试验旨在探究围产期亚临床酮病与泌乳早期奶牛繁殖性能、卵泡发育之间的关系,并检测试验牛血液生化指标的变化。试验在黑龙江某大型集约化牛场开展,根据产后血酮水平确定亚临床酮病组(SCK)和健康组(C)奶牛共60头,根据试验牛产后50 d内发情状况,将SCK组再分为发情组(SCKE,16头)和乏情组(SCKA,14头),C组也同样分为发情组(CE,25头)和乏情组(CA,5头)。所有试验牛在产后50 d通过直肠检查和B超检查了解子宫复旧及卵泡发育状况,记录繁殖性能数据,并进行血液生化指标分析。结果表明:与健康组发情奶牛相比,亚临床酮病发情奶牛产后首次发情日期推迟约10 d(P<0.05);产后50 d卵泡直径差异极显著(差值约4 mm)(P<0.01)。亚临床酮病乏情奶牛子宫复旧延迟发生率显著高于健康组发情奶牛(P<0.05);亚临床酮病奶牛血浆中胰岛素样生长因子-1(IGF-1)含量显著低于健康奶牛(P<0.05),而泌乳量极显著提高(P<0.01)。与发情奶牛相比,乏情奶牛血浆中甘油三酯(TG)含量显著升高(P<0.05);葡萄糖(Glu)、胰岛素(Ins)、雌二醇(E2)、孕酮(P4)含量显著或极显著降低(P<0.05;P<0.01)。综合以上试验结果,奶牛患亚临床酮病而导致能量代谢指标异常是引起奶牛乏情、产后卵泡发育受阻和繁殖障碍的主要因素,进而导致奶牛繁殖力下降。  相似文献

A study was conducted to determine the effect of charcoal-extracted, bovine follicular fluid (CFF) on plasma follicle stimulating hormone (FSH) and luteinizing hormone (LH) concentrations, the interval from luteolysis to estrus, and subsequent luteal function in heifers. Fifteen Angus, Simmental and Hereford heifers were allotted by age, weight and breed to a control (C, n = 8) or a CFF (n = 7) group. Heifers received injections of saline or CFF (iv, 8 ml/injection) every 12 h from d 1 (d 0 estrus) through d 5 of the estrous cycle. On d 6, each heifer was injected (im) with 25 mg of prostaglandin F2 alpha (PGF2 alpha). Blood samples were collected every 12 h by venipuncture starting just before the first saline or CFF injection and continuing until estrus. Thereafter, blood samples were collected every other day during the subsequent estrous cycle and assayed for FSH, LH, estradiol-17 beta and progesterone by radioimmunoassay. Injections of CFF had no effect (P greater than .05) on circulating FSH or LH concentrations from d 1 to 5 relative to the C group; however, there was a transient rise (P less than .05) in FSH concentrations 24 h following cessation of CFF injections. This transient rise in FSH was not immediately followed by an increase in plasma estradiol-17 beta concentrations. Three experiments were conducted on the metabolism of stallion sperm. In experiment 1, whole and washed sperm were incubated under aerobic and anaerobic enviroments and analyzed before and after controlled incubation for motility, pH, lactic acid, glucose, fructose, and O2 comsumption. In experiment 2, whole and washed sperm were incubated aerobically and anaerobically with and without uterine tubal fluids. Experiment 3 was the same as experiment 2, except added substrates of glucose and lactic acid were studied. The same examinations were made in experiments 2 and 3 as for experiment 1. Motility decreased significantly during incubation for all treatments, with the greatest decrease occurring for whole semen where only trace amounts of substrate (fructose) were present. Exogenous glucose plus uterine tubal fluid maintained sperm motility better than did added lactate. However, sperm respiration rates were highest when exogenous lactate was the only substrate in the incubation medium. The mean pH values for gel-free stallion semen at the start of controlled aerobic and anaerobic incubation were 7.08 and 7.34. Lactic acid accummulation for 1 hour increased from 0.05 mg to 0.09 mg/10(9) sperm when uterine tubal fluid was added to the incubation medium. Washed spermatozoa incubated in 0.03 M glucose plus uterine tubal fluid utilized less glucose than did sperm incubated in the glucose medium. These results, along with the increased oxygen utilization (ZO2) values produced by adding uterine tubal fluid to the incubation mediums, might indicate utilization of a uterine tubal substrate. Added uterine tubal fluid resulted in increased ZO2 values (expressed in mul of O2 utilized by 10(8) sperm in 1 hour at 37 C) for whole semen from 10.45 to 12.63. Washed spermatozoa also respired at a significantly greater rate than whole sperm. Respiration rates were greater for sperm incubated with 0.01 M lactic acid than for any other substrate or experiment.  相似文献

