beta-Carotene is a naturally occurring carotenoid reported to have health-promoting effects in several species. Advancing age is known to have a negative impact on various immune variables in several species. This study was conducted in order to assess the effect of age on immune response in dogs and to determine whether beta-carotene is able to reverse this age-associated decline. To test this hypothesis, young and old dogs (n = 36) were fed either a control diet or experimental diets containing supplemental beta-carotene for 2-month periods. Age significantly (P < .05) lowered CD4+ T cell populations (47.2% versus 33.7%; young-control versus old-control, respectively) and beta-carotene restored percent distributions in old dogs to nonsignificance versus younger controls (41.0%). T cell proliferation was lower in old dogs (30,254 +/- 2,248 versus 14,811 +/- 2,497 cCPM; young-control versus old-control, respectively; P < .05), and beta-carotene supplementation significantly improved responses in this age group (21,329 +/- 2,275 cCPM). Although B cell proliferation was depressed with age (17,967 +/- 1,384 versus 7,535 +/- 1,469 cCPM; young-control versus old-control, respectively; P < .05), beta-carotene supplementation improved B cell proliferation in young dogs (23,500 +/- 1,339 cCPM). Old dogs displayed lower delayed-type hypersensitivity test (DTH) responses versus younger controls to both phytohemagglutinin-P (PHA; 11.1 +/- 0.95 versus 7.57 +/- 1.15 mm; young-control versus old-control, respectively; P < .05) and sheep red blood cell (RBC; 9.12 +/- 0.62 versus 8.08 +/- 0.75 mm; young-control versus old-control, respectively; P < .10). beta-Carotene improved these responses, mostly within the first 24-48 hours after injection. In summary, older dogs have lower immunological responses compared with younger controls. beta-Carotene supplementation significantly restored immune responses in older dogs when compared with their age-matched controls and younger counterparts.  相似文献

BACKGROUND: Obesity is a risk factor for cardiovascular disease in people, but overweight and obese human heart failure patients have improved survival compared with normal--or underweight controls--the obesity paradox. The purpose of this study was to determine if there is an association of body weight and body condition with survival in dogs with heart failure. HYPOTHESIS: That body condition and changes in body weight are predictors of survival in dogs with heart failure. ANIMALS: One hundred and eight dogs with heart failure (International Small Animal Cardiac Health Council stages 2, 3a, or 3b) secondary to dilated cardiomyopathy or chronic valvular disease. METHODS: Medical records were reviewed, and data regarding initial body weight and body condition score (BCS), subsequent changes in body weight, and treatment were collected. Survival times were determined for dogs that were discharged from the hospital and lived >24 hours. RESULTS: Survival was significantly different between dogs that gained, lost, or maintained body weight over the course of their disease (P= .04), with dogs that gained weight surviving the longest. BCS and medications were not significantly associated with survival time; however, n-3 fatty acid intake was associated with longer survival time (P= .009). CONCLUSIONS AND CLINICAL IMPORTANCE: These results suggest that changes in body weight might be an important consideration in the survival of dogs with heart failure.  相似文献

There are limited data on glucocorticoid treatment in dogs. The purpose of this study was to investigate whether dogs of higher body weight experienced more adverse events when receiving glucocorticoid therapy. Data pertaining to glucocorticoid therapy was abstracted from the records of 61 dogs that were prescribed glucocorticoids for treatment of immune-mediated thrombocytopenia or hemolytic anemia from 2014 to 2019. The odds of developing muscle atrophy and polyphagia during therapy were increased by 30% for each 5 kg of additional body weight. Almost half of the dogs (44.3%) fluctuated > 15% from baseline weight during therapy. Dogs whose body condition scored as above ideal were at increased risk (odds ratio = 4.2) for being diagnosed with urinary tract infection. Our findings suggest that standard linear glucocorticoid dosing may place higher body weight dogs at increased risk of developing adverse events. Accelerated glucocorticoid tapering and/or alternative dosing schemes in dogs with higher body weights may be prudent in efforts to improve tolerance and client compliance.  相似文献

