Twenty-one healthy Thoroughbred and Quarter Horse foals were studied from birth until 1 year of age. Foals had access to an iron-supplemented creep feed before weaning and were fed an iron-supplemented concentrate as part of their diet after weaning at 4 months of age. Initial blood samples were taken before foals were allowed to nurse. Serum iron concentration, total iron-binding capacity, and PCV decreased during the foal's first 24 hours of life. Serum iron concentration decreased rapidly from 446 +/- 16 micrograms/dl (mean +/- SE) at birth to 105 +/- 11 micrograms/dl at 3 days of age. Serum ferritin concentration increased from a mean of 85 +/- 8 ng/ml at birth to 159 +/- 11 ng/ml at 1 day of age. Thereafter, ferritin concentration decreased gradually to a minimum of 61 +/- 6 ng/ml at 3 weeks of age, and then at 6 months increased to values similar to those from reference adult horses. The ferritin concentration in colostrum at birth was 354 +/- 42 ng/ml, compared with 25 +/- 2 ng/ml in milk 1 day later. The decrease and then increase in serum ferritin concentration occurred concomitantly with opposite changes in serum total iron-binding capacity. The mean PCV decreased gradually to a minimum at 3 months of age. This decrease was associated with an increasing number of microcytes, as determined with a cell-size distribution analyzer.  相似文献

At birth, 24 Standardbred foals were assigned at random to 1 of 2 groups and were given a placebo supplement (group 1) or an iron supplement (248 mg of iron/treatment; group 2). Foals were given iron supplement or placebo 4 times during the second and third weeks after birth. Hematologic variables and general health were monitored until foals were 4 months old. Mean PCV in foals of both groups decreased during the first 2 weeks after birth, but values remained within adult horse reference ranges. During the first 6 weeks after birth, foal erythrocytes were smaller than adult horse erythrocytes, but foal erythrocyte glucose-6-phosphate dehydrogenase activity was greater than that in adult horses. At every measurement, indices of anisocytosis were lower in foals, compared with adult horse reference values, suggesting that foals have a homogeneous population of microcytic erythrocytes during early foalhood. In 2-week-old foals of both groups and in 4-week-old placebo-treated foals, mean serum iron concentration was lower than that in adult horses. In foals at birth and during the first 4 months, total iron-binding capacity values were above the adult reference range. In newborn foals, transferrin saturation percentage values decreased to below the reference range in foals from 2 weeks to 4 months after birth. When foals were born, serum ferritin concentration values were above the adult horse reference range, but decreased to within the reference range by the time foals were 1 day old. The injection of iron-dextran into beef calves at an early age immediately increased values for erythrocyte (RBC) numbers, hemoglobin (Hb) concentration, packed cell volume (PCV), mean corpuscular hemoglobin (MCH) and mean corpuscular volume (MCV), even though the calves were not anemic. There were no significant differences in average daily gain (ADG) from birth through 15 wk between those that were injected and those that were not injected. Hereford calves had lower values for PCV, Hb, MCH and MCV than Angus and crossbred calves, and both Hereford and Angus calves had lesser body weight and slower ADG than crossbred calves. Serum iron (Fe) concentrations were higher in calves born in the spring and pastured in the summer than in calves born in late summer and fall and pastured in the fall. The serum Fe concentration of calves at first sampling (during first week of life) was positively correlated with their weight at that and subsequent times to 15 wk. Mean corpuscular volume and MCH in cows before calving were positively correlated with the MCV and MCH in their calves at first sampling. Serum Fe concentration in cows before calving was negatively correlated with the ADG of their calves through 15 wk.  相似文献