Forty uterine fluid samples were obtained from 4 mares classified as resistant to uterine bacterial infection. The uterus of each mare was flushed with 50 ml of saline solution during estrus and diestrus of successive estrous cycles. Bacteria or fungi were isolated from 4 samples, and 7 additional samples were obtained from a mare with active intrauterine infection. Fluid volumes obtained during estrus (means = 40.3 +/- 11 ml) tended to be greater than those recovered during diestrus (means = 36.8 +/- 7.9 ml), but the difference was not significant. Concentrations and yields of protein in recovered fluid did not vary significantly with the day of the estrous cycle. Protein concentration was significantly increased in samples from the infected uterus (P = less than 0.001). The ability of uterine fluid samples to attract neutrophils was measured using chemotactic chambers. There was no significant difference between distances migrated by neutrophils toward fluids obtained during estrus or diestrus. Chemotaxis scores tended to be higher with samples from the infected uterus, and the difference was significant for samples from 2 mares. Chemotaxis was neither significantly correlated with protein concentration of uterine fluid, nor with serum estrogens or progesterone.  相似文献

This project aimed to determine the effect of Se as inorganic Na-selenite (MSe) or organic Se-yeast (OSe) on antioxidant status, hormonal profile, reproductive performance, and embryo development in first-parity gilts. Forty-nine gilts were allocated to 1 of the 3 dietary treatments starting at first pubertal estrus and lasting up to 30 d after AI: control [CONT: basal diet (Se = 0.2 mg/kg) without added Se; n = 16], MSe (CONT + 0.3 mg/kg of MSe; n = 16), and OSe (CONT + 0.3 mg/kg of OSe; n = 17). Blood was collected from all gilts on the day after each onset of estrus and on d 30 after AI. Blood was also collected daily from d -4 to d +4 of the third onset of estrus (d 0) in 8 CONT, 9 MSe, and 8 OSe cannulated gilts. Gilts had received, after d 14 and 15 of their third estrus, a hormonal challenge to induce super-ovulation. At slaughter, embryos and corpora lutea (CL) were weighed and measured. Blood Se was less (P < 0.01) in CONT than in Se gilts and greater in OSe than in MSe (P < 0.01) from the first estrus until d 30 of gestation. At the same time, blood Se-dependent glutathione peroxidase (GSH-Px) decreased for CONT gilts, whereas it increased for both Se groups. The increase was greater in MSe than in OSe gilts (treatment × time, P = 0.02). Plasma 3,3',5-triiodothyronine and thyroxine concentrations for MSe tended to be less than for OSe gilts (P < 0.06). In cannulated gilts, plasma FSH tended to change among treatments (treatment × time, P = 0.06), and plasma estradiol-17β (E(2)) was less (P = 0.01) for MSe than for OSe. There was no treatment effect on mean litter size or embryonic antioxidant status. The Se content of individual embryos was greater for Se-treated than for CONT gilts (P = 0.03), and Se content of individual embryos and total litter was greater for OSe than for MSe gilts (P < 0.01). The length, weight, and protein content of embryos were greater in OSe than in MSe gilts (P < 0.05). There was no treatment effect on weight, length, Se content, and ferric reducing antioxidant power of CL, but GSH-Px in CL was greater for Se than for CONT gilts (P = 0.02). In summary, the Se status response of gilts to dietary Se was affected by both the quantity and the source of Se dietary supplements. Moreover, the uterine transfer of Se to embryos was improved with OSe as compared with MSe, and this was concomitant with an enhanced development of embryos.  相似文献

