The objectives of this study were to investigate the relationships between body weight and morpho-structural indices to predict body weight from their orthogonal body shape characters using principal component analysis and to morphologically classify the chicken genotypes using multivariate discriminant analysis. Data used were from 273 randomly selected 12-weeks-old indigenous chickens of normal-feathered (NF), frizzle-feathered (FF), naked-neck (NN) and Anak Titan (AT) genotypes. Phenotypic correlation among body weight and most biometric traits ranged from 0.227-0.876, -0.7-0.901, 0.034-0.968 and -0.207-0.849 for NF, NN and AT chickens, respectively. Factor analysis with varimax rotation of interrelated traits revealed three principal components which accounted for 83.1%, 74.4%, 78.8% and 76.5% of the total variance in NF, FF, NN and AT chickens in the order listed. Breast girth, keel length, thigh length, shank length and wing length were found to be the most discriminating variables to separate the chicken genotypes. The longest distance (72.54) occurred between AT and NF genotypes while the shortest distance (4.27) was recorded for FF and NN genotypes. Classification results showed that 85.2% of AT genotype was correctly classified into their source population. However, 22.7% of NF was misclassified as NN, while 33.3% of NN was misclassified as NF chickens. These results suggest that there is high rate of gene flow between these two indigenous chicken genotypes. Information obtained from this study may be considered useful in breed improvement programmes for selection, characterization, conservation and better management of Nigerian indigenous chickens.  相似文献

In commercial pure white leghorn lines, A, B, and C, the effects on resistance against a virulent strain of Marek's disease virus were assessed for B19 and B21 haplotypes of the chicken major histocompatibility complex. B haplotypes were identified by direct hemagglutination using alloantisera raised against erythrocyte antigens. In homozygous B21 female chicks from lines A and B, mortality upon challenge with virus was 16% and 9%, respectively; in B19 chicks, mortality was 42% and 60%, respectively. Intermediate mortality was observed in heterozygous B19/B21 birds. When line A and B hens were crossed with B15/B15 or B5/B19 cocks from line C, differences between B19 and B21 were significant only in the progeny from B5/B19 sires. Therefore, it was concluded that selection for major histocompatibility complex-associated disease resistance markers may be useful only when B haplotypes complement each other in commercial line crosses and when interactions with genetic background do not severely obscure the differential haplotype effects, as are observed within pure lines.  相似文献

German Dahlem Red chickens with three different major genes of tropical interest: Nana- (naked neck), Ff- (frizzled) and dw- (dwarf), respectively, were tested for serum haemolytic complement, which is essential in innate host defence against infectious agents. Eight different combinations of genes for body size and feather coverage were evaluated. Significant differences both for both the calcium-dependent (classical, CPW) and the calcium-independent (alternative, APW) complement titres were found between the phenotypes. Phenotype nanaffDw- showed the highest complement status. The frizzled (Ff-) gene had a negative influence on APW titres, whereas the dwarf (dw-) gene had a negative influence on CPW titres. The naked neck (Nana-) gene had various influences on the haemolytic complement status. All tested hens had MHC (B) 21 haplotypes, whereas the gene for dwarfism appeared to be linked with the B19 haplotype. It was concluded that introducing major genes (Nana-, dw-, Ff-) to conquer environmental stress in hot climates can have a negative impact on certain aspects of the innate immunity of poultry.  相似文献

