A virus was isolated in Italy from Tamus communis in which it caused symptoms of red mosaic and leaf distortion. The virus, for which the name tamus red mosaic virus (TRMV) is suggested, was differentiated from other potexviruses or possible potexviruses on the basis of host range, particle properties and serology. In crude preparations, TRMV had elongated particles of modal length 550–560 nm and diameter of 13 nm. TRMV was easily purified from Chenopodium quinoa and had a buoyant density of 1·34 g/cm3 in CsCl and 1·27 g/cm-3 in Cs2SO4. An antiserum to TRMV was obtained with a titre of 1/1024 in slide precipitin tests. TRMV was related to, but distinct from, tamus latent virus, a potexvirus isolated from Tamus communis in the UK, and more distantly related to potato virus X, cactus virus X and commelina virus X. Polyacrylamide gel electrophoresis (PAGE) indicated that the virus particles contained a single protein species of estimated molecular weight 25·2 kDa. Enzymatic digestions, agarose gel electrophoresis and infectivity of the isolated nucleic acid indicated that the virus has a single major molecule of single-stranded positive-sense RNA of estimated molecular weight 2120 kDa ( ca. 6·4 kb).  相似文献

A spherical virus–like agent ($$32 nm in diameter) was isolated from a plant of Brachypodium sylvaticum (Huds.) Beauv. growing in a botanic garden in England and showing yellow streaks in the leaves. The agent was readily purified and sedimented as a single component in sucrose density rate gradients. The particles had a sedimentation coefficient at infinite dilution of $$122S and a buoyant density of 1.35 g/cm3 in CsCl and 1·33 in CS2SO4. The particles were stable at acid pH but above pH 7·0 in the presence of EDTA dissociated. A protein having a major polypeptide with a molecular weight of $$3·76 × 104 and a species of single stranded RN A with a MW of 1·67 × 106 were detected in the particles. The agent was not transmitted by manual inoculation, by the insects Myzus persicae Sulzer, Rhopalosiphum padi L. or Nephotettix virescens Distant, through soil by leakage from roots or by seed. The particles had physicochemical properties in common with tombus– and sobemoviruses but were not serologically related to 10 members of these groups or to 57 other small spherical RNA plant viruses.  相似文献

A potyvirus that induced stunting and a characteristic bushy appearance at the apical region, due to proliferation of terminal branches with narrowed, reduced and deformed leaflets, was isolated from chickpea in India. The virus was sap-transmissible to 14 species of Chenopodiaceae, Leguminosae, Solanaceae and Malvaceae; Chenopodium amaranticolor was a good local lesion host. Virus particles, trapped by immunosorbent electron microscopy and stained with uranyl acetate, were 710 ×10 nm long. Purified virus preparations contained a single polypeptide species of 32,500 Da and one nucleic acid species of 3.1 · 106 Da. The virus was serologically related to soybean mosaic, azuki bean mosaic and peanut mottle viruses but not to clover yellow vein, pea seed-borne mosaic and bean yellow mosaic viruses.
On the basis of these properties, the virus was identified as a previously undescribed potyvirus in chickpea, for which the name chickpea bushy dwarf virus is proposed.  相似文献