Prior to starting a weight loss programme, target weight (TW) is often estimated, using starting body condition score (BCS). The current study assessed how well such estimates perform in clinical practice. Information on body weight, BCS and body composition was assessed before and after weight loss in 28 obese, client-owned dogs. Median decrease in starting weight per BCS unit was 10% (5–15%), with no significant difference between dogs losing moderate (1–2 BCS points) or marked (3–4 BCS points) amounts of weight (P = 0.627). Mean decrease in body fat per BCS unit change was 5% (3–9%). A model based on a change of 10% of starting weight per unit of BCS above ideal (5/9) most closely estimated actual TW, but marked variability was seen. Therefore, although such calculations may provide a guide to final TW in obese dogs, they can either over- or under-estimate the appropriate end point of weight loss.  相似文献

OBJECTIVE: To evaluate the effect of age and body weight on several neurohumoral variables that are commonly altered in heart failure in Cavalier King Charles Spaniels. ANIMALS: 17 healthy privately owned Cavalier King Charles Spaniels, 10 males and 7 females, ranging in age from 0.4 to 9.7 years, and ranging in body weight from 6.6 to 12.2 kg. PROCEDURE: The clinical condition of the dogs was evaluated by physical examination, thoracic radiography, and echocardiography. Plasma nitrate and nitrite (P-NN), N-terminal atrial natriuretic and brain natriuretic peptides (NT-ANP and BNP, respectively), endothelin (ET-1), urine cyclic guanosine monophosphate (U-cGMP), and urine nitrate and nitrite (U-NN) concentrations were analyzed. RESULTS: Plasma concentrations of NT-ANP and P-NN increased significantly with age, but plasma NT-ANP and P-NN also correlated significantly, irrespective of age. A modest increase of left atrial size did not explain the increase of NT-ANP and P-NN with age. Concentration of ET-1 correlated positively with heart rate; heart rate did not change with age. Weight had a negative impact on NT-ANP, P-NN, and U-cGMP concentrations and left atrial relative size. CONCLUSIONS AND CLINICAL RELEVANCE: Age-matched controls are essential for evaluation of NT-ANP and P-NN concentrations and left atrial size. Weight may alter reference values of plasma NT-ANP, P-NN, and urine cGMP concentrations. Natriuretic peptides can be used as further evidence that heart failure exists. The increased plasma concentrations of NT-ANP (but not BNP) and P-NN with aging reflect neurohumoral physiologic changes that must be distinguished from pathologic changes in patients with heart failure.  相似文献

Cancer cachexia is a well-recognized syndrome in human patients that is characterized by progressive involuntary weight loss. The prevalence of this syndrome in veterinary cancer patients is unknown. This study's objective was to investigate the occurrence of weight loss and cachexia, as characterized by body condition scoring, in dogs presented to a veterinary oncology service. Information collected on 100 dogs included signalment, diagnosis, weight at time of diagnosis, and, when available, weight from a time approximately 12 months before diagnosis. Body condition was assessed by using a 9-point system based on body silhouette and palpation of adipose tissue (4-5 = optimal, 1 = extreme cachexia, 9 = extreme obesity). Muscle wasting was scored based on palpation of skeletal muscle (3 = no wasting, 2 = mild, 1 = moderate, 0 = severe). Only 4% of the dogs exhibited cachexia as defined by a body condition score < or = 3, whereas 29% were classified as markedly overweight (> or = 7). Fifteen percent had evidence of clinically relevant muscle wasting (< or = 1). Body weight from a time before the diagnosis of cancer was available for 64 dogs. At the time of diagnosis, 31% had maintained or gained weight, 31% had lost up to 5%, 14% had lost between 5 and 10%, and 23% had lost >10% of body weight. Overall, the percentage of dogs with signs indicating a decline in nutritional status was less than what has been reported for human cancer patients. Future studies should investigate the extent to which weight loss occurs in canine patients on an appropriate plane of nutrition as well as to establish whether an association exists between poor nutritional status and outcome in canine cancer patients.  相似文献