Natural infections with influenza viruses have been reported in a variety of animal species including humans, pigs, horses, sea mammals, mustelids and birds. Occasionally, devastating pandemics occur in domestic chickens (broiler and layers) and in humans. From November 2003 to March 2004 in many countries in Asia, there were outbreaks of H5N1 avian influenza virus, causing death of infected patients, and devastating the poultry industry. Some groups of Thai indigenous chickens survived and were therefore classified as resistant. These traits were related to immunogenetics, in particular, the major histocompatibility complex (MHC) class I and class II molecules. The chicken MHC class I were investigated as candidate genes for avian influenza virus disease resistance. Seven hundred and thirty Thai indigenous chickens from smallholder farms in the rural area of avian influenza virus disease outbreaks in the central part of Thailand were used in the present study. They were separated into two groups, 340 surviving chickens and 390 dead chickens (resistant and susceptible). Genomic DNA were precipitated from blood samples and feathers. The DNA were used to amplify the MHC class I gene. Data were analyzed using χ2 analysis to test significant differences of influences of MHC class I haplotypes on avian influenza virus disease traits. The results represented nine MHC class I haplotypes: A1, B12, B13, B15, B19, B21, B2, B6, and BA12, and included 10 of their heterozygotes. The homozygous B21 from these collected samples had a 100% survival rate and they were the major survival group. In addition, the heterozygous B21 also had a high survival rate because of co‐dominant expression of these genes. In contrast, the homozygous B13 had a 100% mortality rate and they were the major mortality group. These results confirmed that MHC class I haplotypes influence avian influenza virus disease‐resistant traits in Thai indigenous chicken. The MHC genes can be used as genetic markers to improve disease‐resistant traits in chicken.  相似文献

中国地方鸡种资源丰富,风味较优,但存在生长速度慢、饲料效率低等缺点,因此为了加速我国地方鸡种和肉鸡品种的遗传改良,分子标记技术得到了广泛应用。已知SH3RF2基因的插入/缺失突变与肉鸡生长和体重密切相关,因此本试验对分布在我国各地的15个地方鸡品种进行了SH3RF2基因的等位基因频率检测,以确定SH3RF2基因在我国地方鸡种中的等位基因分布情况,继而将SH3RF2基因用于鸡生长性状的选择的分子育种标记。研究结果发现所检测地方鸡种和肉鸡品种均是非缺失的纯合子,没有发现缺失的等位基因。要利用SH3RF2基因对中国地方鸡种进行选择,可能需要通过杂交等手段将缺失型等位基因导入我国地方鸡种的群体之中。  相似文献

The present study was conducted to assess the performance of indigenous chickens under extensive system in southern Nyanza, Kenya. The study was carried out in two phases in Komolorume and Kawere villages in Rongo and Rachuonyo districts, respectively. The first phase was a cross-sectional study in 81 farms selected by cluster sampling to get the overview of the indigenous chicken production. A four-month prospective longitudinal study in 60 farms randomly selected from the previous 81 farms followed. Mean flock sizes per household were 20 and 18 birds in Komolorume and Kawere, respectively. Overall mean flock size was 19 birds ranging from 1to 64. The mean clutch size, egg weight and hatchability were 12 eggs, 48 g and 81% respectively in Komolorume and 10 eggs, 45 g and 70%, respectively, in Kawere. The chick survival rates to the age of eight weeks were 13% and 10% in Komolorume and Kawere, respectively. Mean live weights for cocks and hens were 2096 g and 1599 g in Komolorume and 2071 g and 1482 g in Kawere, respectively. The mean household cock to hen ratio was 2:5 and 2:4 for Komolorume and Kawere, respectively. The mean chick to grower to adult ratio per household was 8: 6:6 in Komolorume and 8:4:6 in Kawere. Clutch sizes and hatchability rates were significantly higher in Komolorume village (P?<?0.5). The productivity of the indigenous chickens was shown to be low compared to that of the improved chickens in other parts of the world.  相似文献

本试验测定了我校最新育成的隐性白羽肉鸡快慢羽系父系、母系及其配套地方鸡广西黄鸡和文昌鸡主要的血液生化指标。结果表明,四个品系血清总蛋白、白蛋白、葡萄糖、胆固醇、Ca含量接近,血清P、K、Na含量A系最高,显著(P<0.05)或极显著(P<0.01)高于其它三系,各品系内公母之间母鸡的血Ca均极显著(P<0.01)高于公鸡。血清淀粉酶(AMY)活力和肌酸磷酸激酶(CK)活力B系极显著(P<0.01)高于其它三系,血清碱性磷酸酶(AKP)活力品系间差异不显著(P>0.05)。  相似文献

