Manipulation of storage environment to provide favorable conditions for wound healing immediately after harvest reducedAlternaria solani tuber infection. Prestorage of tubers at 15.6°C for three weeks before final storage at 10°C normally maintained for commercial storage of potato chip stocks resulted in fewer and smaller lesions than constant storage at 10°C or 4.4°C or prestorage for varying periods at these temperatures. Prestorage of potatoes at a low temperature (4.4°C) for 1–3 months resulted in moreA. solani infection suggesting that low temperatures may inhibit the wound healing process more than they reduce rates ofA. solani infection or disease development. Variations in relative humidities above 75% did not have an appreciable effect on the wound healing process nor onA. solani infection.  相似文献

The wound response within damaged potato tubers (Solanum tuberosum L.) must be a coordination of suberization with other resistance responses if infection is to be avoided. Previously, we showed that wound healing was affected by wound severity and consequently the type of tissue damaged within the tuber. Using arachidonic acid-induced accumulation of rishitin as a model for phytoalexin accumulation, we now demonstrate that this tuber resistance response is also influenced by wound severity and the type of tissue exposed to the elicitor. Kennebec and Reddale tubers that were superficially wounded by removing a thin tissue slice (0.75 mm thick) from the surface and then treated with the elicitor arachidonic acid produced significantly less rishitin than more severely wounded tubers (e.g., tubers cut in half) and excised tuber tissue disks (17 mm × 4 mm). Excised, elicitortreated tuber tissues accumulated significantly more rishitin in cortical cells than in perimedullary and pith cells. Rishitin accumulation was routinely measured 96 h after wounding and was found to be declining by 144 h regardless of wound severity. Induction of rishitin accumulation was very localized with more than 80 % of the rishitin found within 0.75 mm of the treated wound. These results indicate that excised tuber tissue disks, often used as models in wound research, are not fully representative of intact tubers. Perhaps more important, the results show that superficial wounds of intact tubers, i.e., similar to shallow nick and abrasion type wounds typically incurred during harvest, accumulate very little rishitin. However, rishitin accumulation in the tuber disk model system is much greater than that found in superficially wounded tubers, but is similar to the accumulation that could be elicited in cut (seed) tubers. The complications contributed by these wound-related and tissue-specific interactions must be factored into the model system(s) used in describing the role(s) of phytoalexins in the broad framework of disease resistance for stored potatoes and cut seed.  相似文献

Pre-harvest propane burning of vines and soil surface significantly reduced populations ofAlternaria solani spores on the soil surface in field studies. Infection of potato tubers harvested from burned fields was reduced in most cases. Laboratory studies indicated a minimum temperature of 200°C for 1–2 seconds was required to significantly (P = 0.05) reduce germination ofA. solani spores. This temperature was achieved at the soil surface in the field when plots were burned at tractor speeds of 2, 3, 4 and 5 mph. In practice burning at only 2, 3 and 4 mph significantly reduced spore populations and tuber infection in the field. Recontamination of plots after burning occurred relatively rapidly. The data show that propane burning at tractor speeds of 2–4 mph can effectively reduceA. solani infection of potato tubers by lowering fungus populations on the soil surface.  相似文献

Potato tubers became infected byAlternaria solani at the time of harvest. Infection occurred only on wounded tubers irrespective of their depth in the soil. The presence of free moisture on unwounded tubers did not increase the incidence of infection, and immature unwounded tubers were not susceptible. Tuber infections may be reduced by allowing tubers to fully mature before harvest or possibly, by applying chemicals to the soil surface.  相似文献