Microcytosis is common in dogs with congenital portosystemic shunts (PSS) and acquired liver disease. The objective of this study was to determine if microcytosis could be induced in normal dogs by surgical creation of PSS, and to characterize the changes in hematology and iron status. Hematocrit, mean cell volume, mean cell hemoglobin, and mean cell hemoglobin concentration decreased linearly from 45.5%. 69.1 fL, 22.8 g/dL and 33.1% to 39.5%. 55.9 fL, 17.8 g/dL and 31.9%. respectively, 18 weeks after creation of PSS. The erythrocyte count did not change, but red cell distribution widths indicated a shift to a heterogenous population with decreased volume. Mean cell volume and mean cell hemoglobin decreased rapidly after induction of PSS and were significantly ( P < .05) different from presurgery values within 2 weeks. Serum iron and copper concentrations and total iron binding capacity were decreased in dogs with PSS. Liver iron concentration doubled after creation of PSS, with the majority of stainable iron located in Kupffer cells. The changes in erythrocyte indices and measures of iron status in dogs with surgically induced PSS were similar to those in dogs with congenital PSS. Microcytosis developed rapidly in dogs after induction of PSS. These results indicate that iron deficiency was not the cause of microcytosis in these dogs.  相似文献

Seventeen healthy Thoroughbred and Quarter Horse foals were divided into control (n=8) and treated (n=9) groups and studied from birth until 4 months of age. Foals in the treated group were intravenously injected with an experimental drug containing 0.4 g of iron in the form of a polynuclear complex of carbohydrate and ferric hydroxide on days 7, 14, 21 and 28, for a total dosage of 1.6 g of iron. Foals had access to Bermuda grass hay and Bahia pasture. A creep feed formulated with no trace mineral supplement was provided after two months of age. Hemoglobin concentration, hematecrit, erythrocyte counts, mean cell volume, microcyte numbers, plasma protein, fibrinogen, total leucocyte count, serum iron, total iron binding capacity and serum ferritin values were determined at 8 different ages. On the basis of analysis of variance at P<0.05, no significant differences were found between groups for any of these parameters, however, there were changes in parameters over time in both groups. Echinocytes have been incriminated in the pathogenesis of exertional diseases in horses. To evaluate the hypothesis that echinocytes are dehydrated erythrocytes, we decreased blood sodium and potassium concentrations in 4 horses by administering furosemide (1.0 mg/kg of body weight, q 12 h) for 2 days and we monitored CBC, serum and erythrocyte sodium and potassium concentrations, and echinocyte numbers. Serum sodium concentration decreased progressively over the 48 hours of furosemide administration, then returned to near baseline concentration at 168 hours. A statistically significant decrease (P < 0.05) in serum potassium concentration was observed at 24, 48, and 72 hours after initial furosemide administration, and remained less than the baseline value at the end of the study. Mean erythrocyte potassium concentration decreased rapidly and remained low at the end of the study. Minimal changes were observed in erythrocyte sodium concentration during the first 72 hours after furosemide administration, but the value was significantly (P < 0.05) increased at 168 hours. Type-I and type-II echinocyte numbers increased by 4 hours after furosemide administration and persisted throughout the study. Type-III echinocytes were not seen in baseline samples, but numbers increased only modestly after furosemide administration. Administration of epinephrine to well-hydrated horses increased echinocyte numbers only minimally, indicating that splenic contraction was not the likely cause for the furosemide-associated increase. To determine whether the decrease in erythrocyte potassium concentration and increase in sodium concentration was caused by furosemide acting directly on the erythrocyte membrane, we quantified erythrocyte potassium and sodium concentrations before and after incubation with furosemide in vitro.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Blood samples were collected, at one day of age and at weekly intervals from one through 25 weeks of age, from seven white-tailed fawns (Odocoileus virginianus) to determine the effect of age upon serum biochemical and hematological values. Serum total protein concentration increased continually during the six month period. The rate of anabolism of serum gamma-globulin exceeded the rate of catabolism at about seven weeks of age. Serum cholesterol concentration more than doubled from one day to eight weeks of age. Blood hemoglobin concentration and packed cell volume increased markedly during the first eight weeks of life, but tended to remain constant from eight to 25 weeks of age. Blood erythrocyte count increased throughout the duration of the study; however, the relative increase was greater during the first eight weeks of age than thereafter.  相似文献