The effects of the density and proportion of both volunteer barley ( Hordeum vulgare L.) and Stellaria media (L.) Vill. (common chickweed) on the seed yield of both species was investigated in linseed ( Linum usitatissimum L.) and autumn-sown field bean ( Vicia faba L.). A model was created to estimate these effects. It was a combination of two models. The first was a simple linear model relating weed seed number m2 to weed dry weight m2. The second was an inverse linear model relating weed dry weight per plant to weed density. A very good relationship existed between barley dry weight and number of seeds m2 and between S. media dry weight and number of seed capsules m2. This relationship was relatively consistent between experiments. The inverse linear model provided a good estimation of the relationship between weed density and weed dry weight per plant for both barley and S. media . Model variables, however, differed between experiments. Using the combined model it was estimated that, in the absence of other weed species and at a density of 800 plants m2, S. media would produce between 4000 and 48 000 seeds m2, whereas volunteer barley, at a density of 400 plants m2, would produce between 2000 and 8000 seeds m2. The presence of barley always reduced S. media seed yield and a barley population of 100 plants m2 could reduce S. media seed yield by up to 79%. The presence of S. media reduced barley seed yield in only one of three experiments, in which a S. media density of 800 plants m2 reduced barley seed yield by up to 68%. A potyvirus (designated as no. 656) causing mosaic or vein-banding symptoms on melons was isolated and characterized. The virus was mechanically transmitted to 14 herbaceous plant species, and induced mosaic symptoms in most cucurbitaceous plants. Aphis gossypii transmitted the virus non-persistently, and flexuous filamentous virus particles c. 755 nm in length were consistently observed in extracts of the infected pumpkin leaf tissues. Pinwheel and tubular inclusions were observed in the cytoplasm of infected zucchini leaf tissues. The virus was purified from infected pumpkin leaves by isopycnic centrifugation (Cs2SO4). The molecular weights of purified capsid and cylindrical inclusion proteins as estimated by SDS-PAGE were 32·5 and 73 KDa, respectively. In SDS-immunodiffusion tests, antiserum to virus particles from isolate 656 was serologically unrelated to ZYMV, WMV-2, PRV-W and a type W variant of PRV, but antiserum to its cylindrical inclusion protein did produce spur precipitin bands between homologous and WMV-2 antigen wells. However, neither WMV-2 virus particle nor cylindrical inclusion antisera reacted with this virus. Furthermore, this virus was not serologically related to BCMV, BYMV, CYVV, SMV, WMV-M or ZYFV. Based on test results and symptomatology, this virus appears to be a new potyvirus, for which the name melon vein-banding mosaic virus (MVbMV) is proposed.  相似文献

 自武昌芝麻坏死花叶病株得到1个病毒分离物,根据该分离物生物学特性、粒体形状、血清学性质、外壳蛋白分子量和核酸组分、大小,明确为自然侵染芝麻的一种新病毒,定名为芝麻坏死花叶病毒(Sesame necrotic mosaic virus,SNMV)。在人工接种7科38种植物中,SNMV侵染6科23种;体外稳定性状:稀释限点10-4~10-5,致死温度80~85℃,体外存活期限40d;病毒可通过土壤和接触传播。SNMV病毒粒体球状,表面有粒状突起,直径约32nm。制备病毒抗血清用琼脂双扩散方法测定效价为1:128。SNMV和红三叶草坏死花叶病毒(RCNMV)、甜三叶草坏死花叶病毒(SCNMV)、黄瓜花叶病毒(CMV)和花生矮化病毒(PSV)抗血清没有反应。病毒外壳蛋白亚基分子量为38 100道尔顿;核酸1个组分,大小约4700nt,另含RNA片段,大小约600nt,可能为卫星RNA。依据上述病毒特性,SNMV可能归属于番茄丛矮病毒科(Tombusviridae)。  相似文献

 从自然感病的哈密瓜上分离到的烟草坏死病毒(TNV),其寄主范围较广,在藜科、苋科、豆科和茄科一些植物上呈局部坏死反应,并在葫芦科一些植物上呈局部坏死斑和系统花叶症状。病毒的热灭活点为90~95℃,稀释限点10-5~10-6,体外保毒期在10天以上。病毒颗粒呈六角形,直径约25毫微米。提纯病毒的紫外吸收,最高260毫微米,最低为245毫微米;沉降常数为94s。从病毒提取的核酸的紫外吸收,最高为260毫微米,最低234毫微米;经聚丙烯酰胺凝胶电泳测定,核酸分子量为1.4×106;病毒蛋白的分子量为2.75×104。经血清学试验,该病毒与烟草花叶病毒的A型和D型的抗血清发生沉淀反应,而与黄瓜坏死病毒的抗血清无沉淀反应。  相似文献