Silver scurf, caused byHelminthosporium solani, and black dot, caused byColletotrichum atramentarium, are pathogens of tuber periderm whose presence in Colorado was only recently reported. A field survey conducted in September 1977 revealed thatC. atramentarium was more prevalent (21.8% tuber infection) and had a wider distribution thanH. solani (5.4% tuber infection). A greater incidence of both pathogens was observed on thin skinned tubers of chipping cultivars (49.0% infection) than on thicker skinned tubers of table stock cultivars (9.1% infection). Conidial development ofH. solani is tretic, pleurogenous, and requires 17–21 hours per conidium. Conidial septations appear while conidial elongate, and require 3–5 hours per septum. Light microscopy revealed that at least 11 conidia per conidiophore are produced in culture in 54 hours at 20–25 C (68–77 F) and humidity >90%. Scanning electron microscopy showed that fructifications ofH. solani (conidiophores-conidia) arise from beneath infected tuber periderm. Histological studies indicate some peridermal loosening and sloughing. Heavy deposition of unidentified compounds was observed in infected periderm, and hyphae were restricted to periderm cells. Fresh weight loss of tubers naturally infected withC. atramentarium was significantly greater than fresh weight loss of nearly noninfected (< 1% surface area infected) control tubers. Periderm infected with eitherH. solani orC. atramentarium appeared similar, i.e. shriveled, suggesting infections from either pathogen may result in increased fresh weight loss through alteration of the periderm.  相似文献

Summary In each of the five years 1978 to 1982, seed lots of cv. Bintje, variously infested byPhoma foveata Foister andFusarium solani var.coeruleum (Sacc.) Booth, as found by wound and cut tests, respectively, were planted in a series of field experiments at six sites in Sweden. A statistically significant relationship between seed and progeny infestation was found forP. foveata each year, and in three years forF. solani. Benomyl and thiabendazole mist treatments of seed tubers at grading in spring controlled the development of gangrene and dry rot in treated seed tubers, but did not consistently reduce the levels ofP. foveata andF. solani infestation in progeny from treated seed tubers. The site of cultivation markedly influenced mean levels ofP. foveata infestation, which were found to be related to temperature and rainfall during a 40 day pre-harvest period at the trail sites; however, mean levels ofF. solani infestation were not so related.  相似文献

Potato (Solanum tuberosum L. cv. Russet Burbank) tubers were harvested weekly over a 6 week period from non-water-stressed and water-stressed plants. Discs were cut from fresh tubers and from tubers stored for 10 days at 4°C to determine their wound healing ability as evaluated by disc resistance to water loss. Neither harvest date nor water status significantly influenced wound healing while storage significantly increased wound healing ability. Enhanced wound healing associated with storage was accompanied by reduced variability of disc resistance to water loss and increased levels of suberin as reflected by the diol and alcohol components. This work suggests that plant senescence and harvest date are not good indicators of the potential wound healing ability of tubers. Physiological evidence is presented to support the practice of short periods of storage following harvest to improve tuber wound healing ability and thereby reduce shipping and handling losses due to injury.  相似文献

We demonstrated that porometrically derived wound-healing profiles of whole tubers were greatly affected by the depth or severity of the wound. A rapid decrease in water vapor conductance was detected during the first 24 h of healing after mature, whole tubers were wounded by tangentially cutting into the cortex. The rapid decline in vapor conductance, indicating deposition of soluble waxes, was not concurrent with detectable deposition of polymeric phenolic or polymeric aliphatic components of the suberin polyester; instead these polymerized components were detected after vapor conductance decreased by approximately 80% and was nearly stable. The rate of decline of vapor conductance of wound-healing tubers was hormonally hastened with abscisic acid treatments. Although deeper tangential wounds (3.0 mm) had slower initial rates of wound-healing than did shallow wounds (0.75 mm) through tuber growth and maturation, the ability to wound-heal more rapidly increased as the tubers matured in the field. The vapor conductance of wound healing tubers declined in a log-linear fashion during the first 1 to 3 days after wounding depending upon tuber maturity and genotype.  相似文献