A total of 376 chickens from different ecotypes were immunized with the non-pathogenic multi-determinant antigen sheep red blood cells (SRBC). The ecotypes included indigenous chickens from various locations in Tanzania (n=102), India (n=86) and Bolivia (n=89). In addition, eight German Dahlem Red (GDR) chicken lines with different major genes (dwarf, naked neck and frizzled) of tropical interest were also immunized with SRBC. Immune competence of the breeds was assessed by measuring complement haemolytic activity, both from the classical calcium-dependent complement pathway (CPW) and alternative calcium-independent complement pathway (APW), alongside IgTotal, IgG and IgM antibody responses to SRBC at 7 days post immunization. Large variations in complement activity and antibody responses to SRBC were observed within and between the indigenous breeds. Many indigenous chickens, especially from Bolivia, showed decreased complement activity (APW) following immunization with SRBC. Breeds from India showed the highest CPW activity and humoral (especially IgM) responses to SRBC, suggesting high immune competence. In contrast, Bolivian chickens were characterized by low CPW activity, low APW activity and low antibody levels to SRBC suggesting an overall low immune competence. In the GDR chickens, characterized by high CPW activity and high IgG antibody responses to SRBC, the major genes for naked neck, frizzling and dwarfism had no significant effect on the antibody responses and complement activity to SRBC.  相似文献

To study the relation between serum complement levels and the chicken MHC (B) complex, complement haemolytic activity was measured in sera from hens from seven pure-bred B-typed White and one Brown Leghorn lines, and three ISA-Warren lines that had been divergently selected for antibody responses to sheep red blood cells (SRBC). Significant differences occurred in the serum haemolytic complement activities, both belonging to the classic (CPW) and the alternative (APW) pathways, among the 11 different haplotyped chicken lines. Hens with high CPW and high APW titres predominantly displayed the B2 or B21 haplotypes. Chickens with low CPW and APW were found in B14 and B15 haplotypes. Haplotype B14 appears to be different in complement levels when present into the pure-bred lines or into the ISA-Warren line selected for low antibody responses to SRBC. Otherwise, the presence of B21 in ISA-Warren line selected for high antibody responses to SRBC does not differ with the B21 in the inbred lines (except in the NL-line for CPW values). In general the haplotypes B2 and B21 are found in chicken lines with enhanced disease resistance, and the B15 haplotype has been connected with enhanced disease susceptibility. Our results suggest that levels of haemolytic complement activity, either from the classical or from the alternative pathways, may underlie part of the immunocompetence ascribed to the MHC (B) complex in chickens.  相似文献

There is a variety of professions working with village chickens in developing countries, including farmers, veterinarians and chicken traders. People from all these occupations were involved in a participatory rural appraisal to investigate husbandry practices and trade of village chickens in Myanmar. Data were collected in two climatically different regions of the country, in the Yangon and in the Mandalay divisions. The breeding and training of fighting cocks was practised only in the Mandalay division, with well-trained birds sold for very high prices. Apart from this, chickens were raised in both regions mainly for small disposable income and were generally sold when money was needed, in particular during religious festivals. Chicken traders on bicycles, often called ‘middle men’, usually purchase birds from farmers in about 10 villages per day. Several ‘middle men’ supply birds to wealthier chicken merchants, who sell these birds at larger chicken markets. There is in general limited knowledge among farmers about the prevention of Newcastle disease via vaccination. Commercial indigenous chicken production is practised in Myanmar, but family poultry farming dominates indigenous chicken production in the country.  相似文献

