Bovine rotavirus (BRV) has been detected in both dairy and beef cattle herds worldwide. Stool samples collected from calves in the Gippsland region of Victoria, Australia were screened to determine the presence of BRV. A total of 100 faecal samples were collected from calves with and without diarrhoea across three farms during 2004 and 2005. Group A BRV was detected in 26% of faecal samples (22 from diarrheic calves and four from asymptomatic calves). Genotyping analysis of rotavirus positive samples indicated that G6P[5] was the most prevalent genotype (38.5%) followed by G6P[5 + 11] (15.4%). G10P[11] and G6 + G10P[5] were each detected at a rate of 7.7%, and G6 + G10P[11] was found in a single sample (3.8%). Seven samples (26.9%) could not be G and/or P typed. Thirty percent of the BRV positive samples were mixed infections, indicating that individual calves were co-infected with more than one strain of rotavirus. The G6P[5] strains exhibited high VP7 identity (>97% amino acid identity) with B-60, a G6 strain identified in Victorian calves during 1988. A G10P[11] isolate was closely related (>97% amino acid identity in VP7 and VP4 proteins) to a Victorian G10P[11] strain (B-11) also identified during 1988. This study demonstrates that BRV is a contributing pathogen to diarrhoeal disease in Victorian calves, with sequence analysis suggesting long-term conservation of the VP7 protein over a 16-year period.  相似文献

A total of 113 diarrheic samples comprising of 68 buffalo calves and 45 cow calves were screened by RNA-PAGE for the detection of presence of rotavirus. RNA-PAGE analysis of these samples revealed 11 (9.73%) was found positive for rotavirus. Out of 68 faecal samples of buffalo calves tested for viral gastroenteritis, 8 (11.76%) were found positive for rotavirus. Similarly, out of 45 faecal samples of cattle calves tested for viral gastroenteritis, 3 (6.66%) was found positive for rotavirus. Rotavirus-positive samples represented long electropherotype. All RNA-PAGE-positive faecal samples for rotavirus subjected to RT-PCR for VP7 gene, ten samples yielded a specific product of 1,013 bp of VP7 gene. All the PCR-positive samples of the present study were subjected to genotyping with primers for G6, G8 and G10 genotype. All positive samples showed G10 genotype. This indicates that G10 may be predominant genotype among bovine calves in Mumbai region in India.  相似文献

Clostridium difficile was investigated as a possible cause of enteritis in calves. The organism and its toxins (TcdA and TcdB), respectively, were found in 25.3% and 22.9% of stool samples from diarrheic calves. Culture positive samples were more likely than culture negative samples to be toxin positive. However, toxin positive stools were more common among nondiarrheic calves, but diarrheic calves were nearly twice as likely to be culture positive. Ribotype 078 was dominant among isolates. Salmonella sp. was isolated from both diarrheic and nondiarrheic calves, but large numbers of E. coli were found more commonly in diarrheic calves than in nondiarrheic animals. Prevalence rates for coronavirus and Cryptosporidium sp. were substantially higher in nondiarrheic calves than in diarrheic, but rates of detection of rotavirus and Giardia sp. were more nearly equal between groups. Lesions in naturally infected calves included superficial mucosal erosion with associated fibrinous exudates. Neutrophils and eosinophils infiltrated lamina propria. Large Gram-positive rods morphologically compatible with C. difficile were abundant in the colonic lumen and the organism was isolated by bacteriologic culture. Toxins were found throughout the colon. Purified toxins A and B (individually and conjointly) caused comparable lesions, as well as fluid accumulation, in ligated intestinal loops. AIM: To determine the presence of Group A rotavirus G6, G8, and G10 genotypes in calves in the North Island of New Zealand. METHODS: Faecal samples from 730 calves (<6 weeks old) with diarrhoea were collected during 2006 and 2007 from seven regions in the North Island of New Zealand. The samples were screened for the presence of Group A rotavirus antigen, using a commercial ELISA. Forty-one samples from different farms were randomly selected out of the 385 ELISA-positive samples and tested using PCR for the presence of G6, G8, and G10 genotypes of rotavirus. RESULTS: The PCR analysis of 41 antigen-positive field samples revealed that 37 (90%) contained genotype G6, three (7%) genotype G10, one sample (2%) had both G6 and G10 genotypes, and none contained genotype G8. CONCLUSIONS: Rotavirus genotype G6 was the predominant genotype found in this preliminary study and was present in all seven regions studied. Genotype G10 was also found in some regions of the North Island, whereas genotype G8 was not found in any sample. CLINICAL RELEVANCE: This is the first report on rotavirus G genotypes present in calves in the North Island of New Zealand.  相似文献

