Microbial fermentation of carbohydrates in the hindgut of dairy cattle is responsible for 5 to 10% of total-tract carbohydrate digestion. When dietary, animal, or environmental factors contribute to abnormal, excessive flow of fermentable carbohydrates from the small intestine, hindgut acidosis can occur. Hindgut acidosis is characterized by increased rates of production of short-chain fatty acids including lactic acid, decreased digesta pH, and damage to gut epithelium as evidenced by the appearance of mucin casts in feces. Hindgut acidosis is more likely to occur in high-producing animals fed diets with relatively greater proportions of grains and lesser proportions of forage. In these animals, ruminal acidosis and poor selective retention of fermentable carbohydrates by the rumen will increase carbohydrate flow to the hindgut. In more severe situations, hindgut acidosis is characterized by an inflammatory response; the resulting breach of the barrier between animal and digesta may contribute to laminitis and other disorders. In a research setting, effects of increased hindgut fermentation have been evaluated using pulse-dose or continuous abomasal infusions of varying amounts of fermentable carbohydrates. Continuous small-dose abomasal infusions of 1 kg/d of pectin or fructans into lactating cows resulted in decreased diet digestibility and decreased milk fat percentage without affecting fecal pH or VFA concentrations. The decreased diet digestibility likely resulted from increased bulk in the digestive tract or from increased digesta passage rate, reducing exposure of the digesta to intestinal enzymes and epithelial absorptive surfaces. The same mechanism is proposed to explain the decreased milk fat percentage because only milk concentrations of long-chain fatty acids were decreased. Pulse-dose abomasal fructan infusions (1 g/kg of BW) into steers resulted in watery feces, decreased fecal pH, and increased fecal VFA concentrations, without causing an inflammatory response. Daily 12-h abomasal infusions of a large dose of starch (~4 kg/d) have also induced hindgut acidosis as indicated by decreased fecal pH and watery feces. On the farm, watery or foamy feces or presence of mucin casts in feces may indicate hindgut acidosis. In summary, hindgut acidosis occurs because of relatively high rates of large intestinal fermentation, likely due to digestive dysfunction in other parts of the gut. A better understanding of the relationship of this disorder to other animal health disorders is needed.  相似文献

Highly fermentable diets are rapidly converted to organic acids [i.e., short-chain fatty acids (SCFA) and lactic acid] within the rumen. The resulting release of protons can constitute a challenge to the ruminal ecosystem and animal health. Health disturbances, resulting from acidogenic diets, are classified as subacute and acute acidosis based on the degree of ruminal pH depression. Although increased acid production is a nutritionally desired effect of increased concentrate feeding, the accumulation of protons in the rumen is not. Consequently, mechanisms of proton removal and their quantitative importance are of major interest. Saliva buffers (i.e., bicarbonate, phosphate) have long been identified as important mechanisms for ruminal proton removal. An even larger proportion of protons appears to be removed from the rumen by SCFA absorption across the ruminal epithelium, making efficiency of SCFA absorption a key determinant for the individual susceptibility to subacute ruminal acidosis. Proceeding initially from a model of exclusively diffusional absorption of fermentation acids, several protein-dependent mechanisms have been discovered over the last 2 decades. Although the molecular identity of these proteins is mostly uncertain, apical acetate absorption is mediated, to a major degree, via acetate-bicarbonate exchange in addition to another nitrate-sensitive, bicarbonate-independent transport mechanism and lipophilic diffusion. Propionate and butyrate also show partially bicarbonate-dependent transport modes. Basolateral efflux of SCFA and their metabolites has to be mediated primarily by proteins and probably involves the monocarboxylate transporter (MCT1) and anion channels. Although the ruminal epithelium removes a large fraction of protons from the rumen, it also recycles protons to the rumen via apical sodium-proton exchanger, NHE. The latter is stimulated by ruminal SCFA absorption and salivary Na(+) secretion and protects epithelial integrity. Finally, SCFA absorption also accelerates urea transport into the rumen, which via ammonium recycling, may remove protons from rumen to the blood. Ammonium absorption into the blood is also stimulated by luminal SCFA. It is suggested that the interacting transport processes for SCFA, urea, and ammonia represent evolutionary adaptations of ruminants to actively coordinate energy fermentation, protein assimilation, and pH regulation in the rumen.  相似文献