为掌握禽偏肺病毒(aMPV)在安徽省鸡群中的感染状况,采用酶联免疫吸附试验(ELISA)对安徽省合肥、亳州、定远、舒城等地区的9个鸡场、7个不同品种(系)鸡群的296份血液样本进行了aMPV血清抗体检测。结果表明,所有被检鸡场均有aMPV感染,鸡场阳性率最高达100%,最低为20%;各品种(系)鸡均有感染,感染率最高的是青脚麻肉鸡,其次分别为科宝肉鸡、海兰蛋鸡、禽粤黄蛋鸡、淮南麻黄鸡、黄羽土鸡和新广麻肉鸡;其中蛋用型鸡血清样本总体阳性率为88.7%,明显高于肉用和兼用型鸡;公鸡和母鸡血清抗体阳性率均较高。研究结果表明,安徽省鸡群aMPV的感染已广泛存在,且不同地区、品种(系)、用途和性别的鸡群均较严重,应根据感染状况尽早制定相应的防控对策。  相似文献

通过对线粒体DNA(mtDNA)D-loop区的测序和比对,探讨了中国6个地方鸡品种的母系起源。结果表明,文昌鸡、鲁西斗鸡、寿光鸡、济宁百日鸡和莱芜黑鸡分别有5、5、5、6和7种单倍型,琅琊鸡只有3种单倍型;6个地方鸡品种聚为3个分支:分支A是1个主要分支,共有17种单倍型,有寿光鸡(10只)、鲁西斗鸡(9只)、文昌鸡(5只)、莱芜黑鸡(6只)、济宁百日鸡(6只)和琅琊鸡(2只),分别占所分析各地方鸡品种个体数的100%、90%、84%、75%、60%和20%,与分布于老挝、云南红原鸡的3个大陆亚种(G.g.gallus、G.g.jabouile和G.g.spadi-ceus)关系较近;分支B中包含8只琅琊鸡(80%)和1只济宁百日鸡(10%);分支C中有1只鲁西斗鸡(10%)、1只文昌鸡(16.7%)、2只莱芜黑鸡(25%)和3只济宁百日鸡(30%);分支B和C分别与来自红原鸡G.g.gallus亚种H19单倍型和H32、H33单倍型聚在一起。推测这6个地方鸡品种分别来自云南、老挝和越南附近地区的红原鸡大陆亚种。基因流是上述品种群体间遗传分化的主要因素。错配分布和Fu'sFs检验表明,分布于山东的5个地方鸡品种未发生群体扩张。  相似文献

Eight recently developed 15.B congenic lines of chickens were tested for Marek's disease (MD) resistance by intra-abdominal injection of cell-associated preparations of MD virus of a virulent strain (JM), a very virulent strain (Md5), or Md5 after vaccination with turkey herpesvirus (HVT) strain FC126. Chickens of the 15.N congenic line (B15B21 or B21B21) were very resistant to JM-induced MD, in contrast to chickens homozygous for the B-haplotypes 2, 5, 12, 13, 15, or 19. After Md5 infection, more than 88% of the chickens in all of the congenic lines developed MD. However, when chickens were vaccinated with HVT before being inoculated with Md5, the B5 and B12 homozygotes were more resistant to MD than were the B2, B13, or B19 homozygotes, and B15 and B21 homozygotes had intermediate resistance. B5B5 and B2B5 F2 chicks inoculated with HVT and Md5 had a lower prevalence of MD than B2B2 sibs. These results demonstrate that a protocol involving HVT vaccination of chicks followed by infection with very virulent MD virus will allow the detection of B-haplotypes determining MD resistance, some of which are not detectable in unvaccinated chicks challenged with virulent MD.  相似文献

The residue of aflatoxins in the liver, muscle and eggs of laying ducks, hens and quails and in broiler chickens was examined by conducting 7-day feeding experiments with a diet containing 3 ppm Aflatoxin B1 (AFB1). Birds were sacrificed on the 8th or 11th day of AFB1 feeding. AFB1 and its metabolites in the tissues and eggs were determined by HPLC. The tissue levels of AFB1 and its metabolites were higher in quail than in the other birds. The levels of AFB1 and its metabolites, including acid-hydrolyzable metabolites, were more than 10-fold higher in the liver than in the muscle in all the species. The ratio of AFB1 in the feed to the residual level in the liver was 383 in quail, but was > or = 5769 in the other birds. The corresponding ratios in the egg yolk and albumen of AFB1 were 4615 and 3846, respectively, in chicken hens, and these values were higher than those in the other birds.  相似文献

