本试验旨在探讨注射地塞米松(DEX)模拟应激状态下,应激和饲粮能量水平对肉仔鸡能量采食的影响。选取体重相近的180只23日龄的雄性爱拔益加(AA)肉鸡随机分成6组,每组3个重复,每个重复10只鸡。试验采用2×3析因设计,因素为DEX[处理(注射DEX2 mg/kg)、未处理(注射等剂量生理盐水)]和饲粮能量水平[高能(HE)、低能(LE)以及高能、低能自由采食(H-LE)]。预试期5 d,正试期7 d。结果表明:1)DEX处理极显著降低了肉仔鸡的采食量和体增重(P<0.01),极显著提高了耗料增重比、耗能增重比、腹脂率以及血浆葡萄糖、甘油三酯和尿酸的浓度(P<0.01),显著提高了肠道指数(P<0.05)。2)采用LE饲粮体增重(P<0.05)、腹脂率(P<0.01)显著低于其他2种饲粮,耗料增重比显著低于HE饲粮(P<0.05);采用HE饲粮耗能增重比极显著高于其他2种饲粮(P<0.01);采用H-LE饲粮神经肽Y(NPY)基因表达量显著高于LE饲粮(P<0.05)。3)DEX、饲粮能量水平对耗料增重比(P<0.05)和耗能增重比(P<0.01)的影响存在显著的互作效应。结果提示,DEX应激可使肉仔鸡血浆中葡萄糖、尿酸和甘油三酯的浓度升高;DEX应激和HE饲粮均能增加脂肪在腹部沉积,提高耗料增重比和耗能增重比;H-LE饲粮能上调肉仔鸡下丘脑NPY基因表达。  相似文献

Various lines of evidence suggest that appetite‐related neuropeptides in the hypothalamus are regulated by adiposity signals such as leptin and insulin in mammals. In the present study, we examined age‐dependent changes in the weight of abdominal fat and hypothalamic mRNA levels of neuropeptide Y (NPY, an orexigenic neuropeptide) and proopiomelanocortin (POMC, a precursor of anorexigenic neuropeptides) in growing chickens at 7, 14, 21 and 28 days of age. Hypothalamic NPY mRNA levels were significantly (P < 0.05) decreased after 14 days of age, whereas hypothalamic POMC mRNA levels were significantly (P < 0.05) increased at 28 days of age. The percentage of abdominal fat was significantly increased after 14 days of age in chickens. We next examined the correlation of hypothalamic NPY and POMC mRNA levels and several parameters at 28 days of age. There were no significant correlations between hypothalamic mRNA levels of NPY or POMC and the percentage of abdominal fat. These findings suggest that the gene expressions of NPY and POMC do not depend on adiposity in chickens, at least in 28‐day‐old layer chickens.  相似文献

为了研究日粮能量水平与神经肽Y(neuropeptide,NPY)基因对文昌鸡开产性状的影响,运用PCR- RFLP方法检测3组试验鸡群(Ⅰ组,低能组;Ⅱ组,中能组;Ⅲ组,高能组)NPY基因DraⅠ酶切位点的多态性,共得到3种基因型,并对其与开产性状间进行了方差分析。结果表明,日粮能量水平对开产日龄、开产体重、开产蛋重无显著影响(P>0.05),但是由高能到低能,开产日龄呈现由早到晚的趋势,开产蛋重呈现由低到高的趋势。NPY基因型对开产日龄、开产体重、开产蛋重无显著影响(P>0.05)。  相似文献

