The efficacy of four chemical reagents, iodophor, formalin, hydrogen peroxide and bronopol as fish egg surface disinfectants were evaluated in bluefin sea bream (Sparidentex hasta). Fertilized eggs were counted and subjected to a static bath dip treatment in different concentrations of the above chemicals for 4 min before being incubated at 20 ± 0.5°C for 40 h. Treatment efficacy of the different disinfectants was evaluated by assessing the bactericidal activity, egg hatch percentage and survival of larvae up to 3 days post hatch. Results showed that iodophor at medium concentrations (75 and 100 ppm) was the best of all tested disinfectants in bacterial killing ability (12% reduction in the bacterial counts), egg hatching per cent (99.8% and 99.6% respectively) and larval survival up to 3 days post hatch (50.8% and 54.8% respectively). Formalin was the second best disinfectant at levels of 100 and 150 ppm. Hydrogen peroxide gave good results compared with the control while, bronopol showed discouraging results. In conclusion, iodophor appeared to be suitable for bluefin sea bream eggs disinfection with a 4 min exposure to 75–100 ppm when applied 14–16 h after egg fertilization.  相似文献

Chill tolerance was determined for various larval stages of Artemia salina with and without the addition of single cryoprotective agents (glycerol, glucose, sucrose and dimethylsulfoxide). Synchronous cultures of larvae were removed from the batch culture at post-hatch intervals (0, 3, 6, 12 and 24 h) and instantaneously exposed to solutions at −1°C for 10, 30 or 90 min intervals. Survival of larvae placed in freezing solutions without cryoprotectant (control group) varied with larval age. In zero-hour post-hatch controls larval survival decreased linearily with increasing exposure time. Each older stage (control group), however, was insensitive to the duration of −1°C exposure. The addition of cryoprotectant to the chilled solutions radically modified survival patterns and yielded results dependent on age, cryoprotectant concentration, and exposure time. Survival in cryoprotectant solutions sometimes exceeded 200% of survival in control solutions. Three hour post-hatch specimens were generally most tolerant to chilling in cryoprotectant solutions, while 12 h nauplii were least tolerant. The significance of these results for the development of cryogenic larval banking techniques is discussed.  相似文献

Fungal and parasitic infections of fish can significantly impact the survival of cultured fish. Formalin is currently used to control such infections; however, concern has arisen over its safety to users and to the environment. Hydrogen peroxide has been designated as a low priority fungicidal drug by the United States Food and Drug Administration (FDA), yet, little information is available on treatment concentrations or its toxicity to trout. Rainbow trout Oncorhynchus mykiss and cutthroat trout Oncorhynchus clarki fry and Angerlings were exposed to hydrogen peroxide concentrations of 0, 70, 170, 280, 420 and 540 ppm for 30, 60 or 120 rnin at 15 C to determine the chemical's toxicity. Rainbow trout fry and fingerlings experienced elevated mortalities (>20%) during treatments using 420 and 540 ppm for 30 min; 280, 420 and 540 ppm for 60 min; and >170 ppm for 120 min. Cutthroat trout fry experienced elevated mortalities (>23%) during treatments using 540 ppm for 30 min; 420 and 540 ppm for 60 min; and >170 ppm for 120 min. Cutthroat trout fingerlings experienced elevated mortalities (>60%) during treatments using 540 ppm for 60 min and >280 ppm for 120 min. No control mortalities were encountered for both life stages of either species. The lethal concentrations (LC50) of both age classes and species for each of the three durations ranged from 514–636 ppm for 30 min treatments, 322–506 ppm for 60 min treatments, and 189–280 for 120 min treatments. Mortalities for all four toxicity tests which occurred during a 96-h post-treatment period were centered around the following treatments: 30 min, 540 ppm; 60 min, 280–540 ppm; 120 min, 170 ppm. Tissue damage to gills was found only among fish that did not survive the initial chemical exposure. Test concentrations proved to be relatively stable during a 24-h period, retaining better than 85% of their original strength for all five dilutions. At a water temperature of 15 C concentrations should not exceed 280 ppm for a 30-min treatment.  相似文献