Group A bovine rotavirus (BRV) is one of the main causes of neonatal calf diarrhea. The present study reports the incidence of rotavirus diarrhea and the genotypes of BRV strains circulating in beef and dairy herds from Argentina, during a 10-year period (1994-2003). Group A BRV was detected in 62.5% (250/400) of the total studied cases of diarrhea. Positive cases were analyzed by heminested multiplex RT-PCR for P and G genotypes identification. Sixty percent of them were typed as P[5]G6, 4.4% P[11]G10, 4.4% P[11]G6 and 2.4% P[5]G10. Additionally, 9.2% of the cases were initially typed as G8 combined with P[5] or P[11], but sequence analysis revealed they belonged to genotype G6, lineage Hun4-like. Partial typing was assessed in 12.0% of the cases. One of the partially typed samples was closely related to genotype G15. BRV was detected in 71% and 58% of the outbreaks registered in beef and dairy farms, respectively. A clear differential distribution of G/P types was found according to the herd type. P[5]G6 was the prevalent strain in beef herds, while P[11] was the prevalent P-type in dairy herds (71%), associated in similar proportions with G6 and G10, These findings indicate that BRV genotypes included in the current commercially available rotavirus vaccines (G6, G10, P[5] and P[11]) should protect calves from most Argentinean field strains. Nevertheless, continuous surveillance is necessary to detect the emergence of new variants.  相似文献

Faecal samples were collected from 89 dairy calves to determine the prevalence of rotavirus infection in Tunisia and the genomic diversity of bovine rotavirus strains. After screening of all faecal samples by enzyme-linked immunosorbent assay, rotavirus strains were analysed by RNA polyacrylamide gel electrophoresis and characterized antigenically by monoclonal antibodies to the VP6 subgroup. The VP7 genotype was determined by nested RT-PCR. Of the 89 calves tested, 27 (30%) were positive for rotavirus antigen. Four different long electrophoretypes were identified. All VP6 typeable strains carried the subgroup I specificity. G8 genotype was the most prevalent, but G6 and mixed strains G(6 + 8) were also detected.  相似文献

Clostridium difficile-associated-diarrhea (CDAD) is a nosocomial infection in dogs. Diagnosis of this infection is dependent on clinical signs of disease supported by laboratory detection of C. difficile toxins A or B, or both, in fecal specimens via enzyme-linked immunosorbent assay (ELISA). Unfortunately, to the authors' knowledge, commercially available ELISAs have not been validated in dogs to date. We evaluated 5 ELISAs done on 143 canine fecal specimens (100 diarrheic and 43 nondiarrheic dogs) and on 29 C. difficile isolates. The results of each ELISA were compared with the cytotoxin B tissue culture assay (CTA). Clostridium difficile was isolated from 23% of the fecal specimens. Eighteen of the 143 fecal specimens were toxin positive (15 diarrheic and 3 nondiarrheic dogs). On the basis of multiplex polymerase chain reaction (PCR) analysis for toxin-A and -B genes, 72% of the isolates were toxigenic. The carriage rate of toxigenic isolates in diarrheic dogs was higher than that in the nondiarrheic dogs; however, these differences were not statistically significant. A good correlation was found between CTA, PCR, and culture results. The ELISAs done on fecal specimens collected from diarrheic dogs had low sensitivity (7-33%). In contrast, ELISA for toxin A or B, or both, performed on toxigenic isolates had high sensitivity (93%). These results suggest that commercially available human ELISAs are inadequate for the diagnosis of canine C. difficile-associated diarrhea when tested on fecal specimens. In contrast, the Premier ToxinA/B and Techlab ToxinA/B ELISAs may be useful for the diagnosis of canine CDAD when used on toxigenic isolates.  相似文献

