Postpartum, presuckle, colostrum samples were collected from 100 mares. Colostral specific gravities significantly correlated (r = 0.9) with colostral immunoglobulin (Ig)G concentrations. Foal serum IgG concentrations highly correlated (r = 0.82) with specific gravities of the colostrum each foal ingested. Eight of 48 foals (17%) had serum IgG concentrations less than 400 mg/dl. The dams of these 8 foals had colostral sp gr less than 1.06 and colostral IgG concentrations less than 3,000 mg/dl. Foals had serum IgG concentrations greater than 520 mg/dl 24 hours after parturition, when the colostral specific gravity of the dam was greater than or equal to 1.06. Effects of breed on colostral specific gravity, colostral IgG concentrations, foal serum IgG concentrations, and mare serum IgG concentrations were not significant.  相似文献

OBJECTIVE: To determine whether total serum calcium (tCa) or adjusted tCa concentrations accurately predict ionized calcium (iCa) status in dogs. SAMPLE POPULATION: 1,633 canine serum samples. PROCEDURE: The tCa concentration was adjusted for total protein (TP) or albumin concentration by use of published equations. Correlations between iCa and tCa or adjusted tCa, tCa and TP, and tCa and albumin were calculated. Diagnostic discordance between tCa or adjusted tCa and iCa was determined. Diagnostic discordance in predicting iCa was also determined for 490 dogs with chronic renal failure (CRF). Sensitivity, specificity, positive and negative predictive values, and positive and negative diagnostic likelihood ratios were calculated for tCa, tCa adjusted forTP, and tCa adjusted for albumin. RESULTS: Diagnostic discordance was 27% when tCa concentration was used to predict iCa status. Use of adjusted tCa increased diagnostic discordance to approximately 37% for all dogs and 55% for dogs with CRF. Positive predictive value and positive diagnostic likelihood ratios were poor when tCa concentration was used to predict iCa status. The tCa concentration overestimated normocalcemia and underestimated hypocalcemia. Adjusted tCa overestimated hypercalcemia and underestimated hypocalcemia. CONCLUSIONS AND CLINICAL RELEVANCE: Adjusted tCa or tCa concentrations are unacceptable for predicting iCa status in dogs. Use of adjustment equations is not recommended. Direct measurement of iCa concentration is necessary for accurate assessment of calcium status. Use of tCa or adjusted tCa concentrations to predict iCa status in dogs could cause serious mistakes in diagnosis and case management, especially in dogs with CRF.  相似文献

A simplification of the traditional hemoglobin methods for determining serum pepsinogen concentration was developed. In this method, 10% trichloroacetic acid solution was added to control samples, and hemoglobin substrate was added to controls and active enzyme samples; standards and samples were incubated for 18 hours, the proteins in the active tubes were precipitated with trichloroacetic acid and removed by filtration, and the absorbances of the supernatant of each standard and sample at 280 nm were measured. The major differences between this method and other methods for determining pepsinogen values are that the preacidification of serum with hydrochloric acid was eliminated, the incubation period was reduced to 18 hours (down from 24 hours), the relative pepsinogen concentration was determined by measuring the concentration of hydrolysis products, using ultraviolet, rather than visible absorbance, and a pepsin standard curve was used to determine the serum pepsinogen concentration. Comparison of freshly prepared pepsinogen and pepsin standard curves indicated that the pepsinogen preparations were slightly more active than the pepsin preparations (on a weight-to-weight basis) on the same substrate. Pepsin standards are used because they are more stable than pepsinogen standards. Three linear standard curve ranges were used: O 10 to 100, 50 to 300, and 100 to 500 ng of pepsinogen/ml of serum. The use of pepsin standard curves permits some variability of the incubation conditions without altering the results. For best results, the hemoglobin substrate solution should be prepared daily. This method may be useful in diagnosing ostertagiasis.  相似文献

