Prenatal stress is known to affect several offspring characteristics, but its effects depend among other factors on the period of gestation in which it is applied. In the present study, oral administration of hydrocortisone-acetate (HCA) was used to elevate cortisol concentrations in pregnant sows to levels also observed after psychological stress. HCA was administered during three different periods of gestation (115 days in pigs): period 1: 21-50 (P1, n = 10), period 2: 51-80 (P2, n = 10) and period 3: 81-110 (P3, n = 10) days after insemination. Control sows (n = 11) received vehicle from 21-110 days after insemination. When P1-, P2- and P3-sows did not receive HCA, they also received vehicle. During gestation, weekly saliva samples were taken from the sows to determine salivary cortisol concentrations. Treatment effects on sow, litter and piglet characteristics were determined. In addition, two female piglets per litter were subjected to an ACTH-challenge test at 6 weeks of age to determine the adrenocortical response to ACTH. Pigs were slaughtered at 6 months of age and slaughter weight, back fat thickness and percentage of lean meat were analysed. During the period of treatment with HCA, salivary cortisol concentrations were increased in P1-, P2- and P3-sows compared to control sows (P < 0.01). The total number of piglets born per litter did not differ among treatment groups (P > 0.30), but pooled HCA-litters had a higher percentage of live born piglets (P < 0.05) and fewer mummies than control litters (P < 0.05). Gestation length did not differ among treatment groups (P = 0.21), but did affect treatment effects on birth weight. Overall, HCA-piglets weighed less at birth, and remained lighter until weaning (P < 0.05). The salivary cortisol concentrations after i.m. injection of ACTH (2.5 IU/kg) were lower in P1- and P3-piglets compared to control piglets. At slaughter, HCA-treatment indirectly decreased lean meat percentage and increased back fat thickness. In conclusion, elevated peripheral cortisol concentrations in pregnant sows affect both litter characteristics and piglet physiology, the latter depending on the period of gestation during which concentrations were elevated. Underlying mechanisms require further investigation.  相似文献

本试验利用母猪行为以及唾液皮质醇和淀粉酶含量探索空间限制对母猪生理心理状态造成的影响。试验选择大白妊娠母猪64头,随机平均分成2组,分别采用限位栏(180 cm×60 cm×110 cm)和群养模式(568 cm×448 cm×50 cm)饲养,群养模式下每圈8头母猪,共4圈。于试验1、3、5、6、8、10、11、13、15周第1、2天分别采用10 min间隔扫描取样法记录母猪行为和采集试验母猪唾液样本,检测唾液皮质醇水平和唾液淀粉酶含量。结果表明:限位栏母猪表现无食咀嚼、啃栏、啃槽行为的次数显著高于群养母猪。限位栏母猪的唾液皮质醇和淀粉酶表现出相反的分泌趋势,无食咀嚼与唾液淀粉酶含量之间存在显著的正相关关系(r=0.336,P=0.048)。群养模式下试验母猪的唾液皮质醇水平、淀粉酶含量与无食咀嚼行为之间没有显著性相关的关系。  相似文献

OBJECTIVE: To determine whether concentrations of proinflammatory cytokines, acute-phase proteins, and cortisol differ at parturition among 3 categories of sows (noninoculated, clinically affected and nonaffected following intramammary inoculation with Escherichia coll). ANIMALS: 16 sows. PROCEDURE: Sows were allocated to inoculated (n = 12) or noninoculated (4) groups. Inoculated sows received intramammary administration of E coli (serotype O127) during the 24-hour period preceding parturition. Blood samples were collected from noninoculated and inoculated sows for 3 consecutive days within 3 to 11 days before farrowing and inoculation. Samples were also collected 0, 24, 48, 72, and 96 hours after farrowing and inoculation. Inoculated sows were further categorized as affected (4 sows) or nonaffected (8 sows) based on clinical signs of disease. Serum tumor necrosis factor (TNF)-alpha, plasma interleukin (IL)-6, and serum amyloid A (SAA) concentrations were measured by use of ELISA; serum haptoglobin concentration was assayed by use of a hemoglobin-binding method; and plasma cortisol concentration was determined by use of radioimmunoassay. RESULTS: Plasma or serum concentrations of TNF-alpha, IL-6, and SAA of both categories of inoculated sows were significantly increased by 24 hours after intramammary inoculation of E coli, compared with concentrations in noninoculated sows. Concentrations of serum TNF-alpha and plasma IL-6 were significantly higher in inoculated sows that developed clinical mastitis than in nonaffected inoculated sows. CONCLUSIONS AND CLINICAL RELEVANCE: Concentrations of TNF-alpha and IL-6 are promising markers for the identification of periparturient sows with subclinical coliform mastitis. Identification of such sows should help improve the health and survival of piglets.  相似文献