试验旨在掌握河北省唐山市13个县(市、区)奶牛场奶犊牛病毒性腹泻病原的流行状况,有效防控奶犊牛腹泻。采集唐山市不同地区38个奶牛场腹泻犊牛粪便样品788份,提取腹泻粪便样品中的基因组RNA。根据GenBank公布的牛病毒性腹泻病毒(Bovine viral diarrhea virus,BVDV)E2基因、牛轮状病毒(Bovine rotavirus,BRV)VP6基因、牛冠状病毒(Bovine coronavirus,BCV)N基因序列,利用Primer Premier 5.0软件分别设计特异性引物,采用PCR方法检测粪便样品中这3种基因,利用Excel 2007对不同地区、不同季节和混合感染病原检测结果进行汇总分析。在采样范围内的788份犊牛腹泻粪便样品中,BVDV、BRV和BCV阳性检出率分别为29.82%(235/788)、29.44%(232/788)和18.02%(142/788);在所有被检地区,滦南县BVDV感染率最高,阳性检出率为45.38%(59/130),汉沽区BRV和BCV感染率最高,阳性检出率分别为55.00%(22/40)和37.50%(15/40);从采样季节来看,春季、夏季和秋季BRV感染率最高,阳性检出率分别为27.71%(46/166)、39.58%(114/288)和19.89%(39/196),冬季则以BVDV感染为主,阳性检出率为52.17%(72/138);从混合感染情况来看,以BRV+BCV二重混合感染为主,感染率为8.37%(66/788)。河北省唐山市各地区腹泻犊牛群中均存在BVDV、BRV和BCV感染,以BVDV、BRV单一感染为主。  相似文献

【目的】 试验旨在对新疆某规模牛场患腹泻疾病的犊牛进行病原学鉴定及基因型分析。【方法】 采用抗原诊断试剂盒方法对在新疆某牛场随机采集的15份腹泻犊牛粪便样品进行检测,对抗原检测结果为阳性的样品进行反复冻融和过滤处理,然后将样品接种于Marc-145细胞进行病毒的分离和传代。对分离毒株进行间接免疫荧光试验(IFA)和负染电镜观察进一步确定病原。对分离株的VP6和VP7基因进行PCR扩增测序,并对其进行基因相似性比对和进化树分析。【结果】 抗原诊断试剂盒结果显示,3份粪便样品呈牛轮状病毒(Bovine rotavirus,BRV)抗原阳性。将阳性粪便样品分别接种于Marc-145细胞,仅有1份样品连续盲传至第9代出现明显的细胞病变效应(CPE)。IFA结果显示,接种该分离株的Marc-145细胞有亮绿色荧光,对照组未见荧光。电镜观察可见约65 nm的圆形病毒粒子,并命名为XJ-2022株。经PCR扩增获得VP6和VP7基因的目的条带,长度分别为1 356和342 bp。基因相似性比对和遗传进化分析表明,VP6基因与人源A群轮状病毒参考株DB2015-066(LC367318.1)相似性最高且遗传进化亲缘关系最近;VP7基因与牛源G10轮状病毒参考株XJX2(MN937506.1)相似性最高,遗传进化亲缘关系最近,确定该分离株为A群G10型轮状病毒。【结论】 试验成功分离到基因型为A群G10型BRV XJ-2022株,该毒株为新疆地区首次发现的多宿主来源的基因重配病毒。  相似文献

Molecular epidemiology of bovine rotavirus from the Charolais area (France). Faecal samples from 164 diarrhoeic calves under 60 days of age were collected from the Charolais area of France during winter of 1998. The samples were tested by an enzyme-linked immunosorbent assay (ELISA) to detect the presence of rotavirus antigen. Of 164 dairy calves tested, 45.1% were positive for rotavirus antigen. The presence of rotavirus was confirmed by electrophoresis of genomic segments. Genomic segment 9 coding for the surface glycoprotein VP7 was amplified by RT-PCR using amplimeres corresponding to a conserved sequence located at the 5' and 3' ends. Nucleotides of the region 29 to 320-560 (average 427) was determined by the Taq dye deoxynucleotide cycle sequencing method. By comparison to the 175 sequences of gene 9 previously published, sequence analysis demonstrated that all of the isolates from the present study belong to the genotype G6. This result confirms previously published data indicating the prevalence of rotavirus G6 in bovine, and suggests that a monovalent vaccine based on G6 antigen would be sufficient to elicit a good protection.  相似文献

