The current paper describes aspects of local immunity in the ovary and oviduct, and the significance of immunity to reproductive functions in hens. The immunocompetent cell populations in the ovary and oviduct change with a positive correlation to sexual activity, and gonadal steroid is one of the key factors in the increase. Local immune responses mediated by major histocompatibility complex class II and T cell subsets occur in response to infection by Salmonella enteritidis, which may contaminate eggs. In the ovary, immunocompetent cells are also suggested to play roles in the regulation of ovarian functions. Macrophages and T cells are likely to enhance the regression of atretic follicles to maintain the ovarian tissue microenvironment. Autoantibodies to ovarian tissues appeared in the hens with low egg laying frequency, suggesting that the auto‐antibodies may be one of the factors in the decline of egg production. In the oviduct, local immunity possibly has a role in the selection of sperm, though the immunoreactions may also affect sperm survival leading to the decline in fertility. The concentration of yolk IgY, which plays a role in maternal immunity transmission, significantly decreases with the aging of birds, whereas it is significantly increased by estrogen. Therefore, the immune system plays significant roles not only in defense against infection, but also in the functions of reproductive organs. Investigations on the local immune system in the reproductive organs and factors affecting it are of importance for the production of sterile eggs and improvement of reproductive functions. 相似文献

In cattle, Ureaplasma diversum has been associated with decreased fertility, granular vulvovaginitis, endometritis, salpingitis and spontaneous abortion in cows and seminal vesiculitis, balanoposthitis and changes in bull sperm. The presence of U. diversum within the Australian cattle population has not been established. One of the aims of this study was to determine if U. diversum was present in Australian cattle, using culture and polymerase chain reaction (PCR), both of which are considered to be gold standards for bacterial identification. Of 100 samples collected from 66 male and 34 female cattle, 15 were positive for U. diversum. Therefore, Australia can no longer be considered free of U. diversum. Further studies should be conducted to ascertain the effects of U. diversum within Australian cattle herds and, if warranted, to investigate prevention, treatment and eradication protocols. 相似文献

The antibody levels to leptospiral antigens among dairy herds were determined and correlated with those of the milkers in three areas of the South Island of New Zealand. The milkers filled out an extensive questionnaire giving age and sex of the milker, as well as details of the herd milked and his/her association with it. The serological results are compared with these data and the value of such seroepidemiological studies examined from the point of view of the advantages of vaccinating cattle to protecting the milker from leptospirosis. Antibodies to L. interrogans serovar hardjo were the ones most frequently encountered among the milkers; however, antibodies to other serovars were also noted. Some animals in most herds had antibodies to serovar hardjo. These studies suggest that vaccinating cattle with serovar hardjo would not necessarily reduce the human incidence of leptospirosis in this population 相似文献

Little is known about the safety and efficacy of extra-label use of the modified live porcine reproductive-and-respiratory syndrome (PRRS) virus vaccine in gestating sows. Our purpose was to determine the impact of vaccination on reproductive performance in 54 herds in Ontario, Manitoba (Canada) and the mid-western United States that were PRRS-positive, PRRS-negative, or concurrently affected by an outbreak of PRRS when initially vaccinated. Majority-vaccinated herds vaccinated ≥50% but <100% of sows at one time, and limit-vaccinated herds vaccinated <50% of sows at one time. Most majority-vaccinated herds did not vaccinate sows in late gestation, and none vaccinated during the initial PRRS outbreak. Numbers of pigs born alive and weaned decreased when pregnant sows were vaccinated. The effect of vaccination on productivity in the gestation following vaccination depended on the vaccination protocol. 相似文献

