Summary Improvements in the success of doubled haploid production have been achieved partly by screening new accessions of Hordeum bulbosum. In order to assess levels of variation within stocks of this species, rates of embryo differentiation from the cross between H. vulgare × H. bulbosum were recorded for selections derived from two different stocks of H. bulbosum. There was little difference within stocks for this character despite variation in the morphology and banding patterns of two enzyme systems of one of the stocks. It is proposed that to obtain further increases in success rates a few selections from many accessions of H. bulbosum should be screened rather than many selections from a few stocks.  相似文献

Summary Embryo development in vivo was compared in the two barley genotypes VK 16032 and Vogelsanger Gold after self pollination and after pollination with H. bulbosum. Embryo growth in VK 16032 after crossing with H. bulbosum showed a similar growth pattern to that of the self pollinated embryos, although the size increase was smaller. The hybrid embryos continued to grow until day 18 after pollination. The embryos from the cross between Vogelsanger Gold and H. bulbosum ceased to grow 8 days after pollination. Arrested embryo growth with subsequent abortion in Vogelsanger Gold was associated with a very early depletion and break down of the endosperm. Use of barley genotypes less sensitive to the genomic disturbances after crossing with H. bulbosum in combination with genotypes like Vogelsanger Gold is recommended in haploid breeding work.  相似文献

Summary Seed and embryo development was studied in crosses between H. bulbosum and the barley genotypes VK 16032, Vada and Vogelsanger Gold and subsequently the F1 and F2 progenies from VK 16032 x Vada and VK 16032 x Vogelsanger Gold. Both seed and embryo development are strongly influenced by the barley genotype. Favourable environmental conditions can promote seed and embryo development in genotypes with existing good characters. Dominant inheritance for good seed development and incomplete dominance for large embryos is evident. Linkage between small embryo size and winter habit exists in Vogelsanger Gold.  相似文献

Summary Partial incompatibility has already been reported between Hordeum vulgare and H. bulbosum during the course of doubled haploid production, and in this paper attempts to overcome the breeding barrier are described. The methods which seem to offer most success are those of environmental adjustments and the adoption of new genotypes of H. bulbosum. Summary An attempt was made to produce doubled haploids on 16 winter wheat and six spring and winter triticale genotypes thought to carry genes for interspecific incompatibility. The potential for haploid production was maximized by the use of Hordeum bulbosum genotypes selected for high crossability on crossable wheat genotypes, the use of two post-pollination applications of gibberellic acid and by the pollination of immature florets.A low frequency of seed was set on both the wheat and the triticale genotypes, having mean seed sets of 0.20 per cent and 0.27 per cent respectively. Although the frequency of embryos (seed quality) was high, doubled haploid production was further limited by poor embryo differentiation and regeneration. Haploid plantlets were obtained from the wheat cultivars Moulin and Renard, although successful chromosome doubling and doubled haploid production was achieved in Moulin only.  相似文献

Summary Possible negative effects of tissue culture on qualitative and quantitative characters have been investigated in three crosses of spring barley, using doubled haploid lines produced by anther culture (AC, with maltose as a carbon source) or the Hordeum bulbosum-method (HB). In one cross inbred lines produced by single seed descent (SSD) have also been included. Quantitative characters were investigated in a 2-year field experiment at one location. The results show that although the methods in the majority of cases gave similar results, the genotypic arrays produced were not identical. Different markers deviated in the various cross/method combinations. The ranking of methods as well as the frequencies of lines transgressive for grain yield differed between crosses. A restriction fragment length polymorphism (RFLP) based linkage map of a cross between two diploid Hordeum bulbosum (2n = 2x = 14) clones, PB1 and PB11, was constructed from 46 recombinant progeny clones. Since both parents are heterozygous, separate and combined parental maps were constructed. All of the RFLP markers screened had previously been mapped in barley (H. vulgare L.) so that comparative maps could be produced. The PB1 linkage map consists of 20 RFLP marker loci assigned to four linkage groups covering 94.3 cM. The PB11 linkage map consists of 27 RFLP marker loci assigned to six linkage groups covering 149.1 cM. Thirteen markers polymorphic in both parents were used as ‘anchors’ to create a combined linkage map consisting of 38 loci assigned to six linkage groups and covering a genetic distance of 198 cM. Marker order was highly conserved in a comparison with the linkage map of H. vulgare (Laurie etal., 1995). However, in contrast, the genetic distances for the same markers were very different being 649 cM and 198 cM respectively, a genetic distance ratio of 1: 3.3. Thus although the map was short, it can be presumed to cover half the genome of H. bulbosum. This study provides further confirmation of the close relationship between the two species and gives a basis for the development of marker mediated introgression through interspecific hybridisation between the two species. Summary A assessment of in vitro characters: embryo size, differentiation stage and germination time of haploid embryos, was carried out with a view to developing a selection procedure to increase the efficiency of doubled haploid (DH) production using the Hordeum bulbosum system. The differentiation stage of haploid embryos at the time of excision was found to be the most promising character for selection. In the two spring crosses examined 55% of excised embryos were undifferentiated and 45% were differentiated, with overall success rates, in terms of the percentage of DHs produced, of 7.91% and 25.16%, respectively. 17% of excised embryos produced callus. These embryos performed poorly in terms of regeneration and subsequent growth. It is suggested that only differentiated non-callusing embryos are maintained in order to maximize the efficiency of DH production. It is also shown that the overall success rate depends not only on initial stages such as seed setting but also on genotypic differences in response to colchicine treatment at later stages.  相似文献

