Summary Tubers of clones from a diploid hybrid population ofSolanum phureja andS. stenotomum were screened for resistance to tuber soft rot caused by strains ofErwinia carotovora subsp.atroseptica (Eca),E. carotovora subsp.carotovora (Ecc) andE. chrysanthemi (Ech). Significant, positive correlations between resistance to the different strains and species were observed, indicating that screening for resistance to tuber soft rot could be accomplished by using just one of the three pathogens. Strains of Ecc and Ech were found to be much more virulent than strains of Eca. Inoculating with Ecc and Ech resulted in more distinct differences among susceptible, moderately resistant and resistant clones than inoculating with Eca.  相似文献

The aim of this work is the evaluation of tetraploid potato clones with germplasm fromS. chacoense andS. phureja for chipping and resistance to tuber soft rot (Erwinia carotovora subsp.carotovora). Tetraploid potato clones were obtained via 2n eggs through 2x–4x crosses. Cytological mechanisms were investigated in the diploid hybrid 2n-egg producer used as parent in the unilateral sexual polyploidization scheme; the omission of the second meiotic division and cytokinesis failure at the end of meiosis were identified with a genetic consequence equivalent to second division restitution (SDR). Tuber marketable yield, specific gravity and chip-processing ability evaluated at harvest and after cold storage with and without reconditioning were higher in some clones as compared to theS. tuberosum tetraploid parent. Tuber soft rot resistance, contributed byS. chacoense, was successfully transferred to the4x level. After inoculation withErwinia carotovora subsp.carotovora and estimation of the rotted area, one tetraploid clone proved to be resistant and three partially resistant. The genetic value of exotic germplasm and sexual polyploidization strategy are discussed in relation to potato improvement.  相似文献

Summary The effect on the development of blackleg symptoms and on yield of inoculating seed tubers of 15 cultivars withErwinia carotovora subsp.atroseptica (Eca) was investigated. Tubers were planted either whole or as pieces. Eca inoculation reduced the mean yield per plant by 8% for whole tubers and by 12% for cut tubers. There was no correlation between blackleg symptoms and yield loss.  相似文献

Diploid interspecific hybrids from crosses betweenSolanum tuberosum haploids andS. canasense, S. multidissectum, andS. tarijense, along with tetraploid and hexaploid somatic hybrids S.tuberosum (+)S. commersonii, were screened for resistance to blackleg and tuber soft rot byErwinia carotovora subsp.atroseptica andE. carotovora subsp.carotovora. Among the diploid hybrids, those involving clone mlt la of S.multidissectum and tar 2b ofS. tarijense yielded the highest number of genotypes resistant to both blackleg and tuber soft rot. By contrast, all the hybrids involving clone tar 1lb ofS. tarijense were susceptible to both bacterial diseases. As far as resistance of somatic hybrids is concerned, the most interesting genotype was the tetraploid one which showed resistance to both blackleg and tuber soft rot Significant correlations were found betweenE. carotovora subsp.atroseptica andE. carotovora subsp.carotovora both in resistance to blackleg and in resistance to tuber soft rot, suggesting that resistance to the two subspecies is controlled by the same gene/s or that the relevant genes are linked. On the other hand, correlations between blackleg and tuber soft rot were never significant, indicating that different mechanisms may control resistance in tuber and stems. Alternatively, the resistance could be controlled by the same gene/s under a different spatial/temporal expression pattern. Twelve diploid sexual hybrids and one tetraploid somatic hybrid were selected for resistance to blackleg and/or tuber soft rot and for tuber characteristics.  相似文献

The effect ofFusarium solani (Fsc),F. sambucinum (Fs),Erwinia carotovora subsp.atroseptica (Eca), andE. carotovora subsp.carotovora (Ecc) inoculated singly and in combination (FscFs, FscEca, FscEcc, FsEca, FsEcc, and EcaEcc) on wound-healed potato seed pieces of the cultivar Kennebec was studied. Potato seed pieces wound-healed for 5 days at 13 C and approximately 100% relative humidity, inoculated with Fsc, Fs, Eca, Ecc and their combinations, and incubated for 10 days at 9 C were protected from Eca, Ecc, and EcaEcc. When wound-healed potato seed pieces were inoculated with Fsc, Fs, Eca, Ecc, and their combinations and planted in soil maintained at 7, 10, 15, 20 and 25 C, wound-healing protected seed pieces from all pathogens and their combinations in seed pieces held for 10 days at 10 and 15 C. When seed pieces of cultivars Atlantic, Kennebec, Norchip, Russet Burbank and Superior were inoculated with Fsc, Fs, Eca, Ecc, and their combinations, different levels of protection were observed: Russet Burbank had the highest level of protection, Atlantic and Kennebec were intermediate, and Norchip and Superior were slightly protected. Dusting mancozeb on wound-healed seed pieces before inoculation with Fsc, Fs, Eca, Ecc, and their combinations increased protection against all pathogens and their combinations by 39% or more compared to the nonchemical control.  相似文献