Eight newborn calves were challenged orally with a known enteropathogenic strain of Escherichia coli 0101 K?(A) and two to six hours later each calf was fed a minimum of three pints colostrum. All calves suffered from acute diarrhoea of varying severity within 24 to 48 hours of infection. Immunofluorescent and histological examination of the small intestine demonstrated adherence of the challenge organism to the epithelium and the presence of pathological lesions similar to those seen in colostrum-deprived calves with enteric colibacillosis. It was concluded that in order to be effective prophylactically, colostrum must be fed prior to infection.  相似文献

Research has been conducted as part of the Small Ruminant Collaborative Research Support Program (SR-CRSP) on evaluation of genetic resources and methods of effecting genetic improvement of small ruminants in the participating countries. In Kenya, the focus has been on development of a dual-purpose goat for milk and meat production in farming areas, utilizing two locally adapted breeds, Galla and East African, and two imported dairy breeds, Anglo-Nubian and Toggenburg, into a single stock. That research now has reached the stage of field testing. In Morocco, the performance of a highly prolific breed of sheep, the D'Man, and of a less prolific but larger breed, commonly raised under extensive management, the Sardi, and of their F1, F2 and backcross progeny has been evaluated for several reproduction and growth traits. Prolificacy of the D'Man was transmitted additively, but there was favorable heterosis for age at puberty, fertility and growth rate, resulting in substantial heterosis in total performance. In Indonesia, a high degree of variability in prolificacy in sheep appears to be due to segregation of a gene with large effect on ovulation rate, similar in some respects to the Booroola gene. In Peru, work has involved evaluation of effectiveness of current selection programs and estimation of phenotypic and genetic parameters, in unimproved and improved types of sheep, with a limited amount of work also on alpacas. Fiber production is the principal economic trait in alpacas. There also has been work on evaluation of hair sheep in Brazil, Kenya and Indonesia.  相似文献

单宁是一类广泛存在于植物中的多酚化合物,因其涩味降低反刍动物采食量,故通常被归类为饲料中的抗营养因子。近年来,研究发现单宁可与蛋白质结合,抑制蛋白质被瘤胃过度降解,增加了蛋白质在后肠道的消化吸收,提高了反刍动物对氮元素(N)的利用率。此外,研究表明单宁在调控瘤胃发酵和抑制甲烷排放方面表现出积极作用。本文主要综述了单宁对反刍动物生产性能、瘤胃发酵和微生物区系的影响,旨在为其应用于反刍动物健康养殖提供合理的理论参考依据。  相似文献

The Na(+/)glucose cotransporter (SGLT1) is the major route for the transport of dietary sugars from the lumen of the intestine into enterocytes. Regulation of this protein is essential for the provision of glucose to the body and avoidance of intestinal malabsorption. This has important nutritional implications in particular for young and growing animals. It has been demonstrated that dietary sugars and artificial sweeteners increase SGLT1 expression and the capacity of the gut to absorb monosaccharides. Furthermore, diets supplemented with artificial sweeteners have been shown to improve growth and performance of weaning piglets. In this review, after describing the organization of intestinal epithelium, the type of gut hormones released in response to dietary carbohydrates, the mechanism underlying the transcellular transport of glucose in the intestine is outlined. Next, a historical background to the work carried out in various laboratories aimed at identifying molecular mechanisms involved in regulation of intestinal glucose transporter, SGLT1, is described. Subsequently, the more recent data on the role of intestinal glucose, or sweet, sensor T1R2 + T1R3, a G protein-coupled receptor, required for upregulation of SGLT1 by dietary sugars and artificial sweeteners, are presented. The glucose sensor subunits, T1R2 + T1R3, are members of the taste receptor family 1, T1R, and are expressed in the gut enteroendocrine cells. Sensing of dietary sugars and artificial sweeteners by T1R2 + T1R3 activates a pathway in endocrine cells leading to secretion of gut hormones. Finally, after describing molecular mechanisms by which a specific gut hormone released by endocrine cells may regulate SGLT1 expression in the neighboring absorptive enterocytes, the application of these findings to enhancing intestinal capacity to absorb dietary sugars in weaning piglets is presented. A better understanding of the molecular events involved in regulation of SGLT1 will allow the identification of nutritional targets with attendant promise of avoiding nutrient malabsorption and enhancing growth and well-being of species.  相似文献