Concentration of hyaluronate (HA) in equine serum was determined by a recently developed specific radioassay. The mean +/- SD HA concentration in equine serum was 288 +/- 145 micrograms/L, was age dependent, and varied widely between horses (range, 190 to 760 micrograms/L). Light or moderate exercise increased serum HA concentration from baseline values by 1.5- to 3-fold. In all horses, serum HA concentration returned to or below the original resting values 1 and 2 hours after exercise.  相似文献

Feline serum samples (n = 434) were classified as hypercalcemic, normocalcemic, or hypocalcemic based on both total calcium (tCa) and ionized calcium (iCa) concentrations. Sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), positive diagnostic likelihood ratio (PDLR), and negative diagnostic likelihood ratio (NDLR) were calculated for prediction of hypercalcemia and hypocalcemia in all samples, in hypoalbuminemic cats, and in those with chronic renal failure (CRF) as compared with cats that had other conditions. Diagnostic discordance in prediction of iCa using tCa was 40%. Sensitivity of tCa in prediction of ionized hypercalcemia was low and specificity was high. The PDLR for prediction of ionized hypercalcemia or hypocalcemia was low in all cats, especially in those with CRF. Due to the high level of diagnostic discordance, tCa should not be used to predict iCa concentration. Concentration of iCa should be measured directly when accurate assessment of calcium status is needed.  相似文献

The range for postmortem vitreous humor Mg2+ concentration in 97 healthy cattle was 1.8 mg/dl to 2.72 mg/dl at 23 C for a 48-hour postmortem interval. The postmortem vitreous Mg2+ concentration closely paralleled the antemortem serum Mg2+ concentration at 23 C. Low environmental temperature (4 C) had no effect on postmortem vitreous concentration. However, high environmental temperatures (30 C) significantly (P less than 0.05) reduced postmortem vitreous Mg2+ concentration at the 36-hour postmortem interval. It was concluded that postmortem vitreous humor Mg2+ determination could be a useful diagnostic aid in cattle for detecting Mg2+ imbalances for at least 48 hours after death, provided the postmortem environmental temperature did not exceed 23 C after 24 hours.  相似文献

Objective: To determine endogenous serum insulin concentration in dogs with diabetic ketoacidosis (DKA), and to compare it to endogenous serum insulin concentration in diabetic dogs with ketonuria but no acidosis (KDM), diabetic dogs with uncomplicated diabetes mellitus (DM) that did not have ketonuria or acidosis, and dogs with non‐pancreatic disease (NP). Design: Prospective study. Setting: Veterinary Hospital of the University of Pennsylvania. Animals: Forty‐four client‐owned dogs; 20 dogs with newly diagnosed diabetes mellitus (7 dogs with DKA, 6 dogs with KDM, and 7 dogs with DM) and 24 dogs with non‐pancreatic disease. Interventions: Blood and urine samples were obtained at the time of admission to the hospital. Measurements and main results: Signalment, clinical signs, physical examination findings, and concurrent disease were recorded for all dogs. Blood glucose concentration, venous blood pH, venous blood HCO3? concentration, urinalysis, and endogenous serum insulin concentration were determined in all dogs. Dogs with DKA have significantly decreased endogenous serum insulin concentrations compared to dogs with DM (P = 0.03) and dogs with non‐pancreatic disease (P = 0.0002), but not compared to dogs with KDM (P = 0.2). Five of 7 dogs with DKA had detectable endogenous serum insulin concentrations, and 2 of these dogs had endogenous serum insulin concentration within the normal range. Conclusions: Diabetic dogs with ketoacidosis have significantly decreased endogenous serum insulin concentration compared to dogs with uncomplicated diabetes mellitus. However, most dogs with DKA have detectable endogenous serum insulin concentrations, and some dogs with DKA have endogenous serum insulin concentrations within the normal range.  相似文献

Immunoturbidimetric determination of serum IgG concentration in foals was compared with the reference methods of single radial immunodiffusion and serum protein electrophoresis. High positive correlations were discovered when the technique was compared with either of these reference methods. The zinc sulfate turbidity test for serum IgG estimation was also evaluated. Although a positive correlation was discovered when the latter method was compared with reference methods, it was not as strong as the correlation between reference methods and the immunoturbidimetric method. The immunoturbidimetric method used in this study is specific and precise for equine serum IgG determination. It is rapid and, thus, is advantageous when timely evaluation of critically ill foals is necessary. The technique should be adaptable to various spectrophotometers and microcomputers for widespread application in veterinary medicine.  相似文献

