采用低温诱发肉鸡腹水综合征(AS),并观察早期限饲对常温和低温饲养时肉鸡AS的发病率、肉鸡体内脂质过氧化作用和抗氧化酶活性的影响。试验1,限饲处理组肉鸡分别于7~14日龄或7~21日龄每天给料8h,对照组肉鸡全程自由采食。试验2,限饲组肉鸡分别于7~14日龄给予对照组肉鸡前一天饲料消耗量的60%或80%,对照组肉鸡自由采食。试验过程中连续测定肉鸡血浆超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—Px)、丙二醛(MDA)水平和腹水心脏指数(RV/TV值)。结果显示低温显著升高了肉鸡血浆MDA浓度,降低了血浆SOD和GSH—Px的活性;低温饲养肉鸡5~6周龄时的RV/TV值显著升高,AS的发病率显著增加。限饲处理显著降低了肉鸡血浆MDA含量,并且限饲处理肉鸡生长后期血浆中SOD和GSH—Px活性显著高于自由采食组肉鸡。所有的限饲处理均能降低肉鸡AS的发病率,并且早期限饲处理肉鸡42日龄的RV/TV值显著低于非限饲肉鸡。这表明早期限饲能够减轻肉鸡体内脂质过氧化作用,提高机体抗氧化酶的活性,从而降低AS的发病率。  相似文献

近几年来,在公猪精液冷冻稀释液中添加抗氧化剂以提高冷冻精液质量的研究受到广泛的关注。添加谷胱甘肽、超氧化物歧化酶和过氧化物酶、维生素E、L-半胱氨酸和L-谷氨酰胺、中草药成分等抗氧化剂可以有效地防止氧化损伤,提高解冻后精子活率、精子成活力、质膜与顶体完整性等,进而提高冷冻保存精液的受精能力。作者综述了抗氧化剂的抗精子冷冻损伤的作用机制及不同抗氧化剂的应用效果,并对其在公猪精液冷冻保存中的应用前景进行了展望。  相似文献

本试验旨在研究辛硫磷(Phoxim)对大鼠的毒性作用,探讨辛硫磷中毒的氧化应激机制。将36只SD大鼠分成对照组和2个染毒组,染毒组大鼠分别以30(低剂量组)和300μmol/kg体重剂量(高剂量组)灌服辛硫磷,连续灌服153、0 d后,分别测定血浆和肝脏胆碱酯酶(ChE)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量,并观察肝脏组织学变化。结果表明:辛硫磷染毒后大鼠血浆和肝脏ChE活性均极显著降低(P<0.01),尤其是高剂量染毒组,大鼠血浆ChE最大降低至对照组的22%,肝匀浆中ChE活性降低至对照组的75%。大鼠血浆和肝脏SOD、GSH-Px活性变化随染毒时间延长呈下降趋势,血浆和肝脏MDA含量均呈上升趋势。组织学检查显示辛硫磷可造成肝细胞脂肪变性。本研究表明,大鼠辛硫磷持续染毒可以诱导机体脂质过氧化增强,并导致肝脏结构损伤,说明氧化应激在辛硫磷的肝脏毒性中发挥着重要作用。  相似文献