In order to determine the role of follicle-stimulating hormone (FSH) on the resumption of ovarian function in cows early postpartum (PP), bovine follicular fluid (FF) was used to selectively suppress concentrations of FSH. Calves were removed from all cows within 24 hr of birth. Follicular fluid that was treated with charcoal to remove steroids (15 ml; n = 14) or serum (S) from an ovariectomized cow (15 ml, n = 14) was injected i.m. twice daily from days 1 to 10 PP. Blood samples were collected before each injection and frequent samples (every 15 min for 6 hr) taken on days 5 and 10 PP. Eight cows from each group (FF and S) were slaughtered on the morning of day 11 PP and pituitaries and ovaries collected. The remaining cows (n = 6) were observed for estrus. Treatment with FF delayed follicular growth (P less than 0.01), as evidenced by the largest follicle per cow observed at time of slaughter (3.6 +/- 0.42 vs 11.5 +/- 1.77 mm dia; FF vs S). The intervals from parturition to first estrus (P less than 0.11) and to first progesterone rise (25.3 +/- 1.97 vs 18.0 +/- 3.62 d; P less than 0.06) tended to be delayed by treatment with FF vs S. Many of the cows treated with S ovulated by day 10 PP, they were divided retrospectively into those that had ovulated by (n = 9) or after (n = 5) day 10 PP for analysis. Cows treated with FF had lower (P less than 0.05) and less variable (P less than 0.01) serum FSH concentrations while levels of luteinizing hormone (LH) tended (P less than 0.08) to be greater on days 5 and 10 PP. Follicular fluid decreased levels of FSH (P less than 0.001), but not LH (P less than 0.15), in the samples obtained twice daily compared to S-treated cows that did not ovulate by day 10 PP. Anterior pituitaries were dissociated, and cells from each cow were cultured in order to ascertain whether treatment with FF in vivo would affect gonadotropin secretion in vitro. Estradiol-17 beta (E) was incubated with pituitary cells to determine the effect of E on gonadotropin secretion from cells of PP cows, and to ascertain whether treatment with FF in vivo and with E in vitro would interact to affect secretion of FSH and LH in culture. The hypothesis that subnormal luteal function after induced ovulation in anestrous ewes was the result of uterine influences exerted during the periovulatory period was tested. Crossbred ewes (n = 27) in seasonal anestrus were induced to ovulate by administration of 12 doses of 250 ng of LHRH at 2-h intervals, followed immediately by a bolus injection of LHRH (250 micrograms; d 0). Ewes were unilaterally hysterectomized on either d -3 (PRELHRH) or 2 (POSTLHRH). Daily blood samples were collected and assayed for progesterone (P4) and 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM). All ewes were slaughtered on d 10, and corpora lutea (CL) were collected, weighed, and assayed for concentration of P4. All ewes that ovulated exclusively in the ovary ipsilateral to the remaining uterine horn had a transient increase in plasma P4 of 2 to 3 d (short luteal phase). In ewes with at least one CL in the isolated ovary, elevated plasma P4 was maintained after hysterectomy but was consistently lower (P less than .05) in POSTLHRH ewes than in PRELHRH ewes. Concentrations of PGFM did not differ between treatments. The CL ipsilateral to the remaining uterine horn weighted less (P less than .01) and contained less P4 (P less than .01) than contralateral CL. These data confirm the hypothesis that premature regression of subnormal CL is uterine-dependent in a local fashion. Presence of the uterus during the follicular and(or) early luteal phase inhibited subsequent luteal function in seasonally anestrous ewes.  相似文献

In a study of preferences for diets containing 1% to 20% sucrose over a bland diet, both female and male dogs preferred the diets containing sucrose over a bland diet, but female dogs had a significantly (P less than 0.05) greater preference for 1% sucrose than did males. Differences between sexes were not observed at the higher concentrations. Mean daily caloric intake was calculated for 12 bitches fed ad libitum during metestrus, estrus, and anestrus, as determined from metachrome-stained vaginal smears. Mean daily caloric intake was lowest during estrus (149 +/- 17 kcal/kg of body weight), was higher during metestrus (159 +/- 8 kcal/kg of body weight), and was highest (175 +/- 9 kcal/kg of body weight) during anestrus. Ovariohysterectomized bitches gained significantly (P less than 0.01) more weight (1.3 +/- 0.3 kg) in the first 10 days after surgery than did sham operated controls (0.3 +/- 0.1 kg). Food intake also was significantly greater ( less than 0.01) in the ovariohysterectomized bitches (1,708 kcal/day) than in the sham operated controls (1,423 kcal/day). Depth of subcutaneous fat in shoulder, rib, and rump areas of ovariohysterectomized bitches was not significantly different from that of intact bitches.  相似文献