Pigs from sows fed a diet deficient in Se and low in vitamin E were fed a Torula yeast diet supplemented with 100 IU dl-alpha-tocopheryl acetate/kg of diet. Dietary treatments were levels of supplemental Se of 0, .025, .050, .075 or .100 ppm. Some death loss occurred in pigs receiving no supplemental Se at approximately 5 wk of age. Autopsy revealed liver and heart lesions typical of vitamin E-Se deficiency. Selenium supplement had no significant effect on average daily gain, feed intake or gain to feed ratio for the 4-wk experiment. Selenium status of pigs was determined by serum Se concentration and serum glutathione peroxidase (GSH-Px) activity. Serum Se increased linearly (P less than .01) with increasing supplemental Se. Serum GSH-Px activity increased linearly (P less than .01) and quadratically (P less than .05) with increasing supplemental Se. With time, the level of serum Se and GSH-Px activity decreased in unsupplemental pigs, but increased in pigs fed diets supplemented with Se and resulted in significant interactions (P less than .01) between dietary Se level and time on experiment. The correlation between serum Se concentration and GSH-Px activity was .81 (P less than .01).  相似文献

The injection of iron-dextran into beef calves at an early age immediately increased values for erythrocyte (RBC) numbers, hemoglobin (Hb) concentration, packed cell volume (PCV), mean corpuscular hemoglobin (MCH) and mean corpuscular volume (MCV), even though the calves were not anemic. There were no significant differences in average daily gain (ADG) from birth through 15 wk between those that were injected and those that were not injected. Hereford calves had lower values for PCV, Hb, MCH and MCV than Angus and crossbred calves, and both Hereford and Angus calves had lesser body weight and slower ADG than crossbred calves. Serum iron (Fe) concentrations were higher in calves born in the spring and pastured in the summer than in calves born in late summer and fall and pastured in the fall. The serum Fe concentration of calves at first sampling (during first week of life) was positively correlated with their weight at that and subsequent times to 15 wk. Mean corpuscular volume and MCH in cows before calving were positively correlated with the MCV and MCH in their calves at first sampling. Serum Fe concentration in cows before calving was negatively correlated with the ADG of their calves through 15 wk.  相似文献

The purposes of the study were to obtain a reference interval and to calculate the critical difference between two analytical results for canine serum fructosamine concentration. To obtain a reference interval, the serum fructosamine concentration was measured in blood samples from 29 adult dogs after a 15-h fasting period. To calculate the critical difference, blood samples from 20 apparently clinically healthy dogs were collected once weekly for five consecutive weeks, and the total variance of the analytical results was divided into the component of variance between dogs (S inter 2 ), the component of variance for weeks within dogs (S intra 2 ) and the component of variance for measurements (S anal 2 ), using nested analysis of variance. The critical difference was then calculated fromS intra 2 andS anal 2 .The main conclusions are in summary: The reference interval for canine serum fructosamine concentration is 258.6–343.8 µmol/L, and the critical difference between two consecutive measurements on a week-to-week basis is 32.4 µmol/L. The critical difference may be used as a guideline to indicate potentially important changes in the serum fructosamine concentration, though the analytical results should not be assessed by the critical differences alone, but should also be compared to the corresponding reference intervals.  相似文献