The aim of the present study was to investigate the long-term effect of using bee venom (BV) on the reproductive performance, immune, and health status of rabbit does and its effect on their litters. Sixty mature does, from Spanish V-line rabbit stock, were randomly assigned to four homogeneous groups with 15 does each. The 1st, 2nd and 3rd groups were injected twice weekly under the neck skin with 0.1 ml solution contains 0.1, 0.2 and 0.3 mg BV/rabbit respectively. The 4th group served as a control group. From the results, litter size at birth, litter weight and survival rate at weaning age as well as milk yield were significantly (≤ 0.05) increased in BV groups than in the control group. Serum estradiol 17-β (E2) was significantly (≤ 0.05) higher (15%) in the rabbit does treated with BV compared to the control group. The treated does with BV at any study doses showed a gradual and significant (≤ 0.05) decrease (12%) in serum progesterone levels (P4) compared to the control. They also showed a significant (p≤0.05) increase in conception (17%) and fertility rates (10%) compared to the control does. Treatment of rabbit does with BV caused a gradual and significant (≤ 0.05) reduction in both aspartate aminotransferase (AST) (16%) and alanine aminotransferase (ALT) (37%) liver enzyme activities. Additionally, results have shown that BV resulted in a gradual and significant (≤ 0.05) increase in total antioxidant capacity (TAC), antioxidative enzymes such as glutathione S-transferase (GST) and glutathione peroxidase (GPx), serum IgG, IgM and IgA levels with significant (≤ 0.05) decrease in malondialdehyde (MDA) and thiobarbituric acid reactive substances (TBARS) in BV groups compared to the control group. Results suggest that BV can be used in rabbit farming as an effective and safe alternative to artificial chemical drugs (sexual-stimulants) to improve certain reproductive traits, immune response and health.  相似文献

通过对试验用荷斯坦奶牛全血中白细胞总数(WBC)、中性粒细胞(PMN)百分比变化及血清、子宫分泌物中的一氧化氮(NO)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)的活性进行检测,探讨产后患有不同程度子宫内膜炎奶牛的氧化损伤和抗氧化能力的变化及其机理。结果表明,CE组和SE组全血中WBC总数和PMN百分比显著高于对照组(P0.01,P0.05)。而CE组血清和子宫分泌物中NO、MDA含量明显高于N组(P0.01),SE组则变化不明显,CE组SOD和GSH-Px活性变化比SE组明显,表现差异显著(P0.01,P0.05)。试验证明,子宫内膜炎奶牛血清和子宫分泌物中脂质过氧化物含量升高、抗氧化能力下降,可能是子宫内组织细胞病理损伤,引起不同程度子宫内膜炎的重要原因之一。  相似文献

通过野外取样,对黄土丘陵沟壑区坡沟系统5种立地环境(阳坡沟谷地和沟间地、峁顶、阴坡沟间地和沟谷地)下19种主要物种叶片的膜脂过氧化指标丙二醛(MDA)含量,3种抗氧化酶即超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性,及2种非酶抗氧化物质即还原型谷胱甘肽(GSH)和类胡萝卜素(Car)含量进行了测定与分析,旨在探讨不同物种适应不同立地环境的抗氧化特性。结果表明:各立地环境下物种间的MDA积累量均呈极显著差异(P<0.01),说明各立地环境的胁迫均导致供试物种遭受不同程度的膜脂过氧化作用,其中互生叶醉鱼草的膜脂过氧化水平最高,猪毛蒿的最低;在不同立地环境间,白羊草、达乌里胡枝子、阿尔泰狗娃花和狼牙刺的各抗氧化酶活性和非酶抗氧化物质含量均无显著差异(P>0.05),中华隐子草、糙隐子草、长芒草、草木樨状黄芪、糙叶黄芪、甘草、菊叶委陵菜、猪毛蒿、铁杆蒿、茭蒿、沙棘、杠柳和互生叶醉鱼草存在显著(P<0.05)或极显著(P<0.01)差异,说明在不同立地环境下,前者均采用多种抗氧化酶和非酶抗氧化物质协同抵御胁迫的策略,后者则调用不同抗氧化酶或非酶抗氧化物质发挥主要抵御作用;由于植物科属和碳同化途径的差异,其抗氧化特性也存在差异:菊科和豆科植物分别主要通过CAT和SOD减轻活性氧的伤害,禾本科植物则能以较高的SOD、POD活性和Car含量维持较低的膜脂过氧化水平,C4植物比C3植物具有较高的GSH和Car含量。隶属函数法综合评价表明,中华隐子草的抗氧化能力最强,互生叶醉鱼草的最弱。  相似文献