Blood and saliva samples were taken from groups of pigs maintained in intensive conditions. Multiple samples were taken from two unrestrained pregnant sows fitted with jugular cannulae. Single samples were taken from groups (mixed gilts and entire males; 70–90 kg) which were lightly exercised (7) or restrained (12). The rate of salivary secretion was low and collection of adequate samples took 5 min; in a number of pigs no saliva could be obtained.In order to stimulate salivary secretion, pigs (70–80 kg) were injected with pilocarpine nitrate (25 mg, subcutaneous) which produced a copious flow of saliva persisting for at least 15 min.Resting sows had higher mean levels of cortisol in plasma, ultrafiltrate and saliva than the other groups, which did not differ from each other. Within the pilocarpine group, males had a higher ultrafiltrate level of cortisol than females. In most instances salivary cortisol was significantly greater than ultrafiltrate cortisol.Ultrafiltrate and plasma cortisol were highly correlated (r=0.883) but this correlation was low in the presence of pilocarpine (r=0.260). Salivary cortisol was poorly correlated with either plasma (r=0.167) or ultrafiltrate cortisol (r=0.278) and the correlation with plasma was even lower following the administration of pilocarpine (r=0.086).It was concluded that salivary estimates of cortisol in the pig were not usefully correlated with levels of ultrafiltrate (free) cortisol.  相似文献

Plasma corticosteroid concentrations are altered in pregnancy, during the reproductive cycle and by ovariectomy in many species. This study was designed to examine basal ACTH and cortisol in ewes of four different reproductive statuses: ovariectomized, nonpregnant cycling, nonpregnant noncycling, and pregnant. Blood samples were drawn every 4 hr for 48 hr from ewes during quiet, undisturbed conditions and analyzed for plasma ACTH, cortisol and progesterone concentrations. There were no significant changes in ACTH, cortisol or progesterone over time. Mean progesterone concentrations were significantly greater in the pregnant ewes than in all other ewes, and were greater in cycling ewes than noncycling or ovariectomized ewes. Mean ACTH was significantly greater in pregnant ewes than noncycling ewes, and mean cortisol was significantly greater in cycling ewes than in nonpregnant cycling or noncycling ewes. Ovariectomized ewes also had significantly greater mean cortisol concentrations than cycling ewes. The results demonstrate that there is an increase in basal ACTH and cortisol in ovine pregnancy.  相似文献

OBJECTIVE: To compare well-being, performance, and longevity of gestating sows housed in stalls or in pens with an electronic sow feeder (ESF). ANIMALS: 382 pregnant sows of parities 1 through 6. PROCEDURE: Sows were housed in separate stalls (n = 176) or group pens (206) with an ESF. Well-being of sows was assessed at various time points in terms of injuries, salivary cortisol concentration, and behavior in a novel arena or to a novel object. Farrowing performance and longevity of sows were also assessed. RESULTS: Total injury scores (TIS) of sows in pens were significantly higher at initial introduction and mixing. In stall-housed sows, TIS was significantly higher during late gestation. The TIS and cortisol concentration were significantly lower in stall-housed sows, compared with values for sows in pens. As parity increased, the likelihood of higher median TIS decreased significantly in pen-housed sows and increased significantly in stall-housed sows. The TIS of sows in pens was negatively correlated with body weight and backfat thickness, whereas these correlations were positive in stall-housed sows. Farrowing performance and results for novel arena or objects did not differ. Proportion of sows removed was significantly higher for pens than for stalls; lameness was the major reason for removal for both systems. CONCLUSIONS AND CLINICAL RELEVANCE: Stalls impose space restrictions for larger sows, resulting in injuries during late gestation. Interventions are needed to minimize aggression during initial introduction and mixing and at the ESF in pens to reduce severe injuries or lameness of gestating sows.  相似文献