Tubers of 11 clones ofSolanum tuberosum subsp.andigena (andigena) and 12 cultivars ofS. tuberosum subsp.tuberosum (tuberosum) were inoculated with water suspensions (5.5 × 104 5.5 × 105, 5.5 × 106 CFU/ml) ofErwinia chrysanthemi. Tubers were inoculated immediately after harvest and after 6 and 16 wk of storage at 4 and 23°C. Tuber rot incidence in andigena and tuberosum increased as inoculum concentration increased. Based on tuber rot severity, clones of andigena were classified as resistant, intermediate, or susceptible and cultivars of tuberosum were classified as intermediate or susceptible. Rot severity increased in all tubers stored at 4°C and in tubers of tuberosum stored at 23°C; rot did not increase in tubers of andigena stored at 23°C. Electrolyte leakage (EL), total sugars (TS), reducing sugars (RS), non-reducing sugars (NRS) and dry matter (DM) were determined in non-inoculated tubers. There was a significant positive correlation between the rate of EL, concentration of RS and tuber rot in andigena and tuberosum. Tuber rot and DM were negatively correlated in tubers of andigena; but they were not correlated in tuberosum. Clones of andigena with low EL, TS, RS, and high DM were resistant to tuber rot, and the incidence of tuber rot in these clones was much less influenced by temperature and length of storage. The influence of temperature and length of storage on susceptibility toE. chrysanthemi may be explained by increased cell membrane permeability; increased leakage of accumulated sugars in potatoes stored at 4°C could favor bacterial proliferation resulting in more disease.  相似文献

Resistance to bacterial soft rot in potato tubers maintained aerobically was induced by wounding tubers 4 to 24 h prior to inoculation withErwinia carotovora subsp.carotovora. This wound-induced resistance phenomenon was blocked by hypoxic stress. An hypoxia-induced resistance mechanism also was detected: tubers acclimated to a hypoxic environment were resistant to rot when inoculated with aerobically grownE. c. subsp.carotovora and incubated in atmospheres made hypoxic by argon or nitrogen. With increased pretreatment, hypoxia-induced resistance approached the levels of resistance observed in tubers inoculated with aerobic-adapted bacteria and incubated aerobically. Mechanically harvested tubers of 14 potato cultivars grown on both loamy sand and silt loam soils were evaluated for resistance to bacterial soft rot caused byErwinia carotovora subsp.atroseptica. Cultivars were also assayed for calcium and dry matter content to determine possible correlations with soft rot resistance. Resistance of potato tubers to bacterial soft rot was assayed after harvest by inoculating tubers and determining percent surface area decayed after four days in a mist chamber. Tubers of the cultivars differed widely in resistance to bacterial soft rot. The ranking of the resistance of tubers from different cultivars to bacterial soft rot was similar for both soils, although disease severity in tubers from a particular soil type varied with the season. Of the cultivars tested, tubers of Butte, Russet Burbank, and Nor-gold Russet were ranked as the most resistant, whereas tubers of Redsen, Norchip, Norland and Belchip were rated as the most susceptible. Tubers of Rhinered, Red La Soda, Superior, Pontiac, Atlantic, and Nooksack were intermediate in resistance. Neither calcium concentration in medullar tissues nor dry matter of potato tubers alone could be correlated with ranking of the cultivars with respect to severity of bacterial soft rot. However, a multiple linear regression model incorporating both variables indicated that tubers with a high content of both calcium and dry matter were likely to be less susceptible to bacterial soft rot than were tubers with lower percentages of these variables.  相似文献

Summary Tubers of diploid interspecific hybrids, hexaploid somatic hybrids ofSolanum tuberosum andSolanum brevidens, a fusion parental line (S. tuberosum) and several Polish and American commercial cultivars were screened for resistance toErwinia carotovora subsp.atroseptica andErwinia chrysanthemi, in laboratory tests. Both groups of hybrids showed a high level of resistance to soft rot and gave a similar type of resistant reaction, which distinguished them from the cultivars and fusion parent. Tubers of the diploid interspecific hybrids had a high starch content. Our results indicate the possibility of obtaining breeding material with a high starch content and resistance to bacterial soft rot.  相似文献