Infection of calves by rotavirus could lead to diarrhea or subclinical disease. Therein, kinetic of viral excretion from a group of asymptomatic calves, previously exposed to a virulent strain of rotavirus, is compared to that of a group the calves of which had diarrhea associated with rotavirus infection. As determined by an enzyme-linked immunosorbent assay (ELISA), duration of rotavirus shedding in feces and maximum yield of virus antigen were similar within the two groups of calves. Rotaviruses isolated from the two above groups of animals were antigenically related as shown by neutralization tests, and showed a similar RNA electrophoretic profile. In conclusion, it is likely that both asymptomatic and diarrheic calves infected by rotavirus are equally a major source of contamination to contact healthy calves.  相似文献

The frequency of group A bovine rotavirus (gpA BRV) in calves from 1998 to 2002 was determined by the polyacrylamide gel electrophoresis technique in 2177 faecal samples, of which 1898 samples were diarrhoeic and 279 were of normal consistency (control group) that were collected from asymptomatic calves for comparative purposes. The animals were from beef and dairy cattle herds (n = 321) from 158 counties in seven States (Paraná, São Paulo, Minas Gerais, Mato Grosso do Sul, Mato Grosso, Goiás and Rondônia) and four Brazilian geographical regions (south, south-east, centre-west, and north). GpA BRV was detected in 19.4% (369/1898; p = 0.0001) of the samples collected in calves with diarrhoea and in only 2.2% (6/279; p = 0.0001) of the faeces with normal consistency. The proportion of positive samples collected from beef and dairy cattle herds was 22.8% (205/899; p = 0.0001) and 16.4% (169/999; p = 0.0005), respectively. In relation to age, a higher prevalence of infections was found in calves up to 30 days old, where 33.0% (189/573; p = 0.0001) and 20.2% (138/683; p = 0.0001) of the diarrhoeic faecal samples from beef and dairy cattle herds, respectively, were positive for gpA BRV. These results show the possible importance of inclusion of gpA BRV in the management of neonatal calf diarrhoea in Brazilian cattle herds.  相似文献

Neutralizing antibody titers to four bovine rotavirus strains, representing three serotypes, were measured in 160 sera from cattle of different age groups. Age-specific seroprevalence analysis revealed serotype 6, represented by bovine rotavirus (BRV) NCDV, as the predominant rotavirus serotype infecting German cattle and serotype 10, represented by BRV V1005, as the least prominent. Infections with serotype 8, represented by BRV 678, occurred with intermediate frequency. Antibodies of young calves distinguished between NCDV and UK virus, two serotype 6 BRV strains differing in VP4 antigen.  相似文献

为确定甘肃省临夏州某奶牛场犊牛腹泻的病因,并提供合适的治疗方案和防控措施,试验采集该牛场13头腹泻犊牛的粪便和血清,通过胶体金技术、ELISA方法、细菌分离鉴定、Kirby-Bauer法分别进行病毒病原学检测、病毒血清学抗体检测、病原菌鉴定和药物敏感性试验。病毒学检测结果显示,13份粪样中未检测出牛轮状病毒(BRV)、牛冠状病毒(BCV)的抗原,牛病毒性腹泻病毒(BVDV)抗原阳性率为23.08%(3/13);未检出BRV和BCV的抗体,BVDV血清学抗体阳性率为38.46%(5/13)。病原菌检测结果显示,13份粪便样品中,分离出13株大肠杆菌和7株奇异变形杆菌。药敏试验表明,分离的大肠杆菌和奇异变形杆菌对20种常规药物均产生了不同程度的耐药,且无对两种细菌均有效的药物。此次犊牛腹泻是由BVDV、大肠杆菌、奇异变形杆菌混合感染引起的,且大肠杆菌和奇异变形杆菌的耐药现象严重,本试验结果为该牛场进一步治疗此次的犊牛腹泻病提供了合理有效的依据。  相似文献