Iron deficiency anemia was identified and characterized in three 14 to 29-month-old male llamas (llama Nos. 1-3) from separate herds in Colorado. The identification of iron deficiency anemia was based on hypoferremia (serum iron = 20-60 micrograms/dl), erythrocytic features, and hematologic response to iron therapy. The anemia was moderate and nonregenerative and characterized by erythrocyte hypochromia, microcytosis (mean cell volume = 15-18 fl), and decreased mean corpuscular hemoglobin concentration (36.0-41.0 g/dl). Morphologic features unique to llamas with iron deficiency anemia included irregular distribution of hypochromia within erythrocytes and increased folded cells and dacryocytes. The cause of iron deficiency was not determined. The llamas were treated with various doses and schedules of parenteral iron dextran. Two of the llamas were monitored for up to 14 months after the start of iron therapy and experienced increases in hematocrit and mean cell volume values. In one llama, progressive replacement of microcytic cells with normal cells was visualized on sequential erythrocyte volume distribution histograms following iron therapy.  相似文献

Serum and plasma samples were collected from 92 gray seal (Halichoerus grypus) pups, all less than 5 wk old, and from 21 yearlings during the breeding season at the Isle of May, Firth of Forth, Scotland, for hematologic and clinical chemistry analysis. In the pups, total WBC, total protein, and glucose values increased until weaning and declined during the postweaning fast. Liver enzymes, particularly alkaline phosphatase, were high at birth and decreased steadily with age, most rapidly during the postweaning fast. In the yearlings, total WBC, erythrocyte sedimentation rates, and rectal temperatures were significantly higher than in the pups and glucose levels were significantly lower. Therefore, consideration should be given to the growth phase of gray seal pups (lactation or postweaning fast) or stage of development when hematology and clinical chemistries are used for diagnostic purposes.  相似文献

Twelve calves of the Red Spotted breed and its crossbreds born by first-calvers, kept in a large-capacity stable of a dairy farm, were subject to the study of the ontogenetic development of the values of the blood picture from birth up to the 10th day of age. The erythrocyte and leucocyte count, the packed cell volume and the relative and absolute number of neutrophils reached the significantly highest levels in calves before drinking the colostrum. The amount of haemoglobin, MCV and MCHC reached the highest values on the first day after birth. The values of the red blood picture decreased in the first week of life and reached the minimum on the tenth day. The leucocyte count decreased during the first to second day after birth, the decrease being due to a drop of the absolute number of neutrophils, and on the tenth day the leucocyte count returned to its initial value. Neutrophils prevailed in the peripheral blood of new-born calves whereas lymphocytes prevailed in the peripheral blood from the fourth day of life on. The amount of cosinophiles in the peripheral blood of the calves was about 0.5% up to the fourth day; on the seventh day it exceeded 1%. The relative number of monocytes was at its minimum level just after birth and showed a significant increase during the second to fourth day.  相似文献

