The influence of rearing conditions on Flavobacterium columnare infection of rainbow trout, Oncorhynchus mykiss (Walbaum), was studied experimentally in the laboratory and at a fish farm. In experiment I, the effect of parasitic infection on columnaris disease was studied using F. columnare carrier fish. The fish were exposed to Diplostomum spathaceum cercariae and a set of other stressors in order to induce clinical columnaris infection. Parasitic infection and other stressors failed to induce the disease. Disease occurred when the fish were challenged with F. columnare, but D. spathaceum infection did not enhance the severity of the infection. In experiment II, the influence of rearing density and water temperature was studied. Overall mortality was highest in fish at normal rearing density with high temperature (+23 degrees C). At low temperature (+18 degrees C) mortality was not affected by rearing density, but the transmission of columnaris disease was faster at normal rearing density at both temperatures. This supports the view that reduction of fish density could be used in prevention of columnaris disease especially if water temperature is high. Because the lower rearing density can also decrease the transmission of ectoparasites and penetrating endoparasites, it could be an efficient tool in ecological disease management.  相似文献

The aim of this study was to determine the effects of dietary probiotics on reproductive performance, growth, haematological and biochemical parameters of female rainbow trout broodstock. A total of 60 broodstocks with an average initial weight of 2,267.4 ± 54.9 g were divided into four groups and three replicates in raceway ponds. Fish were fed with diets containing 0 (control), 1 × 109 (P1), 2 × 109 (P2) and 4 × 109 (P3) CFU probiotic per kg diet twice a day for 8 weeks before spawning season. Results revealed that higher levels of dietary probiotics (P3) enhanced protein efficiency ratio and decreased feed conversion ratio (p < .05). There were no significant differences in haematological and other growth parameters among different treatments. According to the results, probiotic‐fed treatments had the lowest levels of cholesterol (237.7 ± 4.3 mg/dl) and the highest levels of total protein (11.3 ± 0.3 g/dl) and albumin (6.9 ± 0.3 g/dl), so that differences were found between P3 and control groups in all cases (p < .05). Based on the observations from reproductive parameters, the highest egg diameter, working and relative fecundities, fertilization and hatching rates, eyed eggs survival and alevins survival rates up to the absorption of the yolk sac were observed in P3 treatment (p < .05). Moreover, eyeing, hatching and yolk sac absorbing stages in P3 treatment were earlier than those in other treatments (p < .05). The results of present study demonstrated that probiotic supplementation improved reproductive performance and some growth and biochemical parameters in female rainbow trout broodstock and the best performance was observed in fish fed 4 × 109 CFU probiotic per kg diet. Due to an increase in egg production and egg size in probiotic‐fed treatments, this feed additive can be used as a suitable ingredient to increase high‐quality egg production in rainbow trout breeders.  相似文献

The effects of weekly anaesthetization with clove oil and tricaine methanesulphonate (MS‐222) on feed intake and growth were examined in juvenile rainbow trout, Oncorhynchus mykiss (Walbaum), held individually. Repeated handling without anaesthetics significantly reduced feed intake and weight gain compared with an unhandled control group during an 8‐week experiment. When anaesthetics were used during handling the feed consumption and weight gain were significantly (MS‐222) or not significantly (clove oil) higher than in fish handled without anaesthesia. When compared with the unhandled control group, neither of these two anaesthetics had significant effects on feed intake but, in contrast to MS‐222, repeated anaesthesia with clove oil had a significant negative effect on growth. However, the effects of MS‐222 and clove oil on the growth were not significantly different from each other. Feed conversion ratio (feed/gain) of MS‐222‐anaesthetized fish was significantly higher compared with unhandled control and handled control fish but was not significantly different from fish anaesthetized with clove oil. These results suggest that both MS‐222 and clove oil alleviate handling stress in juvenile rainbow trout, and that these two anaesthetics are rather similar with respect to their effects during repeated exposures.  相似文献

