples to be quantified in a working day. Subambient GC-MS oven temperatures were most suitable for phosphine residues ranging from 0.001 to 0.005 mg/kg, and a GC oven temperature of 100 degrees C was appropriate for residues >0.005 mg/kg. The method was sufficiently robust to be evaluated for other similar commodities as the need arises.  相似文献

A previously published method for ochratoxin A was evaluated and proved appropriate for simultaneous determination of aflatoxins, ochratoxin A, sterigmatocystin, and zearalenone, with considerable savings in time and reagent costs. The detection limits were 2, 5, 15, and 55 micrograms/kg, respectively. The recoveries and coefficients of variation obtained with artificially contaminated samples were 91-101% and 0-16% for aflatoxin B1, 98-117% and 0-17% for sterigmatocystin, and 96-107% and 0-17% for zearalenone, respectively. The coefficients of variation for naturally contaminated samples (aflatoxins in rice and ochratoxin A in beans) ranged from 0 to 8%. The method was used to survey 296 samples that included 10 cultivars of dried beans, 8 types of corn products, 3 types of cassava flour, and both polished and parboiled rice between May 1985 and June 1986 in Campinas, Brazil. Only aflatoxin B1 (9 samples, 20-52 micrograms/kg), aflatoxin G1 (4 samples, 18-31 micrograms/kg), and ochratoxin A (5 samples, 32-160 micrograms/kg) were found. The average contamination percentage was 4.7%; beans showed the highest (6.6%) and rice showed the lowest (3.3%) incidence rates. Zearalenone and sterigmatocystin were not detected. Positive samples were confirmed by chemical derivatization, corroborated by development in 3 solvent systems.  相似文献

A thorough assessment of Canadian exposure to mercury (Hg) has not been undertaken since 1970. A multimedia approach was used to update estimates of Hg exposure to members of the general population in Canada, based on currently available information. Adult Canadians have an estimated intake of all Hg species via all routes of 7.7μg/day (0.11μg/kg body weight/day), which equated to an absorbed dose of 5.3μg/day (0.076μg/kg bw/day). Fish consumption accounts for much of this exposure (27% of intake, 40% of absorbed dose), in the form of methylmercury. However, dental amalgam appears to account for a greater proportion of total Hg exposure than fish consumption. Exposure from amalgam was estimated for intake and absorbed doses (of Hg0) at 2.81 and 2.25μg/day, respectively. This represents 36% of total Hg intake and 42% of absorbed dose. Hg2+ arises principally from foods other than fish. Intake of Hg2+ by adults was determined to be 1.82μg/day and absorbed dose only 0.18μg/day. Since the discovery of acrylamide in foods, there have been many calculations of dietary exposure. Total diet studies have been commonly used to estimate consumer exposure of acrylamide; however, these often fall short in evaluating true exposure levels because of limitations in small occurrence data sets. Dietary exposure to acrylamide can also be estimated by use of modeling packages. The U.K. Food Standards Agency and the Food Safety Authority of Ireland have prepared estimates for dietary acrylamide exposure using semiprobabilistic and probabilistic modeling. Occurrence data were obtained from the European Union acrylamide monitoring database, whereas consumption data were obtained from the relevant U.K. and Irish National Diet and Nutrition Surveys. The mean adult U.K. consumer exposure was estimated as 0.61 microg/kg of body weight (bw)/day and high-level adult consumer exposure (P97.5) as 1.29 microg/kg of bw/day. The mean adult Irish consumer exposure was estimated as 0.59 microg/kg of bw/day and the high-level adult consumer exposure (P97.5) as 1.75 microg/kg of bw/day. Owing to the wide range of acrylamide levels in foods, semiprobabilistic modeling does not always provide an accurate picture of dietary exposure levels and patterns. Therefore, a comparison of semiprobabilistic assessments to probabilistic assessments of U.K. and Irish dietary exposure estimates of certain food groups is provided.  相似文献