Fasting has severe effects on thyroid metabolism in the chicken: plasma thyroxine (T4) concentrations increase, whereas 3′,5,3-triiodothyronine (T3) concentrations decrease. In the present report we studied the effect of fasting at the level of: 1) the pituitary (plasma thyrotropin (TSH) concentrations; the sensitivity of thyrotrophs to corticotropin-releasing hormone (CRH) and TSH-releasing hormone (TRH)); and 2) the hypothalamus (TRH content). A regulatory role of corticosterone is discussed. One day of fasting resulted in a drop in plasma TSH concentrations. Fed and nonfed animals were treated with ovine CRH (oCRH) or TRH. The sensitivity of thyrotrophs to the respective hypothalamic hormones was increased when animals were subjected to a 1-d period of fasting. A 75% (TRH) and 50% (oCRH) increase in plasma TSH was recorded in fasted animals, whereas both secretagogues did not evoke any response in their fed counterparts. The drop in plasma TSH cannot, therefore, be attributed to a loss in sensitivity of thyrotrophs to hypothalamic stimulatory control. In an identical experiment, plasma TSH concentrations decreased, whereas hypothalamic TRH content was higher in fasted animals, suggesting a decreased hypothalamic TRH release toward the pituitary. In both fasting experiments, plasma corticosterone concentrations were increased after 1 d of fasting. Because an iv injection of corticosterone-elevated hypothalamic TRH contents and decreased plasma TSH concentrations, a corticosterone-induced TSH decrease during fasting is suggested through an action at the level of the hypothalamus.  相似文献

1. An experiment was conducted to study changes in the expression of the hypothalamic leptin receptor, neuropeptide Y (NPY) and proopiomelanocortin (POMC) with age during the early neonatal period in two different strains of chickens: Beijing-You (BY) and Arbor Acres (AA). 2. Compared with BY chickens, AA chickens ate more, and grew faster. Hypothalamic NPY concentrations of both strains increased with age until d 7 followed by a decline. Hypothalamic NPY of BY chickens on d 7 was lower than in AA chickens at the same age. 3. No difference with age was observed in hypothalamic α-melanocyte-stimulating hormone (α-MSH) of BY chickens, while hypothalamic α-MSH in AA chickens on d 0 was higher than on the other days. Compared with AA chickens, BY chickens showed lower hypothalamic α-MSH on d 0. 4. Similar developmental changes between two strains were observed in the expression of leptin receptor, NPY or POMC genes in the hypothalamus. Hypothalamic mRNA of leptin receptor on d 0 was higher than on d 1 and 7. Unlike NPY, hypothalamic NPY mRNA on d 0 was higher than on the other days. 5. Hypothalamic POMC mRNA decreased gradually with age until 7 d followed by a slight increase. 6. The results showed that the developmental changes of hypothalamic signal molecules varied with age and strain. NPY, α-MSH and leptin receptor might be involved in the early programming of feed intake in newly hatched chickens.  相似文献

In order to determine the role of the cecum on energy use in growing chickens, metabolizability of the dietary energy and energy expenditure were examined in the week following bilateral ligation and washing out of the cecum. Apparent metabolizable energy (AME) values were 14.30 and 13.69 kJ/g air‐dry matter for sham‐operated and cecally ligated chickens, respectively. These values were found to be significantly different (P < 0.05). Although AME intake and fasting heat production were decreased by cecal ligation, the distribution of AME (measured as fasting and feeding heat production, as well as heat increment and energy retention, as a proportion of AME intake) was not affected. These results suggest that the cecum helps chickens extract AME from corn‐soybean type diets with little, if any, effect on AME use, based on the present study of growing chickens in the week following cecal ligation.  相似文献

1. Effects of Ascaridia galli infection on the energy and nitrogen (N) metabolism were studied on groups of 5 cross-bred cockerels aged about 5 weeks and given a diet deficient or adequate in vitamin A at two levels of feeding in respiration chambers. 2. Metabolisability of dietary energy was 67% and N retention 33% in infected chickens compared with 71 and 41% respectively, in uninfected chickens. 3. Maintenance energy requirement of vitamin A-deficient birds was 882 kJ/kgW d compared with 998 kJ/kgW d for normal birds. N balance of the deficient chickens was also less when compared at the same energy balance. Infection did not affect maintenance energy requirement nor N balance. 4. Starvation heat production of infected chickens (619 kJ/kgW d) was higher than that of uninfected controls (586 kJ/kgW d). When infection treatments were combined, vitamin A-adequate chickens had a higher heat production (615 kJ/kg d) than the vitamin A-deficient (580 kJ/kgW d). Endogenous N excretion (mg/gW) was less in vitamin A-deficient than in adequate, starved birds. 5. Deficient chickens had undetectable liver reserves of vitamin A and only very low plasma concentrations. There was a difference in the length of larvae (17 d after infection) associated with vitamin A status, and with level of feeding.  相似文献

