A total of 24 newborn pigs were used to determine 1) the relationship between the quantity of colostrum consumed and the capacity for gluconeogenesis and fatty acid oxidation and 2) whether fatty acid oxidation limits gluconeogenesis in isolated hepatocytes. Neonatal pigs were obtained prior to nursing and allotted to one of three treatment groups; fed (ad libitum), limit-fed (25% of fed group), or fasted. Hepatocytes were isolated when pigs were 24 h old. Colostrum intake altered the metabolic status of neonates such that the capacity for glucose synthesis and oxidation of octanoate increased with intake. Glucose synthesis with lactate as the substrate was greater (P less than .01) for fed pigs (10.79 mumol glucose.h-1.mg DNA-1) than for either limit-fed (6.56) or fasted counterparts (4.78), which were similar (P greater than .10). Colostrum intake failed to stimulate synthesis from alanine. The oxidation rate for octanoate was similar for fed and limit-fed pigs (.62 and .61 nmol CO2.h-1.mg DNA-1, respectively) but greater (P less than .05) than that observed for fasted counterparts (.36). Oxidation of octanoate (2 mM) was approximately 30-fold greater than for oleate (1 mM); oxidation of the latter was not affected by either colostrum intake or the addition of carnitine (1 mM). The increase in octanoate oxidation, however, did not elicit an increase in glucose synthesis by fasting pigs with either lactate or alanine as precursors. Thus, we conclude that gluconeogenesis is a function of colostrum intake and that reducing equivalents and(or) ATP may not be primary factors limiting glucose synthesis in pigs fasted from birth.  相似文献

A total of 48 neonatal pigs were used to determine whether intubation with colostrum or medium-chain triglyceride (MCT) would enhance glucose homeostasis and survival. Pigs were removed from the sow prior to nursing and alloted to three treatment groups. Fasted pigs received only water for 30 h, whereas those allotted to supplemented groups received either 30 ml of colostrum or 15 ml of MCT at 6 and 16 h after birth (t6 and t16). Supplementation with MCT (t6) resulted in a 2.4-fold elevation in plasma non-esterified fatty acid (NEFA) concentration compared with fasted pigs (260 vs 109 microEq/liter at t8; P less than .05). This difference increased following the second MCT dose (436 vs 117 microEq/liter at t18; P less than .05). Colostrum supplementation also elevated plasma NEFA (201 and 259 microEq/liter at t8 and t18, respectively); however, less triglyceride fatty acid was presented via colostrum compared with MCT. Supplementation with MCT resulted in a greater increase in plasma glucose concentration, relative to fasting levels (75 vs 56 mg/ml at t8; 76 vs 62 mg/ml at t18), than was obtained with colostrum (68 and 65 mg/ml at t8 and t18, respectively). Residual effects of supplementation to t30 were evident for both MCT and colostrum pigs in NEFA levels, but only the MCT group had a greater (P less than .05) concentration of plasma glucose at t30 compared with the fasted group (63 vs 49; P less than .05). 1. The oral administration of glucose or dietary glucose reduces fasting plasma mannose concentrations in mammals. On the other hand, there have been no reports on plasma mannose levels in birds. We have analysed chicken plasma mannose and glucose by an original high-performance liquid chromatography (HPLC) method, together with plasma non-esterified fatty acid (NEFA) concentrations in chickens.

2. Plasma glucose concentrations of chickens did not differ among three different age groups (0, 18 and 150 d). However, the plasma mannose concentrations of chicks at the age of 0 d were higher than those of chickens at the ages of 18 and 150 d.

3. At the age of 18 and 150 d, plasma glucose concentrations were elevated and plasma mannose and NEFA concentrations were decreased after regular feeding, compared to fasting levels.  相似文献