Development of potato cultivars resistant to Erwinia species (Erwinia carotovora subspeciesatroseptica, E. carotovora subspeciescarotovora, andErwinia chrysanthemi) causing soft rot of stems and tubers involves identification and transfer of the resistance to breeding lines possessing desirable agronomic traits. In total, 363 accessions of six wildSolanum species, three somatic hybrids and nine sexual progeny of somatic hybrids were screened for stem soft rot resistance. Thirty-one, 50, and 67 percent of accessions ofSolanum boliviense, Solanum chacoense, and Solanumsanctarosae, respectively, were resistant or highly resistant, whereas, 100, 97, and 95 percent of accessions ofSolanum canasense, Solanum tarijense, andSolanum spegazzinii were susceptible or highly susceptible, respectively. Two tuber soft-rot resistant somatic hybrids, A937 and T355-11 produced earlier by the fusion ofSolanum brevidens (PI 218228) andSolanum tuberosum (PI203900) orS. tuberosum cv. Russet Burbank, respectively, and the sexual progeny of A937 andS. tuberosum cv. Katahdin were also highly resistant. Transfer of resistance fromS. brevidens to somatic hybrid (A937) and subsequently to the progeny suggests that the resistance is simply inherited. In addition to the identification of 65 resistant or highly resistant clones of wildSolanum species, this study has identified three tuber soft rot-resistant clones that are also resistant to stem soft rot. Potato cultivars showed a range of responses to the pathogen.  相似文献

The Quaggy Joe potato variety is high-yielding and medium to medium-late maturing with attractive, round-oval, white-skinned, white-fleshed tubers with medium-shallow pink eyes. Its major use is expected to be as a maincrop tablestock variety. Quaggy Joe is resistant to the potato leafroll virus, and is also moderately resistant to early blight (Alternaria solani), dry rot (Fusarium sambucinum), and soft rot (Erwinia carotovora subsp.carotovora). Quaggy Joe tubers have shown very little shatter or blackspot bruising, but sprout earlier than Atlantic. Young plants have shown sensitivity to the herbicide metribuzin.  相似文献

Summary The efficiency of acetylsalicylic acid (ASA) in inducing localised acquired resistance against infection byErwinia carotovora subsp.carotovora was evaluated by treating potato tubers with ASA at three concentrations. Three days after treatments, tubers were inoculated withE. carotovora subsp.carotovora by wound inoculation or by irrigation with a bacterial suspension. Experiments were performed for two consecutive years. Statistical analysis revealed that treatment of tubers by immersion in ASA solutions at low concentrations induced a significant reduction in the soft rot incidence. Wounding of the tubers was the most effective inoculation method and ASA at concentration of 0.0125% (w/v), pH 7 was more efficient than at 0.025 or 0.05%. No phytotoxicity of such treatment was observed.  相似文献

Blackleg and soft rot diseases, caused by the bacteriumErwinia carotovora, are among the diseases that cause important losses in culture and storage of potato. In this paper, we introduced bacterial resistance into potato, via genes encoding for proteins with antibacterial activity. For this purpose, potato clones were transformed either with the gene encoding the acidic attacin protein fromHyalophora cecropia, or with the gene encoding the cecropin analog peptide SB37. These clones were evaluated for soft rot and blackleg resistance, after inoculation with the bacterial strainErwinia carotovora subsp.atroseptica T7. Results reported in this paper indicate that a considerable percentage of the potato clones (15–22%) showed increased resistance to bacterial infection, revealed by reduced severity of blackleg or soft rot symptoms. Expression of the transgenes was demonstrated in some of the clones by Northern blot analysis. This is the first report indicating that expression of the gene encoding for an attacin protein and for the cecropin SB-37 peptide in transgenic potato confers increased resistance to bacterial infection.  相似文献

Infection of potato plants and tubers with the bacteriumErwinia carotovora subsp.atroseptica produces blackleg and soft rot diseases, which cause significant losses to crops and stored potatoes. In order to obtain resistance against this bacterium, the genechly encoding the enzyme lysozyme from chicken was introduced into potato plants (cv. Desirée) viaAgrobacterium- mediated transformation. Sixty-three and 69 transgenic potato clones were evaluated in the greenhouse for resistance to blackleg and soft rot diseases, respectively. Results reported in this paper indicate that 21%-29% of the potato clones showed increased resistance to infection by the bacteriumE. c. subsp.atroseptica T7, as revealed by a reduced severity of blackleg or soft rot symptoms. Nine clones showing different levels of resistance were selected for further molecular analysis. The number of copies of the transgene integrated in the plant genome of these clones was estimated by Southern blot analysis. The level of transgene expression, detected by Northern blot analysis, correlated with the level of resistance detected in these clones.  相似文献