The present experiment was conducted to determine the influence of dietary fatty acids C18:2n-6 and C18:3n-3 on the modulation of intrauterine synthesis of prostaglandin E2 (PGE2) and F2alpha (PGF2alpha) during early pregnancy in pigs. Prostaglandin E2 in uterine fluid has been previously reported to be associated with embryo survival and development. Thirty-two Yorkshire-Landrace nulliparous gilts were randomly allocated to four diets containing 5% supplemental fat. The four dietary treatments were: HT, hydrogenated tallow (26.5% C16:0 and 54.8% C18:0); SO, sunflower oil (61.3% C18:2n-6); LO, linseed oil (50.4% C18:3n-3); and SO(CLA), a mixture of sunflower oil and conjugated linoleic acids to provide 20% CLA. Treatments started 2 d after the first pubertal estrus (d -21) and lasted for 36 d (slaughter), which was 15 d after the second estrus (d 0; insemination). Fatty acids and PGE2 were measured in the peripheral blood plasma on d -19, d -7, d 0, and d 14. Fatty acids in endometrial tissues and PGE2 and PGF2alpha in the uterine fluid collected on d 15 were also measured. Concentrations of fatty acids in the plasma reflected the content of fatty acids in the diet as early as d -7. From d -7, PGE2 concentrations in the plasma were higher in gilts fed SO compared with HT (P < 0.05). Plasma PGE2 concentrations were lower (P < 0.01) on d 14 in gilts fed LO compared with HT. Total PGF2alpha contents in the uterine fluid of gilts fed LO were more than 70% lower (P < 0.05) than for the HT group. A similar trend was observed for total PGE2 content and for the ratio PGF2alpha:PGE2, but the effect (LO vs HT) was less marked (P < 0.07 and P < 0.10, respectively). There was no effect of SO or SO(CLA) on total PGE2 contents in the uterine fluid. Dietary enrichment in C18:2n-6 and/or C18:3n-3 for early pregnant gilts can influence fatty acids in plasma and endometrial tissue and can modulate circulatory and intrauterine prostaglandins.  相似文献

Effects of recombinant bovine somatotropin (rbST) on ovarian and uterine function and the production of components of the insulin-like growth factor (IGF) system were examined during the period of maternal recognition of pregnancy in cattle. Lactating dairy cows were treated with 25 mg/d rbST (n = 8) or saline (n = 8) for 16 d after estrus. Ovaries, uteri, and conceptuses were collected on Day 17 after estrus. The length (millimeters) of the conceptus was recorded. The concentration of IGF-I and the content of IGF-binding proteins (BP) in uterine flushings were determined. Corpora lutea (CL) were weighed, and the number of follicles (⩾2 mm in diameter) were counted. Follicular fluid from the largest and second-largest follicles was assayed for the concentration of IGF-I, IGFBP, progesterone, and estradiol. The length of the conceptus and the total amount of IGF-I in uterine fluid were similar for rbST and control. Recombinant bST increased 1) the weight of the CL, 2) the number of largest follicles (10 to 15 mm in diameter), 3) the concentration of IGF-I in the follicular fluid, 4) the follicular fluid content of IGFBP of the largest estrogenic follicle, and 5) the quantity of IGFBP in uterine flushings. The concentration of progesterone in the follicular fluid tended to be increased in rbST-treated cows, whereas the concentration of estradiol was similar to that of control cows. The concentration of progesterone in plasma was similar for rbST compared with control. In conclusion, the administration of rbST in lactating dairy cows for 16 d after estrus did not alter the growth of the conceptus collected on Day 17. The greatest responses to rbST were found within the ovary, where rbST increased the weight of the CL and altered the amount of IGF-I and IGFBP in the follicular fluid.  相似文献