A study was undertaken to investigate the changes of plasma non-esterified fatty acids (NEFA), glucose and alpha-amino nitrogen and their relationship with age, body weight (BW) and plasma growth hormone (GH) in growing buffaloes. For the purpose, six growing female Murrah buffalo calves of 6-8 months of age were selected on the basis of their BW and fed according to Kearl standard (Nutrient Requirements of Ruminants in Developing Countries, International Feedstuffs Institute, Utah State University, Utah, USA, 1982, p. 89) for growing buffaloes (target growth rate 500 g/day) to meet energy and protein requirement of the animals. Blood samples collected at fortnight intervals for 1 year were analysed for plasma NEFA, glucose, alpha-amino nitrogen and GH. The animals were also weighed at fortnight intervals. Plasma NEFA and glucose levels were found to decrease (p < 0.01) with age. Unlike plasma NEFA and glucose, plasma alpha-amino nitrogen level increased (p < 0.01) as the buffaloes become older. Plasma NEFA and glucose concentrations in growing buffaloes were found to be positively correlated with plasma GH (r = 0.379 and 0.420 respectively), but these were non-significant (p > 0.01). However, plasma NEFA and glucose showed a good correlation (p < 0.01; r = 0.780 and 0.652 respectively) with plasma GH per 100 kg live weight. Plasma alpha-amino nitrogen exhibited non-significant (p > 0.01) negative correlation (r = -0.295) with plasma GH but a negative correlation (p < 0.01; r = -0.641) with GH per 100 kg BW. So, plasma metabolites showed a definite pattern of change during growth and these have a significant (p < 0.01) correlation with plasma GH per 100 kg BW than GH.  相似文献

Non-esterified fatty acid (NEFA) levels in housed, 3-month-old calves were monitored in two experiments designed to establish (i) the effect of diet and time of sampling in relation to feeding on blood levels; and (ii) the relationship between appetite and plasma NEFA in animals on chopped hay. In the first experiment, a significant drop in NEFA levels was recorded after feeding. Furthermore, the NEFA levels of calves maintained on concentrate were significantly lower than those on hay throughout the post-feeding period. In the second study, a significant negative correlation was established between NEFA levels and intake of chopped hay in (i) ad-lib-fed calves infected with the abomasal nematode Ostertagia ostertagi; (ii) worm-free animals pair-fed with those in group (i); and (iii) ad-lib-fed worm-free calves. Regression analysis of raw and transformed data from all three groups combined also yielded significant correlations. It is suggested that the measurement of blood NEFA may be a useful indirect indicator of feed intake in conditioned housed calves offered hay diets.  相似文献

The dose-dependent effects of naloxone on feed intake, and plasma chemicals (insulin, glucose, FFA) purportedly involved in feed intake regulation, were determined in 16-hr fasted sheep that were lean and chronically fed maintenance. Dorset ewes (n = 5) were treated with 0 (saline), 0.3, 1 or 3 mg/kg of naloxone in a generalized randomized block experiment with at least 7 d between successive doses. Feed intakes and plasma insulin, glucose and FFA were determined frequently during 24 hr of ad libitum intake after each naloxone treatment. The 0.3, 1 and 3 mg/kg doses of naloxone reduced (P less than 0.01) the 4-hr feed intake by 30, 40, and 60% respectively, whereas the initial feed intake (10 min) was decreased (P less than 0.05) 45% only by 3 mg/kg naloxone. However, total 24-hr intakes were similar across all doses because intakes between 4 and 24 hr of feeding in sheep treated with 0.3 (839 g), 1.0 (802 g) and 3.0 (1330 g) mg/kg naloxone exceeded (P less than 0.01) that in saline-treated sheep (391 g). Feeding-induced changes in plasma insulin, glucose and FFA concentrations were independent of naloxone treatment, suggesting that endorphinergic control of feed intake may not involve coincidental changes in plasma insulin, glucose and FFA levels which are thought to play a role in systemic regulation of appetite in animals. The endorphinergic regulation of appetite in sheep may involve the central nervous system, rather than peripheral opiate mechanisms that utilize blood-borne signals. Further, the ability of naloxone to suppress appetite in sheep appears inversely related to the duration of fasting or severity of negative energy balance.  相似文献

The plasma concentration of glucose was monitored for six hours in lambs fed either ewe colostrum, cow colostrum or milk replacer by stomach tube at one hour of age. The feeding of ewe colostrum resulted in an elevation of the mean (+/- se) plasma glucose concentration from 2 . 6 +/- 0 . 3 mmol/litre before feeding to 7 . 3 +/- 0 . 92 mmol/litre three hours after feeding and this elevation was maintained. A similar rise was observed in the lambs fed cow colostrum but this was of shorter duration. A marked hyperglycaemia was observed after the feeding of milk replacer with a maximum value of 23 . 4 +/- 1 . 33 mmol/litre three hours after feeding. Effect of colostrum intake on diarrhoea incidence in new-born calves   总被引:4,自引:0,他引:4  
In a survey which lasted one year and included data of 73 dairy cows with their calves, colostrum immunoglobulin G (IgG) of 22 primiparous cows and serum IgG of their calves were lower than the corresponding IgG levels of 51 multiparous cows and their calves. Serum IgG concentration was not correlated with diarrhoea incidence. Although there were no seasonal differences in the IgG concentration of colostrum and calf serum, neonatal diarrhoea incidence was higher in calves which were born in winter than in calves which were born in summer (P < 0.01). Thus the high diarrhoea incidence in winter was not a consequence of an insufficient IgG transfer to the calves. The 60 calves of the second study were fed colostrum on the first day of life. From the second to the tenth day 28 experimental calves received milk and 0.5 l of surplus colostrum of the first and second milking twice a day, whereas 32 control calves received milk only twice a day. Two of the 28 experimental and 11 of the 32 control calves suffered from diarrhoea during the first ten days of life (P < 0.05). These results show that the ingestion of surplus colostrum in addition to milk after the first day of life protects the new-born calf against infectious diarrhoea.  相似文献