The aim of this study was to investigate the effect of dietary lactoferrin (Lf) on growth, haematology and non‐specific immune response of rainbow trout Oncorhynchus mykiss (Walbaum). Fish were fed an experimental diet containing 0 (as control), 50, 100, 200 and 400 mg Lf kg?1 diet twice daily for 8 weeks and sampled at 2, 4, 6 (immune function and growth), and 8 weeks (immune function, haematology and growth). Statistical analyses revealed no significant effects of dietary Lf on growth performance (specific growth rate, weight gain, feed conversion ratio, feed intake and condition factor) or haematological parameters (red and white blood cell count, haemoglobin, haematocrit, serum iron and total iron binding capacity [TIBC]). Among the serum non‐specific immune parameters, lysozyme activity increased significantly in fish fed 100, 200, or 400 mg Lf kg?1 feed for 8 weeks, whereas haemolytic complement activity increased in fish fed 100 and 400 mg Lf kg?1 diet after 6 weeks. The antiprotease activity increased in groups fed 100, 200 or 400 mg Lf kg?1 diet after 8 weeks. However, no significant effect was observed on serum peroxidase level. It can be concluded that feeding of rainbow trout on the diet supplemented with 100 mg kg?1 or higher for 8 weeks enhances the non‐specific immune response.  相似文献

A feed budget model, which enables farmers to adapt a general model to local conditions, was tested on rainbow trout, Oncorhynchus mykiss (Walbaum). The model has two major components; component 1 is used to calculate the energy required for one unit of biomass gain (g or kg) and component 2 estimates the daily growth increment (g or kg). Both components can be adjusted in relation to prevailing environmental conditions and, together, they express the daily energetic requirement of the specific species or strain. Fish fed using the model as a guide had a high rate of growth and good feed conversion. The model accurately predicted growth over the experiment, and is presented as a means of calculating the daily feed budget in fish farming. Growth of fish fed using self-feeders is presented for comparison with the results obtained using the model.  相似文献

The aim of the present study was to evaluate the effects of three formulated diets: wheat gluten protein‐based diets supplemented with Lys–Gly dipeptide (PP) or free lisine and glycine (AA), and a control diet without lysine supplementation (CON), two commercial starters: Aller Futura (AF), Aglo Norse (AN) and frozen Chironomidae larvae (CH) for on the growth and digestive system development of rainbow trout. The experiment lasted 28 days and survival at the end of the experiment was the highest in the group fed AF (87.9±4.2%) and the lowest in the group fed the CON diet (74.0±4.3%). Fish fed the CON diet showed a significantly lower crude protein content, and the AA group had a significantly higher lipid level (P≤0.05) in body carcass compared with the other groups. The highest density of intestinal goblet cells (P≤0.05) was observed in fish fed the CON diet, while the lowest density was seen in the distal intestine in the AF group. The hepatocyte cytoplasm of all fish stored more glycogen than lipids. These results indicate that wheat gluten protein‐based diets supplemented with dipeptide Lys–Gly (PP) or free lysine and glycine (AA) supported growth, protein efficiency rate and intestine epithelium homeostasis. On the other hand, fish fed CON (lysine‐deficient diet) showed lower survival and growth rate, and disturbances of intestine epithelium homeostasis, probably as a result of lysine deficiency.  相似文献

Increased use of plant oils with different origins and quality in fish feed needs to be approached from a food safety and fish welfare point of view. Plant oils contain a number of bioactive minor lipid compounds that may affect the fish’s metabolism and taste perception. This study focuses on the effect of replacing fish oil (FO) with different levels of cold‐pressed rapeseed oil (RO) on the lipid composition in muscle and liver as well as on the preference by the fish. Rainbow trout (Oncorhynchus mykiss) were fed diets with a FO : RO ratio of 100 : 0, 75 : 25, 50 : 50 and 25 : 75 until twofold weight increase. In self‐selecting feed trials of single rainbow trout, fish preferred the diet composed of only FO compared with the diets with RO but did not discriminate between different levels of RO. Plant sterols and their metabolites were found in liver of the fish fed RO diets, suggesting an effect on the sterol metabolism different from fish fed a 100% FO diet. The largest effects were seen in the fatty acid composition of the edible tissue of the fish with a decrease in 22:6n‐3 and 20:5n‐3 and an increase in 18:2n‐6 and 18:1n‐9.  相似文献