Brain stem auditory-evoked responses were recorded in 9 male and 11 female clinically normal mature dogs, weighing between 2 and 36 kg. Mean wave latency for the entire group of dogs, using 60-dB hearing level click stimuli at 11/s for waves I to VII was: 1.41, 2.21, 2.85, 3.31, 3.71, 5.12, and 6.46 ms, respectively. The mean interpeak latency for waves I and V (IPLIV) was 2.32 ms. Neither gender nor ear effect was detectable. Positive correlation was observed between cranium length, cranium width, nasion-external auditory meatus interval, and body weight for wave-V latency and IPLIV. Such correlation was not documented for wave I. The regression equations for their effects on IPLIV were: cranium length, y = 0.05x + 1.85; cranium width, y = 0.07x + 1.32; nasion-external auditory meatus interval, y = 0.05x + 1.79; and body weight, y = 0.01x + 2.15. On the basis of any of the 3 variables of cranium size or body weight, the study population could be classified into groups of large and small dogs, with the large group having significantly (P less than 0.05) longer latency for wave V and IPLIV. It is recommended that the effect of size variation in dogs on brain stem auditory-evoked responses should be compensated for by use of the regression equation based on cranium length.  相似文献

OBJECTIVE: To evaluate the effect of carprofen on hemostatic variables in clinically normal dogs. ANIMALS: 12 clinically normal Labrador Retrievers. PROCEDURE: 10 dogs (6 females, 4 males) received carprofen (2.2 mg/kg of body weight, PO, q 12 h) for 5 days. Two dogs (untreated control group; 1 female, 1 male) did not receive carprofen. Hemostatic variables (platelet count, activated partial thromboplastin time, prothrombin time, fibrinogen, platelet aggregation, and bleeding time) were assessed for all dogs prior to treatment, on day 5 of treatment, and 2 and 7 days after discontinuation of the drug (days 7 and 12). Serum biochemical variables and Hct were assessed prior to treatment and on days 5 and 12. RESULTS: In dogs receiving carprofen, platelet aggregation was significantly decreased, and onset of aggregation was significantly delayed on days 5, 7, and 12, compared with pretreatment values. Activated partial thromboplastin time was significantly increased on days 5, 7, and 12 over pretreatment values in treated dogs, but values remained within reference ranges. Significant differences were not detected in buccal mucosal bleeding time, other serum biochemical and hemostatic variables, or Hct, compared with pretreatment values and the internal control group. CONCLUSIONS AND CLINICAL RELEVANCE: Administration of carprofen for 5 days causes minor but not clinically important alterations in hemostatic and serum biochemical variables in clinically normal Labrador Retrievers. Carprofen is commonly used to treat osteoarthritis and chronic pain in dogs, but prior to this study, its effect on platelet aggregation and hemostatic variables was unknown.  相似文献

A significant (P less than 0.0001) positive correlation was demonstrated between left ventricular internal chamber dimension in diastole or systole and body weight, body surface area, cycle length, and the square root of cycle length. On the basis of adjusted coefficients of determination, multiple regression analysis, using body weight or body surface area and cycle length or the square root of cycle length, was superior to separate simple regression with these variables in accounting for variations in left ventricular internal chamber dimensions. Shortening fraction had a significant (P less than 0.0001) negative correlation and left ventricular free wall measurements had a significant (P less than 0.0001) positive correlation to body weight and body surface area. For these echocardiographic variables, correlation to the square root of cycle length was insignificant (P greater than 0.05), and a multiple regression model was not helpful in developing confidence intervals. Septal wall measurements were not correlated with body weight, body surface area, cycle length, or the square root of cycle length.  相似文献

