Lungworm-infected seeder calves were used on two 1.41 ha paddocks to ensure that groups of 11 susceptible trial calves would be exposed to heavy early season challenge with Dictyocaulus viviparus. This produced conditions for an artificially severe test of two control strategies. The first employed a front-loaded oxfendazole pulse release bolus, ie, an intraruminal device which released one therapeutic anthelmintic dose immediately and five subsequent pulses at approximately three-weekly intervals. These front-loaded boluses were given to five of 11 calves on one paddock as soon as parasitic bronchitis had become clinically obvious (34 days after turnout) while the remaining six calves were kept as untreated controls. Clinical signs quickly subsided in the treated animals and no further respiratory problems occurred despite continued exposure to reinfection. The other control strategy involved the administration at turnout of an oxfendazole pulse release device which released the first of five anthelmintic doses approximately three weeks after administration, to all 11 calves on the other paddock. This strategy was almost completely successful in preventing patent infections from establishing and reduced the infectivity of the pasture in August and September by 94.1 per cent as shown by tracer calf studies. A novel intraruminal bolus developed for the sustained delivery of the anthelmintic morantel tartrate was evaluated in the seasonal control of parasitic gastroenteritis in first season grazing calves. The morantel sustained release trilaminate is a trilaminate sheet consisting of a central lamina of a morantel tartrate/ethylene vinyl acetate matrix coated on both sides with a thin impermeable layer of ethylene vinyl acetate. A symmetrical pattern of circular perforations punched through the device controls the release of morantel. Administration of the trilaminate to calves significantly reduced their faecal egg output compared with untreated controls and thus reduced pasture larval contamination. Clinical parasitic gastroenteritis was prevented in the treated calves and there were significant reductions in their worm burdens compared with the untreated control calves both during and at the end of the grazing season. The control of parasitic gastroenteritis resulted in a significantly greater (P less than 0.0001) weight gain, of 45 kg, by the treated calves.  相似文献

The efficacy of the first of a new generation of intraruminal devices for cattle, the morantel sustained release trilaminate, was assessed in two field trials. In each trial the animals were divided into a control group and a treated group. The device was administered to each calf before turn out in the spring and the reduction of gastrointestinal parasitism resulted in a substantial reduction in the level of pasture contamination with infective helminth larvae later in the season. Compared with the control calves the treated calves had a 94 per cent reduction of worm burdens acquired over the entire grazing season in the first trial, despite the controls being treated for clinical disease in September. In the second trial four anthelmintic treatments were administered to the control calves during the grazing season, but nevertheless a 64 per cent reduction of worm burdens in the treated group compared to the control group was recorded. The control of parasitic infection by the sustained-release devices resulted in mean weight gain advantages of 28.3 kg and 34.7 kg by the treated animals in the first and second trials respectively.  相似文献

A high, constant and sustained release of drug appears to be a major requirement to avoid a late season rise in pasture infectivity that may result in production losses in calves in autumn or Type II osteragiasis in the following spring. The timing of administration is of crucial importance and will vary in regions of different epidemiology, such as between northern and southern United States. Lack of a standard and reliable technique for pasture larval counts has resulted in some negative or erratic results which are open to question. Although controlled release anthelmintics offer advantages of convenience, a comparison of the economic benefits at present favours prophylactic treatment of dairy heifers with conventional anthelmintics 3 and 6 weeks after spring turnout in northern regions of the Northern Hemisphere. There is a serious risk that boluses based on controlled release by diffusion will behave like slow decaying insecticides and select strongly for drug resistance, especially if farmers administer them in succession throughout the grazing season. There are, however, 2 features in the design of a controlled release device that in theory may minimize the risk of rapid selection for resistance: a high and constant release of anthelmintic followed by a rapid decline to zero as the device becomes exhausted. Under these conditions, the device may even prolong the useful life of an anthelmintic to which resistance has already developed. Two experiments were performed to study the effect of an oxfendazole pulse release intraruminal device (OPRB) on the development of lungworm infection and immunity in calves. Infections were monitored in OPRB-treated and control calves by faecal and pastural larval counts. Levels of immunity of grazing groups and helminth-free housed groups were compared by challenge infection. Even though signs of parasitic bronchitis were seen only in control animals, sufficient immunity had developed in OPRB-treated calves to prevent disease after a substantial challenge infection. However, the degree of immunity was less than in untreated grazing control groups and there were indications that it was related to initial infection levels. These results imply that the OPRB may successfully control parasitic bronchitis, provided that pasture larval infections are sufficient to allow the development of immunity.  相似文献

