Adhesion of Staphylococcus aureus and Escherichia coli to bovine udder epithelial cells 总被引:1,自引:0,他引:1
An assay for the adhesion of tritiated thymidine-labelled Staphylococcus aureus and Escherichia coli to bovine mammary ductular epithelial cell lines was developed. The relative adhesion of 15 strains of S. aureus to these cell lines was examined. Four strains did not adhere and the remaining 11 adhered at variable levels. Adhesion to different cell lines was generally similar. Adhesion to freshly collected bovine mammary epithelial cells was significantly greater than that to cells maintained in tissue culture. The system described was demonstrated to be a suitable model for studying adhesion of mastitis-causing organisms to bovine mammary epithelial cells. 相似文献

The sequences of the ccrAB genes from bovine-, canine- and chicken-originating methicillin-resistant Staphylococcus (S.) epidermidis (MRSE) and bovine methicillin-resistant Staphylococcus (S.) aureus (MRSA) were compared to investigate the frequency of intra-species horizontal transfer of the staphylococcal cassette chromosome mec (SCCmec) complex. Nineteen MRSE strains were isolated from bovine milk, chickens, and dogs, and their genetic characteristics were investigated by multilocus sequence typing and SCCmec typing. Among the animal MRSE strains, the most frequent SCCmec type was type IV, which consisted of the type B mec complex and ccrAB type 2. The ccrA2 and ccrB2 genes were sequenced from the bovine, chicken and canine MRSE strains and compared with those of the bovine MRSA strains. The sequences generally clustered as MRSA and MRSE groups, regardless of the animal source. Additionally, no bovine MRSE sequence was associated with the bovine MRSA groups. Although most of the bovine MRSE and MRSA isolates possessed SCCmec type IV sequences, our results suggest that the intra-species gene transfer of the SCCmec complex between bovine S. aureus and bovine S. epidermidis strains is not a frequent event. 相似文献

Data from a mastitis control programme involving 158 herds in northern Norway are presented. The association between housing factors and indoor climate variables, and the prevalence of subclinical mastitis and herd cell count was calculated from a least squares model by a backward elimination procedure. Ten to 23% of the herd udder health variation at the start of the programme was associated with environmental variables. The following variables were found to be of significance: herd size, hoof trimming, lactation age, stall floor surface, type of stall, stall width, stall length, presence or absence of a grating over dung channel, single-/double-glazed windows and ambient humidity. 相似文献

为了探讨金黄色葡萄球菌对氟喹诺酮耐药性与gyrA、gyrB突变的关系,本研究对氟喹诺酮敏感金黄色葡萄球菌进行耐药诱导,获得一系列不同氟喹诺酮耐药水平的人工诱导耐药菌,应用PCR扩增野生菌和人工诱导菌的gyrA、gyrB基因并测序,经核苷酸和氨基酸序列分析表明:氟喹诺酮耐药菌的gyrB基因无突变,氟喹诺酮耐药菌的gyrA有Ser85→Leu和Ser84→Pro的突变。实验结果表明:gyrA突变是金黄色葡萄球菌氟喹诺酮耐药水平增高的主要原因。 相似文献

The aims of the present study were to relate intramammary infection (IMI) occurrence and somatic cell count (SCC) with teat-end condition (TEC) and udder cleanliness (UC). Milk samples from 1931 teats were evaluated according to the presence of IMI and SCC. Scores were applied to teats according to the TEC and to UC. Teats ends with a very rough ring had the largest number of IMI when compared to the other three categories, as well as animals with dirtier udders. The change in a TEC score increased by around 30% the chance of IMI. Also, the chance of the animal developing IMI increased by approximately 47% when the UC score increased. No significant association between both scores and quarter SCC was found. It can be concluded that animals with very rough teat end rings and very dirty udders have a greater predisposition to IMI. 相似文献

The udders and teats of cows were examined by brightness mode (B-mode) ultrasonography. A 5-MHz linear array transducer and a 5-MHz or 10-MHz mechanical sector transducer were used in the examinations. In lactating animals, the teat sinus, gland sinus, and lactiferous ducts were imaged easily. The scans also allowed visualization of the layers of the teat wall. The annular fold and the folding of the mucosa at the junction of the teat sinus and the papillary duct were seen best with the 5-MHz and 10-MHz mechanical sector transducers. In one cow lacking milk flow from one quarter, a mass at the junction of the papillary duct and the teat sinus was observed ultrasonographically. Surgical removal of the mass resulted in a return of milk flow from that quarter. 相似文献

Bovine udders, together with the attached section of the ventral abdominal wall, as well as the bony pelvic floor were studied. The parts of the suspensory apparatus and attachment to the body were investigated. The location of the medial and lateral laminae served as the basis for classifying them into abdominal and pelvic parts. The udder base is attached to the ventral abdominal wall by means of the dorsal connective tissue plate. The abdominal part of the medial lamina and the pelvic part of the lateral lamina are the functionally more important bands. Sixty per cent of the udder base is attached to the ventral abdominal wall and 40% to the pelvic floor by the symphysial tendon. The symphysial tendon is attached to the ventral pubic tubercle and the symphysial eminence as well as to the symphysial crest, which develops by the fourth year. In order to study the osseous changes, bovine pelvises from the Middle Ages, the beginning of the 20th century, and the present time were compared. Increased udder mass has brought about changes in the structures of the suspensory apparatus of the udder and in the structure of the pelvis. 相似文献