The influence of dietary restriction on the negative feedback potency of 17-beta-estradiol (E2) was evaluated in both castrated male (wethers) and female sheep (OVX ewes) during the breeding season. In study 1, OVX ewes received maintenance or restricted dietary energy for 7 weeks or maintenance energy for 6 weeks prior to a 5 day fast (n=12ewes/feeding group). Estradiol (0.31microg E2/50kg/h) or vehicle (10% EtOH-saline) was continuously infused into half the animals in each dietary treatment for the final 54h of the study. The dynamic pattern of LH secretion was assessed during the final 6h of infusion. Estradiol inhibited luteinizing hormone (LH) pulse amplitude independent of nutrition (P=0.02); fasting increased mean LH, LH peak height, and LH nadir in the absence of E2 (P=0.004, P=0.02, and P=0.02, respectively); while E2 inhibited pulse frequency (P=0.02) and increased peak width (P=0.04) in restricted ewes. Interestingly, despite uniform E2 delivery, serum concentrations of E2 differed with feeding status. Therefore, 12 wethers were infused with 0.31microg E2/50kg/h (6 fed, 6 fasted) and six wethers received 0.19microg E2/50kg/h (fasted) to establish similar serum concentrations of E2 in fed (0.31microg/50kg/h) and fasted (0.19microg/50kg/h) wethers. When fed and fasted wethers had uniform serum concentrations of E2 LH pulse frequency was suppressed (P<0.05) in fasted relative to fed animals, supporting the postulate that energy restriction enhances the E2 negative feedback potency. Collectively, these studies demonstrate that nutrition affects E2 feedback potency and clearance.  相似文献

Thirty-two Ross 308 male broiler chickens were used in a precision feeding assay to investigate the effect of exogenous phytase (EC 3.1.3.26) on dietary apparent metabolisable energy (AME), dry matter digestibility (DMD) coefficient, nitrogen (NR), amino acid and mineral retentions. The excretion of endogenous losses measured as sialic acid (SA) was also determined. Four dietary treatments (control (C), C + 250 FTU (phytase units per kg feed), C + 500 FTU, and C + 2500 FTU) were studied with each treatment replicated eight times in randomised complete block design. Diets were formulated to be nutritionally adequate with the exception of available P content (2.3 g/kg non-phytate P). Over the 48-h collection period, the phytase fed birds retained 29.3 mg more Na and 2.3 mg more Zn (p < 0.05) than the control fed birds, with the relationship between phytase dose and Na and Zn retention being best described by a linear function (p < 0.05 and p < 0.001, for Na and Zn, respectively). Phytase supplementation did not have an effect on dietary AME, DMD and NR. However, increasing the dose of phytase led to a linear increase in dietary amino acid retention (p < 0.05). Dietary phytase decreased total sialic acid excretion in a linear fashion (p < 0.05). It can be concluded that supplementary phytase increases the retention (reduces the excretion) of dietary Zn and Na in broiler chickens. The beneficial effects of the addition of exogenous phytases to poultry diets seems to be mediated through improved dietary nutrients absorption and reduced endogenous losses.  相似文献