An experiment was conducted to determine whether morphological and functional characteristics of follicles differed at a similar stage of pubertal (first) and third estrus in the same gilts. Nine prepubertal gilts were checked three times daily for estrus and laparotomized 6 h after detected first and third estrus. Samples of vena cava and ovarian venous blood were collected, follicle numbers and diameters were recorded, and follicular fluid (FF) was aspirated from all follicles 8 to 12 mm in diameter. Sera and(or) FF were analyzed for progesterone (P4), estradiol-17 beta (E2), testosterone (T), androstenedione (A4), 5 alpha-dihydrotestosterone (DHT), plasminogen activator (PA), and plasmin (PLM). Overall mean number of follicles > or = 8 mm in diameter did not differ between gilts at first and third estrus (P > .05) but gilts at first estrus had more follicles 4 to 8 (P < .05) and 8.1 to 10 mm in diameter (P < .01) and fewer 10.1 to 12 mm in diameter (P < .07) than at third estrus. Mean FF concentrations of E2, T, and A4 at third estrus were significantly greater than at first estrus, whereas FF concentrations of P4, DHT, PA, and PLM were similar at first and third estrus (P > .05). Mean concentrations of E2 in systemic and ovarian venous sera were also greater in gilts at third than at first estrus (both P < .05). To investigate some biochemical changes during bovine follicle development, ovaries were obtained from cyclic heifers (7 to 11 heifers/d on each day of the 21-d estrous cycle; N = 152). Follicular fluid from the two largest follicles from both ovaries and a pool from small follicles (N = 30/cow) were collected from each animal and analyzed for ionic, enzymatic and endocrine changes in relation to day of the estrous cycle, follicle size, rank and atretic or growing status. Follicular fluid alkaline phosphatase activity and ascorbate concentrations were highest in all follicular sizes during the earlier portion of the estrous cycle (d 1 to 12; P less than .05), then decreased to the lowest levels (d 13 to 21). As follicular size (diameter) increased lactate dehydrogenase (LDH), acid and alkaline phosphatase activity was reduced in follicular fluid (P less than .05). Alkaline phosphatase and LDH activity tended to be increased in atretic follicles (P less than .10), and was correlated with increased progesterone and androgen concentrations of follicular fluid (r = .4, P less than .05). Both albumin and total protein concentrations decreased as follicular diameter increased (P less than .05). Sodium concentrations in follicular fluid were greater in growing-antral than atretic follicles, and increased with follicular enlargement (P less than .05). Follicular potassium concentrations increased as the estrous cycle progressed (P less than .05), and tended to be elevated in atretic follicles (nonsignificant). Both Ca and Mg concentrations increased with follicular enlargement (P less than .05). Dehydroepiandrosterone and testosterone were the predominant androgens in follicular fluid (androstenedione, the lowest concentration); their concentration decreased with follicle development (P less than .05), but were quite variable. Estradiol was increased in growing follicles (P less than .01). Estrone and estradiol concentrations increased as ovulation approached, particularly in small follicles (less than or equal to 4 mm diameter). Changes of biochemical components found in follicular fluid that relate to the growth and atresia process may provide a more sensitive and accurate method to classify follicle status, and thus aid in understanding the complexity of events associated with maturation of the bovine follicle and oocyte.  相似文献