Earlier studies have shown that the B haplotype has a significant influence on the protective efficacy of vaccines against Marek's disease (MD) and that the level of protection varies dependent on the serotype of MD virus (MDV) used in the vaccine. To determine if the protective glycoprotein gene gB is a basis for this association, we compared recombinant fowlpox virus (rFPV) containing a single gB gene from three serotypes of MDV. The rFPV were used to vaccinate 15.B congenic lines. Nonvaccinated chickens from all three haplotypes had 84%-97% MD after challenge. The rFPV containing gB1 provides better protection than rFPV containing gB2 or gB3 in all three B genotypes. Moreover, the gB proteins were critical, since the B*21/*21 chickens had better protection than chickens with B*13/*13 or B*5/*5 using rFPV with gB1, gB2, or gB3. A newly described combined rFPV/gB1gEgIUL32 + HVT vaccine was analyzed in chickens of lines 15 x 7 (B*2/*15) and N (B*21/*21) challenged with two vv+ strains of MDV. There were line differences in protection by the vaccines and line N had better protection with the rFPV/gB1gEgIUL32 + HVT vaccines (92%-100%) following either MDV challenge, but protection was significantly lower in 15 X 7 chickens (35%) when compared with the vaccine CVI988/Rispens (94%) and 301B1 + HVT (65%). Another experiment used four lines of chickens receiving the new rFPV + HVT vaccine or CVI988/Rispens and challenge with 648A MDV. The CVI 988/Rispens generally provided better protection in lines P and 15 X 7 and in one replicate with line TK. The combined rFPV/gB1gEgIUL32 + HVT vaccines protected line N chickens (90%) better than did CVI988/Rispens (73%). These data indicate that rFPV + HVT vaccines may provide protection against MD that is equivalent to or superior to CVI988/ Rispens in some chicken strains. It is not clear whether the rFPV/gB1gEgIUL32 + HVT vaccine will offer high levels of protection to commercial strains, but this vaccine, when used in line N chickens, may be a useful model to study interactions between vaccines and chicken genotypes and may thereby improve future MD vaccines.  相似文献

The efficacy of three commercial Mycoplasma gallisepticum (MG) immunizing agents-a bacterin, a recombinant fowlpox-MG vaccine, and a live F-strain vaccine-was compared in specific-pathogen-free hens in egg production. Three groups of 25 chickens were vaccinated with one of the vaccines at 10 wk of age and 25 birds were not vaccinated. At 25 wk of age (and approximately 50% egg production), 20 birds from each of the three vaccinated groups and 15 nonvaccinated controls were challenged with virulent R-strain via aerosol; the birds were necropsied and evaluated at 10 days post-challenge. The MG bacterin and live F-strain vaccinations were both protective and resulted in significant differences in air sac lesions, tracheal lesions, and ovarian regression compared to the nonvaccinated controls and the recombinant fowlpox-MG vaccine (P < or = 0.05). The evaluation of ovarian regression is a useful method of testing the efficacy of MG vaccines in laying hens.  相似文献

Previous evidence for the presence of chicken anemia virus (CAV) in the gonads of immune specific-pathogen-free chickens raised the question whether this occurs also in commercial breeders. The presence of CAV was investigated by nested PCR in the gonads and spleens of hens from two 55- and 59-week-old, CAV-vaccinated (flocks 2 and 3), and two 48- and 31-week-old non-vaccinated broiler breeder flocks (flocks 1 and 4). In addition, lymphoid tissues of 20-day-old embryos from these hens were also investigated for the presence of CAV. CAV was detected in the gonads and of 5/6 and 11/22 of the vaccinated hens and in some hens also in the spleen alone. Embryos from 7/8 and 5/18 of these hens were positive. In the non-vaccinated flocks, CAV was detected in the gonads of 11/34 and 10/10 hens in flocks 1 and 4, respectively. In addition, 11 birds in flock 1 had positive spleens. CAV DNA was detected in 3/11 and 2/10 of their embryos. CAV-positive gonads and embryos were detected in samples from hens with moderate as well as high VN antibody titers. Vaccinated chickens positive for CAV in the gonads and in their embryos had VN titers ranging from >1:512 to <1:2048. In non-vaccinated chickens, the VN titers of CAV positive chickens ranged from 1:128 to 1:4096. These results demonstrate that CAV genome can remain present in the gonads of hens in commercial broiler breeder flocks even in the presence of high neutralizing antibody titers that have been associated with protection against CAV vertical transmission. It also suggests that transmission to the progeny may occur irrespectively of the level of the humoral immune response in the hens.  相似文献