The aim of the present study was to estimate the between-dog, within-dog and analytical components of variance for serum thyrotropin (TSH) in healthy dogs, and to use these components of variance to 1) estimate the critical difference for significance between serial results; 2) assess the utility of the conventional population-based reference interval; 3) set a desirable performance standard for analytical imprecision; and 4) estimate the number of samples required for determination of the true mean value for an individual dog. Using the Immulite test system, TSH was measured in serum samples collected weekly for five weeks from eight clinically healthy dogs. Results were subjected to nested analysis of variance. Between-dog variation was 43.6%, within-dog variation was 13.6%, analytical variation was 8.8%, the one-sided critical difference was 37.8%, the index of individuality was 0.4, the maximum allowable analytical imprecision was 6.8%, and the number of samples required to determine the true mean value in a single dog was 40. In practical terms, the present study indicated that the analytical imprecision of canine serum TSH measurement should be < 7%, and that comparing a single serum TSH measurement from an individual dog to the conventional population-based reference range may be too insensitive to detect small but important changes in the serum TSH level of that particular dog. In addition, when treating a hypothyroid dog, serum TSH, measured on a weekly basis, should decrease by at least one-third before any effect of exogenous thyroxine supplementation can be said to have influenced the serum TSH level.  相似文献

The cholesterol concentration of uncooked and cooked longissimus muscle, uncooked subcutaneous fat and serum, as affected by days on feed, breed type and sex class in a factorial arrangement, and the relationship between serum cholesterol and tissue cholesterol were studied. Days on feed, breed type and sex class had no effect (P greater than .05) on tissue cholesterol concentrations. Overall least-squares means for uncooked and cooked longissimus muscle and subcutaneous fat were 63.32, 80.27 and 98.90 mg of cholesterol/100 g of tissue, respectively. Cholesterol concentration was 26.8% higher in cooked vs uncooked muscle and 56.2% higher in uncooked subcutaneous fat compared with uncooked muscle. Serum cholesterol concentration was lower (P less than .05) for fullblood Chianinas and steers (122.6 and 133.9 mg/dl) compared with crossbreds and heifers (162.3 and 151.0 mg/dl), respectively. Serum cholesterol concentration showed a cubic response (P less than .05) over days on feed; it was lowest at 0 d (79.6 mg/dl), increased up to 77 d (156.3 mg/dl), leveled off (154.7 mg/dl), then increased again (179.3 mg/dl). Serum cholesterol had low and insignificant correlation coefficients with all tissue cholesterol concentrations (r = .08 to .22; P greater than .05). Results indicate that muscle and fat tissue cholesterol concentrations do not vary in response to the differences in breed type, sex class or time on feed represented in the present study and thus may be an inherent characteristic dependent upon quantitative needs for cellular membrane functions. However, serum cholesterol concentration was affected by treatments evaluated in this study and may be related to factors affecting lipid metabolism.  相似文献

Prednisone was give norally(2mg/kg b.i.d.) to seven healthy mixed breed dogs for 3 consecutive days. Serum iron concentration increased significantly (p < 0.05) from 142 +/- 26 micro g/dl (mean +/- SE) before a drug adminis- tration on Day 0 to a maximum of 307 +/- 47 micro g/dl on Day 2, and returned to the Day 0 value by Day 5. Mean total iron binding capacity did not vary more than 25% from the Day 0 value during the 9 day long study. The percent saturation of transferrin with iron increased from 33 +/- 6% on Day 0 to a maximum of 71 +/- 9% on Day 3. This determination had decreased to 34 +/- 3% on Day 5. No statistically significant changes occurred in these parameters studied in six control dogs that were not given the drug. To determine whether serum iron concentration might be correlated with endogenous serum cortisol concentration, these tests were determined in serum collected from nine dogs at 7 a.m., 3 p.m., and 11 p.m. each day for 3 consecutive days. Serum iron concentration was lower at 7 a.m. (147 +/- 9 micro g/dl) than at 3 p.m. (164 +/- 9 micro g/dl) or 11 p.m. (159 +/- 10 micro g/dl). Likewise serum cortisol was lower at 7 a.m. (1.29 +/- 0.18 micro g/dl) than at 3 p.m. (1.49 +/- 0.19 micro g/dl) or 11 p.m. (1.51 +/- 0.22 micro g/dl). There was a significant positive linear correlation between serum iron and serum cortisol concentrations when they were compared using mean values for each dog. From these studies, it appears that exogenously administered glucocorticoids and endogenous increases in serum cortisol concentrations may result in increased serum iron concentrations in dogs.  相似文献