Mature and healthy male house rats, Rattus rattus ( n= 160) were fed on bait (cracked wheat: powdered sugar, 98:2) containing different concentrations of triptolide (0.1, 0.05, 0.025 and 0%) for 7 and 14 days in no‐choice and bi‐choice feeding tests in the laboratory. The objective of the study was to record the antifertility affects of triptolide after 30 and 60 days of termination of treatment. Results revealed no significant effect of triptolide treatment on weights of testis, epididymis, seminal vesicles and prostate gland of rats. Overall, sperm motility, live sperm count, sperm density and sperm morphology in the cauda epididymal fluid were found to differ significantly ( P≤ 0.05) between untreated and treated groups of rats. The major effect of triptolide on sperm morphology was in the form of sperm head tail separation, which was up to 56.0% in rats treated for 14 days in no‐choice and autopsied after 30 days. A significant effect ( P≤ 0.05) of triptolide treatment was observed on the histomorphology of the testis, which included a dose‐dependent decrease in diameter of seminiferous tubules, thickness of germinal epithelium and numbers of various spermatogenic cells. Cell associations in the seminiferous epithelial cycle were poorly developed in rats ingesting medium (4.7–5.1 mg/100 g bw) and high doses (6.9–7.2 mg/100 g bw) of triptolide than rats ingesting low doses (1.8–2.3 mg/100 g bw) and untreated rats. The cell stages affected had not recovered fully within the 60 day period following triptolide withdrawal. The present study suggests the potential of triptolide in reproductive management of Rattus rattus. 相似文献

Experiments were undertaken in six Holstein-Friesian cattle to determine whether secretory IgA (SIgA) could be transported from serum into exocrine body fluids. Preliminary data indicated that when IgG 1, IgG 2, IgM and SIgA were administered i.v., only SIgA and IgG 1 appeared in bile 90 min later at concentrations equal to or exceeding those in serum at the same time. Two hours post-injection, 70% of the SIgA recovered in bile was intact however only 30% co-precipitable with anti-secretory component (SC) while >90% of the administered IgA was precipitated by this method. All recovered IgG 1 was of low molecular weight. More detailed studies indicated that the IgA recovered in bile 7 h post-injection or in milk 3 h post-injection, was predominately lower molecular weight than intact SIgA. Most of this low molecular weight radioactivity was TCA precipitable and ca 50% was dialyzable; these data indicate that TCA-precipitability is an inadequate criterion for determining whether intact SIgA is transported. The radioactivity recovered in parotid saliva was almost entirely non-TCA precipitable and dialyzable. Almost all SIgA recovered in bovine serum remained intact and had a
of 15.7 h. When transport into milk and bile was calculated from total, recovered radioactivity (i.e. 29% and 2.7, respectively), data compared favorably with those conducted in sheep in which dimeric IgA (without SC) was administered i.v. When we calculated transport on the basis of recovered intact IgA, only 1.47 and 0.54% of the injected dose had been transported into milk and bile, respectively, 24 h later. Serum neutralization tests conducted by microtiter procedure on 2970 serum samples from calves, heifers and adult cows from representative regions of Italy indicate that rotaviruses, and the infection which they cause, existed in Italy long before the virus was isolated. Non-typhoid Salmonella serovars remain a potential threat to human health, and beef cattle and broiler chickens are possible sources of these organisms on Prince Edward Island (PEI). In this study, the ceca of beef cattle belonging to fasted and non-fasted groups, and broiler chickens were examined for Salmonella at the time of slaughter. The characteristics of the isolates, including antimicrobial resistance patterns and virulence genes, were studied along with the isolates obtained from cases of human salmonellosis on PEI during the study period (1996–97). The prevalence of Salmonella in beef cattle was 4.6% (11/240). The rate was significantly higher in fasted cattle (7.46%), than in non-fasted cattle (0.94%). The prevalence rate in chickens was 32.5% (39/120). In beef cattle, Salmonella typhimurium phage type (PT) or definitive type (DT) 104 which was resistant to ampicillin, chloramphenicol, streptomycin, sulfisoxazole and tetracycline, was the most predominant type (64%). In chickens, S. heidelberg, with resistance to gentamicin, streptomycin and sulfisoxazole, predominated. Of 26 isolates from humans, the most common serovar was S. typhimurium, including a multidrug-resistant strain of DT104. Examination by PCR revealed presence of the virulence gene invA in all serovars, and the spvC gene in all S. typhimurium isolates, of both beef cattle and human origin. Among the other serovars the latter gene was found in 7 human isolates, but in none of the chicken or beef isolates. All but 3 of the spvC-positive isolates possessed a 90 kilobasepair (kbp) plasmid suggesting that the 3 isolates had the spvC gene on their chromosome. These findings were confirmed by plasmid DNA isolation using 3 different protocols and by sequence analysis of the spvC-PCR product. 相似文献