A total of 147 simple sequence repeat (SSR) markers (including86 barley and 61 wheat microsatellite markers) were tested for their segregation in a doubled haploid (DH) and an F2 population of barley. The DH population consisted of 71 doubled haploid lines, developed from F1 plants of a cross between Tadmor and WI2291using isolated microspore culture technique. A genetic linkage map consisting of 43 microsatellite markers was constructed using the DH population. Particularly on chromosome 4H microsatellite markers showed distorted segregation ratios. Segregation of DH lines based on molecular markers were compared with segregation of 92 F2 lines from the same cross. The proportion of loci deviating from the expected monogenic segregation ratios in the DH population was significantly higher (19/43loci, 44%) than in the F2 population (7/43 loci, 16%). The deviation was biased towards the WI2291 parent alleles. In line with this observation, WI2291 was found to perform better than Tadmor in regenerating green plantlets with the isolated microspore-culture technique. Summary Certain cultivars of Hordeum vulgare L. gave low seed set (average <30%) after pollination with H. bulbosum, while others consistently gave high seed set (average >80%). The reduced seed setting occurred in crosses with cultivars which had H. distichum var. laevigatum in their ancestry. The results show that the reduced setting is probably controlled by a single dominant gene. This finding is of particular importance to breeders concerned with the use of Vada or some of its high yielding derivatives for producing haploids by interspecific hybridization.  相似文献

Summary Two tetraploid and two diploid clones of Hordeum bulbosum were screened for resistance to five isolates of powdery mildew which are virulent on cultivated barley. All were resistant and this resistance was also expressed in hybrids with H. vulgare. The tetraploid genotypes were also resistant to isolates of yellow rust and brown rust. These results show that H. bulbosum contains useful genes for resistance to these diseases and that there is a potential to transfer these into cultivated barley.  相似文献

Summary To produce hexaploid (or other polyploid) hybrids, diploid or tetraploid Hordeum vulgare was crossed with hexaploid or octoploid H. bulbosum, and perennial triploid hybrids between the two species were treated with colchicine. The crosses did not yield viable plants: seedset was low, the seed aborted and embryo culture was unsuccessful. The colchicine treatments geve rise to plants in which hexaploid chromosome numbers were observed. At the hexaploid level chromosomal instability occurred, resulting in chromosome elimination.The colchicine-treated triploid hybrids showed in the first years after the treatment better fertility after open flowering than untreated plants, but the level of fertility remained very low. The offspring consisted of haploid, diploid and approximately triploid plants like H. vulgare, tetraploid and approximately tetraploid plants like H. bulbosum, and plants with hybrid morphology and unstable chromosome number, which were highly sterile. Thus the crossing barrier between H. vulgare and H. bulbosum could not be broken down at higher ploidy level.  相似文献

Summary Diploid Hordeum bulbosum L. (2n=2x=14), collected for the first time in south-west Spain, is described. One hundred and ten plants from the original seed were grown to maturity to determine the type and extent of phenotypic variation within the population. Cytological study verified that all the plants were diploids. The plants proved to be variable for traits such as growth type, tiller number, culm and spike length, flag leaf length and width and bulb characteristic and size. The variation in the diploids was found to be greater than previously reported. Summary Four six-rowed uniculm lines of spring barley were grown in two yield trials at different sowing densities. One trial was grown on light sandy soil in 1990. The second trial was grown on clay soil in 1991 and included the two-rowed, tillering cultivar Golf. In the trial on sandy soil, the grain yield of the uniculm lines approached that of Golf grown in an adjacent trial, but in 1991 when the growing conditions were more favourable, Golf yielded significantly more than the uniculm lines. Uniculm lines apparently perform relatively better under marginal growing conditions than in high yielding environments.The uniculm lines do not conform to the ideotype proposed for wheat by Donald (1968) but the results show that a drastic change in plant type need not to imply a large drop in grain yield.  相似文献