The clinical efficacy for weight loss and safety of dirlotapide in dogs were evaluated in two multi-centre studies with parallel designs. Overweight, adult dogs ( n  = 245) of various breeds were randomized to treatment with dirlotapide or placebo in a 2:1 ratio. Dirlotapide was administered orally once daily to dogs at an initial dose of 0.05 mg/kg/day commencing on day 0 and doubled after 14 days. Every 28 days, dogs were examined, weighed, body condition scores (BCS) were recorded, and dose was adjusted to meet weight loss targets. Each study comprised three consecutive phases: weight-loss (up to day 196); weight-stabilization (84 days); and post-treatment (28 days). pre-treatment feeding and exercise regimens were continued during treatment. Dirlotapide-treated dogs showed mean weight loss of 15.9% (study A) and 14.0% (study B) by the end of weight loss phase (up to day 196). Percentage weekly weight losses for dirlotapide were significantly greater than for placebo ( P  ≤ 0.0002). Emesis and diarrhoea were experienced in both treatments but were more frequent with dirlotapide; resolution was spontaneous. BCS improved for 75.7–82.5% of dogs on dirlotapide treatment compared with 15.4–41.4% for placebo. Mean dirlotapide dosage at end of weight-loss phase was 0.38 (study A) and 0.29 (study B) mg/kg initial body weight/day. Dirlotapide was found to be clinically safe and effective in the reduction of body weight in overweight dogs.  相似文献

Height measurements of the mandible and the mandibular first molar tooth were obtained from 234 dogs. Statistical analysis was performed to determine possible associations between these measurements and body weight, sex, age, and body condition. There was a statistically significant association between the mandible: mandibular first molar tooth height measurement ratio and body weight (p < 0.0001) in all body weight groups. Small dogs had proportionally larger mandibular first molar teeth relative to mandibular height compared with larger dogs. This relationship may contribute to an increased susceptibility in small dogs for periodontitis and subsequent tooth loss. There was no statistically significant association between the mandible: mandibular first molar tooth height measurement ratio and other parameters evaluated in this study.  相似文献

Circulating antibody titers (1:20 to 1:2560) against thyroglobulin were demonstrated in 48% of pet dogs with hypothyroidism by the chromic chloride passive hemagglutination test. Four of six dogs with acanthosis nigricans (1:20) and one of six male dogs with hyperestrogenism (1:40) had low titers of antibody against thyroglobulin whereas clinically normal pet dogs and dogs with other selected endocrinopathies (hypoadrenocorticism, cortisol-excess, diabetes mellitus) or obesity were consistently negative. Circulating immune complexes evaluated by the mastocytoma cell-assay were present in the sera of 20% of pet dogs with hypothyroidism but were absent in clinically normal dogs. Although variations in dose significantly altered the quantitative response of the thyroid gland to thyrotropin the qualitative pattern of response was similar for T3 but not T4 in clinically normal laboratory beagles. The peak increases for serum triiodothyronine and thyroxine were observed either at eight (0.1 and 0.2 I.U bTSH/5 lbs) or 12 (1.0 I.U. bTYSH/5 lbs) hours postthyrotropin. Dogs with naturally occurring hypothyroidism had a decreased serum T3 and T4 at baseline and eight hours postthyrotropin (1.0 I.U. bTSH/5 lbs) compared to clinically normal pet dogs, laboratory beagles and dogs with other clinical endocrinopathies. The consistent lack of a significant increase of serum T3 and T4 in response to thyrotropin was necessary for the separation of certain hypothyroid from euthyroid pet dogs in which the baseline level of thyroid hormones were equivocal.  相似文献