A 12‐week feeding trial was conducted to evaluate the effects of fish oil replacement by soybean oil, on lipid distribution and liver histology of two commercially important finfish species: rainbow trout (Oncorhynchus mykiss) and European sea bass (Dicentrarchus labrax). Sea bass (16.2 ± 0.5 g; mean ± SD) and rainbow trout (52.1 ± 0.5 g) juveniles were fed one of three isonitrogenous (500 g kg?1 CP) and isoenergetic (19 kJ g?1) diets, containing 0% (control, diet A), 25% (diet B) and 50% (diet C) soybean oil. At the end of the experiment, lipid deposition was evaluated in muscle, liver and viscera. Cholesterol and triglycerides levels were also determined in plasma. Tissue total, neutral and polar lipid composition (g kg?1 total lipids) showed no significant differences within species, regardless the dietary treatment. The same trend was observed for plasma parameters (P > 0.05). Viscera were the preferential tissue of lipid deposition, with 252–276 and 469–513 g kg?1 total lipid content in trout and sea bass, respectively. Dietary fish oil replacement had no effect on either hepatic lipid droplets accumulation or degree and pattern of vacuolization in the observed liver sections. These data suggest that both sea bass and trout can be fed diets containing up to 50% soybean oil without adverse effects on tissue lipid composition or liver histology.  相似文献

Juvenile rainbow trout, held at 12°C on a 12 h light :12 h dark photocycle, were fed a constant ration (1 % of body wt day ?1) of isonitrogenous and isoenergetic diets that varied in either arginine content (3.6-56.1 g kg?1 dry matter; experiment 1), or glycine (3.3–118 g kg?1 dry matter) and alanine (5.0-42.3 g kg?1 dry matter) content (experiment 2). In experiment 1, the lowest dietary level of arginine depressed growth, feed efficiency, plasma l -thyroxine (T4) and 3,5,3′-triiodo-l -thyronine (T3) levels and hepatic T4 5’monodeiodinase (5'D) activity responsible for T4-to-T3 conversion. Over the dietary range of 7.1–56.1 g arginine kg?1, there was no change in 5'D activity, despite an arginine stimulation of growth. The optimum level of arginine for growth was within the range of 14.1–28.1 g kg?1 of the diet or 32–63 g kg?1 of dietary protein. In experiment 2, an increase in dietary glycine level, at the expense of glutamic acid, increased 5'D activity without attendant elevation of the plasma T3 level. The latter finding suggests that glycine also induced a compensatory increase in T3 degradation rate. This may explain why the glycine-induced increase in 5'D activity was unaccompanied by any changes in growth indices. Alteration of dietary alanine content did not affect growth or thyroid function. We conclude that of the various dietary amino acids tested, only glycine led to a progressive stimulation of hepatic T4 5'D activity. However, because glycine likely enhanced T3 degradation, no increases in plasma T3 or growth indices were found. Glycine may serve as an advance signal that activates thyroid function immediately preceding or coincident with energy and nutrient (especially protein) intake. This, in turn, may improve the efficiency of nutrient absorption and/or post-absorptive anabolic events.  相似文献

Juvenile rainbow trout Oncorhynchus mykiss (Walbaum) were exposed to therapeutic, and higher concentrations of chloramine‐T (Cl‐T) to assess the effects of this chemical on the antioxidant enzyme system and genetic structure. Red blood cells acetylcholinesterase, ?‐aminolevulinic acid dehydratase, paraoxonase and liver glutathione S‐transferase activity were increased at 10 and 20 mg L?1 Cl‐T‐exposed fish, while they were decreased at 30 mg L?1 Cl‐T‐exposed fish. On the other hand, liver catalase activity and liver protein levels increased at 10 mg L?1 and decreased at 20 and 30 mg L?1 concentrations of Cl‐T. Liver super‐oxide dismutase activity decreased at 10 mg L?1 and 20 mg L?1 Cl‐T and increased at 30 mg L?1 of Cl‐T. Compared to control, comet assay indicated that Cl‐T did not cause significant DNA damage to red blood cells of the fish. Results indicate that 10 or 20 mg L?1 Cl‐T can be safely used to prevent or treat external parasitic and bacterial infection of rainbow trout.  相似文献