Colostrum feeding and glucocorticoid administration affect glucose metabolism and insulin release in calves. We have tested the hypothesis that dexamethasone as well as colostrum feeding influence insulin-dependent glucose metabolism in neonatal calves using the euglycemic-hyperinsulinemic clamp technique. Newborn calves were fed either colostrum or a milk-based formula (n=14 per group) and in each feeding group, half of the calves were treated with dexamethasone (30 microg/[kg body weight per day]). Preprandial blood samples were taken on days 1, 2, and 4. On day 5, insulin was infused for 3h and plasma glucose concentrations were kept at 5 mmol/L+/-10%. Clamps were combined with [(13)C]-bicarbonate and [6,6-(2)H]-glucose infusions for 5.5h (i.e., from -150 to 180 min, relative to insulin infusion) to determine glucose turnover, glucose appearance rate (Ra), endogenous glucose production (eGP), and gluconeogenesis before and at the end of the clamp. After the clamp liver biopsies were taken to measure mRNA levels of phosphoenolpyruvate carboxykinase (PEPCK) and pyruvate carboxylase (PC). Dexamethasone increased plasma glucose, insulin, and glucagon concentrations in the pre-clamp period thus necessitating a reduction in the rate of glucose infusion to maintain euglycemia during the clamp. Glucose turnover and Ra increased during the clamp and were lower at the end of the clamp in dexamethasone-treated calves. Dexamethasone treatment did not affect basal gluconeogenesis or eGP. At the end of the clamp, dexamethasone reduced eGP and PC mRNA levels, whereas mitochondrial PEPCK mRNA levels increased. In conclusion, insulin increased glucose turnover and dexamethasone impaired insulin-dependent glucose metabolism, and this was independent of different feeding.  相似文献

The effects of estrogen and progesterone on the blood levels of glucose, non-esterified fatty acids and cholesterol in ovariectomized sheep.The effects of estradiol benzoate and progesterone on blood glucose, NEFA and cholesterol were studied in ovariectomized sheep. Intramuscular injection of 2.5 mg estradiol benzoate gave biphasic changes in NEFA. After 2 hrs. NEFA was decreased, but thereafter an increase occurred and maximum levels were reached after 24 hrs. Blood glucose was significantly increased from 12 to 48 hrs. after the injection. Serum cholesterol was lowered after 24 hrs., but thereafter the level increased. Maximum values were obtained after 120 hrs. Progesterone at the same dose did not change any of the measured parameters. Simultaneous administration of estradiol benzoate and progesterone gave similar responses as estradiol benzoate alone.Blood glucose and NEFA were followed during heat in a lactating cow. Both parameters increased after ovulation.Since NEFA was increased during so long time after the injection of estradiol benzoate, the mechanism behind this effect was discussed. No lipolytic hormone has been reported to give a response of this duration. Estrogen is known to increase plasma GH, and since GH is strongly lipolytic in sheep it seemed possible that the elevated NEFA levels were caused by increased GH secretion. There is now evidence that also estrogen-induced changes in serum cholesterol are pituitary dependant. It was therefore considered possible that all the noted metabolic changes were mediated by the pituitary.  相似文献

A series of experiments has been conducted with chicks to study the effects of excess lysine, leucine and incomplete mixtures of amino acids on growth, food intake and plasma amino acid patterns.