Two hundred sixty-two sows were used to investigate the effects of boar exposure during the last week of nursing (BPRE) and after weaning (BPOST) on the return to estrus. Because approximately one-half of the sows were weaning their first litter, a third factor, primiparous vs multiparous (LITT), was considered in the statistical analysis. To evaluate the effect of treatment on ovarian activity, the sows were also blood-sampled twice weekly for 3 wk after weaning for the measurement of plasma progesterone concentrations as an index of ovulation. Boar exposure after weaning was the most important stimulus of early ovulation and estrus after weaning (P less than .001). A greater proportion of first-litter sows exhibited estrus later (P less than .02) and ovulated later (P less than .09) than did multiparous sows. First-litter sows were unaffected (P greater than .10) by boar exposure before weaning. Multiparous sows were sensitive to boar exposure during nursing. Maximal boar exposure for these sows (BPRE + BPOST) resulted in 95% of sows in estrus and ovulating within 20 d of weaning. No boar exposure resulted in 45% and 38% anestrus and anovulatory sows. Boar exposure, either before or after weaning, was effective in reducing the number of anestrus and anovulatory sows to between 15 to 30%. The effects of BPRE and BPOST on return to ovulation were additive and approximately equal.  相似文献

Twenty-two nonlactating Hereford cows exhibiting normal estrous cycles were fed either maintenance (M) or restricted (R) diets until most of the R cows became anestrus; R cows then were fed 160% of the M diet until estrous cycles resumed. Concentrations of progesterone, glucose, insulin and nonesterified fatty acids (NEFA) were determined in weekly blood samples. Blood also was collected frequently, before and after i.v. infusion of 300 ml of a 40% glucose solution, to evaluate responses in blood concentrations of glucose and insulin when cows were exhibiting normal estrous cycles, when R cows were initiating anestrus, during anestrus, and at the reinitiation of estrous cycles. Losses in BW and body condition score in R cows were associated with reduced (P less than .01) concentrations of glucose and insulin and greater (P less than .01) concentrations of NEFA in blood plasma compared with those of M cows. During normal estrous cycles, disappearance of infused glucose from plasma and concentrations of insulin in serum were similar for R and M cows. Glucose disappearance from plasma was retarded and serum concentrations of insulin remained increased for a longer time after glucose infusion in R at the start of anestrus compared with M cows (P less than .01). Similarly, during anestrus, the rate of glucose disappearance was slower for R cows (P less than .01). During refeeding of R cows, disappearance of infused glucose was similar for R and M cows. In conclusion, reduced concentrations of glucose and insulin and increased concentrations of NEFA in blood were associated with nutritional anestrus and the glucoregulatory effects of insulin were compromised during nutritional anestrus.  相似文献

The objective of the present study was to evaluate changes in equine follicular fluid insulin-like growth factor binding protein (IGFBP) proteolytic activity as well as steroid, IGF, and IGFBP concentrations during follicular development in the mare. Mares (n = 14) were classified as either in the follicular phase (n = 8) or luteal phase (n = 6). Follicles (n = 92) were categorized as small (6 to 15 mm; n = 54), medium (16 to 25 mm; n = 23), or large (> 25 mm; n = 15), and follicular fluid was collected. Estradiol and androstenedione levels in follicular fluid were greater (P < 0.05), and IGFBP-3 concentrations tended to be greater (P < 0.10) in large than in small or medium follicles, whereas IGFBP-2, -4, and -5 levels were less (P < 0.05) in large than in small or medium follicles. Estradiol and androstenedione concentrations were negatively correlated (P < 0.01) with IGFBP-2, -4, and -5 but not IGFBP-3 concentrations. To evaluate proteolysis of IGFBP, follicular fluid was