 对从北京郊区菜豆皱缩花叶病株上获得的一个病毒分离物进行了系统研究。根据该分离物的寄主范围、传播特性、病毒颗粒形态、血清学性质和理化特性等,确定为花生矮化病毒(Peanut Stunt Virus,PSV),并称为PSV菜豆分离物,即PSV Bean Isolate,简称PSV-B。这是国内从菜豆上分离PSV的首次报道。在测定的5科28种植物中,PSV-B侵染5科21种。PSV-B致死温度60~65℃,稀释限点10-4~10-5,存活期限6 d以上。PSV-B能被豆蚜(Aphis craccivora Koch)以非持久性方式传播。提纯病毒颗粒球形,直径30 nm。制备的PSV-B抗血清,采用琼脂双扩散法测定效价为1 128。在血清学关系上,P SV-B与已报道的中国PSV-Mi株系相近,与美国PSV-E株系差异明显。PSV-B外壳蛋白亚基分子量为25 000道尔顿;含有4个RNA片段。  相似文献

Forty-two percent of leaf samples taken from hop cv. Qingdao Dahua growing in plantations in northeastern China in 1983-1984 were infected with hop mosaic virus (HMV) and/or hop latent virus (HLV). Mosaic or line-pattern symptoms were seen in some plants but only in spring and it is uncertain if the symptoms were caused by either virus which appeared in our studies to be similar, both serologically and in host range, to HMV and HLV described in Europe and North America. In host range studies, HMV infected Nicotiana clevelandii systemically without inducing symptoms. Occasionally it caused chlorotic spots in the inoculated leaves of Chenopodium quinoa and brown star-point lesions in inoculated leaves of Phaseolus vulgaris cv. Topcrop, The virus was purified by PEG precipitation from extracts of N. clevelandii plants; yields were about 40-120 mg/kg fresh leaves. The virus contained a single protein (estimated molecular weight 34 200) that was estimated to contain 272 amino acids with no methionine, and a single RNA species (estimated molecular weight 295 x 106) that represented c. 6% of the particle weight. HLV sometimes caused inconspicuous chlorotic spots in inoculated leaves of C. murale and local pinpoint lesions in P. vulgaris in winter only. A virus similar to alfalfa mosaic virus was isolated from leaves of hop cv. Golding and caused systemic necrotic symptoms in C. quinoa and C. amaranticolor , and systemic chlorotic spots in hop cv. A new virus, subterranean clover mottle virus (SCMoV) was found at several locations in Western Australia in Trifolium subterraneum L. (subterranean clover) with severe disease symptoms. Paiticles of the virus were shown to share many physical properties with members of the sobemo-virus group. However, in addition to the single-stranded RNA of MJ about 1.5 × 106 which is characteristic of sobemoviruses, SCMoV particles also contained circular and linear viroid-like RNAs of two size classes, one about 400 and the other about 300 nucleotides long. At present it is not clear if the two types of viroid-like RNAs are components of the same SCMoV strain or of closely related strains of the virus which are able to confect subterranean clover.
SCMoV was shown to be serologically related to lucerne transient streak virus (LTSV). However, the relationship was remote and whereas most high-titred antisera to LTSV reacted with SCMoV particles, antisera to SCMoV failed to recogaize those of LTSV. Neither SCMoV nor LTSV are serologically related to velvet tobacco mottle virus (VTMoV) or Solanum nodiflorum mottle virus (SNMV). However, particles of all four viruses have many physical and chemical properties in common, including RNA complements consisting of both virus-like and viroid-like RNAs. Because of their affinities and unique RNA complements, we suggest that SCMoV, LTSV, VTMoV and SNMV may warrant inclusion in a new taxonomic group.  相似文献

 采用多种方法对雀麦花叶病毒德国株系(BMV-G)及英国株系(BMV-E)粒子的提纯及RNAs的提取效果进行了比较,结果分别以聚乙二醇(PEG)沉淀法及两相酚提取法为佳。聚丙烯酰胺凝胶(PAA)电泳证明BMV-G具有5个RNA组份,比BMV-E (代表典型株系)多一个RNA-3a,其分子量为0.52×106,介于RNA-3及RNA-4之间。另外变性及不变性RNAs分子量的测定结果也基本相同。薄层电泳分析指出BMV二种株系RNAs的碱基比也基本一致,同于Paul及Huth的报道。经34% CsCl平衡等密度离心得知BMV-G及BMV-E均具有4种粒子组份,其中最重的粒子的浮力比重值分别为1.3513及1.3534,而最轻的则分别达1.2993及1.3045。  相似文献