Larval sinking causes larval mass mortality during seed production in the horsehair crab, Erimacrus isenbeckii. Under normal light conditions, horsehair crab larvae generally show negative phototactic behaviour and sink in their rearing seawater. It has been proposed that culturing larvae in the dark may prevent larval sinking. Herein, we examined the effect of photoperiods on horsehair crab larval survival and development to facilitate the development of larval rearing techniques that prevent sinking. Newly hatched larvae were reared with Artemia to the first crab stage in 2‐L beakers under five photoperiods: 0L:24D, 6L:18D, 12L:12D, 18L:6D and 24L:0D. Larvae survived and molted to the first crab stage under all tested photoperiod conditions. The survival rate improved with increasing light period, whereas the developmental period for each larval stage decreased with increasing light period. Longer light periods increased the carapace length at the first crab stage. Our results suggest that larvae could be cultured to the first crab stage in large‐scale tanks under constant darkness. However, significantly improved larval performance under longer photoperiodic conditions indicates a need for developing alternative culture techniques to control larval behaviour in the seed production tank.  相似文献

为了得到壳型规则、大小均一的单体牡蛎,本研究采用肾上腺素对岩牡蛎单体苗种生产的诱导条件进行研究,阐明了岩牡蛎单体苗种的最适诱导浓度、诱导时间和诱导密度。结果显示,肾上腺素能显著诱导岩牡蛎幼虫的不固着变态,最适诱导浓度为5×10–5 mol/L,最适诱导时间为1 h,提高诱导浓度和延长诱导时间导致岩牡蛎幼虫的不固着变态率、稚贝壳高和存活率显著降低;肾上腺素对低于8个/mL幼虫密度的诱导效果差异不显著,但稚贝壳高和存活率在8个/mL的培育密度下显著低于0.5~4个/mL,研究表明利用肾上腺素诱导岩牡蛎单体时,可大批量处理眼点幼虫,但稚贝充气培养的最适培养密度应不高于4个/mL。  相似文献

To determine the optimal condition of photoperiod for embryo development and larval culture of tawny puffer, Takifugu flavidus, the effects of photoperiod on egg hatching and the growth and survival of larvae from 3 to 23 d after hatching (dah) were assessed. The results show that photoperiodic responses varied with the developmental stage during the early development of tawny puffer. Photoperiod did not significantly affect the hatch rate, viability of 24 h post‐hatch larvae, and total mortality rate of tawny puffer embryos; however, it affected the incubation period, which significantly increased with increasing photoperiod from 6 to 24 h. The shortest incubation period was predicted to occur at 3.56 h day length from the quadratic relationship. However, photoperiod strongly affected the growth and survival of the tawny puffer larvae from 3 to 23 dah. Increasing the day length resulted in improvement of the larval growth and survival within the 0–12 h day length range; however, beyond the 12 h day length, a longer day length was not beneficial, and a 24 h continuous light had a negative effect on growth. From the quadratic relationships, the highest growth and survival rates were predicted to occur at 14.16 h and 17.74 h day length, respectively. Therefore, the optimal photoperiods for tawny puffer embryos and larvae were 3–6 and 12–18 h day length, respectively. The results of this study are useful in increasing the production of the species during incubation and larval culture.  相似文献

Chlorine and iodine solutions were effective at inactivating Ichthyophonus spores in vitro. Inactivation in sea water increased directly with halogen concentration and exposure duration, with significant differences (P < 0.05) from controls occurring at all chlorine concentrations and exposure durations tested (1.5–13.3 ppm for 1–60 min) and at most iodine concentrations and exposure durations tested (1.2 ppm for 60 min and 5.9–10.7 ppm for 1–60 min). However, 10‐fold reductions in spore viability occurred only after exposure to halogen solutions at higher concentrations and/or longer durations (13 ppm total chlorine for 1–60 min, 5.9 ppm total iodine for 60 min, and 10.7 ppm total iodine for 1–60 min). Inactivation efficacy was greater when halogen solutions were prepared in fresh water, presumably because of combined effects of halogen‐induced inactivation and general spore instability in fresh water. The results have practical implications for disinfection and biocontainment in research laboratories and other facilities that handle live Ichthyophonus cultures and/or infected fish.  相似文献