试验旨在研究不同日粮代谢能水平对北京油鸡脂肪沉积及脂蛋白酯酶(LPL)基因mRNA表达影响。试验以3周龄北京油鸡公鸡为研究对象,将480只公鸡随机分为4组,分别饲喂4个代谢能水平的日粮(12.122、12.540、12.958和13.376 MJ/kg),每个处理组5个重复,每个重复24只鸡。试验期为10周。采用荧光定量PCR方法检测LPL基因mRNA表达水平。结果表明,日粮代谢能水平对鸡体脂沉积各项指标和LPL基因mRNA表达水平影响极显著,随日粮代谢能水平提高,各项体脂沉积指标极显著上升,胸肌和腿肌IMF、皮脂重和皮脂率在ME3达到峰值,LPL基因mRNA表达水平在ME2组达到峰值(P<0.01)。LPL基因mRNA表达水平与腹脂率、皮脂重和皮脂率呈显著正相关(P<0.05)。研究结果提示,日粮代谢能水平显著影响北京油鸡体脂沉积和LPL基因mRNA表达;LPL基因表达影响北京油鸡体脂沉积状况。  相似文献

ABSTRACT

1. This study was conducted to assess the effects of graded levels of dietary corn resistant starch (RS) on growth performance, carcass traits, nutrient retention, digestive organ index, intestinal morphology, digestive enzyme activities, and mRNA expression of certain nutrient transporters in broiler chickens.

2. A total of 320, 1-d-old Arbor Acres broiler chickens were randomly assigned to five dietary treatments, with eight replicates of eight birds in each. These treatments included one corn-soybean control diet, a corn-soybean based diet containing 20% corn starch, and three diets supplemented with 4%, 8% and 12% RS by replacing corn starch with 6.67%, 13.33% and 20% of Hi-Maize 260® (identified as control, RS1, RS2, RS3 and RS4, respectively). The feeding period lasted 42 days.

3. Performance parameters including feed consumption, feed conversion, body weight gain and percentage of abdominal fat at d 42 of age, nutrient retention (including dry matter, fat, total starch and nitrogen free extract), and apparent metabolisable energy was measured from d 18 to 20 and d 39 to 41 and showed negative linear responses to increasing dietary RS level (P < 0.05). Birds fed the RS3 and RS4 diets showed higher relative weight of duodenum, jejunum and ileum, as well as lower villus height and villus height/crypt depth compared to the control (P < 0.05). The activity of pancreatic trypsin of birds at d 21 and 42 of age decreased linearly in response to the increase of dietary RS level (P < 0.01). There were linear changes in up-regulated mRNA expression of SGLT-1 and down-regulated mRNA expression of GLUT-2 with increasing proportion of RS at d 21 and 42 of age (P < 0.05), respectively.

4. It was concluded that feeding broilers with diets containing higher concentrations of RS impaired the development of small intestine, which resulted in lower apparent total tract retention of nutrients and poorer body weight gain, feed efficiency and carcass traits of broiler chickens.  相似文献

Adipose triglyceride lipase (ATGL), a newly identified lipase, is a rate-limiting enzyme for triglyceride hydrolysis in adipocytes. The regulatory proteins involved in ATGL-mediated lipolysis in fat tissue are not fully identified and understood. The G(0)/G(1) switch gene 2 (G0S2) is an inhibitor of ATGL activity by interacting with ATGL through the hydrophobic domain of G0S2. Here, for the first time, we have cloned the coding sequence of G0S2 cDNA for the chicken, turkey, and quail. Sequence comparisons with mammals revealed that the avian G0S2 also have a conserved hydrophobic domain. Avian G0S2 is predominantly expressed in adipose tissues relative to other tested tissues. Within the adipose tissue, G0S2 is expressed 20-fold greater in the adipocyte than in the stromal-vascular (SV) fraction (P < 0.001). Expression of G0S2 mRNA gradually increased during differentiation of chicken adipocytes in culture (P < 0.05). However, there is G0S2 expression in embryonic adipose tissue, SV fraction, and primary preadipocytes before confluence that generally have an increased capacity of cell proliferation, which indicates it has an important role in adipocyte differentiation rather than proliferation. For a better understanding of how G0S2 responds to environmental stimuli, chickens were fasted for 24 h and then refed. Expression of G0S2 in adipose tissue was dramatically decreased (P < 0.05) in the chickens and quail after a 24-h fasting period, and increased to the control level after refeeding. In contrast to G0S2 expression, ATGL expression was induced (P < 0.05) after the 24-h fasting period and rapidly returned to the control level during the refeeding period. These data indicate that changes in lipolytic activities of adipose tissue in vivo can be regulated by G0S2 expression, as an inhibitor of ATGL.  相似文献