Serum antibody analyses for bovine herpesvirus type 1 (BHV-1), bovine viral diarrhea virus (BVDV), bovine respiratory syncytial virus (BRSV), bovine coronavirus (BCV), and bovine rotavirus (BRV) were performed on 527 randomly selected cows, before calving, and on 407 three-week-old calves. In cows and calves, BCV and BRV were the most seroprevalent viruses (80% to 100% according to virus and vaccination status). Bovine respiratory syncytial virus was the least seroprevalent in the cows, independent of the vaccination status. In nonvaccinated cows the seroprevalence to BRSV was 36.7%, and 53.5% in cows vaccinated less than two weeks prior to collecting blood, and 67.6% in cows vaccinated two weeks or more prior to blood collection. In their calves, BHV-1 was the least seroprevalent, independent of the vaccination status. The serological status and antibody titers in calves were generally associated with those of the dam. The occurrence of respiratory diseases in the calves was associated with cow and calf serological profiles (BHV-1, BRSV and BCV in the nonvaccinated group, BHV-1, BVDV and BCV in the vaccinated group). The occurrence of diarrhea was not associated with cow and calf serological profiles but was negatively associated with high level calf serum IgG in the nonvaccinated group (odds ratio = 0.73). Bovine coronavirus and BRV were shed by 1.4% and 4.9% of calves in the nonvaccinated group, and by 0% and 9.9% of calves in the vaccinated group, respectively. Bovine rotavirus shedding was associated with fecal diarrheic consistency at the moment of fecal sampling but not with previous occurrence of diarrhea.  相似文献

Reports of rotavirus excretion in calves usually result from cross-sectional studies, and in face of the conflicting results regarding protection of calves born to vaccinated dams against diarrhea, the aim of the present study was to evaluate rotavirus excretion in dairy calves born to vaccinated or unvaccinated dams, to identify the genotypes of bovine rotavirus group A (RVA) strains isolated from these animals as well as to investigate characteristics of the disease in naturally occurring circumstances throughout the first month of life. Five hundred fifty-two fecal samples were taken from 56 calves, 28 from each farm and, in the vaccinated herd, 11/281 samples (3.91%) taken from six different calves tested positive for RVA while in the unvaccinated herd, 3/271 samples (1.11%) taken from 3 different calves tested positive. The genotyping of the VP7 genes showed 91.2% nucleotide sequence identity to G6 genotype (NCDV strain), and for the VP4 gene, strains from the vaccinated herd were 96.6% related to B223 strain, while strains from the unvaccinated herd were 88% related to P[5] genotype (UK strain). Genotypes found in this study were G6P[11] in the vaccinated herd and G6P[5] in the unvaccinated herd. All calves infected with rotavirus presented an episode of diarrhea in the first month of life, and the discrepancy between the genotypes found in the commercial vaccine (G6P[1] and G10P[11]) and the rotavirus strains circulating in both vaccinated and unvaccinated herds show the importance of keeping constant surveillance in order to avoid potential causes of vaccination failure.  相似文献

The occurrence and distribution of certain enteropathogenic organisms and the manifestation of diarrhea, especially in relation to colostral management, were studied in neonatal calves of eight southern Finnish dairy herds with recurrent outbreaks of enteric disturbances. The fecal samples were obtained from the calves and were provided by the farmers. Potentially pathogenic agents were demonstrated in 52 of the 68 calves studied. Cryptosporidium was found in 36 of them while rotavirus was found in 29 and Escherichia coli in 15 of them, either singly or in combination. Salmonella, coronavirus or parvovirus were not detected in any of the calves. Twenty-six of the 36 calves contracted their Cryptosporidium infection and 18 of the 29 calves their rotavirus infection within the first week of life.

Eighteen of the 52 infected calves developed diarrhea of microbial origin within 20 days of age; the most frequent finding was mixed infection with Cryptosporidium and rotavirus. In addition there were six diarrheic calves where no infection could be demonstrated. Only 10 of the 68 calves had neither infections nor diarrhea. There was a highly significant difference between the mean times of the first colostral feeding of the uninfected non-diarrheic and diarrheic calves and a significant difference between the mean times of the first colostral feeding of the subclinically infected and diarrheic calves. The importance of sucking of colostrum at the first feeding was observed. The smallest amount protecting the calves from infections was 0.75 1 given within 1 h of delivery.  相似文献

Enzyme-linked immunosorbent assay (ELISA) was compared with electron microscopy in the examination of faeces from experimental calves and showed 100 per cent agreement in the detection of 19 bovine coronavirus and 15 bovine rotavirus electron microscope positive samples. In a limited field survey of calf diarrhoea 75 selected faeces were examined independently by ELISA and electron microscopy and the agreement between the two tests was 95 per cent for coronavirus and 84 per cent for rotavirus. A further comparison was made with 74 samples submitted for routine diagnosis and this yielded agreements of 82 per cent (coronavirus) and 89 per cent (rotavirus). Factors contributing to discrepant results were examined and the relative advantages and disadvantages of the two tests for routine detection of these enteric viruses are discussed.  相似文献