Eighteen 9- to 10-week old Beagles were fed casein-based diets (4,710 kcal of metabolizable energy/kg of body weight) containing either 12, 80, or 160 mg of iron/kg of diet. Growth and feed consumption were monitored throughout the 47-day study. Hematocrit (Hct), hemoglobin (Hb) concentration, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), RBC numbers, erythrocyte protoporphyrin (EP) concentration, serum iron concentration, serum total iron-binding capacity (TIBC), and serum ferritin concentration were determined weekly. Growth rate and feed efficiency were not significantly influenced by dietary iron content. At 14 days, Hb concentration, Hct, MCV, MCH, RBC numbers, and serum iron concentration were significantly (P less than 0.05) lower in dogs fed the 12 mg/kg diet, and remained significantly low for the remainder of the study. Erythrocyte protoporphyrin concentration increased significantly (P less than 0.05) by 14 days in dogs fed the basal diet, and remained significantly high relative to that in dogs of the other dietary groups for the remainder of the study. Serum ferritin concentration decreased in dogs of the group fed the basal diet, with a significant (P less than 0.05) difference beyond day 42. Differences in Hct, MCH, MCV, or hemoglobin, serum iron, serum ferritin, or EP concentration were not found between groups fed 80 and 160 mg of iron/kg of diet. Infectious diseases are common in foals aged 1-5 months. The objectives of this investigation were to evaluate immunologic parameters in foals from birth to weaning to establish reference values for the proportion of circulating lymphocytes that were helper (CD4+) or cytotoxic (CD8+) T cells, or B cells; to measure serum immunoglobulin (IgM and IgG) concentrations; and to compare these immunologic parameters to values in foals with naturally occurring Rhodococcus equi pneumonia and in adult horses. Peripheral blood lymphocyte subpopulations were determined by flow cytometric analysis, and serum IgG and IgM concentrations were determined by radial immunodiffusion. Flow cytometric analysis of lymphocyte subpopulations suggested age-related changes in the cell-mediated immune system in horses. Absolute circulating CD4+ and CD8+ T lymphocytes and B cells increased linearly up to 3 months of age. Circulating B cell concentrations from birth to 6 months of age were greater than values in adult horses and the lymphocyte differences among the age groups are mainly due to variation in B lymphocytes. Both absolute and proportional B cell concentrations were greater in foals with R equi pneumonia than in healthy foals at the same age. The increase in absolute cell counts of each subpopulation was dependent on the increase of absolute peripheral blood lymphocyte count. Serum IgG concentration increased linearly from 1 to 3 months of age, and serum IgM concentrations increased from 1 to 6 months of age. These data suggest age-dependent cell-mediated and humoral development in young foals.  相似文献

Erythrocyte counts, haematocrit, haemoglobin concentration, red blood cell indices, total and differential leukocyte counts were determined in 202 cross-bred and 14 indigenous East African goats aged 6–12 months and also in 59 Norwegian dairy goats, of which 24 were 15–45 days old, 8 were 8 months old and 27 were over 3 years and pregnant. These were reared in Tanzania. Comparisons were made with 24 Dwarf and 57 Danish Landrace goats at 6–12 months of age and 76 adult pregnant Danish Landrace goats reared in Denmark. The purpose was to determine reference ranges for cross-bred and indigenous East African goats and to compare these with those of other breeds. The haemoglobin concentration, haematocrit, erythrocyte and white blood cells were lowest in the Norwegian kids. The highest values were observed in 6–12-month-old goats in all the breeds, whereafter they decreased to relatively constant adult levels. The mean corpuscular volumes were highest in kids followed by adult pregnant Norwegian and Danish Landrace goats, and lowest in 6–12 months old goats. East African and cross-bred goats had the smallest mean corpuscular volumes. The haemoglobin concentration, erythrocyte and leukocyte counts were highest in indigenous East African, followed by young Norwegian and cross-bred goats. The mean corpuscular haemoglobin concentration was highest in the cross-breds, while the mean corpuscular haemoglobin was higher in pregnant than in other goats. Number of platelets, leucocytes and packed red cell volume (PGV) have been determined in normal piglets during the first 2 weeks of life.A mean platelet count of 337 ± 79 at birth decreased during the first 2 days to 241 ± 48, followed by increasing numbers the next week to a maximum of 578 ±128 at 10 days after birth. Then a slight decrease to 492 ±115 on the 15th day was observed.The leucocyte counts were highest the first 30 hrs. of life, the mean count being 16.9. During this period, however, there was as wide a range as from 3.6 to 46.3. For the rest of the period the mean varied from 8.0 to 11.8, the counts ranging from 4.5 to 20.0.The mean PGV, which was highest at birth, decreased from a mean of 40.8 ± 6.1 to 32.9 ± 5.0 during the first 2 days. At 7 days of age the mean was 31.5 ± 3.8 but increased during the following week to 39.9 ± 3.6 on the 15th day.  相似文献