The efficacy of keyhole limpet haemocyanin (KLH) as a test antigen was determined in calves. Humoral and cellular (in vivo and in vitro) responses were compared. Calves were immunized with KLH at either 3 weeks or up to 5 months of age and immune responses were subsequently tested. Class and subclass antibody responses were detected by ELISA, lymphocyte blastogenesis was measured using a whole blood culture technique (LTT) and skin sensitivity responses were measured as an increase in skin thickness following intradermal injection. In young calves, skin test responses were maximal at 24 h and were found to correlate with IgG1 and IgG2 responses (P less than 0.01), with IgA (P less than 0.05) but not with IgM or LTT. Histological examination of skin swellings found a sequence of cellular events, with polymorphonuclear cells dominating until 48 h after intradermal injection, when mononuclear cells became involved. However, in older calves, skin test responses correlated not only with IgG1 and IgA responses but also with lymphocyte transformation (P less than 0.05). These findings suggest that, while immune responses to KLH may be a useful indicator of immune competence in calves, interpretation should be made with caution particularly in young calves.  相似文献

For lactating dairy cows, we need management tools, that are "clean, green and ethical", cost-effective and easy to use. Specific tools are needed for artificial insemination (AI) after oestrus detection within a few months of calving, and for managing the complex nutritional requirements of cows between successive calvings. Assessment of energy deficit by measurement of body condition score (BCS) has been useful in the past but we now need more sophisticated ways to measure the relationship between adipose tissue and fertility. For this reason, we have focused our attention on the cells of the adipose tissue, the adipocytes, and the role of the hormone that they produce, leptin. This hormone affects pulsatile LH release and, in dairy cows, it seems to be linked to the first postpartum ovulation. Adipocytes are always sensing energy status and they control leptin secretion dynamically, so blood leptin concentrations can change acutely, even when there is no detectable change in BCS. Leptin secretion seems to be determined by the secretory activity of each adipocyte as well as the total mass of adipocytes in the body of the animal (as measured by BCS). The strong relationship between BCS, leptin concentration and reproductive function in dairy cows suggests that we should reconsider the interval of the recovery from prepartum and postpartum damages, the need for high milk yields at the last lactation causing the dry-off stress and the subsequent troubles. We should also re-assess the current drive to reduce calving interval because milk yields during the early stages of lactation are economically very important but high yields seem to cause several metabolic and reproductive disorders in modern dairy cows. In general, the thinking has been that calving interval must be short because short intervals are more profitable. However, if we remember that main product from dairy cows is milk and that a short calving interval is very difficult without reproductive problems, then a longer calving interval might be more sensible and also more profitable. We have example of an extended calving interval in Japan, Supercows which are very rare cows yielding remarkable high milk. Finally, we probably need to improve dairy cows genetically if we are to achieve the goal of "clean, green and ethical" dairy farming. This paper reviews data relevant to these strategies and we conclude that more basic and applied research will be required if we are to find ways to reach that goal.  相似文献