Surveys among wild marine fish have revealed occurrence of viral haemorrhagic septicaemia virus (VHSV) infections in a high number of diverse fish species. In marine aquaculture of rainbow trout, preying on invading wild fish might thus be a risk factor for introduction and adaptation of VHSV and subsequent disease outbreaks. Our objective was to determine whether an oral transmission route for VHSV in rainbow trout exists. Juvenile trout were infected through oral, waterborne and cohabitation transmission routes, using a recombinant virus strain harbouring Renilla luciferase as reporter gene. Viral replication in stomach and kidney tissue was detected through bioluminescence activity of luciferase and qRT-PCR. Replication was detected in both tissues, irrespective of transmission route. Replication patterns, however, differed among transmission routes. In trout infected through oral transmission, replication was detected in the stomach prior to kidney tissue. In trout infected through waterborne or cohabitation transmission, replication was detected in kidney prior to stomach or in both tissues simultaneously. We demonstrate the existence of an oral transmission route for VHSV in rainbow trout. This implies that preying on invading infected wild fish is a risk factor for introduction of VHSV into marine cultures of rainbow trout.  相似文献

Diludine is used as an effective agent reducing mutagenic effects of environmental pollutants, increasing productivity and protecting reproductive system of parental individuals and quality of their offspring. A 10‐week study was conducted to evaluate potential effects of diludine on rainbow trout (Oncorhynchus mykiss) juveniles growth performance, feed utilization, survival rate, liver antioxidant enzyme activities (superoxide dismutase, SOD, EC1.15.1.1 ; catalase, CAT, EC1.11.1.16 ; glutathione peroxidase, GPx, EC 1.11.1.9 ; glutathione reductase, GR, EC1.6.4.2 ; glucose‐6‐phosphate dehydrogenase, G6PD, EC1.1.1.49 and glutathione S‐transferase, GST, EC1.5.1.18 ), the levels of malondialdehyde (MDA) as a lipid peroxidation marker and muscular trace elements at a low water temperature (9 °C). Diludine was added at 0.2, 0.5 and 1 g kg?1 to a fish meal–based control diet, and each diet was fed to triplicate groups of rainbow trout juveniles. After 10 weeks, there were no differences in survival rate between fish fed experimental diets (P > 0.05). It was observed that a significant improvability existed for both growth and feed utilization in fish fed diets supplemented with diludine (P < 0.05). Antioxidant enzymes showed significantly increased activity in liver tissues (P < 0.05) and then a decrease to initial activity levels during the experimental time. (GPx activity was not determined in the juvenile livers.) SOD and GST activities in liver of fish fed the diets with diludine tended to be higher and GR activity tended to be lower than that in fish fed the basal diet. However, in general, it was determined that different concentration of diludine did not affect the studied antioxidant enzyme activities except G6PD in the liver of juveniles fed the diet with 1 g kg?1 diludine. On the other hand, the significant reduction was found in MDA levels in the fish fed the diets with diludine compared with basal diet on the 14th and 28th days of experiment. It was also observed that Cu, Fe, Mg, Zn and Se levels were significantly higher in the juveniles fed the diet with 1 g kg?1 diludine than those fed the basal diet. In conclusion, the results of this trial indicate that 1 g kg?1 dietary supplementation with diludine seems to be most positive for rainbow trout production in cold‐water adaptation.  相似文献