The association between sole ulcer found at routine claw trimming 2–6 months after calving and reproductive, health and productive traits in the same lactation, was investigated in an observational study of 2368 dairy cows in 102 Swedish herds. The data were analysed by mixed multivariable linear-, logistic- and Poisson-regression modelling at the cow level, accounting for clustering within herd. Associations were found between sole ulcer and first-service conception risk in the first study year (OR 0.59), calving interval (2% longer), treatment for anoestrus (OR 1.61), and 305-day milk yield (479 kg ECM higher). No associations could be shown between sole ulcer and first-service conception risk in the second study year, the number of services per conception, clinical mastitis, high milk somatic-cell counts or culling. 相似文献

The development of bacteriological systematics during the latest three decades makes us also revaluate the data reported on the finding of certain microbial species in farm and laboratory animals. These experiments were upon the occurrence of pasteurellae in the nasal or nasopharyngeal mucus, the samples of which came from farms showing different epizootic situations.The biochemical analyses of suspected strains obtained by the direct cultivation of samples on blood agar yielded the following results: 1. 1. Pasteurellae occurred with different frequency in all of the animal species and their age groups independently of the health condition on the particular farm. 2. 2. The total findings suggest the specific occurrence of pasteurellae according to animal hosts, namely: in cattle, pigs and rabbits P. multocida; in domestic fowl P. multocida and P. gallinarum; in albino rats and mice P. pneumotropica and P. ureae; and in guinea-pigs P. ureae. 3. 3. On infested farms, significantly higher frequency of pasteurellae was observed in calves, runners, piglets and hens (P. multocida, in hens also P. gallinarum), while in sows and rabbits the findings varied at about the same level as on healthy farms. 4. 4. The average frequency of pasteurellae in individual host species can be expressed as the percentage of bacilli carriers, but it should be pointed out that these largely randomized examinations also covered farms where the results were invariably negative. 5. 5. The relation of carrier strains to infections by the pasteurellae is discussed and finally the chances of raising biologically healthy animals are considered.
RésuméLe développement de la systématique bactériologique dans les trois dernières décennies oblige à revoir les données concernant la présence de plusieurs espèces microbiennes chez les animaux domestiques et de laboratoire. Au cours de ces expériences, nous avons recherché la présence des pasteurellae dans les secrétions nasales et du nasopharynx, dans des fermes ayant des situations epizootiques différentes.En examinant, à l'aide des tests biochimiques actuels, les souches suspectes obtenues par la culture directe des échantillons sur gélose au sang, nous avons obtenu les résultats suivants: 1. 1. Nous avons trouvé des pasteurellae, à des fréquences différentes, chez toutes les espèces et chez tous les groupes d'âge des animaux observés, sans correlation avec la situation sanitaire de la ferme. 2. 2. De ces examens généraux, nous pouvons conclure qu'il existe une relation entre l'hôte animal et les espèces microbiennes identifiées; ches les bovins, les porcins et les lapins, P. multocida; chez les gallinacés domestiques, P. multocida et P. gallinarum; chez les rats et les souris, P. pneumotropica et P. ureae; et chez les cobayes, P. ureae. 3. 3. Dans les fermes infectées, la fréquence des isolements de pasteurellea était significativement plus grande chez les veaux, les porcs, les porcelets et les poules (P. multocida avec, chez les poules, P. gallinarum), tandis que chez les truies et les lapins la fréquence constatée était à peu près du même niveau que celui des fermes saines. 4. 4. La fréquence moyenne des pasteurellae chez les différentes espèces d'hôtes peut être exprimée par le pourcentage des porteurs de ces germes, mais il faut encore mentionner qu'il y avait aussi des fermes où les résultats furent complètement négatifs, les examens ayant été faits au hasard en général. 5. 5. On discute des relations entre les souches isolées des porteurs et les infections causées par les pasteurellae ainsi que de la possibilité d'élever les animaux sains du point de vue biologique.