Exposing a pregnant sow to stress has been shown to affect the resulting offspring. Our objective was to determine if rough handling of pregnant sows altered the physiology of her offspring and if these alterations were different from an experimentally induced model of prenatal stress. Sow treatments consisted of i.v. injections of ACTH (1 IU/kg of BW), exposure to rough handling for 10 min (Rough), or no treatment (Control) once a week during d 42 to 77 of gestation. To determine the plasma cortisol response to treatments, blood (5 mL) was collected from 30 sows after treatment administration. To conduct the prenatal stress study, a separate group of 56 sows was used in 1 of 4 replicates. At birth, production data were collected for each litter, including birth weight, number born, anogenital distance, and pig viability. At weaning, pigs were blocked by BW and sex, and placed in a nursery pen of 6 pigs, with 2 pigs from each treatment group. To assess the effect of treatments on cortisol, corticosteroid-binding globulin (CBG), and hematological cell profiles, blood was collected every other day for 10 d after weaning. Application of treatments caused plasma cortisol concentrations to be greatest in ACTH sows compared with Control sows (P < 0.001), with Rough sows having intermediate values (P = 0.07). Treatments did not affect the number of pigs born, number of stillborn, or pig viability (P > 0.40). The ratio of cortisol to CBG did not differ between treatments (P = 0.09). Hematological variables did not differ between treatments (P > 0.19). Pigs born to ACTH sows had a smaller anogenital distance compared with controls (P < 0.03), with pigs from Rough sows being intermediate. Our data indicate that swine exposed to prenatal stress (ACTH injection) can have alterations in sexual morphology without effects on growth or the immune cell populations measured in this study.  相似文献

The objective of this study was to determine the effects of boar presence on aggression, shoulder scratches, and salivary cortisol in group-housed sows during the period after mixing, which is when dominance hierarchies are formed. A total of 225 York-shire sows were used. Five groups of 15 sows were each exposed to 1 of 3 levels of boar presence (n = 15): physical (boar in pen with sows; 2.15 m2 per sow), fenceline (boar housed adjacent to sows; 2.3 m2 per sow), or control (no boar in the room; 2.3 m2 per sow). The experiment was divided into 2 phases. In phase 1, behavioral measurements were taken for 48 h after mixing and boar introduction. In phase 2, behavioral measurements were taken beginning on d 6 after mixing and included 24 h before and 48 h after boar removal. In phases 1 and 2, shoulder scratches were scored 24 h before and 24 h after mixing and boar removal, respectively. Saliva samples were collected each morning (0600 to 0700) and afternoon (1600 to 1700) during both phases. Frequencies of intersow aggressive contact (bite, body knock, and head knock) and threats as well as frequencies and durations of fighting bouts were determined from video recordings during daily feeding (49.6 +/- 1.4 min) and nonfeeding (21 +/- 0.2 h) periods. During phase 1, boar presence did not affect the frequency of threats during either the feeding or nonfeeding periods. Control groups had fewer aggressive contacts at feeding during the 25- to 48-h period compared with the fenceline groups (P = 0.01). Total duration of fighting and average fighting bout duration were unaffected by treatment. However, the number of fights occurring during feeding 25 to 48 h postmixing was lower in the physical groups than in the fenceline groups (P = 0.023); measures for the control groups were intermediate. Physical sows also received fewer shoulder scratches postmixing than did control sows (P = 0.048). After mixing, physical sows showed greater morning (P = 0.08) and afternoon (6 h postmixing, P = 0.01; 32 h postmixing, P = 0.08) salivary cortisol concentrations compared with fenceline sows, although they were only numerically greater than those for control sows. During phase 2, removing the boar did not increase fighting among sows or affect shoulder scratches or salivary cortisol concentrations. The presence of a boar was minimally effective at reducing fighting and scratches during the postmixing period, and sows showed a greater stress response in the presence of a boar.  相似文献