Hematologic abnormalities consistent with iron deficiency anemia were experimentally induced in two healthy llamas by repeated phlebotomy. Hematologic abnormalities included erythrocyte microcytosis and hypochromia, decreased hemoglobin concentration, hypoferremia, and decreased transferrin saturation. Erythrocyte volume distribution histograms were more sensitive than mean corpuscular volume values for detection of microcytosis. Hypochromia, which was often eccentric, was morphologically observed on Wright-Giemsa-stained blood films. Frequent folded erythrocytes and dacryocytes were also noted on the blood films. Hematologic abnormalities resolved rapidly after cessation of blood removal, without parenteral iron supplementation.  相似文献

OBJECTIVE: To characterize age-associated changes in lymphocyte population subsets and immunoglobulin isotypes. ANIMALS: 30 healthy young light-breed horses (5 to 12 years old) and 30 healthy aged light-breed horses (> 20 years old). PROCEDURE: Lymphocyte subset populations were identified, using monoclonal antibodies to cell surface markers CD5, CD4, CD8, and IgG. Subset populations were quantitated by use of flow cytometric analysis of antibody-stained cells. Serum immunoglobulin concentration was determined using single radial immunodiffusion. RESULTS: Absolute cell counts of total lymphocytes, T cells, CD4+ and CD8+ T cells, and B cells were decreased in aged horses, compared with young horses. There was a significant decrease in the percentage of CD8+ cells and an increase in the CD4+-to-CD8+ cell ratio in the aged population, compared with young horses. However, serum concentration of IgG, IgG(T), IgM, or IgA did not differ with age. CONCLUSIONS AND CLINICAL RELEVANCE: In horses, total lymphocyte count and lymphocyte subset cell counts decrease with age. Age-matched control values are necessary for optimal evaluation of hematologic variables in aged horses. The decrease in lymphocyte subset cell counts in healthy aged horses mimics that seen in other species and may contribute to an age-associated decrease in immunocompetency.  相似文献

Half-life and apparent specific volume of distribution of hexobarbital, sulphadimidine and chloramphenicol were investigated in newborn, 1, 3, 5 and 8 weeks old pigs. Hexobarbital sleeping time and plasma concentration of hexobarbital at recovery were measured in the same age groups. The half-life of hexobarbital and chloramphenicol was long in newborn pigs but decreased fast during the first week after birth. From 1 to 8 weeks after birth the decrease was less pronounced. The half-life of sulphadimidine increased during the first 3 weeks of life, but in 1 and 3 weeks old pigs the amount of N4-acetylated sulphadimidine in plasma at 200 min. after the injection was higher than in the newborn pigs.The apparent specific volume of distribution of hexobarbital, sulphadimidine and chloramphenicol was changed in different ways from birth to 8 weeks of age.The hexobarbital sleeping time was very long in the newborn pigs and decreased until 3 weeks of age. The concentration of hexobarbital in plasma at recovery was unchanged from birth to 8 weeks of age.The concentration of chloramphenicol metabolites in plasma 100 min. after the injection increased very fast during the 8 weeks of observation. The concentration of N4-acetylated sulphadimidine in plasma at 200 min. after the injection increased from birth to 1 week of age, then it decreased.The data are stressing that the neonatal pig is a convenient model for pharmacokinetic testing of drugs used as pharmacotherapeutics in neonatal life.  相似文献