The number of loci encoding an enzyme system, tissue specificity of gene expression and the degree of gene expression in various tissues are often not mentioned in evolutionary studies, but could indirectly provide evidence of evolutionary relationships. Protein electrophoresis was used to study the distributions and tissue specificity of gene expression of enzymes encoded by 42 loci in Rhinolophus clivosus and R. landeri, the genetically most divergent of the ten species of southern African horseshoe bats. No differences in gene expression were found between R. clivosus and R. landeri and isozyme patterns may be compared with other bat species to derive possible phylogenetic relationships. 相似文献

Even after using the first arms against tumour (surgery and radiotherapy) 70% of solid tumours reappear. Chemotherapy cannot eliminate ‘the last cell’. Specific and/or non specific immunotherapy helps the organism to eliminate the last cells by stimulating its immune capacity. The different generations of adjuvants, agents of immunotherapy were studied successively. BCG belongs to the first generation of adjuvants. We considered its association with specific immunotherapy (of irradiated tumour cells) and/or chemotherapy (cyclophosphamide), then we analysed the problems of dose and the ways of administration, advantages and disadvantages (stimulation of suppressor cells). Second generation of adjuvant (Corynebacterium parvum, nucleotids and levamisole) allowed to specify the parameters of the efficiency of adjuvants. They have the same disadvantages as BCG. Finally the third generation share the following characteristics: their side-effects will be minimal (they will not induce suppressor cells), their action will be limited to stimulate one population of cells only and they will be well defined chemical compounds. Thymosin, Bestatin and MDP belong to this generation. Combinations of adjuvants may also be envisaged with the objective of stimulating a particular mechanism of antitumoural activity. 相似文献

Humoral and cell-mediated immunity (CMI) to herpesvirus saimiri (HVS), an oncogenic lymphotropic herpesvirus, was studied in squirrel and rhesus monkeys. Natural antibody to HVS was found in five of six squirrel monkeys but there was no evidence of specific CMI directed against HVS. Rhesus monkeys did not show natural antibody or CMI against HVS antigens. Immunization with HVS, however, produced both antibody and specific CMI in the rhesus monkeys, but no CMI developed in the squirrel monkeys. These findings are important in the development of animal models for the treatment of tumors associated with lymphotropic herpesviruses. 相似文献

Vaginal mucus during estrus was examined with the Metricheck device and the relationship to the reproduction of high‐yielding dairy cows was studied. The study was conducted in 99 dairy herds located in Western Germany and 1348 Holstein‐Friesian heifers and cows showing spontaneous estrus were examined. Independent of the Metricheck result, the animals were inspected by professional insemination technicians and those suitable for insemination ( n = 989) were bred by artificial insemination (AI). Reproductive performance was characterized by non‐return rate at 90 days (NRR90). The discharge of the animals predominantly had a clear appearance (70%) and a stringy consistency (80%). Animals with clear vaginal discharge had higher NRR90 (56%; n = 697) than animals with abnormal (turbid, mucopurulent, purulent, sanguineous) vaginal secretion (48%, n = 292; P < 0.05). NRR90 was lower in animals with short calving to AI interval (< 70 days; 39%) than with medium (70–130 days; 54%) or long (> 130 days; 62%) intervals ( P < 0.05). NRR90 decreased by 12% from the lowest (< 15 kg) to the highest (> 45 kg) milk yield class. In conclusion, the use of the Metricheck device integrated into the insemination procedure is recommended to identify dairy cows suffering severely from uterine disease. 相似文献

Le test de Habel utilisé dans le monde entier a rendu d'inestimables services. Il continuera à en rendre longtemps encore puisqu'il permet de caractériser de façon simple et rapide un très bon ou un très mauvais vaccin. Grâce à une interprétation critique et raisonnée des résultats obtenus il est possible de distinguer des vaccins de valeur moyenne ou médiocre mais, dans ce cas, l'imprécision de ce test est grande, ce qui pose à l'expérimentateur des problèmes évidents. En raison, d'une part de la nécessité d'utiliser un test aussi simple que le test de Habel, mais plus fiable, d'interprétation plus aisée et répondant mieux aux conditions d'utilisation des vaccins préventifs sur le terrain, et d'autre part, de la difficulté à généraliser actuellement tout au moins un test qui implique l'emploi d'un vaccin de référence, il nous semble possible et souhaitable de définir dès maintenant, en tenant compte des travaux antérieurs sur cette question, une technique qui réponde à ces impératifs. 相似文献