The effects of Cd and Cu on embryos and larvae of the ide Leuciscus idus were evaluated. The ide is an European cyprinid fish, natural populations of which tend to decrease. The ide is also used as a bioindicator organism to evaluate water quality. However, sensitivity of ide early developmental stages to heavy metal intoxication is not known. Fish were exposed to Cd or Cu (100 μg/L) during embryonic, larval or both developmental periods. Survival of the embryos, time of hatching, size and quality of newly hatched larvae were evaluated at the end of embryonic period. Correctly developed larvae from the control and Cd or Cu-exposed groups were transferred to clean water, Cd or Cu solutions (100 μg/L) immediately after hatching. Larval development was observed, and the larvae were photographed. Time of yolk sac resorption, onset of active feeding and swim bladder inflation were evaluated, and the measurements were done on body and swim bladder size. The results showed that exposure of embryos to Cd and Cu significantly reduced embryonic survival and increased frequency of body malformations and death in newly hatched larvae and delayed hatching. Exposure to Cd and Cu during larval period reduced larval survival, growth and delayed development (yolk utilization, beginning of active feeding and swim bladder inflation). Cadmium was more toxic to the ide embryos and larvae than copper. Exposures to metals during embryonic period alone caused adverse impact on larval performance even when larval development took place in clean water. However, exposure of embryos to Cu reduced toxic impact of metal on larvae in continuous Cu exposure compared to the non-preexposed fish, but no such an effect occurred in case of Cd exposure. The results show that even a short-term exposure to Cd or Cu during early development of ide may adversely affect recruitment of this species. Among the measured endpoints, quality of newly hatched larvae (frequency of body malformations and larvae dead immediately after hatching) and swim bladder size were the most sensitive to intoxication with both metals. Embryos were more sensitive to Cu intoxication than larvae, while in case of Cd, sensitivity of both stages was similar.  相似文献

Exposure of brown trout to a single dose of malachite green (2.27 ppm) caused a significant elevation of blood cortisol levels within 1 h. After 4 weeks of daily exposure to malachite green, however, this stress response was totally abolished. Daily treatment had no long-term effects on basal plasma cortisol levels, or on the numbers of circulating erythrocytes, thrombocytes and lymphocytes. Blood neutrophil numbers were significantly reduced in malachite green-treated fish, an effect thought to be due to a decreased ectoparasitic load.  相似文献

As alternative to formalin, the antifungal effect of a plant product [Origanum onites L. (Lamiaceae) oil] was investigated for use in the artificial incubation of narrow‐clawed crayfish (Astacus leptodactylus Eschscholtz) eggs. For this purpose, this study was conducted as two experiments. In experiment I, the eggs were artificially incubated for 40 days. In experiment II, juveniles were cultured to determine effects of O. onites oil on juveniles for 30 days. The experimental groups were as follows: formalin (3500 ppm for 15 min), O. onites oil (300 ppm for 15 min, 700 ppm for 2 min and 1000 ppm as a dip treatment 15 split‐second) and a control (no treatment). In the experiment I, the highest hatching rate (86%) and survival rate of stage II juveniles (80%) were observed in 1000 ppm dip group. These results were similar to that of formalin group (85% and 79%) respectively. The control group exhibited the lowest hatching rate (49%) and stage II rate (42%) compared with the 1000 ppm dip group and 3500 ppm formalin treatments. However, other concentrations (300 and 700 ppm) of O. onites showed toxic effects on the eggs and there was no hatching. In the experiment II, the survival rate and growth performance of the crayfish juveniles were similar in all groups. This study indicated that the 1000 ppm O. onites dip treatment could be a good alternative to formalin for improved egg hatchability in the artificial incubation of crayfish eggs.  相似文献