incubated with human 125I-labeled IGFBP-2, -3, and -5 and protein separated by 12% SDS-PAGE. Follicular fluid caused little or no proteolysis of 125I-lableled IGFBP-2 or -3, and the small amount of proteolysis of IGFBP-2 and -3 did not differ (P > 0.10) among follicle classes. However, more 125I-labeled IGFBP-5 was cleaved (P < 0.05) by follicular fluid from large follicles collected during the follicular phase than large follicles during the luteal phase, and small or medium follicles from follicular and luteal phase mares indicating that a protease to IGFBP-5 exists in estrogen-dominant equine follicles. This IGFBP-5 protease was inhibited by kallikrein/serine protease and metalloprotease inhibitors. We conclude that the tendency of estrogen-dominant follicles of mares to have greater levels of IGFBP-3 and lesser levels of IGFBP-2 does not appear to be due to differences in proteolysis, whereas changes in IGFBP-5 levels are likely due to changes in activity of a serine protease or metalloprotease. Changes in IGFBP may alter levels of bioavailable IGF that stimulate steroidogenesis and mitogenesis in developing mare follicles.  相似文献

The objective of this study was to determine the effects of BCS at calving and the amount of postpartum protein supplementation on the dominant follicle (DF) and behavioral characteristics at the first postpartum estrus of mature beef cows. Multiparous Angus x Hereford cows (n = 45) were fed to calve in thin (T; < 5) or moderate (M; >or=5) BCS. Cows were stratified by BCS and calving date, and randomly assigned to receive lesser (L; 1.2 kg/d) or greater (G; 2.5 kg/d) amounts of a 42% CP supplement. All cows grazed the same native grass pasture and were fed in individual stalls for 49 +/- 2 d. Beginning 20 d after calving, blood samples were collected from each cow thrice weekly, and estrous behavior was monitored continuously with a radiotelemetry system. At 4 to 16 h after the onset of estrus, size of the DF was determined by ultrasonography. Body condition score of T cows was less (P < 0.01) at calving than M cows; L and G cows had similar BCS at calving and at the end of the feeding period. Body weight gains during treatment did not differ for L or G cows. Duration from calving to first estrus was greater (P < 0.01) for T than M cows. The incidence of a short luteal phase before first estrus was not influenced by BCS or protein supplement. Concentrations of IGF-I in plasma tended (P < 0.07) to be greater and size of the DF was greater (P < 0.01) for M than T cows. Size of the DF tended (P < 0.06) to be greater for G than L cows. Duration and number of mounts received at the first estrus were not influenced by BCS or supplement. Pregnancy rate of M cows during the breeding season was greater (P < 0.05) than T cows. Postpartum protein intake and BCS at calving influenced the size of the DF at the first postpartum estrus in mature suckled beef cows. Cows should be managed to calve in moderate BCS and maintain BW after parturition to decrease the interval to first estrus, increase follicular development, and maximize pregnancy rate.  相似文献

Effects of an increased level of dietary energy (flushing) on plasma concentrations of FSH, LH, insulin, progesterone and estradiol-17 beta and ovulation rate were studied in 16 gilts. Gilts received 5,400 kcal ME/d for one estrous cycle and the first 7 d of a second. On d 8 of the second estrous cycle, gilts received either 5,400 kcal ME/d (control [C], n = 8) or 11,000 kcal ME/d (flushed [F], n = 8) for the remainder of the estrous cycle. Blood was collected daily at 15-min intervals for 6 h from d 8 through estrus. Gilts were examined by laparotomy 6 d after estrus. Ovulation rate was greater (P less than .05) in F than C gilts (16.0 vs 9.4). Mean daily concentrations of FSH were greater (P less than .05) in F gilts at 5 d, 4 d and 3 d prior to estrus compared with C females. In both C and F gilts, FSH decreased (P less than .05) prior to estrus. Mean daily concentrations of LH and