Staphylococci are a major cause of intramammary infections (IMI) in ruminants. The main aim of this study was to investigate staphylococcal IMI in dairy cattle with emphasis on persistence and distribution of staphylococcal species and genotypes. With a sampling interval of 4-8 weeks, over a year, 4030 samples from 206 cows in 4 herds were collected. Coagulase-negative staphylococci (CNS) and Staphylococcus aureus were detected in 13.2% and 4.2% of the samples, respectively. Selected CNS isolates from quarter milk samples were identified to species level using sodA sequencing. Staphylococcus chromogenes (32%) and Staphylococcus simulans (25%) predominated. The proportion of S. chromogenes was greater in primiparous (52%) than in multiparous cows (12%), while the opposite was the case for Staphylococcus epidermidis (6% and 21%, respectively). Isolates from possibly persistent IMI were selected for pulsed-field gel electrophoresis (PFGE). Six staphylococcal species were found to cause persistent IMI; S. aureus, S. chromogenes, S. simulans, S. epidermidis, Staphylococcus haemolyticus and Staphylococcus warneri. It was shown that several pulsotypes (PTs) within each species were associated with persistent infections, but only a few were spread and caused persistent IMI in multiple cows within a herd. Of special interest was the observation that only one, or a few, strains of each species caused persistent IMI in multiple cows within a same herd. This paper presents data from 158 dairy herds in northern Norway. The influence of the milking machine and milking management on the prevalence of subclinical mastitis and herd cell counts was calculated from a least squares model by a backward elimination procedure. It was shown that 16–45% of the herd udder health variation found at the start of the programme could be associated with milking machine and milking management variables. The following variables were found to be of significance: number of clusters in the herd, vaccum, function and vaccum regulator, undulation of pipeline, limping of pulsator, rate of pulsator, type of claw-piece, hygienic/-technical condition of liner, preparation time, air admission during application of teat cups, overmilking and machine-on time. 相似文献

Pregnant non-lactating cows were immunised by intramammary infusion with killed Streptococcus uberis into one quarter and ovalbumin into another, at one week (group 2) or one week and two weeks (group 1) before the expected date of parturition. A small IgG1 and IgG2 antibody response to ovalbumin was detected in the serum of these cows. There was also a small increase in IgG1 and IgA serum antibody activity to S uberis. In whey the response was restricted to IgA with activity to S uberis. The IgA antibody response to S uberis in group 1 was significantly greater in the quarter immunised with bacteria than that of the control quarters for up to two months after calving. In contrast, the serum IgA response was short or absent in a number of animals. 相似文献

Twenty bovine udder quarters colonized with Corynebacterium bovis SR6 and 20 uncolonized quarters were challenged by inoculation of Staphylococcus aureus Newbould 305 (ATCC 29740) into the teat cistern. The percentage of infection in quarters colonized with C. bovis (50%) was significantly lower than that in controls (100%). By similar challenge no significant difference was observed between the percentage of infection with Streptococcus agalactiae ATCC 27956 in 33 quarters colonized with C. bovis (70%) compared to 33 controls (87.9%). A total of 37 quarters colonized with C. bovis and 37 control quarters were challenged with Staph. aureus Newbould 305 (ATCC 29740) and Maxi (ATCC 27543) and Strep. agalactiae (ATCC 27956) by exposure of the teat orifice. The percentage of teat ducts colonized with C. bovis which became infected with either pathogen was not different from that for controls. 相似文献

The susceptibility of lactating bovine udder quarters fitted with a polyethylene intramammary device to infection was investigated. Following experimental challenge with Streptococcus agalactiae or Staphylococcus aureus, the incidence of infection was significantly (p less than 0.05) lower in intramammary device-fitted quarters compared to control quarters. In general, total foremilk and strippings milk somatic cell counts for intramammary device-fitted and control quarters were not significantly (p less than 0.05) different. Differential foremilk and strippings milk somatic cell counts were significantly (p less than 0.05) higher in samples from intramammary device-fitted quarters compared to control quarters. 相似文献

Growth of the secretory epithelium during prepartum time, and for a short period after calving, is under hormonal control by estrogen, progesterone and prolactin. The mechanism(s) by which these hormones act is not known but colostrum and milk have been shown to contain different growth promoting substances. In an attempt to unravel these relationships the effect of bovine colostrum on cellular proliferation in vitro have been characterized. Colostral thermostable factors not present in milk nor associated with fat, potently induce the proliferation associated enzyme, ornithine decarboxylase, in fibroblast cell lines. However, mammary epithelial cells appear to proliferate in response to different colostral heat sensitive factor(s) that await further characterization. 相似文献

Samples of bulk tank milk and cow-composite milk from 23,138 dairy cows from 50 California dairies were examined by use of microbiologic procedures. The number of colonies of mastitis pathogens isolated per milliliter of bulk tank milk (used as a predictor of the percentage of infected cows in the herd) was evaluated, using simple regression analysis and Spearman's rank correlation. Correlations between the pathogens and the percentage of cows in each herd shedding the pathogens were found for Streptococcus agalactiae (r = 0.71) and mycoplasma (r = 0.59), but were considerably lower for other pathogens. When greater than or equal to 4,000 colonies of Streptococcus agalactiae were found per milliliter of bulk tank milk, at least 7% of the cows in the herd was shedding this organism. However, a pattern was not found between the number of mycoplasma colonies per milliliter of bulk tank milk and the percentage of infected cows in the herd. 相似文献