Single aphids ( Myzus persicae ) were allowed sequential access to two leaves which were either free from infection or infected with potato virus Y° (PVY°). PVYN. beet mosaic virus (BMV), or tobacco mosaic virus (TMV). Aphids given prior or subsequent access to PVYN-infected leaves transmitted PVY° and BMV less frequently than aphids given prior or subsequent access to virus-free leaves; those given prior or subsequent access to BMV also transmitted PVY° less frequently. However, prior or subsequent access to PVY° did not decrease transmission of either BMV or PVYN, and access to BMV did not decrease transmission of PVYN. Access to the non-aphid-transmissible TMV did not affect transmission of either PVY° or PVYN. PVYN had the greatest electrophoretic mobility. BMV was least mobile and PVY° was intermediate, so ability to decrease transmission was not directly related to the total anionic charge on the virus particles.  相似文献

Genetic variation of the barley powdery mildew fungus ( Erysiphe graminis f.sp. hordei ) was estimated in three Danish local populations. Genetic variation was estimated from the variation amongst clones of Egh , and was therefore an estimate of the maximum genetic variation in the local populations. The average gene diversity, Ĥ , was estimated as 0.84. The effective population size was estimated as: log10 ( N^ e ) = 0.64 − log10(μ), or 4.4 × 109, assuming a nucleotide mutation rate ( μ) of 10−9 per base per generation. A study was carried out to demonstrate that Rice yellow mottle virus (RYMV), a virus known to be transmitted by beetles, can spread between rice plants by direct leaf contact caused by wind. Almost all healthy plants surrounding an infected plant became infected when exposed to a fan blowing for 15 min at a distance of 50 cm. Spread of RYMV by plant contact, mediated by wind, was also demonstrated in field experiments, the extent of spread depending on plant density. Infection was almost 10 times higher in plots with a density of 33 plants m−2 than in plots with 16 plants m−2. Less spread was observed in plots protected by 1·5 m high windscreens. It is suggested that wind-mediated spread of RYMV may result from abrasive contact between leaves of plants.  相似文献

 山东省田间侵染大豆的花叶病毒,粒体线条状,大小714-729×12-13nm,粗提纯液与SMV抗血清呈阳性反应。寄主范围窄,接种5科14种植物只侵染大豆,呈系统花叶症状。体外抗性。钝化温度65-70℃,稀释限点10-3-10-4,体外保毒划3-4天。传毒方式:可通过种子、蚜虫和摩擦接种传毒。根据以上特性,侵染大豆的花叶病毒,毒原应为大豆花叶病毒(SMV),属于马铃薯y病毒组(Potyvirus group)的成员。  相似文献

 为害华南番木瓜的病毒以番木瓜科(Caricaceae)和葫芦科(Cucurlitaceae)中若干植物为寄主,范围狭窄。病毒的稀释终点为10-2~10-3;热灭活点为50~55℃;体外存活期为8~16小时。病毒颗粒长约600~800nm、宽约10~15nm的略为弯曲的细丝。在感病番木瓜组织的细胞内,电镜观察到典型的风轮状内含体(pinwheel inclusion body)。病毒具有马铃薯Y病毒组成员的典型特征,可认为是国外报道的番木瓜环斑病毒的中国株系,初步命名为华南番木瓜环斑病毒。  相似文献

The mutual effects of redroot pigweed ( Amaranthus retroflexus ) on corn ( Zea mays ) were evaluated in an experiment conducted in 2005 at the Iranian Plant Protection Research Institute at Qazvin, considering the different densities of redroot pigweed against four different corn densities. Redroot pigweed, at 0, 35, 50, 65, and 80 plants m−1 row−1, was arranged factorially with corn at four, five, six, and seven plants m−1 row−1 in a randomized complete block design. Crop–weed competition resulted in a reduction in the total dry matter, Leaf Area Index, and crop growth rate of corn. Furthermore, an increasing weed density ≤65 plants m−1 row−1 reduced the corn grain yield and biological yield. Overall, six corn plants m−1 row−1 was suggested as the optimum density of this crop in competition with redroot pigweed.  相似文献