Protective effects of nanochitosan/zeolite composite besides zeolite and chitosan/zeolite composite on rainbow trout growth, digestive enzyme activities and biochemical parameters were studied. Nanochitosan/zeolite hybrid composites with three different ratio of nanochitosan:zeolite (35:100, 3.5:100 and 0.35:100) were synthesized and analysed by X‐ray powder diffraction (XRD), thermal gravim (TG) analysis and scanning electron microscope (SEM) methods. Prepared composites as well as zeolite and control diets were fed to rainbow trout (50 g) for a period of 60 days. The addition of treated diets significantly improved growth performance compared to the control diet. Supplemental zeolite could only enhance the amylase activity in fish intestine, whereas other treatment groups could increase the pepsin activity besides intestinal alkaline phosphatase, trypsin and amylase activities. No differences were observed for intestinal acid phosphatase and lipase activities among the experimental diets. Meanwhile, serum total antioxidant activity and lipid peroxidation product, indicated by malondialdehyde (MDA), significantly increased and decreased, respectively, with some doses of administration, indicating the elevated antioxidant status in treatment groups. Serum‐specific marker enzymes, namely aspartate aminotransferase (AST) and alkaline phosphatase (ALP), were not affected in all groups. Meanwhile serum total protein in most treatment groups was significantly higher than the control group. Results showed that synthesized composites especially nanochitosan/zeolite composite at 5 g/kg had potential to enhance growth performance, digestive enzyme activities and some biochemical parameters in rainbow trout.  相似文献

This study was conducted to evaluate the effect of soybean lecithin (SBL) and chicken egg lecithin (EGL) on growth performance; pancreatic and intestine enzymes, cholecystokinin (CCK) and lipoprotein fractions of rainbow trout, Oncorhynchus mykiss (Walbaum) fry. Seven isonitrogenic and isolipidic diets were prepared by replacing 0 (as a control), 2%, 4% and 6% soybean and chicken egg lecithin instead of soybean oil in diets. Trout fry with initial average weight of 120 ± 4.08 mg, were randomly distributed in 21 90 L tanks. Triplicate groups (165 fish for each tank) of fry were fed each diet to visual satiation at six meals per day for 40 days. Dietary supplements of 2% and 4% chicken egg lecithin led to increase in fry final weight, weight gain and specific growth rate suggesting that moderate PL levels are needed during this stage of rainbow trout. Also 2% chicken egg lecithin caused significantly higher activity of amylase, lipase, phospholipase A2, alkaline phosphatase (AP), aminopeptidase‐N (AN) and lower activity of leucine alanine peptidase (leu‐ala) in the digestive tract of fry than those in control group. Furthermore, fry fed 2% chicken egg lecithin had a higher ratio of AN/leu‐ala and AP/leu‐ala than that in the control group. Chicken egg lecithin increased significantly cholecystokinin (CCK) and chylomicron fractions of fry compared to control group. It is noteworthy that soybean lecithin had moderate effect on final growth, weight gain and specific growth rate of fry and induced significantly higher activity of amylase, lipase, phospholipase A2 and secretion of CCK compared to control group. The results of this study indicate that dietary supplementation 2% chicken egg lecithin indirectly through some biochemical parameters improves growth performance of rainbow trout fry.  相似文献

In recirculating aquaculture systems (RAS)s, off-flavors and odors, mainly caused by geosmin (GSM) and 2-methylisoborneol (MIB), can accumulate in the flesh of fish from RAS water, reducing the profitability of production. In this study, peracetic acid (PAA) was applied in three application intervals to pump sumps of rainbow trout (Oncorhynchus mykiss) reared in RAS. Using a real-time polymerase chain reaction (qPCR), the potential off-flavor producers were quantified using geoA and MIB synthase genes. Streptomyces was identified as the major GSM producer, and biofilters showed the highest number of potential off-flavor producers. Concentrations of GSM and MIB were analyzed in the circulating water and in the lateral part of the fish fillet. In water, concentrations up to 51 ng L−1 (GSM) and 60.3 ng L−1 (MIB) were found, while in the fillet, these were up to 9.8 ng g−1 (GSM) and 10.2 ng g−1 (MIB), decreasing with increasing number of PAA applications. PAA applications reduced the levels of off-flavor compounds, although this was insufficient to fully prevent the accumulation of GSM and MIB.  相似文献