Bacterial soft rot is a serious disease in potato (Solanum tuberosum L.), causing rapid tuber tissue maceration and, consequently, marketable yield loss. Soft rot bacteria, including Pectobacterium carotovorum subsp. carotovorum (Pbc), are favored by moist conditions, which are prevalent in large potato storage facilities. However, although most potatoes in North America are stored before use, there are no published surveys of soft rot resistance in cultivars exposed to long-term storage conditions. Thus, we tested 65 cultivars and 13 breeding lines for soft rot resistance after 6 months of storage. There was a significant effect of cultivar and production environment on soft rot resistance score. During 6 months of storage, tuber soft rot resistance in resistant clones did not change, while it changed in susceptible clones. The three most resistant cultivars to soft rot were Freedom Russet, Anett, and Alaska Red Eye.  相似文献

Summary Tubers of 17 cultivars were inoculated withErwinia carotovora subsp.atroseptica (ECA) and planted in 1986 and 1987 in the Negev in randomised plots in three blocks where eitherVerticillium dahliae orAlternaria solani were present or where both were absent. ECA reduced the yield in the control block and exacerbated the yield losses in the disease blocks, but the intensity of symptom expression did not always accord with the extent of yield loss. ECA had a greater effect on plants infected withA. solani than on those withV. dahliae. Fungi of the genusFusarium Link isolated in Perú from potatoes (Solanum tuberosum L. subsp.andigena Juz. & Buk.) affected by: i) wilt, ii) dry rot of stored tubers, iii) secondary rot in tubers affected by “cork” disease (potato mop-top virus?), caused dry rot when inoculated to tubers of Renaacimiento variety. The identification of some of theseFusaria suggests thatFusarium solani Snyder & Hansen is associated with wilt and dry rot, andFusarium oxysporum Snyder & Hansen with rot secondary to cork disease. Tubers of eight potato varieties inoculated with fourFusaria representative of those isolated showed a great range in levels of susceptibility to colonization by theseFusaria, Sapa being the most susceptible variety, and Mantaro appearing to be the most resistant. These results suggest that useful levels of resistance toFusarium tuber rot are available in the Peruvian germ-plasm.  相似文献

Summary The susceptibility of six new Austrian cultivars to soft rot, stem rot and blackleg caused byErwinia carotovora subsp.atroseptica was assessed over 2 years in two locations. Soft rot susceptibility was assessed by whole tuber and half tuber tests, stem rot by stem inoculation and blackleg by planting inoculated tubers in the glasshouse. The susceptibility of the cultivars tested with the tuber and blackleg test methods were in general agreement. The half tuber test was more precise in differentiating intermediate levels of susceptibility than the whole tuber test. The reaction of the cultivars to soft rot could be classified as susceptible (Gina and Romina), moderately susceptible (Ditta, Sonja and Bintje), or slightly susceptible (Treff, Komet and Désirée). Results from the stem rot test did not agree with this classification and also depended upon the physiological state of plants as influenced by growing season and glasshouse conditions.  相似文献

Summary A cultivar collection was assessed for tuber resistance to the soft rot organism,Erwinia carotovora subsp.atroseptica (=Eca), and for the strength of the defence reaction associated with wound healing, by measuring the inoculum density needed to initiate infection (effective dose). Wound healing was more efficient in susceptible genotypes than in resistant ones (Fig. 1b), and resulted in changes of the ranking of cultivars, and a narrowing of the variation for soft rot resistance (Fig. 1a). No relationship was found between enzymic maceration of tissue and effective dose (Fig. 2a). The resistance ratings of cultivars were in general agreement with the effective dose and with symptom expression (Fig. 2b). Resistance behaviour was modified by growing conditions (harvest year) (Table 1), and cultivars need therefore to be assessed over several years.

Summary In the Netherlands seed potato crops are harvested when still green, the haulm being destroyed before harvest. We compared the effect on the contamination of seed potatoes byErwinia carotovora subsp.atroseptica (Eca) andE. chrysanthemi (Ech) of the common method of haulm destruction by flailing and chemically destroying remaining stems, with the recently developed green crop lifting method. After chenical haulm destruction the levels of contamination with both Eca and Ech surviving cold storage were found not to differ significantly from those after green crop lifting. Losses due to watery wound rot (Pythium ultimum) were considerable in the warm and humid conditions during 1991, especially after green crop lifting.  相似文献