LH pulse amplitude were not different (P greater than .05) between treatments. Mean number of LH pulses/6 h at 4 d, 3 d and 2 d prior to estrus were greater (P less than .05) in F than in C gilts. In both treatments, LH pulse amplitude decreased (P less than .05) and pulse frequency increased (P less than .07) prior to estrus. Delaying the onset of estrus after weaning and adding fat to the postweaning diet were studied for their effects on estrus and fertility in 232 crossbred primiparous sows on a commercial swine farm. Sows were assigned randomly to the following treatments after weaning in June, July, August, or September, 1983: 1) altrenogest (20 mg/d) was fed for 7 d after weaning (n = 76), 2) altrenogest was fed for 7 d plus .53 kg dried animal and vegetable fat product (.45 kg actual fat/d) for 14 d after weaning (n = 78), or 3) no treatment (controls, n = 78). While a similar proportion of sows came into heat after weaning (lactation length = 4 wk), sows fed altrenogest (14.4 +/- .2 d) returned to estrus about 9 d later (P less than .01) than controls (5.6 +/- .2 d). Serum progesterone concentration was assayed in blood samples collected from a subgroup (74%) of the cows not observed in estrus by 3 wk after weaning to determine possible causes of anestrus. If serum progesterone (greater than 5 ng/ml) was elevated, we assumed that sows had ovulated without expressing estrus (behavioral anestrus) or ovulated with undetected estrus (less overt estrus or error in estrous detection), whereas low progesterone (less than or equal to 5 ng/ml) indicated that sows were anovulatory. About 53% of the sows not observed in estrus across all treatments had luteal function, probably resulting from post-weaning ovulation. Incidence of anovulation without estrus was 47%. Serine proteases help to regulate the ovarian cycle at different levels and they are subjected to the control of gonadotropic hormones and protease inhibitors. Superovulation stimulations influence the activities of trypsin inhibitors (model serine protease) in blood plasma (BP) and in follicular fluid (FF), and also in dependence on the breed. Trypsin inhibiting activities were determined from the reduced rate of trypsin hydrolysis of a chromogenic substrate (TAPA) and they were determined in percent. A change in absorbancy at 405 nm = 1.0 after 10-minute incubation at 25 degrees C and pH = 8.05 was taken as 100%. The incubation mixture as a sample contained 100 microliters blood plasma or 10 microliters follicular fluid, diluted with gammaglobulin at 1:10. The differences in the trypsin inhibiting activities (TIA) of BP in ewes of the Merino, Tsigai and Wallachian breeds were insignificant, but Agelin synchronization (20 mg chlorsuperlutin per vaginal swab) induced statistically significant differences. The lowest TIA BP was recorded in the Tsigai breed (T), P less than 0.001 in comparison with the Wallachian (W) and Merino (M) breeds. Following the administration of 1,500 IU PMSG, the TIA BP within 120 hours decreased in W (P less than 0.001), it increased in T (P less than 0.1) and in M the changes in the TIA BP were insignificant. The average numbers of ovulations increased from 2.25 +/- 2.5 to 3.0 +/- 1.2 in W; from 0.25 +/- 0.43 to 2.5 +/- 1.6 in T and from 0.00 +/- 0.0 to 2.5 +/- 2.3 in M. Following the single administration of 2,000 IU PMSG after Agelin synchronization, the changes in M ewes were also insignificant, and there were no different responses in pregnant (1st to 2nd month) and nonpregnant ewes. In pregnant T ewes the TIA BP increased after Agelin synchronization and stimulation (P less than 0.01), in nonpregnant ewes these changes were not significant. In W lambing ewes the TIA BP increased (P less than 0.001), the effects of Agelin were greater than those of PMSG. The TIA of follicular fluid (FF) of antral follicles were on average tenfold if compared with BP. After hormonal treatment of ovaries, the TIA FF mostly increased at different levels of statistical significance.