Thiabendazole-resistant strainsof Fusarium sambucinum andF. solani var.coeruleum threaten to negate chemical control options for post harvest treatment of Fusarium dry rot. Biological control of dry rot of storage tubers is feasible using bacterial antagonists (25). The impact of yeasts on dry rot has not been investigated. Initial biological control tests employed strains of twenty species of yeasts from the ARS Culture Collection (NRRL). Strain selection was based on strain isolation from plant matter or from environments that would indicate a high likelihood of strain survival in soil. The control potential of these and 29 additional strains isolated from soil adhering to recently harvested tubers was evaluated using a whole Russet Burbank tuber bioassay. At 2xl06 cells/ml, only two unidentified strains andCryptococcus laurentii strain NRRL Y-2536 reduced disease (P=0.05, P=0.10, respectively) while bacterial strainPseudomonas fluorescens 2-79 (NRRL B-15132) was more effective (P=0.01). Conversely,Debaryomyces robertsiae increased disease (P=0.05). No yeast strains significantly controlled disease in a subsequent trial. One of six additional strains ofC. laurentii (P=0.10), none of five strains ofPichia farinosa and neither unidentified strain controlled disease at 5 x 107 cells/ml whereasP. fluorescens again reduced disease (P=0.01). After 6 h, four yeast strains decreased and three increased conidial germination ofF. sambucinum R-6380 though there were no differences after 18 h. Five yeast strains, including two strains ofC. laurentii (NRRL Y-2536, NRRL Y-7139) were marginally effective in controlling disease incited byF. solani var.coeruleum S-1257. Though additional testing may identify yeast strains with considerable promise as biological control agents active against Fusarium dry rot, evidence to date indicates bacterial agents have a greater potential for commercial development.  相似文献

Thiabendazole insensitive strains ofHelminthosporium solani, the causal agent of silver scurf, make controlling the disease with seed treatment difficult. Potato tuber seed treatments and environmental storage management practices were investigated as means to minimize silver scurf. Fungicide seed treatments were evaluated for control ofH. solani; disease was evaluated during the growing season, at harvest, and after 5 months of storage. Silver scurf was observed on progeny tubers eleven weeks after planting. Fungicides that reduced silver scurf incidence and severity on the seed resulted in reduced incidence and severity of the disease in the progeny tubers at harvest and significantly lower disease ratings after storage. Only small increases in disease incidence (0-8%) were seen after storage. Thiophanate-methyl with mancozeb, Captan with mancozeb, and fludioxonil were among the most effective in reducing the incidence and severity of silver scurf on seed and in progeny tubers (Incidence on progeny tubers at harvest for these three treatments were 3%, 9%, and 8% respectively). Thiophanatemethyl alone was not effective for control of silver scurf (48% incidence compared to 43% incidence for the untreated control). Environmental conditions in storage affected disease development. Reduced humidity (85%) during the curing period (0–3 weeks after harvest) significantly reduced (11%) the surface area of tubers infected with silver scurf. Free moisture on the tuber surfaces during storage significantly increased (15%) tuber surface area infection.H. solani was shown to survive in soil and on some potato storage building materials for up to 9 months. The silver scurf disease of potatoes can be suppressed using effective seed treatment and storage management.  相似文献

Two tillage practices, chisel plowing (30 cm) and deep moldboard plowing (22 cm), and five rotation crops (oats, lupine, buckwheat, broccoli and peas) were studied for their effects on the soil population ofRhizoctonia solani AG-3 and on Rhizoctonia disease on potato. All rotation crops were harvested except buckwheat, which was treated as a green manure crop. Chisel plowing significantly reduced (p = 0.05) the incidence and severity of stem lesions on potato caused byR. solani AG-3. In 1990, oats after moldboard plowing significantly increased disease when compared to other crops and broccoli after chisel plowing decreased disease severity. Soil populations ofR. solani AG-3 were significantly lower with chisel plowing. No interactions between tillage and rotation crops were observed. Rhizoctonia solani Kühn is a soil inhabiting plant pathogen found worldwide that affects many plant species including white potato (Solanum tuberosum L.).R. solani attacks potato at one or more stages in development resulting in distinct disease symptoms (25) often termed the Rhizoctonia disease complex of potato. In Maine, only strains AG-3 and AG-5 ofR. solani (4, 5) have been identified as attacking potato and causing four distinct types of symptoms: 1) black scurf (sclerotia) on tubers, 2) stem cankers, 3) aerial tubers and top rosetting, and 4) killing of sprouts. Crop rotation has been reported to reduce the incidence and severity ofR. solani on potato, but no single rotation method controls completely or to a high degree of reliability (11, 23, 24, 26). Deep moldboard plowing has been shown to reduce diseases caused byR. solani andSclerotium rolfsii Sacc. in crops other than potatoes (2, 7, 15, 19, 20). However, Gudmestadet al. (6) reported that deep moldboard plowing increased the severity ofR. solani on stems and stolons of potatoes. The reduction of diseases caused byR. solani andS. rolfsii by moldboard plowing is attributed to low inoculum densities in the upper soil layer by the burial of sclerotia to depths where germination and infection were prevented (14, 15, 19, 20). However, disking to a depth of 5–7 cm did not affect disease as the inoculum remained in the root zone (14, 15). Gurkin (7) states that the rationale for deep moldboard plowing is to promote decay of organic matter, remove organic matter from the infection court and to bury the sclerotia below the infection court. Cultural control techniques are largely preventive and are designed to reduce the quantity or the activity of inoculum by means of crop rotation, tillage practices, green manure crops, etc. (22). This study was conducted to determine individual effects and possible interactions of deep moldboard plowing versus chisel plowing in various rotation crops on the presence ofR. solani AG-3 in the soil and on the incidence of Rhizoctonia disease complex of potato.  相似文献