Four groups, each of six male Friesian calves, were set-stocked on separate 0.66 ha paddocks from May 7 until October 23 1986. Each of the animals in groups 1 and 4 was dosed with an oxfendazole pulse release bolus at turn out whereas the animals in groups 2 and 3 were left untreated. Parasite-free naive tracer calves were introduced into each paddock for a limited period 12 days after turn out and again at the end of the trial. No adverse reactions or clinical signs were observed in either of the groups of calves which received boluses. The development of clinical parasitic gastroenteritis in both the untreated groups necessitated the humane slaughter of two animals and emergency anthelmintic treatment of the remainder. The lower plasma pepsinogen concentrations, and lower faecal egg and larval counts and worm burdens post mortem, together with the absence of clinical signs of parasitic gastroenteritis and bronchitis in the treated calves, confirmed the high efficacy of the bolus treatment.  相似文献

During the first grazing season a group of calves treated with an oxfendazole pulse release bolus achieved a mean (+/- sem) weight gain of 140.7 (+/- 6.7) kg compared with 106.5 (+/- 5.7) kg by a group treated once with ivermectin mid-season, and 116.9 (+/- 6.9) kg by a group which received no treatment. This economic advantage was maintained during the period of winter housing. By the end of the second grazing season, during which the animals received no anthelmintic medication, they weighed on average 20 kg more than the wholly untreated group, a difference which was not statistically significant. No signs of clinical disease were observed in either the animals dosed with a pulse release bolus or the undosed control animals during the two year trial period. The treatment with the oxfendazole pulse release bolus greatly reduced the degree of pasture contamination in the first year but in the second year those animals that had been treated in the first year developed higher worm egg counts (P less than 0.001) and thus augmented the levels of pasture contamination compared with the untreated control animals. Nematodirus battus and N filicollis both produced low grade but fertile infestations in the calves.  相似文献

Sequential in vivo measurements of total body water, exchangeable sodium and exchangeable potassium, made by radioisotopic dilution techniques, were used to determine changes in body composition in calves exposed to natural infections with gastrointestinal nematodes during their first grazing season. Two groups of calves were studied, one of which received a sustained release anthelmintic bolus at turn out. Over the grazing season the bolus-treated calves showed a significantly improved pattern of bodyweight gain, compared with the untreated control calves. There were also significant increases in both total body water and exchangeable potassium, as a percentage of the bodyweight, in the bolus calves compared with the controls. These findings indicated that body protein as a percentage of bodyweight was increased in the bolus animals. The bolus-treated calves also had significantly lower serum pepsinogen concentrations, faecal egg counts and worm burdens.  相似文献

Friesian calves given a low level infection of the abomasal parasite Ostertagia ostertagi over a six week period displayed a mild diarrhoea with high faecal egg counts and elevated plasma pepsinogen values. At necropsy on day 23 abomasal lesions characteristic of ostertagiasis were widespread. At 42 and 84 days oedema and congestion were also prominent. Total worm burdens on days 23 and 42 were similar but a marked decrease had occurred by day 84. Feed digestibility and nitrogen economy were not markedly affected but radioisotopic measurements demonstrated an increase in albumin disappearance and catabolic rates, and plasma faecal clearance during the course of the infection. Prior administration of a morantel sustained release bolus to a group of similarly infected calves reduced the total worm burdens to less than 50 per cent of those recorded in the infected calves on days 23 and 42 and this fell to 3 per cent on day 84. Abomasal damage and the adverse pathophysiological changes associated with infection were prevented in this group.  相似文献