The southern flounder (Paralichthys lethostigma) is a commercially important marine flatfish from the southeastern Atlantic and Gulf Coasts of the USA and an attractive candidate for aquaculture. Hatchery methods are relatively well developed for southern flounder; however, knowledge of the optimum environmental conditions for culturing the larval stages is needed to make these technologies more cost effective. The objectives of this study were to determine the effects of water turbulence (as controlled by varying rates of diffused aeration) on growth, survival, and whole‐body osmolality of larval southern flounder from hatching through day 16 posthatching (d16ph). Embryos were stocked into black 15‐L cylindrical tanks under four turbulence levels (20, 90, 170, and 250 mL/min of diffused aeration) and two salinities (24 and 35 ppt) in a 4 × 2 factorial design. Larvae were provided with enriched s‐type rotifers from d2ph at a density of 10 individuals/mL. Temperature was 19 C, light intensity was 390 lx, and photoperiod was 18 L:6 D. Significant (P < 0.05) effects of turbulence on growth (notochord length [NL], wet weight, and dry weight) were observed. On d16ph, NL (μm) increased with decreasing turbulence level and was significantly greater at 20 mL/min (64.2) and 90 mL/min (58.2) than at 170 mL/min (56.3) and 250 mL/min (57.2). Survival declined primarily during the prefeeding and first‐feeding stages from d0 to d8ph, then stabilized from d8 to d16ph. In contrast to growth trends, survival (%) on d16ph increased with increasing turbulence levels and was significantly greater at 170 mL/min (57.9) and 250 mL/min (54.0) than at 20 and 90 mL/min (21.4 and 26.2, respectively). Mean rotifer concentrations (individuals/mL) at 24 h postfeeding were significantly higher (P < 0.05) in the low‐turbulence treatments of 20 mL/min (4.48) and 90 mL/min (4.23) than in the high‐turbulence treatments of 170 and 250 mL/min (2.28 and 2.45, respectively). Under both salinities, larval whole‐body osmolality (mOsm/kg) increased with increasing turbulence levels and was significantly higher at 250 mL/min (427) than at 20 mL/min (381), indicating osmoregulatory stress at the higher turbulence levels. On d14ph, larvae in all treatments were positively buoyant in 35 ppt and negatively buoyant in 24 ppt. Results showed that growth of southern flounder larvae in 15‐L tanks was maximized under low turbulence levels of 20 and 90 mL/min, while survival was maximized at high turbulence levels of 170 and 250 mL/min. The data suggested that, in prefeeding‐ and early‐feeding‐stage larvae (which have weak swimming ability), higher turbulence levels improved buoyancy and prevented sinking. In feeding‐stage larvae (which are relatively strong swimmers), higher turbulence levels caused excessive swimming, osmoregulatory stress, and slower growth. Based on these results, we recommend that turbulence levels be maintained relatively high during prefeeding (yolk sac) and first‐feeding stages to maintain buoyancy and survival and then decreased for mid‐ to late‐feeding‐ and premetamorphic stage larvae to optimize prey encounters and feeding efficiency.  相似文献

Abstract. A novel method using 6-dimethylaminopurine (6-DMAP) has recently been demonstrated to be a good triploid inductor for bivalve molluscs. The effects of 6-DMAP concentration, timing of treatment after fertilization, and treatment duration on survival and triploid induction of Crassostrea gigas (Thunberg) embryos were examined. An influence of sperm concentration was also detected and discussed. Survival to D-stage was inversely related to 6-DMAP concentration and the percentage of triploids was shown to be 6-DMAP dose dependent. Two optimal moments for treatment application were determined, corresponding to extrusions of the first and second polar bodies. Optimal treatment of 450μmol/1 6-DMAP beginning 15min after fertilization over a 10min period yielded mean survival to D-stage of 64% with a mean of 85% triploid production. As a comparison, a sample treated with 1 mg/1 of cytochalasin B (CB), 20 min after fertilization over a 15 min period, yielded a mean survival to D-stage of only 36% with a mean of 95% triploid production. The advantages of 6-DMAP over CB are clearly identified: this chemical is not carcinogenic, cheaper than cytochalasin B and much easier to use as it is water soluble.  相似文献