OBJECTIVE: To compare adrenal gland stimulation achieved following administration of cosyntropin (5 microg/kg [2.3 microg/lb]) IM versus IV in healthy dogs and dogs with hyperadrenocorticism. DESIGN: Clinical trial. Animals-9 healthy dogs and 9 dogs with hyperadrenocorticism. PROCEDURES: In both groups, ACTH stimulation was performed twice. Healthy dogs were randomly assigned to receive cosyntropin IM or IV first, but all dogs with hyperadrenocorticism received cosyntropin IV first. In healthy dogs, serum cortisol concentration was measured before (baseline) and 30, 60, 90, and 120 minutes after cosyntropin administration. In dogs with hyperadrenocorticism, serum cortisol concentration was measured before and 60 minutes after cosyntropin administration. RESULTS: In the healthy dogs, serum cortisol concentration increased significantly after administration of cosyntropin, regardless of route of administration, and serum cortisol concentrations after IM administration were not significantly different from concentrations after IV administration. For both routes of administration, serum cortisol concentration peaked 60 or 90 minutes after cosyntropin administration. In dogs with hyperadrenocorticism, serum cortisol concentration was significantly increased 60 minutes after cosyntropin administration, compared with baseline concentration, and concentrations after IM administration were not significantly different from concentrations after IV administration. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that in healthy dogs and dogs with hyperadrenocorticism, administration of cosyntropin at a dose of 5 microg/kg, IV or IM, resulted in equivalent adrenal gland stimulation.  相似文献

选用5头妊娠后期母猪,于妊娠111d上午9∶00肌肉注射氯前列烯醇(PG)0.20mg。5头母猪皆于次日白天分娩。从注射前至分娩结束,在整个诱导过程中多次采血,监测了血清中孕酮(P4)、硫酸雌酮(E1S)、催乳素(PRL)和皮质醇(cortisol)4种激素浓度的变化。结果表明,诱导分娩过程中各种激素的变化和自然分娩基本一样。这就解释了用国产前列腺素诱导母猪分娩,只要处理时间合适(妊娠111d或112d),就会有良好效果,不会产生不良副作用。  相似文献

The site within the hypothalamic-pituitary axis at which cortisol acts to inhibit luteinizing hormone (LH) secretion was investigated in female pigs. Six ovariectomized, hypophysial stalk-transected (HST) gilts were given 1 microgram pulses of gonadotropin releasing-hormone (GnRH) iv every 45 min from day 0 to 12. On days 6-12, each of 3 gilts received either hydrocortisone acetate (HCA; 3.2 mg/kg body weight) or oil vehicle im at 12-hr intervals. Four ovariectomized, pituitary stalk-intact gilts served as controls and received HCA and pulses of 3.5% sodium citrate. Jugular blood was sampled daily and every 15 min for 5 hr on days 5 and 12. Treatment with HCA decreased serum LH concentrations and LH pulse frequency in stalk-intact animals. In contrast, serum LH concentrations, as well as the frequency and amplitude of LH pulses, were unaffected by HCA in HST gilts and were similar to those observed in oil-treated HST gilts. We suggest that chronically elevated concentrations of circulating cortisol inhibit LH secretion in pigs by acting at the level of the hypothalamus.  相似文献

OBJECTIVE: To evaluate effect of alternate-day oral administration of prednisolone on endogenous plasma ACTH concentration and adrenocortical response to exogenous ACTH in dogs. ANIMALS: 12 Beagles. PROCEDURE: Dogs were allotted to 2 groups (group 1, 8 dogs treated with 1 mg of prednisolone/kg of body weight; group 2, 4 dogs given excipient only). During a 30-day period, blood samples were collected for determination of plasma ACTH and cortisol concentrations before, during, and after treatment with prednisolone. From day 7 to 23, prednisolone or excipient was given on alternate days. Sample collection (48-hour period with 6-hour intervals) was performed on days 1, 7, 15, 21, and 28; on other days, sample collection was performed at 24-hour intervals. Pre- and post-ACTH plasma cortisol concentrations were determined on days 3, 9, 17, 23, and 30. RESULTS: A significant difference was detected between treatment and time for group 1. Plasma ACTH concentrations significantly decreased for 18 to 24 hours after prednisolone treatment in group-1 dogs. At 24 to 48 hours, ACTH concentrations were numerically higher but not significantly different in group-1 dogs. Post-ACTH plasma cortisol concentration significantly decreased after 1 dose of prednisolone and became more profound during the treatment period. However, post-ACTH cortisol concentration returned to the reference range 1 week after prednisolone administration was discontinued. CONCLUSIONS AND CLINICAL RELEVANCE: Single oral administration of 1 mg of prednisolone/kg significantly suppressed plasma ACTH concentration in dogs for 18 to 24 hours after treatment. Alternate-day treatment did not prevent suppression, as documented by the response to ACTH.  相似文献