One group of first-season calves was dosed with an oxfendazole pulse release bolus at spring turnout (April 30) and on July 15 a second group received the front-loaded oxfendazole pulse release bolus. The objective was to test the boluses for the prophylaxis or control of nematodiasis. The control group consisted of calves to which no bolus was administered. The three groups occupied separate but adjacent plots. For the first five weeks of the trial, three calves, artificially infected with Dictyocaulus viviparus grazed in each plot. Parasitic bronchitis severely affected the control calves, necessitating repeated emergency treatment, whereas administration of the bolus at turnout almost completely prevented this condition. D viviparus infection increased markedly on the control herbage in July and August but was eliminated by the end of June on pasture grazed by bolus treated calves. Treatment in mid-season with the front-loaded bolus brought an outbreak of parasitic bronchitis under control. Gastrointestinal worm egg output was satisfactorily suppressed after the administration of both boluses, resulting in reduced levels of herbage infection. Calves treated with a bolus at turnout gained significantly more weight than either the controls (P less than 0.001) or the calves treated with a front-loaded bolus in mid-season (P less than 0.01). The weight-gain of the calves treated with a front-loaded bolus was slightly but not significantly greater than that of the control calves. The interaction of the morantel sustained release bolus with the development of immunity in calves vaccinated with two doses of gamma irradiated (40 Kr) Dictyocaulus viviparus larvae was investigated under laboratory conditions. A total of 37 helminth-naive calves were used. Eight calves were used in the first part of the study to test the efficacy of a larval vaccine prepared by using gamma rays delivered from a cobalt source. In the second part of the study, four groups of four groups of four calves each were vaccinated and of these, all the animals in two groups each received a bolus. The remaining three groups (two groups of four and one group of five calves each) remained nonvaccinated with each animal in one group receiving a bolus. All the calves were challenged with approximately 2000 lungworm larvae four months postvaccination. In order to simulate possible field conditions, two of the vaccinated groups and two of the nonvaccinated groups were given a trickle infection of 800 lungworm larvae over a four-week period, three months prior to challenge. Based on a comparison of clinical signs, pathology and lungworm burdens at necropsy, the vaccination of the calves conferred a significant degree of protection (P less than 0.001) to a subsequent challenge compared with controls. The introduction of a morantel sustained release bolus and/or a trickle infection had no effect on the high degree of protection engendered by the vaccination. Nonvaccinated calves given a trickle infection, with or without a bolus, were also highly immune to challenge.  相似文献

The morantel sustained release bolus was administered at turnout to first-season grazing calves in order to assess its efficacy in the seasonal control of infection by nematode parasites in Ireland. The pastures grazed by control calves showed a marked increase in gastrointestinal trichostrongylid infective larvae by September, while numbers of infective larvae on pasture grazed by bolus-treated calves remained at a low level throughout the grazing season. In consequence, the controls showed significantly higher worm egg counts in late season and significantly higher worm burdens (mainly Ostertagia spp) at necropsy carried out in November on representative number of principal animals selected from each group. These reduced worm burdens were attributed to the suppression of egg output during the early part of the season as a result of treatment with the morantel sustained release bolus at turnout in the spring. Pasture contamination with Dictyocaulus viviparus larvae was present on all treatment pastures. The bolus-treated calves however were subjected to an increase in D. viviparus infection which occurred on their pasture in late season after the active life of the bolus had expired. It was concluded that bolus treatment delayed (rather than prevented) the buildup of D. viviparus infection on the pasture by 60-90 days.  相似文献

