Twelve populations of Girella punctata, from widespread locations of the species’ range in Japan and Korea, were screened for sequence variability within the mitochondrial DNA (mtDNA) control region (n = 128) and at five polymorphic microsatellite loci (n = 547) to determine the genetic structure maintaining population integrity. mtDNA variability of 132 variable sites within a 334-bp region reveals shallow genetic differentiation across populations. The weak differentiation of G. punctata was partly supported by the screening of five polymorphic microsatellite loci. However, hierarchical analysis of molecular variance and principal component analysis on the basis of allele frequencies in microsatellite loci extracted a subtle substructure in a subsidiary population and in near-subsidiary populations in the semi-enclosed Seto Inland Sea.  相似文献

The genetic structure of two native Silurus aristotelis (one of Europe's oldest surviving freshwater fish species) populations was investigated using eight microsatellite loci. Average values of population heterozygosity were very high, even approaching values reported for marine fishes, possibly due to stable population sizes and the prolonged undisturbed conditions prevailing in the lakes that represent the native distribution of S. aristotelis. The success of an attempt to introduce the species into a new environment was also evaluated. No loss of genetic variability in the introduced population was detected. Additionally, assignment tests and trees based on genetic distances among individuals indicated the low differentiation of the translocated population from its donor population, and, differentiated the two autochthonous populations successfully. Comparisons with previous allozyme and mitochondrial RFLP studies were also made and showed that results on relative levels of genetic variability among populations were in good agreement among all methods. However, microsatellite analysis exhibited higher power of statistical tests for differentiation among population samples compared to allozymes.  相似文献

云斑尖塘鳢微卫星分子标记的筛选与检测   总被引:1,自引:0,他引:1  
用磁珠富集法分离云斑尖塘鳢的微卫星序列,由所获得的1032个克隆中筛选出146个阳性克隆,经测序81个含有微卫星序列,52个为完美型,27个为非完美型,2个为复合型,其中43个微卫星序列重复次数在10以上。所获得的云斑尖塘鳢微卫星序列中除探针中使用的CA重复单元和GA重复单元外,还有TAC等其他类型的重复单元。设计合成38对微卫星引物,其中29对引物可稳定扩增出条带,使用这些引物对云斑尖塘鳢48个个体进行检测显示:观测杂合度平均值为0.63,期望杂合度平均值为0.43。29对引物中1对引物表现为单态,7对表现为高度多态,14对表现为中度多态,7对表现为低度多态,多态性较为丰富,说明本研究开发的绝大部分微卫星分子标记较为理想。  相似文献

Nibea albiflora is a commercially important fish species in China. Herein we report 16 novel polymorphic microsatellite markers in Nibea albiflora by using the 5′ anchored polymerase chain reaction (PCR) technique. The characteristics of these loci were estimated by using a sample of 30 individuals. A total of 79 alleles were detected with an average of 4.9 alleles per locus. The number of alleles per locus ranged from three to nine. The polymorphism information content (PIC) values for the 16 microsatellite loci ranged from 0.3131 to 0.7910. The observed and expected heterozygosity per locus ranged from 0.2333 to 1.000 and from 0.3452 to 0.8421, with an average of 0.7248 and 0.6592, respectively. Four loci significantly deviated from Hardy–Weinberg equilibrium after Bonferroni correction (P < 0.0031), and no significant linkage disequilibrium between pairs of loci was found. This study provides useful information for studies on genetic diversity and structure, construction of genetic linkage maps of N. albiflora, and effective management of this fish resource.  相似文献

采用磁珠富集和PCR筛选相结合的方法,得到番红砗磲的19个多态性微卫星标记。利用新开发微卫星标记对西沙群岛2个野生群体的遗传多样性进行比较分析,七连屿海域野生群体和永兴岛海域野生群体的平均观测等位基因数(Na)分别为11.105、11.895,平均有效等位基因数(Ne)分别为6.274、6.173,平均期望杂合度(He)分别为0.776、0.788,平均多态性信息含量分别为0.730、0.744,发现2个野生群体的遗传多样性都处于高度多态水平,说明其有效群体大小仍然保持在较高水平。Bonferroni校正后,在2个群体中各有4个位点偏离Hardy-Weinberg平衡。另外分析了这些引物在近缘种中的通用性情况,发现鳞砗磲中有7对微卫星引物具有通用性,6对具有多态性;无鳞砗磲中有3对微卫星引物具有通用性,1对具有多态性;诺瓦砗磲中有5对微卫星引物具有通用性,5对具有多态性;长砗磲中有9对微卫星引物具有通用性,8对具有多态性;砗蚝中有2对引物具有通用性,2对具有多态性。  相似文献