达乌里胡枝子抗氧化防御系统对增强UV-B辐射的动态响应   总被引:1,自引:0,他引:1  
为探究增强UV-B辐射对达乌里胡枝子(Lespedeza davrica)抗氧化能力的影响,采用盆栽试验研究其在不同UV-B辐射强度(CK=0,T1=1.944 kJ·m-2·d-1,T2=3.002 kJ·m-2·d-1)下的抗氧化保护系统相关指标的动态响应,UV-B辐射处理持续15 d,每3 d取样一次至结束后第3 d再取样一次。结果表明:随着处理时间的延长,增强UV-B辐射使超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性降低,抗坏血酸过氧化物酶(APX)和过氧化物酶(POD)活性升高,且SOD,POD和CAT的活性随着辐射强度的增加而降低,APX活性在开始处理9 d内随着辐射强度的增加而增加,此后随着辐射强度的增加而降低;增强UV-B辐射使抗坏血酸(AsA)含量增加,类胡萝卜素(Car)含量降低,且二者的含量均随着辐射强度的增加而降低;增强UV-B辐射使超氧阴离子(O2·-)、丙二醛(MDA)含量升高,O2·-含量和MDA含量均随着辐射强度的增加而增加。UV-B处理结束后,除SOD活性、POD活性、AsA含量呈下降趋势外,其他指标都呈上升趋势。通过隶属函数法综合评价得出:增强UV-B辐射降低了达乌里胡枝子的抗氧化能力,随着UV-B辐射强度的增加,抗氧化能力降低。  相似文献

在培养液中加入不同浓度的Cd(10、20、40μmol/L)、Pb(10、20、40μmol/L)和Pb-Cd联合(分别为5、10、20μmol/L),来探讨铅镉对肾细胞的作用及可能机制。在显微镜下观察细胞形态、测定细胞凋亡率和细胞周期及上清液脂质过氧化指标。结果表明:20μmol/L以上染毒组细胞皱缩、体积变小,表面有细胞碎片,甚至呈泡沫状或树枝状;铅、镉单独和联合染毒组GSH—Px、SOD活性随染毒剂量的增加,呈逐渐下降趋势(P〈0.05).而MDA含量和凋亡率则呈升高趋势(P〈0.05),并存在剂量和时间效应。镉和铅也能使细胞周期停滞。Pb、Cd联合可加剧细胞损伤。  相似文献

研究比较了天门冬氨酸镁和氧化镁对肉仔鸡镁营养状态及肝脏和脾脏等组织抗氧化酶能力的影响。将252只1日龄的AA肉仔鸡随机分为7个处理组,每个处理6个重复,每个重复6只鸡。天门冬氨酸镁和氧化镁的添加水平为0.9、1.8、2.7g/kg日粮,基础日粮组作为不添加镁的对照组。日粮中添加镁能显著提高血清、肝脏和脾脏中的镁浓度(P<0.01),天门冬氨酸镁比氧化镁更能有效地提高血清和肝脏中的镁含量(P<0.01)。肝脏过氧化氢酶(CAT)的活性,随天门冬氨酸镁添加量的增加线性增加(P<0.01),随氧化镁添加量的增加呈二次曲线增加(P<0.05)。天门冬氨酸镁比氧化镁更有效地增强肝脏中CAT的活性(P<0.01)。脾脏CAT的活性随天门冬氨酸镁(P<0.05)或氧化镁(P<0.01)添加量的增加线性增加。肝脏(P<0.05)和脾脏(P<0.01)丙二醛MDA的浓度随氧化镁或天门冬氨酸镁添加量的增加线性降低。天门冬氨酸镁比氧化镁更有效地降低肝脏中的MDA浓度(P<0.01)。结果表明:日粮中添加镁能显著改善肉仔鸡各种组织中镁的含量,并能提高肝脏和脾脏等组织中抗氧化酶的活性,降低脂质过氧化水平。  相似文献