Rainbow trout (Oncorhynchus mykiss) males were fed, during a 9‐month period, five experimental diets where fishmeal proteins were gradually replaced with cottonseed meal (CS) proteins (0, 25, 50, 75 and 100%; diets 1–5, respectively). This study was carried out to evaluate the action and tissue concentrations of gossypol. Growth performance of fish was not affected with the increasing levels of CS in the diets. Haemoglobin concentration and haematocrit significantly decreased in fish fed 100% CS compared with other dietary treatments. Gonadosomatic index, plasma sex steroids (testosterone, 11‐ketotestosterone and 17,20β‐dihydroxy‐4‐pregnen‐3‐one) and sperm characteristics (concentration, motility, protein concentration and lactate dehydrogenase activity) were not negatively affected by increasing levels of CS. For the first time, a comprehensive study of gossypol concentrations, total and (+) and (?)‐enantiomers, in several tissues (liver, blood plasma, spermatozoa and seminal plasma) was performed. The concentrations of both (+) and (?)‐enantiomers significantly increased with increasing levels of CS in the diet. The highest concentrations were found in the liver of fish fed diet 5 (185 ± 18 μg g?1). In blood plasma, the concentrations of total gossypol were 10 times lower than the one found in the liver, but 10 and 100 times higher than the concentrations in the spermatozoa and seminal plasma, respectively. In all tissues studied, the concentration of (+)‐enantiomer was higher than the (?)‐enantiomer. The ratio (?)‐enantiomer/total gossypol did not change significantly with the increasing levels of CS in the diet. The results of the present study indicate that CS can be used over a period of 9 months to replace fishmeal proteins completely without compromising growth and reproduction of rainbow trout males.  相似文献

Two trials were designed to quantify the effect of feeding ratio and fish size on the cohabitation transmission of Loma salmonae, the causative agent of microsporidial gill disease (MGD) in salmonids, Oncorhynchus spp. To evaluate the effect of feeding ratio on disease onset, groups of 45 rainbow trout, O. mykiss (Walbaum) (RBT), were fed daily at 1% (low), 2% (medium) or 4% (high) of the fish biomass in the tank. There were three tanks at each feeding level: two tanks were exposed to the pathogen and one was a control. For the second objective, 300 RBT were separated into seven tanks so that the weight classifications were small (17-23 g), medium (32-38 g) and large (57-63 g). Each size class was done in duplicate with one control tank containing medium-sized fish. Separately for each trial, on day 0 post-exposure (PE) five highly infectious RBT were added to each tank (not including the control tanks) to begin the cohabitation exposure period. Beginning on day 21 PE and continuing biweekly until days 70 and 77 PE for the feeding and fish size trials, respectively, each fish was evaluated for visible branchial xenomas to determine disease onset time. Using survival analysis, the survival curves for the low, medium and high feedings were not significantly different from each other. However, there were significant differences amongst the small, medium and large weight classes in the size trial. The median numbers of days to the development of branchial xenomas was 31, 38 and 42 for small, medium and large size fish, respectively. On any given day, a medium or large sized fish had a hazard ratio for developing branchial xenomas of 0.66 and 0.63, respectively, compared with a small fish. In addition to host species and host strain differences, fish size is now considered a host risk factor for the development of MGD.  相似文献

This study evaluated the regulatory effects of water temperature on the development of branchial xenomas caused by Loma salmonae using a high-dose per os-challenge model compared with a low-dose cohabitation-challenge model. Approximately 275 juvenile rainbow trout (RBT), Oncorhynchus mykiss, were randomly distributed to six tanks with two tanks each maintained at 11, 15 and 19 degrees C. Fish in one tank from each temperature setting were exposed per os to macerated L. salmonae-infected gill material and fish in the other tank from each temperature setting were exposed to L. salmonae using the cohabitation-challenge model. Fish were monitored for the development of branchial xenomas beginning at day 21 post-exposure. Survival analyses were used to evaluate the effect of water temperature and challenge model on the number of days until the first visible branchial xenoma was detected. The survivor curves for the per os-challenge model revealed that there was at least one significant difference, whereas the cohabitation challenge did not reveal any significant differences amongst the temperature settings. The proportional hazards model revealed a significant interaction between the challenge model used and water temperature. This indicated that the effect of water temperature was different depending on challenge model. Additionally, from the mean xenoma intensities, on average, the per os-challenged fish showed higher xenoma intensity compared with the cohabitation-challenged fish. Overall, the impact of water temperature on disease pathogenesis was greater when the RBT were per os challenged compared with using the cohabitation model.  相似文献