The objective of this study was to examine effects of increased gut fill and diverse developing environments on pregnant gilts' behavior and physiology. Gilts were cross-fostered at 1 d of age and transferred to either an indoor or outdoor production unit. Littermate gilts remained in their different environments during development and were moved into individual gestation crates in an indoor gestation unit. Of the 42 gilts, 19 were fed a control diet of fortified sorghum-soybean meal and 23 were fed the same diet with 25% beet pulp (high fiber). Control sows ate 2.0 kg/d and high-fiber sows ate 2.67 kg/d in a large pellet (thus resulting in approximately equal energy intake and differing total dietary intakes). Pregnant gilts had behavior and immune measures sampled at 30, 60, and 90 d of gestation. The day x diet interaction was significant (P = 0.01) for duration of standing: sows fed high-fiber diets stood less on d 30, but on d 60 and 90 they and the control sows stood for a similar duration. Sham chewing duration and frequency showed significant (P < 0.05) effects of gestation stage x diet x environment. Gilts reared outdoors and fed high fiber increased sham chewing over gestation, whereas all other treatment groups decreased this behavior over time. Outdoor-reared gilts had greater (P < 0.05) frequency and duration of drinking behavior than indoor-reared gilts. White blood cell numbers were higher (P < 0.05) for gilts fed high-fiber diets than for gilts fed the control diet. Immune (humoral and cellular systems) and reproductive measures (farrowing rate and litter size) and plasma cortisol concentrations were generally not influenced (P > 0.10) by diets and rearing environments, suggesting that in spite of significant changes in behavior and feed intake gilts' immune systems were not suppressed or enhanced. Behavioral data alone suggested that indoor-reared gilts showed fewer behavioral adaptations to the crates than outdoor-reared gilts. However, immune measures did not indicate that any treatments resulted in physiological effects indicative of stress.  相似文献

Spring calving Angus and Angus x Hereford multiparous cows were utilized to determine the effects of intramammary treatment with penicillin G procaine (200,000 IU) and novobiocin (400 mg) at the time of weaning on udder health and calf growth after the subsequent calving. Cows were stratified by age and breed and assigned randomly to receive intramammary treatment (n = 99) at weaning or as untreated controls (n = 97). Quarter milk samples were collected at weaning and at 8 to 14 d after calving. Milk samples were analyzed for somatic cell counts (SCC) and mastitis-causing bacteria. Dry cow treatment decreased (P = 0.005) the number of cows infected after calving. Treatment decreased (P = 0.04) the number of cows that developed new infections and reduced (P = 0.03) the number of quarters with mastitis-causing bacteria after calving that were infected at weaning. Somatic cell counts after calving were greatest (P = 0.008) for cows infected with Staphylococcus aureus. Treatment did not alter (P = 0.19) SCC of quarters after calving that were infected with S. aureus at weaning but reduced (P = 0.002) SCC after calving of quarters that were infected with coagulase-negative staphylococci at weaning. Body weight of calves during early lactation was increased (P = 0.006) if cows with intramammary infection were treated at weaning. Treatment of noninfected cows at weaning increased (P = 0.008) adjusted 205-d weaning weights of calves after the subsequent lactation when compared with untreated noninfected cows. We conclude that treatment of beef cows at weaning with intramammary antibiotics decreased intramammary infections after calving, improved udder health during the subsequent lactation, and increased BW gain of the calves.  相似文献

The neonate's development and survival is dependent upon being vigorous at birth and receiving appropriate maternal care. However, difficulty at delivery can result in less vigorous offspring and maternal care can be altered, probably as a consequence of exhaustion, pain and human intervention. The first 3 h after expulsion of the calf were observed continuously from videos following twelve natural calvings and sixteen calvings assisted by farm staff (including four malpresentations) from Holstein cows. Calvings were balanced within groups for parity of the dam, genetic group, sex and birth weight of the calf, calving pen and calving season. Assisted calves were less vigorous with higher latencies to attempt to stand, achieve standing, walk and reach the udder than unassisted calves (P < 0.05). Furthermore, assisted calves also tended to be less likely to stand and walk within the first 3 h after birth (P < 0.1), spent more time lying on their flank (P = 0.019) and had more frequent bouts of this behaviour (P = 0.033). Assisted dams did not take longer to lick the calf and performed as much licking as unassisted dams (P > 0.05), indicating no delayed onset or impaired expression of maternal behaviour in dams given assistance at delivery. Study of potential pain-related behaviours revealed that assisted dams spent less time self-grooming (P = 0.033) than dams delivering naturally, which could suggest greater pain. However, there were no significant differences in any of the other pain-related behaviours. Our results suggest that, although maternal behaviour was unaffected by a difficult delivery, dairy calves born following difficult calvings have lower vigour in the first 3 h after birth than unassisted calves. This might have longer-term effects on the health and survival of the calves.  相似文献