In four Kenyan pig breeding units the pregnancy diagnosis of sows has been carried out in two groups: Group 1 (n = 1911): the sows were transrectaly pregnancy tested between Days 17-22 post-mating by ultrasound. Sows testing non-pregnant immediately received one dose of 400 IU pregnant mare serum gonadotropin (PMSG) (equine chorion gonadotropin, eCG) and 200 IU human chorion gonadotropin (hCG). On showing signs of oestrous, the animals were subsequently artificially inseminated (AI). Group 2 (n = 1923): sows were pregnancy tested by serum progesterone (P4)-based enzyme-linked immunosorbent assay (ELISA) on Day 17 post-breeding. P4 concentrations were categorized as positive (> 5 ng/ml) or negative (< 5 ng/ml). Sows testing nonpregnant immediately received one dose of 400 IU PMSG and 200 IU hCG by injection, and were subsequently artificially inseminated. The following parameters were evaluated: sows diagnosed non-pregnant, days from first post-weaning insemination until the sows were inseminated at their first return to oestrus; farrowing rate and total piglets born and number of live-born piglets in litters. The percentage of sows diagnosed non-pregnant in the two groups, as well as the totals of born piglets and of live-born piglets in litters did not differ significantly between the two groups. The number of days from the first post-weaning mating until the sows were artificially inseminated at their first return to oestrus and the administration of eCG and hCG was shorter (P < 0.01) and farrowing rate was higher (P< 0.01) in the ELISA-tested sows.  相似文献

Full term crossbred sows were selected to study the interaction of the immune system, hypothalamus-pituitary-adrenal axis, and growth in pigs born by Cesarean section (c-section; n=4 sows) or vaginal birth (n=4 sows). Gestation length and birth weight did not differ between vaginal birth and c-section pigs (P=0.34 and 0.62, respectively). Blood and tissue samples were collected from 44 pigs at birth. Forty-five pigs were weaned at 13 d. On d 14, pigs received an i.p. injection of lipopolysaccaride (LPS; 150 microg/kg) or saline at min 0, and blood samples were collected at -20, -10, 0, 5, 10, 20, 40, 60, 90, and 120 min. Vaginal birth pigs had 21% greater average daily gain than c-section pigs on d 14 (P<0.01). Basal serum concentrations of adrenocorticotrophin (ACTH) and cortisol were greater in c-section than vaginal birth pigs at birth (P<0.01) but were not different at 14 d (P=0.99 and 0.80, respectively). LPS increased serum concentrations of ACTH, cortisol, interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha; P<0.01) but the response was not different between c-section and vaginal birth (P>0.22). Basal serum concentrations of TNF-alpha tended to be greater in c-section vs vaginal birth pigs at 14 d (P=0.0967); however, basal serum concentrations of IFN-gamma tended to be lower in c-section pigs vs vaginal birth pigs at 14 d (P=0.0787). Expression of interleukin (IL)-6, IL-6 receptor, IL-1beta, and TNF-alpha mRNA did not differ between vaginal birth and c-section pigs but changed in an age and tissue dependent manner. Thus, reduced growth rate of c-section pigs is associated with altered immune system function.  相似文献