Seed potatoes from Maine, New Brunswick (Canada), Wisconsin, Minnesota and North Dakota were examined during 4 years for contaminating fusarial propagules on tuber surfaces. The degree of contamination was estimated by counting the number of infections that developed on cut surfaces of 100 pieces cut from 50 tubers removed from the tops of 6 cwt bags. Lesion counts were made after 2-weeks incubation at 13 to 16 C. Of seventy-seven seed stocks examined, 29 developed lesions on 91–100% of the seed pieces. Twenty-four of these heavily contaminated stocks came from Maine and New Brunswick. For the different years,Fusarium solani was obtained from 70 to 90%, andF. roseum from 5 to 28% of the lesions cultured each year. BothFusaria were on potatoes from the different production areas. The infectious propagules were in soil washed or brushed from tuber surfaces, and surface-borne inoculum from the tubers. A preliminary report on identifying contamibefore cutting. Plant emergence and productivity were correlated with tuber contamination.  相似文献

As part of a project to investigate the possibility that teratogenic factors are formed in potato tubers infected with the late blight fungus,Phytophthora infestans, a method was devised which allowed quick, efficient inoculation of large numbers of potato tubers. Tubers were placed in specially constructed trays, wounded by puncturing the periderm with flower “pin frogs”, and sprayed with a zoospore suspensionof P. infestans prepared from cultures grown on rye agar at 20C (68F) for 10 days. Approximately 95% of Katahdin and 93% of Russet Sebago tubers inoculated by this method exhibited typical late blight lesions after two weeks incubation at 20C, 80–90% RH. Silver scurf, caused by Helminthosporium solani, and black dot, caused by Colletotrichum coccodes, cause tuber blemishes on potato (Solanum tuberosum L.) which affect processing and fresh market trade. Tubers from ten cultivars were collected at harvest from three organic farms in Wisconsin and categorized as symptomatic or asymptomatic based on visual symptoms of silver scurf and black dot and/or signs of H. solani and C. coccodes. Tuber incubation and PCR assays were performed on asymptomatic tubers to detect H. solani and C. coccodes. Tuber incubation and PCR assays were in slight to fair agreement (kappa coefficient <0.4) for detecting both pathogens. Most asymptomatic tubers tested were positive by one or both assays for H. solani (75 %) or C. coccodes (94 %). Minituber inoculation assays were also performed to screen potato lines for resistance to silver scurf. Of the 14 lines tested, a diploid interspecific hybrid, C287, had consistently low sporulation, suggesting it has partial resistance to silver scurf. Since the majority of tubers harvested are already infected with one or both pathogens further research should focus on organically acceptable management practices that may inhibit disease development in field and in storage.  相似文献