The functioning of the gastrointestinal tract is under the control of the most extensive system of peripheral neurons in the body, the enteric nervous system, and the largest endocrine system of the body, the GEP endocrine system. The enteric nervous system in large mammals contains 500 million neurons, and the GEP endocrine system produces more than 30 hormones. Numerous enteric neuropathies affecting both humans and animals have been described and digestive disorders affect commercially important species, such as horses and cattle. The most severe enteric neuropathies (e.g., lethal white syndrome in horses or Hirschsprung's disease in humans) can be fatal. Also, horses with ileus or other digestive disorders are commonly euthanized. In this review we discuss examples of enteric neuropathies that affect agricultural animals and humans: prion disease, postoperative ileus, distal enteric aganglionosis, and infective diarrhea. Enteric neurons and glia are a location of prion proteins and are involved in transmission of the infection from gut to brain and brain to gut. Postoperative ileus is a complex disorder involving the local inhibitory effects of sympathetic nervous system activation and the release of opioids, presumably from enteric neurons. Intestinal inflammation, especially of the external muscle that includes enteric ganglia, also occurs in ileus. Congenital distal bowel aganglionosis, responsible for lethal white syndrome in horses, Hirschsprung's disease in humans, and similar conditions in mice and rats, is a fatal condition if untreated. Mutations of the same genes can cause the condition in each of these species. The only effective current treatment is surgical removal of the aganglionic bowel. Infectious diarrheas involve activation of enteric secretomotor neurons by pathogens and the toxins they produce, which causes substantial fluid loss. Strategies to target enteric neurons in the treatment of secretory diarrheas have not been developed. Disorders of enteroendocrine cells, other than GEP endocrine tumors, are less well documented. However, evidence for the involvement of gut endocrine cells in a subset of patients with irritable bowel syndrome, and in the symptomology of celiac disease, has been demonstrated. Further investigation of the involvement of enteric neural and endocrine signaling systems in digestive disorders, especially in agricultural and companion animals, may lead to diagnostic and therapeutic advances.  相似文献

An observational study was conducted to comparatively assess the efficacy of three different oral rehydration and buffering solutions, differentiated by their strong ion difference (SID) concentration, for treatment of neonatal calves with naturally acquired diarrhea. The SID concentrations tested were 100 mM, 170 mM and 230 mM for treatments SID100, SID170 and SID230, respectively. Clinical assessment and blood gas analysis were completed for 18 diarrheic calves once pre- and twice post- (6 and 24 hr after) oral administration with one of the three treatments. A repeated measure mixed model approach was used to analyze (a) the within-group efficacy of each treatment over time and (b) the between-group comparison at each timepoint. SID230 treatment resulted in a significant increase in blood pH, HCO3, BE, SID and Na+ at 6 and 24 hr after treatment, and a significant decrease in AG and K+ by 24 hr after treatment. There were no significant changes in any of the blood gas parameters after treatment with SID100 and SID170. SID230 treatment also resulted in blood gas parameter changes that were significantly different to the other two groups. These results suggest that the optimum SID concentration for the treatment of calves with diarrhea is likely to be higher than current recommendations.  相似文献

采用传统抽滤、ANKOM滤袋和CAU滤袋技术测定16种反刍动物饲料的中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)和木质素(PL,高锰酸钾法)含量。结果表明:3种方法测定的大部分饲料的NDF、ADF和PL含量间没有差异(P>0.05),但在测定谷物性饲料NDF含量时,ANKOM滤袋技术测定值明显低于其他2种方法(P<0.05);在测定动物性蛋白质饲料时,CAU滤袋测定的NDF值明显高于其他2种方法(P<0.05)。3种方法测定的NDF、ADF和木质素含量均具有显著的线性相关关系(R2=0.990~0.996),其中2个滤袋技术测定值之间的相关程度最高(R2=0.992~0.996);2个滤袋技术测定值的变异系数(CV=2.20%~3.96%)均小于传统抽滤(CV=3.21%~4.91%)。  相似文献