In December 1996, a questionnaire about farm management and parasite control measures in calves was sent to 956 randomly chosen dairy cattle farmers in The Netherlands. Another 150 farmers in the vicinity of Deventer who had vaccinated their calves in 1995 against lungworm were approached with the same questions. Our objective was to investigate the consequences on worm control of the withdrawal of the lungworm vaccine from the market for reasons of possible BSE contamination of the vaccine. Of the returned questionnaires, 411 (43%) of the `at random' group and 89 (59.3%) of the `Deventer' group were valid. The most important data with regard to the farms of the `at random' group (411) were: mean area 31.6 ha, mean number of calves 23, heifers 23 and milking cows 53. Sheep (mean 37) were present on 18.3% of the farms. With regard to management: 74.5% of the farmers turned the calves in their first year onto pasture, 25.5% kept them indoors. The average time on pasture was ca. 5 months. Rotational grazing was practised on 81.4% of the farms, on 18.6% calves were set stocked. The first pasture of the calves was mown before turn-out on 72.9% of the farms. On 48.2% of these farms, calves were always moved to mown pastures. With regard to treatments: 33.8% of the farmers vaccinated their calves against lungworm in the years 1993, 1994 and 1995. Despite the withdrawal of the vaccine from the market in 1996, 7.2% of the farmers vaccinated their calves as recommended, with two doses, and 13.1% with a single dose. At turn-out, 41.5% of the farmers gave the calves a preventive anthelmintic treatment. Of these treatments, 66.9% were sustained of pulse release long acting devices. During the grazing season, 36.6% of the farmers treated their calves. After housing, 50.3% of the farmers gave a treatment. Signs of lungworm infection were noticed on 18.6% of the farms. Of the `Deventer' group (89 farmers), 96.6% turned the calves out. Of these farmers, 86.0% had used the lungworm vaccine in 1995. In 1996, 52.7% of the farmers had vaccinated the calves: 36.5% with a single dose and 16.2% with the double dose. Of the 35 farmers who did not vaccinate in 1996, 62.9% gave a preventive treatment at turn-out. Clinical signs of lungworm infection were not observed on the 12 farms which vaccinated the calves twice. On 11% of the farms which vaccinated once and on 14% of the farms which did not vaccinate, signs of lungworm infection were observed. It is concluded that more than 80% of Dutch dairy cattle farmers take appropriate measures to control gastrointestinal nematode and lungworm infections in calves in their first grazing season by grazing on aftermath, rotational grazing on mown pastures combined or not with preventive anthelmintic treatments. However, combinations of aftermath grazing and preventive treatment occurred on 30% of the farms. This may be overprotective and may prevent sufficient build up of immunity, causing worm problems at a later age. The withdrawal of the lungworm vaccine from the market did not cause a rise in lungworm problems. Some farmers did vaccinate, despite the withdrawal. The majority used other preventive treatment measures, mainly the application of long acting boli.  相似文献

The efficacy of the morantel sustained release bolus (MSRB) in reducing gastrointestinal parasitism in first season grazing calves was evaluated during the summer--autumn grazing seasons of 1982 and 1983 in western Oregon. Each of 38 calves (1982) and 40 calves (1983) were randomly assigned to either control or treatment groups which were given MSRB on the day of turnout onto pasture. Mean worm burdens from tracer calves grazed with treated animals in 1982 and 1983 showed overall reductions of 86.4% (P greater than 0.05) and 84.3% (P less than 0.01), respectively, compared to tracers grazed with controls. Ostertagia ostertagi, Cooperia oncophora and Nematodirus helvetianus were the primary nematodes collected at necropsy. Twelve full-season 1982 tracer animals (6 treated and 6 control) indicated an 88.1% (P less than 0.05) overall reduction in mean worm burdens. Mean fecal worm egg per gram (EPG) counts of treated animals reflected a reduction of 69% (P less than 0.05) in 1982 and 90% (P less than 0.05) in 1983. Autumn inhibition of O. ostertagi was observed. In the 1982 trial the control animals showed a slight mean weight gain advantage over the treated group from Day 84 until Day 160 (trial termination) when the mean difference was 7.9 kg. The final mean weight gain advantage of treated animals in 1983 was 13.5 kg (P less than 0.05). These trials demonstrated that the MSRB was an effective anthelmintic for reducing gastrointestinal parasitism in grazing calves and for decreasing pasture larval contamination.  相似文献

Calves were first infected with 5000 Taenia saginata eggs at six to 10 weeks old and treated with praziquantel 12 weeks later. Complete immunity against challenge lasted for at least 12 weeks following anthelmintic treatment. Six months after drug treatment over 90 per cent of the cysticerci had been completely absorbed but some were still detectable especially in the heart. An increase was observed in the ELISA values of sera from infected calves following treatment with praziquantel, but no such rise was detected in sera from resistant calves after challenge infection.  相似文献