The objective of the study was to evaluate the protective effect of Calendula officinalis propylene glycol extracts against oxidative DNA damage and lipid peroxidation induced by high polyunsaturated fatty acid (PUFA) intake in young growing pigs. Forty young growing pigs were assigned to five treatment groups: control; oil (linseed oil supplementation); C. officinalis 1 and 2 groups (linseed oil plus 3 ml/day of C. officinalis propylene glycol extracts); and vitamin E group (linseed oil plus 100 mg/kg of vitamin E). Lymphocyte DNA fragmentation and 24-h urinary 8-hydroxy-2'-deoxyguanosine (8-OHdG) excretion were measured to determine DNA damage. Lipid peroxidation was studied by analysing plasma and urine malondialdehyde (MDA), and urine isoprostane concentrations (iPF2α-VI), total antioxidant status of plasma and glutathione peroxidase (GPx) assays. C. officinalis 1 (extract from petals) effectively protected DNA from oxidative damage. It indicated a numerical trend towards the reduction of plasma MDA and urinary iPF2α-VI excretion. Its effect was comparable with that of vitamin E. C. officinalis 2 (extract from flower tops) showed less antioxidant potential than the extract from petals. We can conclude that the amount of C. officinalis extracts proposed for internal use by traditional medicine protects the organism against DNA damage induced by high PUFA intake.  相似文献

A three‐variable central composite design coupled with surface‐response analysis was used to examine the effects of dietary α‐tocopherol + ascorbic acid (TOCAA), selenium (Se), and iron (Fe) on indices of oxidative stress in juvenile spring Chinook salmon. Each dietary factor was tested at five levels for a total of fifteen dietary combinations (diets). Oxidative damage in liver and kidney (lipid peroxidation, protein carbonyls) and erythrocytes (erythrocyte resistance to peroxidative lysis, ERPL) was determined after feeding experimental diets for 16 (early December) and 28 (early March) weeks. Only TOCAA influenced oxidative stress in this study, with most measures of oxidative damage decreasing (liver lipid peroxidation in December and March; ERPL in December; liver protein carbonyl in March) with increasing levels of TOCAA. We also observed a TOCAA‐stimulated increase in susceptibility of erythrocytes to peroxidative lysis in March at the highest levels of TOCAA. The data suggest that under most circumstances a progressive decrease in oxidative stress occurs as dietary TOCAA increases, but higher TOCAA concentrations can stimulate oxidative damage in some situations. Higher levels of TOCAA in the diet were required in March than in December to achieve comparable levels of protection against oxidative damage, which may have been due to physiological changes associated with the parr‐smolt transformation. Erythrocytes appeared to be more sensitive to variation in dietary levels of TOCAA than liver and kidney tissues. Using the March ERPL assay results as a baseline, a TOCAA level of approximately 350–600 mg/kg diet would provide adequate protection against lipid peroxidation under most circumstances in juvenile Chinook salmon.  相似文献

A randomized blocked factorial experiment was conducted with 90 young pigs. From 4 to 11 weeks of age the pigs were kept in individual pens and fed a selenium supplemented basal diet consisting mostly of propionic acid treated barley, soybean meal and dried skim milk, and containing < 0.5 mg vitamin E per kg. The treatment factors during this period were 3 dietary levels of added vitamin E (nil, 10 and 30 mg/kg) and a 6 % supplement of fresh or oxidized fat (2/3 lard and 1/3 herring oil). From 11 weeks of age until slaughter at 90 kg the pigs received the vitamin E supplements but no fat or dried skim milk. The basal diet for this later experimental period was based on untreated dry barley. Blood samples collected during the period of investigation were examined for vitamin E and for resistance against erythrocyte lipid peroxidation (ELP) in order to evaluate the antioxidant status.Analysis of variance and Student’s t-test on least squares means showed the ELP to be influenced independently by the vitamin E supplement and, during the fat feeding period, by the quality of the fat supplement, with the highest peroxidation resistance (low ELP) in the groups fed fresh fat and a high level of vitamin E. Blood vitamin E level was only influenced — positively — by the vitamin E supplement although variations in the feed vitamin E level below 10–15 mg vitamin E per kg did not result in corresponding variations in measurable blood vitamin E concentrations. In the same low range of vitamin E in the feed there was a statistically significant difference in ELP values between the different vitamin E treatment groups. No clinical manifestations of selenium-vitamin E deficiency were observed in the pigs.The ELP and the plasma vitamin E levels observed would seem to suggest that a total of 15 mg vitamin E per kg barley-based feed will not always be sufficient for growing pigs.  相似文献