In the present study, blood samples were taken from clinically healthy puppies of the breeds Beagle, German Shepherd, and Golden Retriever between days 1 and 3 (n = 146), 8 and 10 (n = 137), 28 and 33 (n = 151), and 50 and 58 (n = 129) post natum. Measurements for red blood cell count, haemoglobin concentration, haematocrit, mean erythrocyte volume (MCV), mean corpuscular haemoglobin (MCH), and mean corpuscular haemoglobin concentration (MCHC) were performed by a semi-automatic blood cell counter; the normoblast number was counted visually. Between the 1st and 3rd day of life, the erythrocyte number of the puppies was 4.57 +/- 0.68 10(6)/microliter and, as such, was clearly below the reference range for adult animals. It further decreased by the 2nd measurement (8th to 10th day of life) to 3.59 +/- 0.41 10(6)/microliter, and then increased again to 4.75 +/- 0.68 10(6)/microliter (reference range: 3.73-6.25 10(6)/microliter, 2.5% to 97.5% percentile) by the final measurement (50th to 58th day of life). The measurement values of the haemoglobin concentration (13.5 +/- 2.0 g/dl) and haematocrit (41.0 +/- 6.5%) after birth were only insignificantly below or around the lower limit of the reference range for adult animals. Both parameters decreased to a more pronounced extent than did the erythrocyte count. They reached a minimum of 8.4 +/- 1.0 g/dl and 26.8 +/- 3.2%, respectively, between the 28th and 33rd day of life. Even at the end of the examination period (50th to 58th day of life), the values of these parameters (10.1 +/- 1.1 g/dl, reference range: 7.5-11.8 g/dl; 32.1 +/- 4.2%, reference range: 24.8 to 40.8%) were remarkably lower than the minimum of reference range for adult dogs. At the 1st sampling (between 1st and 3rd day of life), MCV (89.8 +/- 6.7 fl) and MCH (29.6 +/- 1.9 pg) were distinctly higher than the reference values for adult dogs. Both parameters decreased with increasing age. Thus, from the 50th-58th day of life, the results were comparable to those of adults. No considerable age dependence was found for MCHC. During the first days of life a relatively high number of normoblasts (8 +/- 7/100 Leukozyten) was found; it decreased rapidly. The study revealed significant differences between the breeds, e.g. German Shepherd dogs had lower initial values of erythrocyte count, haemoglobin concentration, and haematocrit when compared to the other breeds. Puppies of this breed also had higher normoblast numbers than the Beagle and Golden Retriever puppies at the 2nd and 3rd samplings. No clear sex differences in the studied parameters were observed. The results of this study reflect the replacement of fetal erythrocytes by postnatal erythrocytes. Moreover, they illustrate the need to use age as well as breed-specific reference ranges.  相似文献

Heparin-induced agglutination of erythrocytes in horses   总被引:2,自引:0,他引:2  
Heparin was administered subcutaneously 2 times a day for 4 days to 5 horses. An additional group of 5 horses was used as time-matched controls. Significant decreases in PCV, erythrocyte count, and hemoglobin concentration were observed during heparin therapy. The mean corpuscular volume (MCV) of the heparin-treated horses increased to a peak value of 66.1 fl on the last day of treatment. Erythrocyte creatine concentration and glucose 6-phosphate dehydrogenase activity increased moderately during the treatment. These data indicated that the rapid, profound increase in MCV during heparin therapy was not primarily a result of release of large immature erythrocytes from the bone marrow. A second experiment was subsequently performed, using 3 horses. These horses were given heparin 2 times a day, as was done in the first experiment. Saline wet mounts of erythrocyte suspensions were examined once a day for the presence of agglutination. Cell suspensions were examined with or without exposure to a dilute trypsin solution, and erythrocyte counts were done on each suspension, using an electronic cell counter. Agglutination of erythrocytes was evident on the first day of treatment and became more pronounced as treatment progressed. Exposure to trypsin solution reversed the agglutination. The apparent erythrocyte count decreased and MCV increased sharply in the samples processed normally, but there was little change in those suspensions exposed to trypsin.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