本文研究了青霉素钾盐和硫酸链霉素(双抗)混合使用对紫彩血蛤(Nutallia olivacea)幼虫发育的影响。结果表明:1~80ppm的双抗均有很好的抑菌效果,均可显著地提高幼虫的存活率,最高可提高52%,最低也可提高38%,同时对幼虫生长和变态均有明显的促进作用。其中以2~4ppm效果最好,幼虫生长最快,变态率最高;高浓度组对幼虫也无明显的副作用,幼虫生长速度、变态率和存活率也都高于对照组。  相似文献

采用高度不饱和脂肪酸(HUFA)营养强化的轮虫、轮虫与卤虫组织投喂中华绒螯蟹幼体,以各项幼体的存活率为评判指标,对不同饵料及其组合的投喂效果进行了探讨。研究结果表明:轮虫是中华绒螯解早期幼体(Ⅰ、Ⅱ期)的适口饵料,幼体存活率随轮虫密度的增加而逐渐上升,但当轮虫数量超过最适密度时,幼体存活率反而有所下降。其中,未强化的轮虫达投喂密度为60ind.mL^-1,强化轮虫最适密度为40ind.mL^-1。Zao状Ⅰ、Ⅱ期投喂40ind.mL^-1轮虫,从Ⅲ期开始投喂10ing.mL^-1卤虫无节幼体,能较好地满足中华绒螯蟹幼体发育的营养需求,提高大眼幼体的存活率。表明强化幼体饵料HUFA特别是EPA和DHA能有效地促进幼体的发育与存活和脱壳率。  相似文献

The sodium adsorption ratio (SAR) is used to describe the ratio of sodium to calcium and magnesium in water that is very important in both aquaculture and agriculture. SAR treatments including of 20, 30, 40, 50 and 60 were adjusted in 10‐L containers. Initial larval density was fixed at 100 larvae L?1 and during the culture period, various larval quality parameters were evaluated. Our findings illustrated that at initial stages of larval development, different SAR treatments affected the larval condition index (LCI), but no influences on larval stage index (LSI) were apparent. Importantly, this process at the final larval stages became reversed. In addition, survival at SAR 30 was 12.4% greater than among any other treatments. The highest larval resistance to stress (191 ± 5 ppm), as measured by the 24‐h LC50 formalin test also was observed for SAR 30, at the 11th larval stage. Analysis indicated that calcium absorption during larval developmental stages is affected by SAR levels in the environment and also showed that the optimal SAR treatment was 30 for Macrobrachium rosenbergii larviculture in inland hatchery brackish water. According to our findings and desired SAR for the environment and agricultural soil (<15), effluent of M. rosenbergii inland hatcheries must be treated at least two times by dilution and adding calcium and magnesium salts.  相似文献

The aim of this study was to evaluate the effects of hatchery‐tank colours (white, yellow, red, blue, green and black) on the performance of larval culture of Macrobrachium amazonicum. The larvae were fed daily with newly hatched Artemia nauplii. The hatchery‐tank colours affected the light level inside the tanks, the consumption of Artemia nauplii (AN), larval development, survival, mass gain and productivity of postlarvae (PL). The overall consumption of Artemia nauplii per larva during the larval cycle was 30% and 45% higher in the green and red tanks respectively. The significant variation of AN consumption among tank colours (= 0.0006) indicates that M. amazonicum larvae are visual predators. Survival was higher in the black, blue and green tanks, reaching more than 75%. However, the highest productivity was obtained in the black tanks (80.1 PL L?1). Lighter coloured tanks and excess luminosity (more than 2 μmol s?1 m2 at tank bottom) appear to be important stress factors for larvae, contributing to reduce survival and productivity. The results indicate that rearing M. amazonicum in black tanks will improve larvae condition, ensure greater productivity of postlarvae and lower Artemia consumption, increasing technological and economic viability.  相似文献