Adrenal and/or thyroid gland function tests were evaluated in horses at various times during short-term therapy with phenylbutazone, stanozolol, and boldenone undecylenate. There were no significant treatment or time effects on mean basal plasma cortisol concentrations in horses during treatment with the following: phenylbutazone, given twice daily (4 to 5 mg/kg, IV) for 5 days; stanozolol, given twice weekly (0.55 mg/kg, IM) for 12 days; boldenone undecylenate, given twice weekly (1.1 mg/kg, IM) for 12 days; or nothing. There was no significant effect of phenylbutazone treatment on the changes in plasma cortisol concentration during the combined dexamethasone-suppression adrenocorticotropic hormone (ACTH)-stimulation test. Plasma cortisol concentration was significantly decreased from base line at 3 hours after dexamethasone administration and was significantly increased from base line at 2 hours after ACTH in all horses (P less than 0.05). Likewise, the stimulation of basal plasma cortisol concentrations at 2 hours after administration of ACTH (P less than 0.05) was not affected by treatment with stanozolol or boldenone undecylenate. There were no significant treatment effects on mean basal plasma concentrations of thyroxine (T4) or triiodothyronine (T3) among horses during the following treatments: stanozolol, given twice weekly (0.55 mg/kg, IM) for 12 days; boldenone undecylenate, given twice weekly (1.1 mg/kg, IM) for 12 days; or nothing. At weaning, 162 sows were assigned randomly to six treatments (27 in each treatment) according to a 2 X 3 factorial arrangement: two levels of supplementary folic acid (0 and 5 mg/kg of diet) and three treatments to stimulate ovulation (none, flushing and pregnant mare serum gonadotropin [PMSG] injection). All sows were mated twice within 7 d after weaning. Of the 162 animals originally selected, 123 sows were pregnant and used in this trial. The flushing treatment consisted of allowing sows ad libitum access to feed from the day after weaning through the 1st day of behavioral estrus, whereas control animals received 2.4 kg of feed daily. The hormonal treatment consisted of one i.m. injection of 1,250 IU of PMSG the day after weaning. The commercial-type diet used as the control was computed to contain .6 mg folates per kilogram. Folic acid supplementation elevated (P less than .001) serum folates between weaning and 30 d of gestation. Fetuses of sows fed the diet supplemented with folic acid had a higher (P less than .05) total protein concentration than fetuses of control sows, whereas RNA and DNA concentrations and protein:DNA ratio were not affected. The PMSG treatment elevated (P less than .05) ovulation rate, whereas the flushing or folic acid treatments had no effect on this trait. The addition of 5 mg/kg folic acid to the commercial-type diet improved (P less than .05) the survival rate of fetuses during early gestation and tended (P = .096) to increase the number of fetuses presumably living at 30 d of gestation when this treatment was associated with high ovulation rate.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Three corticosteroid products (triamcinolone acetonide, fluocinonide, betamethasone valerate) and a control product composed of water, petrolatum, mineral oil, cetyl alcohol, steryl alcohol, sodium lauryl sulfate, cholesterol, and methylparaben each were applied topically to healthy dogs (5 dogs/product) once daily for 5 consecutive days. Plasma concentrations of immunoreactive adrenocorticotropic hormone (iACTH) and cortisol were determined before 1 microgram of ACTH/kg of body weight was given intravenously (pre-ACTH values) and cortisol was again measured 60 minutes after ACTH was given (post-ACTH values). Cortisol and iACTH concentrations were determined in each dog before, during, and after administration of the corticosteroid products. All 3 corticosteroids caused prompt and sustained pituitary-adrenocortical suppression. Compared with control applications, the application of corticosteroids resulted in significant reduction of plasma cortisol and iACTH concentrations by day 2 of treatment, and the lower concentrations continued to day 5. One week after the last application of the corticosteroids, plasma iACTH concentrations in the corticosteroid-treated dogs had returned to the range of values for the control dogs; however, pre- and post-ACTH cortisol concentrations remained suppressed in all corticosteroid-treated dogs. Two weeks after the last treatment, the pre-ACTH plasma cortisol concentrations of corticosteroid-treated dogs returned to those of the control dogs, but the post-ACTH plasma cortisol concentrations remained suppressed. By 3 weeks after the last treatment, post-ACTH plasma cortisol concentrations of dogs treated with triamcinolone acetonide had returned to the range of values for the control dogs, but remained suppressed in the other 2 groups of dogs. Ten multiparous lactating sows were used to investigate whether intramammary infusion of lipopolysaccharides (LPS; Escherichia coli 0111:B4; 2.0 microg/kg of body weight) would affect the circulating concentrations of Ca, P, 25-hydroxyvitamin D (25-OHD), tumour necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6) and cortisol. The sows were randomly allotted to either control group (control) or LPS-treated group with five individuals per group and were infused with either physiological saline solution or LPS solution. The rectal temperature and udder quarter appearance were recorded at 0 (just before infusion), 1, 3, 7, 12 or 24 h after infusion. Blood samples were taken at 0, 1, 3, 7, 12 or 24 h after infusion. Before infusion, the rectal temperatures of all sows were below 39.2 degrees C. At 3 and 7 h after infusion, the sows in the LPS group had a rectal temperature over 39.4 degrees C. At 24 h after infusion, the rectal temperatures returned to pre-infusion levels. Serum Ca and P concentrations in the LPS group decreased (P < 0.05) after LPS infusion compared with the control group at 1 h after infusion. No significant differences (P > 0.05) in the concentrations of 25-OHD were observed between groups control and LPS at any sampling time. Increased (P < 0.01) concentrations of serum TNF-alpha, IL-6 and cortisol were observed in the LPS group compared with the control group at 3 and 7 h after infusion respectively. In conclusion, the elevation of serum concentrations of TNF-alpha, IL-6 and cortisol and the alterations of circulating concentrations of Ca and P following LPS infusion indicate that the immune system has been activated and immune activation may affect macromineral homeostatic regulation, which might have important implications for metabolic health of lactating sows. Lowered serum Ca and P following immune activation also shows a causative mechanism whereby immune activation increases the risk of secondary disorders such as mastitis-metritis-agalactia syndrome. However, immune activation did not affect circulating concentrations of vitamin D metabolites.  相似文献