The effect of hyperthyroidism on some oxidative stress parameters is reported. The hyperthyroid state was induced by intraperitoneal injection of triiodothyronine (T3)(10 μg/kg body weight) for 14 days in two groups of female rabbits (3 and 12 months old). The T3 injection caused increase by 1.5-fold to 1.7-fold in T3 serum level, and 2-fold to 3-fold decrease (age-dependent) in body weight gain at the end of experimental period. The induced hyperthyroidism caused a significant increase in the serum concentration of the lipid peroxidation end-product malondialdehyde and lowered erythrocyte resistance to oxidative stress when subjected to the free radical generator 2,2'-azobis(2-amidinopropane) hydrochloride in vitro. The half maximum haemolysis time (HT50) decreased in the both experimental groups of rabbits, by about 12 min in the 3-month-old animals and 27 min in the 12-month-old animals. The study showed for the first time that hyperthyroidism enhances the ability of plasma to protect against iron-binding and iron oxidizing organic radicals. The scavenging property and antioxidant capacity of plasma against iron-binding inorganic radicals also increased. Measurement of erythrocyte resistance to oxidative stress and the protective ability of plasma against oxygen radicals discriminates the thyroid hormone modulatory effects in defence mechanisms against lipid peroxidation.  相似文献

通过2K1C法制作肾性高血压(RH)大鼠模型,研究向天果提取物(SME)对肾性高血压大鼠的降压和靶器官保护等治疗作用。将造模成功的大鼠随机分为模型组、阳性组(25mg/kg卡托普利)、SME高剂量组(500mg/kg)、SME低剂量组(250mg/kg)。给药8周后,血液生化测定肾功能指标,并采用ELISA测定肾素-血管紧张素-醛固酮系统(RAAS)活性。取右肾进行组织切片,HE染色,组织学观察。结果表明,与模型组相比,SME能显著降低RH大鼠收缩压(P<0.01)。与模型组相比,SME高、低剂量组明显降低RH大鼠RAAS水平(P<0.01)。各治疗组对RH大鼠肾脏损伤有不同程度的改善。说明向天果提取物有降压、保护肾脏的作用,其机制可能与抑制RAAS兴奋,抑制血管收缩,以及水钠重吸收有关。  相似文献

为了观察玉米肽对微波辐射引起动物肝组织抗氧化系统损伤和肝细胞形态变化的保护作用。将36只Wistar大鼠随机分为正常对照组、辐射模型组和辐射给药组,辐射模型组和辐射给药组大鼠给予100 mW/cm2的微波辐照6 min,辐射给药组辐射后给予600 mg/kg·d玉米肽,连续7 d。7 d后处死大鼠,光学显微镜观察肝细胞形态学变化,检测肝组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性。结果与对照组比较,辐射模型组肝细胞体积增大,细胞核形态不规则,细胞水肿严重,细胞之间界限不明显,胞质淡染。肝组织TSOD[(242.98±27.79)U/mg]和GSH-Px活性[(718.82±169.14)U/mg]均极明显降低(P<0.01),MDA含量[(18.62±1.44)nmol/mg]明显升高(P<0.05);与辐射模型组比较,辐射给药组肝组织结构有明显恢复,肝细胞水肿明显减轻,细胞边界较清晰,肝组织TSOD活性[(373.14±15.74)nmol/mg]明显升高(P<0.01),GSH-Px活性[(943.08±274.54)U/mg]明显升高(P<0.05),MDA含量[(17.32±1.01)nmol/mg]明显降低(P<0.05)。表明玉米肽对微波辐射引起的肝细胞形态变化和肝组织抗氧化系统损伤有较好的治疗效果。  相似文献