Poultry diets are formulated with additional animal fat or vegetable oils to improve growth rate and feed conversion efficiency. High‐fat diet feeding in rats and fish has been shown to result in alterations in the phospholipid composition and cholesterol content of the erythrocyte membrane, in turn affecting erythrocyte osmotic fragility. In contrast, the few studies performed using high‐fat diet feeding in avian species show no changes in erythrocyte osmotic fragility. This study made use of the Japanese quail as no data exists on investigation of this species with respect to high‐fat diet feeding and erythrocyte osmotic fragility. Fifty‐seven male quail were randomly divided into six groups and fed either a standard diet (commercial poultry feed) or one of five high‐fat diets (commercial poultry feed with 22% of either coconut oil, lard, palm oil, soya bean oil or sunflower oil on a weight/weight basis) for 12 weeks. All birds on the high‐fat diets were significantly heavier (p < 0.05) after the 12‐week feeding period, than when commencing the dietary intervention. Serum triglyceride concentrations of birds in all high‐fat diet groups were significantly lower (p < 0.05) than birds in the standard diet group, whereas only birds in the palm oil group had significantly lower (p < 0.05) serum cholesterol concentrations compared to the standard diet group. Fragiligrams of erythrocytes from birds in the various dietary groups were similar. High‐fat diet feeding with different types of additional fat did not affect the osmotic fragility of the quail erythrocytes. Feeding quail high‐energy diets of varying degrees of fatty acid saturation was well tolerated and did not seem to affect the overall health status of the birds. Resistance of avian erythrocytes to modification by excess dietary fat may be a general characteristic of avian erythrocytes.  相似文献

Two experiments were conducted to study the interaction between dietary lutein and fat levels in broiler chicks hatched from lutein depleted (Experiment I) and repleted (Experiment II) eggs. In both experiments, a 2 x 3 factorial arrangement of treatments resulted in six dietary treatments (fat at 3% and 6% and lutein at 0, 25 and 50 mg/kg feed) that were fed for 23 days to 18 birds per treatment (in three replications). In Experiment I, the anti-dinitrophenyl-keyhole-lympet-hemocyanin (anti-DNP-KLM) serum antibody response at day 22 and macrophage phagocytotic index at day 17 did not differ among treatment groups (p > 0.05). The concavalin A and phytohaemagglutinin-P lymphocyte proliferation index at day 19 was greater in birds fed 50 mg of lutein and 3% fat than in birds fed all other diets (p < 0.05). Independent of the level of dietary fat, dietary lutein increased macrophage (day 23) nitrite production measured 46 h after in vitro stimulation with LPS (p < 0.05). Among the birds fed lutein at 25 or 50 mg/kg feed, birds fed 3% fat had higher LPS-induced nitrite production compared to the birds fed 6% fat after 46 (p = 0.014) or 70 h (p < 0.001). In Experiment II, macrophage nitrite production was measured at 54 h after LPS stimulation on days 11, 15, 19 and 23. An interaction between dietary lutein and fat levels on nitrite production was observed on day 19 (p = 0.012), where macrophages from birds fed 0 mg lutein and 3% fat had the highest nitrite production (p = 0.012). Macrophages from birds fed lutein at 25 and 50 mg/kg diet and 3% fat had higher (p = 0.012) nitrite production than birds fed 6% fat. Thus, in birds hatched from lutein deplete and replete eggs, modulation of macrophage nitrite production by lutein is dependent on the level of dietary fat.  相似文献

ABSTRACT

1. Two experiments were conducted, the first to determine the optimum inclusion of chitosan oligosaccharide (COS) in broiler diets to support growth performance, digestive functions, intestinal morphology, and immune organs. The second experiment evaluated the immune-protective properties of COS on broiler chickens during coccidia challenge (CC).