Excess lysine induced a marked depression in growth within 48 h. Towards the end of the experiment, the chicks lost weight and showed no signs of adaptation to the excess dose of lysine. The plasma concentration of arginine fell within 24 h of feeding excess lysine but the reduction in food intake did not occur until 6 d of the experiment had elapsed. Excess leucine similarly caused a rapid retardation of growth which was followed subsequently by a depression in food intake. Mixtures of amino acids lacking in leucine, isoleucine and valine also induced marked growth inhibitions the severity of which were enhanced by the inclusion of leucine and isoleucine in the mixtures. Growth tended to be adaptive in chicks fed on the incomplete amino acid mixtures but non‐adaptive in those fed on excess leucine.

Pair‐feeding studies indicated that appreciable differences in weight gain still occurred when food intake was equated between control groups and groups receiving excess lysine or leucine.

The results of the present study indicate that the depression in food intake cannot account completely for the ill‐effects of excess lysine and leucine. They also suggest that the adverse effects of lysine and leucine are distinct from those caused by incomplete mixtures of amino acids.  相似文献

Background

The physiological levels of endocrine and metabolic parameters in Slovene autochthonous breeds of sheep are not yet well known, nor are the mechanisms of their adaptability and responses to climate and environmental factors.Therefore, the aim of this study was to evaluate fluctuations of cortisol, insulin and non-esterified fatty acids (NEFA) in growing ewes over an one-year period. Blood samples were collected monthly from 10 yearling Jezersko-Solchava, 10 Bovec and 10 Istrian ewes. Serum cortisol, insulin and NEFA were measured with commercial kits.

Results

Mean monthly cortisol values fluctuated with low levels in summer and high levels in autumn. Significant peaked cortisol values of 25.69 ± 6.89, 14.67 ± 2.43 and 21.11 ± 7.25 μg/L in Jezersko-Solchava, Bovec and Istrian breed, respectively, were found in September (Bovec breed) and October (Jezersko-Solchava and Istrian breed). Mean monthly insulin values increased during the observation period. The highest levels of 14.60 ± 3.15, 16.03 ± 5.35 and 12.56 ± 2.52 μIU/mL in Jezersko-Solchava, Bovec and Istrian breed, respectively, were observed in the last sample collection in May. NEFA concentrations were found to be low except in some autumn and spring months. The peak values were observed in March for Jezersko-Solchava and Istrian breed (0.60 ± 0.05 and 0.66 ± 0.10 mmol/L), and in April for Bovec breed (0.71 ± 0.11 mmol/L).

Conclusions

Monthly fluctuations of cortisol, insulin and NEFA were measured in all observed sheep breeds, but between-breed differences in monthly values of examined parameters were insignificant. Significantly increased serum cortisol levels were found in autumn for all breeds and were probably associated with the onset of puberty and low environmental temperature. A gradual increase of insulin level in the examined ewes was in parallel with their growth. Significantly higher NEFA values in spring suggest qualitatively insufficient feed supply during that period.  相似文献

Forty-eight Duroc-cross gilts (40 kg initial BW) were fed a control or a linseed diet containing 60 g of whole crushed linseed/kg. Both diets were supplemented with 150 mg of vitamin E/kg. Eight pigs from each dietary treatment were slaughtered at 20, 60, or 100 d after the start of the experiment. There was no effect (P > 0.05) of diet on growth, carcass characteristics, or foreloin tissue composition. Feeding the linseed diet increased (P < 0.05) the content of n-3 PUFA in plasma, muscle, and adipose tissue, but docosahexaenoic acid was not (P > 0.05) altered by diet. The proportions of n-3 PUFA were highest (P < 0.01) in pigs fed the linseed-diet for 60 d, regardless of tissue (plasma, muscle, or adipose tissue) or lipid (neutral lipids and phospholipids) class. The linseed diet produced a PUFA:saturated fatty acid ratio > or = 0.4 in all groups and tissues, which is close to the recommended value for the entire diet of humans, as well as a robust decrease in the n-6:n-3 ratio. The decrease (P < 0.01) in the percentage of oleic acid in adipose tissue of pigs fed the linseed diet for 60 d could be attributed to a 40% decrease (P < 0.001) in stearoyl-CoA-desaturase activity. Diet did not (P > 0.05) affect the activities of acetyl-CoA-carboxylase, malic enzyme, or glucose-6-phosphate-dehydrogenase in any tissues. Muscle vitamin E content was decreased (P < 0.001) 30% in pigs fed crushed linseed for 60 d, whereas lower (P < 0.001) concentrations of skatole in pork fat were observed in linseed-fed pigs at all slaughter times. Inclusion of linseed (flaxseed) in swine diets is a valid method of improving the nutritional value of pork without deleteriously affecting organoleptic characteristics, oxidation, or color stability.  相似文献