OBJECTIVE: To determine blood glucose clearance in 2 species of New World camelids after IV challenge and to examine mechanisms of this clearance. ANIMALS: 5 adult female llamas and 5 adult gelded alpacas. PROCEDURE: After food was withheld for 12 hours, camelids received 0.5 g of glucose/kg of body weight by rapid IV infusion. Serum concentrations of glucose, nonesterified fatty acids, cortisol, and insulin, and plasma concentrations of lactate were determined before and 0, 1, 2, 3, 4, 5, 15, 30, 60, 90, 120, 180, and 240 minutes after infusion. Ratios of insulin to glucose and insulin to cortisol were calculated for each time point. RESULTS: Postinfusion glucose concentrations were significantly higher in llamas than alpacas for the first 15 minutes and remained significantly higher than baseline values in both species for 180 minutes. Lactate and cortisol concentrations did not change significantly; nonesterified fatty acid concentrations decreased in both species 30 minutes after infusion. Baseline insulin concentrations were < 6 microU/ml in both species and increased only to 10.1 +/- 0.7 microU/ml in llamas. Insulin concentrations did not change significantly in alpacas. CONCLUSIONS AND CLINICAL RELEVANCE: Llamas and alpacas clear glucose more slowly than other domestic species after challenge, mainly because of a weak insulin response and slow cellular uptake. This response may impair the assimilation of exogenous glucose as well as make llamas and alpacas prone to diabetes-like disorders when an abundance of endogenous or exogenous glucogenic agents are present.  相似文献

Reasons for performing study: The foal requires an active hypothalamo‐pituitary‐adrenal (HPA) axis for organ maturation and post natal survival. Prenatal administration of synthetic glucocorticoids may provide an effective method for inducing fetal maturation safely in the mare. Objectives: To determine whether dexamethasone administered to late pregnant mares: 1) will induce fetal maturation and precocious delivery; 2) is safe to use and 3) to identify endocrine responses in the mare and foal. Methods: Pregnant Thoroughbred mares received either 100 mg dexamethasone i.m. (treated n = 5) or 50 ml saline i.m. (control n = 5) at 315, 316 and 317 days of gestation. Plasma progestagens, cortisol and prostaglandin F metabolite (PGFM) concentrations were measured before and after treatment. The foals were weighed, the crown‐rump length (CRL) measured and an adrenal stimulation test performed on Day 1. Results: Dexamethasone significantly (P<0.01) reduced gestation length in treated mares without apparent adverse effects. Plasma progestagens increased (P<0.05), and cortisol and PGFM (P<0.05) decreased, following dexamethasone treatment compared with control mares. Foals were clinically mature but those from dexamethasone treated mares had reduced (P<0.05) CRL, but not bodyweights, compared with controls. Their cortisol concentrations increased following exogenous adrenocorticotrophic hormone stimulation but 2 foals from dexamethasone treated mares showed evidence of adrenal suppression. Conclusions: Dexamethasone stimulates precocious fetal maturation and delivery in healthy late pregnant mares. However, fetal HPA activity may be suppressed. Potential relevance: Dexamethasone treatment could be used to improve foal viability in mares at risk of preterm delivery. The endocrine effects of such a therapy must be evaluated before clinical intervention with glucocorticoids can be recommended.  相似文献