In recent years, an increasing interest in the farming of marine ‘white’ fish, such as cod, has been observed in the aquaculture industry. One species being considered for aquaculture development is whiting (Merlangius merlangus L.). With little data being available on the domestication of this species, our study aimed to characterize spawning behaviour and rhythmicity in captive broodstock and provide original data on egg incubation, embryonic development and larval rearing. The results observed through video recording showed that spawning mainly occurred at night with active courtship behaviour. Daily egg batches were collected from two separate spawning seasons, with an average fertilization success declining from 77% to 53%. Time to hatch was inversely related to the incubation temperature and occurred after 78 degree‐days (9 days at 8 °C), with emerging larvae ranging in size from 3.3 to 3.46 mm total length and hatch success ranging from 48.3% to 99.9%. In comparison with a clear water environment, a green water regime during the first days of larval rearing significantly improved larval growth and survival. These results are the first to describe spawning activity, early development and larval performances in whiting and are promising for the potential future development of whiting aquaculture.  相似文献

A series of ingestion trials were conducted to determine the ingestion rate of Artemia nauplii by Eriocheir sinensis zoea larvae with increasing densities of Artemia and with or without rotifers as a co-feed. At each zoeal stage, 10 groups of 10 larvae were reared individually in glass beakers and fed with increasing densities of newly hatched Artemia nauplii (0.5, 2.5, 5, 10 and 20 individual (ind.) mL−1) with or without rotifers (15–25 ind. mL−1) as a co-feed. The average number of ingested Artemia was measured over 24 h. In addition, the average larval development rate (Larval Stage Index, LSI) over a longer period (time needed for the best treatment to reach 100% moult or metamorphosis to the next larval stage) was compared. The results showed that Artemia ingestion rate of E. sinensis larvae increased with increasing prey densities and larval development, and had a significantly negative correlation with rotifer consumption for all zoeal stages. Rotifers as an alternative prey significantly affected the intake of Artemia at early larval stages (Z1 and Z2) and promoted LSI at a lower Artemia density. Further experiments are needed to clarify the effect of prey density on survival and larval development when larvae are reared communally.  相似文献

在室内越冬的中华绒螯蟹亲蟹上检出一种嗜腐性纤毛虫,经鉴定为蟹栖拟阿脑虫Paranophrgs carci-ni。在感染初期,多寄生于新蟹残肢等伤口组织处;在感染后期,侵蚀到蟹体,进入血淋巴,甚至可分布全身,致使器官组织受到严重损害,造成亲蟹死亡。对越冬亲蟹危害严重,短期内使亲蟹死亡率达40%左右。用15~30ppm的福尔马林或0.1~0.5ppm的孔雀石绿短期药治无效,改用25ppm的福尔马林或0.3~0.5ppm的孔雀石绿每天药浴8h,连续10~15d,病情基本得到控制。  相似文献

Microorganisms present in the rearing water colonize the gut of first feeding larvae and represent the first barrier against opportunistic pathogens. The aim of the experiments presented herein was to standardize a protocol for the management of rearing water and microalgae suitable for the larval rearing of common dentex. In Experiment 1, bacteria–algae interactions were tested using a monospecific microalgal community, Tetraselmis chuii, suitable for nutritional experiments and with known antibacterial activity. In Experiment 2, the evolution of the bacterial community and larval performance (growth and survival) were monitored daily, in three conditions: (1) mature water: T. chuii was added 5 d before the rearing of common dentex larvae, (2) green water: T. chuii was added 1 d before, and (3) clear water: no T. chuii addition. The results show the influence of the presence of T. chuii on the evolution of the bacterial communities, both in terms of bacterial density and morphology, and indicate green water is the most suitable water treatment for management of larval rearing for common dentex.  相似文献