2. Experiment 1 investigated the effect of graded dietary concentration of COS in the diets of broiler chickens using eight cage replicates for each of the six diets. A corn-soybean meal-based diet was used as the basal diet and supplemented with 0.0, 0.5, 1.0, 1.5, 2.0, or 2.5 g of COS/kg feed to form the six treatments.

3. The diet supplemented with 1.0 g COS/kg of feed provided the optimal inclusion level for broiler chickens regarding body weight (BW) gain, jejunal villus height, villus height to crypt depth ratio, and ileal energy digestibility at d 22 of age.

4. Experiment 2 investigated the immune-protective properties of COS in broiler chickens during CC. A total of 224 male broiler chicks were randomly assigned to eight replicate cages in a 2 × 2 factorial arrangement of treatments with two COS concentrations (0 or 1 g of COS/kg of diet), with or without CC.

5. On d 18 of age, birds in the CC group received twice the recommended coccidia vaccine dose of 30 doses/kg BW.

6. Coccidia challenge reduced (P < 0.05) and dietary COS increased (P < 0.05) BW gain, and feed intake. Dietary COS mitigated (P < 0.05) the CC-induced effects on gain:feed. Dietary COS supplementation attenuated the CC-induced effects (P < 0.05) on the expression of occludin genes.

7. In conclusion, dietary COS improved performance, and the immune-related beneficial impact of COS supplementation was associated with reduced expression of pro-inflammatory cytokine genes.  相似文献

1. The effects of daily injections of corticosterone (1 or 5 mg/bird) on growth, fat deposition, liver lipid and plasma concentrations of uric acid, glucose, insulin and growth hormone were studied using genetically selected lines of fat (FL) and lean (LL) chickens. 2. Both doses of corticosterone depressed body weight gain and increased the liver lipid and the abdominal fat to the same extent in both lines. 3. In both lines, corticosterone caused a dose-dependent increase in the plasma concentrations of uric acid, glucose and insulin in the fasted and refed states. 4. In untreated birds, plasma concentrations of growth hormone (GH) were slightly higher in FL than in LL chickens and slightly decreased during refeeding. The response was not modified by injection of 1 mg corticosterone. Injections of 5 mg decreased plasma GH in both lines in the fasting state and in LL chickens during refeeding. In contrast, the same dose increased GH in FL chickens during refeeding. This contradiction remains unexplained. 5. The results suggest that corticosterone sensitivity is not involved in difference of fattening between FL and LL chickens.  相似文献

A high-oleic-acid peanut breeding line was used in a study designed to determine the effects of feeding swine diets containing elevated levels of monounsaturated fatty acids as a means to increase the level of monounsaturates and total unsaturates in the resulting carcass fat. Forty-eight pigs were allotted to four treatments that consisted of corn-soybean meal diets that contained 1) high-oleic peanuts (HOP), 2) regular commercial peanuts (RP), or 3) canola oil (CO), each added at a dietary level to provide 10% added fat/oil, and 4) a control diet with no added fat/oil. The oil of HOP averaged 75% oleic acid vs 60% for CO and 53% for RP. The pigs were fed the experimental diets from 33 to 102 kg BW, after which all pigs were slaughtered. All three dietary oil sources resulted in increases (P < .01) of monounsaturates in the backfat; the HOP diet resulted in the greatest increase (32% greater than control). Both CO and RP increased (P < .01) the level of polyunsaturates by nearly twofold; HOP resulted in a small decrease. Total unsaturates increased (P < .01) by 24, 24, and 27% for HOP, RP, and CO treatments, respectively, over that obtained from the control treatment. Carcass fat was softer/oilier (P < .05) from pigs fed CO and RP diets, but not from those fed HOP diets, compared with carcass fat of pigs fed the control diet. Dietary fat/oil source had no effect (P > .05) on other carcass compositional traits and various meat quality attributes.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