Summary The effects on quantitative characters of the ml-o5 allele has been investigated by means of doubled haploid lines produced by the Hordeum bulbosum method. In one series of experiments the genetic background in each cross was found significantly to modify the negative pleiotropic effects. Doubled haploids were also produced from a F1 cross and tested in the field during two years, one with strong and one with weak mildew infections. In the first year only an insignificant reduction in grain yield was detected in the mlo lines, in the second it was significantly reduced with 5.9%. Thousand grain weights were, however, higher in the resistant lines. In both series of experiments the possibility to extract very high yielding mlo genotypes was demonstrated. Doubled-haploid breeding systems are typically based on sampling gametes from F1 plants. However, in the case of repulsion linkages, additional recombination could be advantageous. Pre-selection of gamete donors might also shift progeny performance in a desired direction. The objectives of this study were to quantify the effects of an additional round of recombination and assess the effectiveness of pre-anthesis selection in the production of barley doubled haploids. Assessments were conducted on: 1. 100 F1-derived lines representing a subset of lines used in previous genome-mapping studies; 2. 100 random F2-derived lines; and 3. 50 F2-derived lines from gamete donors selected for early heading. An additional round of recombination had only a modest effect on generating more favourable genotypes. Pre-anthesis selection was ineffective in generating an earlier heading population. According to published quantitative-trait locus (QTL) analyses based on the F1 -derived population, there are few repulsion linkages between QTL determining the traits measured in this experiment. Any advantages to be gained from postponing the generation of derivation of doubled haploids must be weighed against the delay and additional cost.  相似文献

Summary The objective of this study was to evaluate traits which can facilitate and improve selection for grain yield of spring barley. Five experiments were conducted in different environments to measure grain yield and yield related traits of breeding lines and exotic varieties. Differences for rate of canopy expansion were significant and offer the opportunity to select for a high weed suppressing potential but there was no relation to grain yield. Dry matter yield/m2 at anthesis and its water-soluble-carbohydrate content were not correlated with grain yield/m2 and number of grains/m2. Variation in biomass among breeding lines with a similar development and plant height was small. Biomass standardized for plant height was stable across environments and showed a good correlation with number of grains and grain yield. The contribution of pre-anthesis assimilation to grain yield was only important under low yielding experimental conditions, but the differences among the genotypes for this trait were inconsistent. It may be difficult to select genotypes with a high potential contribution of pre-anthesis assimilation to grain yield.  相似文献

Summary A haploid breeding program was initiated to develop doubled haploid salt tolerant rice breeding line via anther culture. Two sensitive breeding lines BR4608-R1-R2 and BR4909-R1-R2 were crossed with a salt tolerant line IR13146-13-3-3 to transfer its salt tolerant character to the doubled haploids.Anther from confirmed F1s of the two crosses were cultured in defined medium for callus induction and eventual plant regeneration. Fifteen doubled haploid (DH) lines were obtained from two crosses. Test for salt tolerance were done in vitro. Five out of 15 lines were found tolerant at the level of 8–10 decisiemens/m (ds/m) while the rests were sensitive to that level of salinity.Field experiment was conducted to evaluate the doubled haploids under saline and non saline soil. Summary The effect of anther orientation on culture response has been examined in a range of barley cultivars of agronomic importance. Anther orientation did not significantly affect the percentage of anthers responding. However, orientation of the anthers in the up position with one lobe in contact with the medium significantly increased the number of green plants produced when compared to anthers cultured in the flat position. Green plantlet production was observed directly from embryoids i.e. without an intermediate callus phase. Cytological examination of the regenerants indicated that the majority of the plantlets possessed the normal barley karyotype. The implications of these results for Barley breeding are discussed.  相似文献

Summary Progeny lines of chromosome-doubled haploids from crosses between one winter and four spring barley varieties were analysed for winter hardiness and vernalization requirement in the field. About one quarter or less of the offspring lines required vernalization under field conditions showing that winter versus spring habit of growth is controlled by at least two pairs of genes. The vernalization requirement could be measured quantitatively by testing a few plants in a greenhouse after artificial cold treatment during germination. The individual winter lines tested required from two to seven weeks of vernalization in order to flower as early as possible. The cold treatment had no effect on the time of flowering in the spring lines.Winter hardiness varied from zero to 100% plant survival in the field. The majority of the lines requiring vernalization survived better than 90% and the survival of the rest was at least 60%. The winter survival of the spring lines varied from zero per cent (about one third of the lines) to 100%. Hence, it is possible to obtain lines of good winter hardiness without requirements for cold treatment. Conversely, a selection for vernalization requirement has a positive effect on winter hardiness.  相似文献