Improving survival is a continuous objective in swine breeding. The aim of this study was to record 22 blood variables and BW gain on the first day of life in Landrace-Yorkshire-Duroc crossbred piglets and to find associations between these variables and survival at weaning. All live piglets from 18 litters were weighed and blood sampled at birth and on d 1 and were monitored to weaning at the age of 5 wk. A total of 261 piglets were born, of which 8.8% were stillborn. Additionally, 15.1% died before weaning. The blood variables glucose, immunoglobulins, and white blood cells increased from birth to d 1 (P < 0.001), whereas α(1)- and β(1)-globulin, red blood cells, hemoglobin, and hematocrit decreased (P < 0.001). At birth, concentrations of lactate (P = 0.004), pH (P = 0.007), red blood cells (P = 0.017), hemoglobin (P = 0.018), and hematocrit (P = 0.052) were associated with survival to weaning. Also, concentrations of lactate increased (P = 0.030) and pH decreased (P < 0.001) when piglets were born in the last third of a litter. On d 1, concentrations of glucose (P = 0.015), hemoglobin (P = 0.025), and BW gain (P = 0.001) were all decreased in piglets that did not survive to weaning. Body weight gain also decreased (P = 0.005) when piglets were born in the last third of a litter. Concentrations of IgG on d 1 was not associated with survival at weaning (P = 0.230) but decreased (P < 0.001) when piglets were born in the last third of a litter. We conclude that several blood variables recorded at birth and on d 1 and BW gain on d 1 were highly associated with survival at weaning and that piglets born in the last third of the litter had less favorable vitality.  相似文献

Body weight, heart rate, and 19 M-mode echocardiographic variables were measured in 41 nonanesthetized healthy cats. Estimated limits were determined for the echocardiographic variables, and each variable was then correlated to body weight, heart rate, and the 18 other variables. A significant (P less than 0.05) positive correlation to body weight was found with aortic diameter, left atrial dimension, septal and left ventricular systolic and left ventricular diastolic wall thicknesses, and left and right ventricular diastolic and right ventricular systolic internal dimensions. Significant inverse correlation (P less than 0.05) to heart rate was found with body weight, left ventricular systolic and diastolic and right ventricular systolic internal dimensions, left atrial dimension, left atrial dimension to aortic ratio, mitral valve E point to ventricular septal separation, and left ventricular ejection time. Left ventricular shortening fraction in the short axis and velocity of circumferential fiber shortening were significantly correlated (P less than 0.05) to heart rate. Significant correlation (P less than 0.05) was also found between many echocardiographic variables.  相似文献

Objective: To characterize the relationship between clinical estimates of hydration in dogs and cats admitted to an intensive care unit (ICU) and changes in their body weight following 24–48 hours of fluid therapy. Design: Outcome study. Setting: ICU at a veterinary teaching hospital (VTH). Animals: A total of 151 dogs and 42 cats with various medical disorders that had not had surgery within 48 hours of admission into the ICU were consecutively admitted into the study. Animals with any condition predisposing to excess fluid loss or retention were excluded: heart disease, sepsis, trauma, pancreatitis, pleural or pericardial effusion, ascites, and pathologic oliguria. Animals that acquired any of the following during the observation period were excluded: gastrointestinal fluid loss, edema or diseases predisposing to edema, oliguria, diuretic therapy, and body fluid drainage or hemorrhage. Fluid therapy was ordered based on estimate of hydration at admission. Other treatments were not modified or withheld. Interventions: Physiologic data were collected at the time of admission and 24–48 hours later. Measurements and main results: Hydration was estimated on admission to the ICU using clinical judgement with no supporting laboratory data. Each admitting clinician used this estimate to plan fluid therapy. Fluid therapy was defined as the administration of any enteral or parenteral fluids as well as any decision to withhold fluids. Paired measurements taken on admission and at 24–48 hours included packed cell volume (PCV), total plasma solids (TS), and body weight. Amount and type of fluids or blood products administered were noted. Neither clinician estimates of dehydration nor baseline PCV or TS predicted clinically significant changes in body weight following fluid therapy, and there was no relationship between weight change and changes in PCV or TS. Conclusions: A clinical diagnosis of dehydration in our ICU does not predict weight gain following fluid therapy. Neither baseline PCV/TS nor changes in these measurements following 24–48 hours of fluid therapy predicted changes in body weight.  相似文献