研究高氟低碘对老年大鼠肺脏细胞DNA损伤的影响。健康离乳Wistar大鼠21只,随机分为4组:对照组(5只),饲喂大鼠标准日粮,饮自来水;高氟组(5只),饲喂大鼠对照日粮,饮100 mg/L氟化钠(NaF)去离子水;低碘组(5只),饲喂低碘日粮(定做),饮去离子水;高氟低碘组(6只),饲喂低碘日粮,饮100 mg/L氟化钠(NaF)去离子水。在大鼠20月龄时,处死采取肺脏组织,检查其DNA的损伤情况。高氟组、低碘组和高氟低碘组的老年大鼠肺细胞拖尾率比对照组显著增高(P<0.01),高氟低碘组大鼠肺细胞拖尾率比高氟组、低碘组高,并且均有差异显著性(分别为P<0.01和P<0.05);高氟组、低碘组和高氟低碘组老年大鼠肺细胞的拖尾长度比对照组显著增长,显著性差异分别为P<0.01、P<0.05 和P<0.01。高氟低碘组大鼠肺细胞的拖尾长度比高氟组及低碘组增长,与低碘组有差异显著性(P<0.01),但与高氟组差异不显著(P>0.05);高氟组和高氟低碘组老年大鼠肺细胞DNA损伤主要是Ⅲ级损伤(Ⅲ级分别为70.00%和79.31%)。低碘组的肺细胞DNA损伤进入Ⅱ级和Ⅲ级范围的比较多,并且Ⅲ级占有率较高(Ⅱ级为31.25%;Ⅲ级为40.62%)。结果表明,高氟、低碘、高氟低碘都影响了大鼠肺脏细胞的DNA损伤。  相似文献

试验选取具有典型内脏型痛风特征的雏鹅5只和相同日龄及生长环境下的健康雏鹅6只,利用RNA-seq技术分析其肾组织转录表达差异,旨在了解雏鹅痛风发生的分子机制。结果表明:痛风雏鹅肾组织转录表达显著区别于健康雏鹅。试验共筛选出差异表达基因1 749个,上调基因501个,下调基因1 248个。GO(Gene Ontology)和KEGG(Kyoto Encyclopedia of Genes and Genomes)分析发现,这些差异基因主要具有趋化因子活性、T细胞受体结合、抗原结合等分子功能,参与免疫反应、趋化因子介导信号通路、炎症反应等生物过程,并主要富集于细胞因子受体相互作用、肠道IgA免疫网络、细胞黏附等信号通路。  相似文献

以北方蚕区现行主要桑树品种选792、沙2、新一之舽、陕305、湖桑32号、农桑14号的1年生绿枝扦插苗为实验材料,研究了水分胁迫对不同桑树品种的膜伤害和保护酶活性的影响。在水分胁迫下,细胞膜受到伤害,细胞膜透性增加,电导率随之增强,作为膜脂过氧化产物的丙二醛(MDA)含量增加,各品种细胞膜受到的伤害度有差异。保护酶SOD、POD、CAT活性都表现出先升后降的变化趋势,但变化规律又有所不同。推测各品种抗旱性存在较大差异,6个供试桑品种抗旱能力从大到小依次为选792>陕305>农桑14号>湖桑32号>新一之舽>沙2。  相似文献

Coriander and white mustard, an annual plants originated in the Mediterranean region, have been cultivated and used as spices for a long time. Recent studies have shown that they may constitute a potential source of phenolic compounds. The aim of this study was to evaluate the content of polyphenols in coriander and white mustard water extracts and to investigate their antioxidant activity in C2C12 mouse skeletal muscle cells, which serve as a good model of cells with intensive metabolism. HPLC analysis showed that polyphenols were able to permeate from the water extracts of studied plants into the undifferentiated myoblasts as well as myocytes undergoing differentiation, increasing the concentration of reduced glutathione and upregulating glutathione reductase and peroxidase activity. White mustard and coriander extracts also decreased the levels of oxysterols and sum of tiobarbituric acid reactive substances (TBARS) in both: myoblasts and differentiating myocytes, demonstrating protective effect on cell membranes. The obtained results indicate that polyphenols synthesized by both herbs may have beneficial effects on muscle tissue.  相似文献