Surgical wounds are common injuries of skin and tissues and usually become a clinical problem. Until now, various synthetic and natural peptides have been widely explored as potential drug candidates for wound healing. Inhibition of the TNF-α signaling pathway and promotion of angiogenesis are suggested to be involved in their effects. Angiogenesis at the wound site is one of the essential requisites for rapid healing. In the present study, a novel peptide extract derived from the natural source Lates calcarifer, commonly known as sea bass or barramundi, was evaluated for its wound healing property. The specific acidic and enzymatic approaches were employed for producing sea bass extract containing small size peptides (molecular weight ranging from 1 kD to 5 kD). The cytotoxicity of the extract was examined in HaCaT and NIH3T3. After this, the effects of enzyme digested peptide extracts of sea bass on wound healing in mice were investigated. The peptide extracts (660 and 1320 mg/kg/day) and control protein (1320 mg/kg/day) was orally given to the wounded mice, respectively, for 12 days. The surgical method was improved by implanting a silicone ring at the wound site. The ring avoided the contracting effect in murine wounds, making it more closely related to a clinical condition. The results showed promising improvement at the wound site in mice. Sea bass peptide extracts accelerated the wound healing process and enhanced the microvessel formation at the wound site. The remarkable effects of this novel sea bass peptide extract in healing traumatic injuries revealed a new option for developing wound management.  相似文献

The effect ofRhizoctonia solani infection on the yield and quality of tubers destined for processing was evaluated in a 10 × 2 factorial experiment. Inoculation of 10 cultivars withR. solani significantly decreased total and marketable yields and significantly increased the number of malformed and fissured tubers as well as the number of tubers with black scurf. Specific gravity was significantly decreased in 3 of the 4 years. Chip color at harvest was adversely affected in 2 of the 4 years and over the 4 years combined; whereas chip color three months after harvest was adversely affected in all 4 years.  相似文献

Silver scurf, caused byHelminthosporium solani, is considered to be a tuberborne disease of potato. The ability ofH. solani to overwinter in soil was evaluated in field studies at two locations in New York during 1992. Field plots were established at sites whereH. solani infected potatoes had been produced in 1991. Plants produced from pathogen-free tubers of four cultivars yielded tubers infected with silver scurf at both locations. Up to 61% of tubers were infected in some plots. The ability ofH. solani to colonize leaf tissue was evaluated inin vitro assays. Detached leaves of ten crops were sprayed with a spore suspension ofH. solani and incubated for 20 days.H. solani colonized and sporulated on senescent leaf tissue of alfalfa, sorghum, rye, oats, corn and wheat, and only colonized senescent tissue of rapeseed, red clover and buckwheat. No growth was observed on potato leaf tissue. These results indicate that soil survival and saprophytic ability may be important in the epidemiology of silver scurf of potato.  相似文献

Summary The viability of five pathogens was decreased by treatment with hot water when tested in vitro.Polyscytalum pustulans was most sensitive andRhizoctonia solani least sensitive. Potato tubers were exposed to 55°C for 5 min in a commercial continuous hot water treatment plant using naturally contaminated seed tubers and tubers which had been inoculated by dipping in comminuted cultures. The frequency of eyes colonised byP. pustulans, Helminthosporium solani, andR. solani was reduced to virtually zero and the effect persisted on tubers subsequently stored at 4°C and at 15°C for up to 16 wk. Results withColletotrichum coccodes were inconclusive. Treatment suppressedPenicillium spp. which, however, rapidly recolonised the eyes during storage, leading to higher contamination levels in the treated than in the untreated tubers. With tubers inoculated withPhoma foveata, good control was achieved when the incubation period before treatment was 10 d but not when the period was 42 d.  相似文献

Field and laboratory studies were conducted in 1985–1986 to evaluate the relative severity ofRhizoctonia solani damage among 26 clones and five potato (Solanum tuberosum) cultivars. A test with true potato seeds showed that resistance toR. solani could be identified in two years. One highly resistant line was identified by this method. Moderate degrees of resistance were found in lines from the USDA Potato Breeding Program (BARC) with russet types producing a higher percentage of highly resistant lines than round whites. A laboratory test revealed that apical sprout damage (nipping) was variable within and between lines and that it was an important phase of disease development to measure when determining resistance because of its effects on the resulting plant growth and production of marketable tubers. It was also determined that to accurately define resistance toR. solani in potato, all phases of the disease must be evaluated to insure against resistance “breakdown” at some point in plant development.  相似文献