White leghorn chickens from seven 15.B congenic lines (genetically similar except for genes linked to the major histocompatibility complex [MHC] B haplotype) and two Line 0.B semicongenic lines were infected at hatch with strain ADOL Hc-1 of subgroup J avian leukosis virus (ALV-J). At 5, 8, 16, and 36 wk of age, chickens were tested for viremia, serum-neutralizing antibody, and cloacal shedding. Chickens were also monitored for development of neoplasia. In the 15.B congenic lines (B*2, B*5, B*12, B*13, B*15, B*19, and B*21) there were no significant differences in the incidence of viremia between B haplotypes. In fact, infection at hatch in all of the 15.B congenic lines induced tolerance to ALV-J because 100% of these chickens were viremic and transient circulating serum-neutralizing antibody was detected in only a few chickens throughout the 36 wk experiment. However, at 16 wk of age more B*15 chickens had antibody and fewer B*15 chickens shed virus than did the 16-wk-old B*2, B*5, or B*13 chickens. Moreover, compared with B*15 chickens, a higher percentage of B*13 chickens consistently shed virus from 8 wk postinfection to termination at 36 wk postinfection. The B haplotype had a transient effect on viral clearance in Line 0.B semicongenics, as more B*13 than B*21 chickens remained viremic through 5 wk of age. Very few (0%-18%) of the Line 0.B semicongenic chickens shed virus. By 36 wk of age, all Line 0 B*13 and B*21 chickens produced serum-neutralizing antibodies and cleared the virus. These results show that following ALV-J infection at hatch the immune response is influenced transiently by the B haplotype and strongly by the line of chicken. Although this study was not designed to study the effect of endogenous virus on ALV-J infection, the data suggest that endogenous virus expression reduced immunity to ALV-J in Line 15I5, compared with Line 0, a line known to lack endogenous virus genes.  相似文献

Brain tissues of 716 slaughtered domestic chickens (524 broilers and 192 hens) were bioassayed for viable toxoplasms. Each tissue was homogenized and subcutaneously injected into 4 SPF mice. Six weeks later the mice were euthanatized and their brains microscopically examined for Toxoplasma gondii tissue cysts. Three (0.4%) out of a total of 716 birds were positive. All positive cases were hens. This is the first isolation of T. gondii from chickens in Croatia.  相似文献

Avian influenza (AI) virus A/chicken/Alabama/7395/75 (H4N8), a putatively non-pathogenic virus associated with a self-limiting outbreak of severe disease in commercial layers, was selectively passed in chickens or in cell cultures and then in chickens to determine whether virus with increased pathogenicity would emerge. When 20 derivatives of the parental virus were each inoculated intranasally and intratracheally in leghorn hens, mortality rates ranged from zero (0/24) to 25% (6/24); mortality was 4% (1/24) for hens inoculated with the parental virus. Many virus reisolates (51/144) from hens that died exhibited high pathogenicity, killing at least six of eight intravenously inoculated 4-week-old chickens. Most derivatives examined produced plaques in trypsin-free cell cultures more efficiently than the parental virus, but the highest plaquing efficiencies observed (10%) were lower than would be expected (100%) for highly pathogenic subtype H5 or H7 AI viruses. These results confirm that the Alabama H4N8 virus can acquire increased pathogenicity upon passage in chickens and suggest that it may have acted alone in producing the severe disease observed in laying chickens in Alabama.  相似文献