An experiment was conducted in calves to investigate the efficacy of a morantel sustained release trilaminate bolus (MSRT) to control gastrointestinal parasitism and to assess the development of immunity during the use of MSRT. Two groups (M and U) of four calves each were infected three times a week with a mixed Ostertagia ostertagi and Cooperia oncophora infection for 12 weeks. Calves of Group M received an MSRT at the start of the experiment. Twenty weeks after the start of the experiment, all animals, including a previously uninfected control group (C), received a challenge with 100,000 Ostertagia and 100,000 Cooperia. After a further 4 weeks all calves were necropsied for worm counts. During the trial calves were weighed and faecal egg counts, larval differentiation and pepsinogen concentrations were determined. The results demonstrated the high level of efficacy of the MSRT in reducing the faecal egg output and preventing parasitic gastroenteritis under conditions of a continuous high rate of infection. Efficacy of treatment was higher for Cooperia than for Ostertagia. Post-mortem worm counts suggested a partially impaired immunity build-up in Group M, at least for Cooperia.  相似文献

When they were turned out to grass in May 1987 for their first season, 10 calves were dosed with a 5 x 750 mg oxfendazole pulse release bolus (OPRB) and a monensin sodium rumen delivery device (RDD); eight calves received one OPRB; 10 calves received one RDD and eight calves received neither bolus. Each group was set-stocked on individual paddocks which had been grazed during the previous season by cattle which developed clinical parasitic gastroenteritis and bronchitis (husk). In July, before they were due to be moved to new pastures in mid-summer, and before they were dosed strategically with levamisole HCl, some of the calves not dosed with an OPRB succumbed to clinical parasitic gastroenteritis and husk and received emergency anthelmintic treatment, after which no further clinical episodes occurred. The 'dose and move' strategy was implemented in early August after which both groups not dosed with an OPRB were set-stocked together until the trial ended on October 14, 147 days after turn out. The two groups of calves which had received the OPRB were also moved to new pasture and set-stocked together until the end of the trial. No evidence of clinical helminthiases developed in either of the two groups of calves dosed with OPRBs and their faecal worm egg and larval counts, and plasma pepsinogen activities remained low. They gained significantly more weight than the two groups of calves not dosed with OPRBs (P less than 0.001). The bolus types were compatible and induced no untoward side-effects when used together.  相似文献

Pathophysiologic effects of Ostertagia ostertagi infection and their prevention by strategic anthelmintic treatments were studied in 3 groups each of 6 steer calves. Group-1 calves were noninfected controls. Group-2 calves were inoculated with 100,000 third-stage larvae on the 1st and 28th days of the experiment and grazed on pasture initially free of contamination. Group-3 calves were on a similar regimen as those in group 2, but were also treated with ivermectin 9 days after each larval inoculation. Group-2 calves had increased plasma pepsinogen and gastrin values and decreased weight gains, and total serum protein and albumin concentrations from the 2nd week of infection onward. They were anemic at 10 to 12 weeks and had lower carcass and meat quality at slaughter. Strategic anthelmintic treatments were effective in preventing these effects and calves in groups 1 and 3 had similar performances. On the basis of our findings, high pepsinogen values were related to worm burdens, whereas high gastrin concentrations were related to gastric lesions.  相似文献

The direct and indirect anthelmintic efficacies of the morantel sustained-release bolus given to calves were assessed in a 154-day controlled field trial. A permanent calf pasture (divided into 2 lots) and naturally parasitized calves were used. The medicated calves were given the bolus at the time they were placed on the pastures. Control calves did not receive anthelmintic therapy. The effectiveness of the bolus to control parasitic gastroenteritis was determined by monitoring various parasitologic determinants. The treated calves had significantly (P less than 0.01) reduced numbers of fecal nematode eggs for every posttreatment sampling period when compared with the control calves. Tracer calves, used periodically during the study to indicate pasture larval infectivities, had equivalent worm burdens at the beginning of the trial (treated vs control pasture). Tracer calves, added later in the study to the lot with treated calves, harbored 83% to 94% fewer nematodes than did their counterparts in the lot with the controls. Plasma pepsinogen concentrations, reflective of abomasal worm burden size and/or activity, were significantly higher (P less than 0.01) in the control than in the treated calves from day 54 until trial termination. At trial termination, the treated calves weighed an average of 27.8 kg more and harbored 80.9% fewer nematodes than the control calves. The morantel sustained-release bolus is an anthelmintic delivery device that has therapeutic and prophylactic antinematode activities. To achieve its optimum performance, the bolus must be used so that the epizootiologic patterns of the predominate parasitic nematodes are effectively disrupted.