采用磁珠富集法筛选适合漠斑牙鲆遗传多样性分析的微卫星分子标记。筛选共获得43条序列,其中完美型26个,占60.5%;非完美型14个,占32.6%;复合型3个,占6.9%。选取其中14对特异性好且扩增效率高的微卫星引物,对采自美国北卡罗来纳州沿海的漠斑牙鲆野生群体和养殖群体进行遗传多样性及遗传结构比较分析。研究结果表明,12对引物的扩增产物具有多态性,其中7个位点为高度多态(PIC>0.5)。两个群体中共检测到90个等位基因。12个多态性微卫星位点在两个群体中的平均观察杂合度(Ho)和期望杂合度(He)分别为0.36和0.57。9个位点在整个群体中呈现出不同程度的偏离遗传平衡(P<0.05),且偏离平衡的位点均表现为杂合子缺失(Fis>0)。野生群体和养殖群体间的遗传距离为0.1115,群体间的遗传分化微弱(Fst=0.0438)。  相似文献

利用微卫星标记对文蛤4个壳色花纹品系的遗传分析   总被引:3,自引:1,他引:2  
利用9个微卫星位点对文蛤红壳(RS)、黑斑(BS)、细纹(TC)、暗纹(DF)4个壳色花纹品系共120个个体进行遗传差异分析。结果共检测到105个等位基因,每个位点平均观察等位基因数为11.7个,其中WG07位点等位基因数最多(21个),WG11位点最少(7个)。每个品系均具有特异等位基因类型,不同品系中相同微卫星位点的等位基因分布规律不同。4个文蛤品系的观测杂合度、期望杂合度和多态信息含量变异范围分别为0.110~1.000、0.486~0.867、0.433~0.834,均属高度多态性位点。Hardy-Weinberg平衡的卡方检测发现,大多数位点偏离平衡状态;聚类分析表明,4个品系间存在较明显的遗传差异,TC品系与其它品系的遗传距离最大,DF品系和RS品系的遗传距离相对较小。  相似文献

为研究野生琼枝与养殖琼枝的遗传差异,利用EST-SSR技术分析了海南东北部地区9株野生和9株养殖琼枝的遗传差异性。结果显示,用5对EST-SSR引物对18个个体的基因组进行扩增,扩增出的片段大小为250~2 500 bp,琼枝野生群体和养殖群体的多态性比例分别为74.29%和70.00%;野生群体之间的平均遗传距离为0.46,养殖琼枝群体之间的平均遗传距离为0.22。聚类分析图显示,在相似系数0.49处,18株群体按野生与养殖分为2大类,在相似系数0.66附近,野生群体分成3类,养殖琼枝群体分为2类,在相似系数0.73附近,野生琼枝群体分成5类,而养殖群体为3类,相似系数越高,野生群体比养殖群体分类单元越多。研究表明,野生琼枝群体的遗传多样性比养殖琼枝群体高。  相似文献

The aim of this study was to compare, levels of genetic polymorphism between Iranian and Azeri bream (Abramis brama orientalis) populations using five microsatellite loci. The reduced number of alleles in the Iranian populations compared with the Azeri population led us to infer that the genetic variability of the Iranian stocks has been reduced due to inbreeding and genetic drift. Significant population differentiation (Fst) was observed between the Azeri and the Iranian populations, which could be explained by the higher number of alleles in the Azeri population. Significant deviations from Hardy–Weinberg equilibrium were found at more loci in the Iranian populations than the Azeri population. Beyond the hypothesis of null alleles, the heterozygote deficiency may have arisen due to breeding between related individuals in the Iranian populations. This investigation represents the first approach to the knowledge of the genetic variability of Iranian and Azeri bream populations using microsatellite markers, and the reported results could be of interest for management and conservation programmes of this species in Iran.  相似文献

三角帆蚌(Hyriopsis cumingii)是我国特有种,它形成的珍珠具有珠质光滑细腻、色泽鲜艳等方面的优点,是淡水蚌中育珠质量最佳者,已成为最主要的淡水养殖珍珠蚌。本研究选取已发表的32对太平洋牡蛎的微卫星引物在三角帆蚌基因组DNA中进行PCR扩增,探讨太平洋牡蛎的引物用于三角帆蚌基因组微卫星分析的可能性。通过优化PCR反应条件,筛选13对引物可在三角帆蚌基因组中扩增出特异性条带(占总数的4l%);其中10对引物(占总数的3l%)在洞庭湖三角帆蚌小群体中即检测到了个体间等位基因的多态性,共出现40条多态性条带。10个位点杂合度大小在0.125到0.693之间,其中7个微卫星位点(Cgi-10,Cgi-22,Cgi-24,Cgi-26。Cgi-29,Cgi-30,Cgi-32)为高度多态性位点,杂合度大于0.500,而其余3个微卫星位点(Cgi-1,Cgi-18 and Cgi-25)杂合度小于0.500,为低度多态性位点。初步的结果表明,部分太平洋牡蛎的微卫星引物可以用于三角帆蚌基因组的分析,这为其它贝类微卫星标记的开发奠定了基础。  相似文献

泥蚶34个EST-SSR标记的开发及在格粗饰蚶中的通用性检测   总被引:1,自引:0,他引:1  
利用泥蚶转录组高通量测序、拼接获得的大量EST序列开发SSR标记,在1123条EST序列里筛查到73条含有SSR位点的EST序列,其中54个位点适合设计引物,在位点两侧设计引物并进行PCR扩增.结果显示,46对引物获得稳定扩增的位点,引物在泥蚶奉化群体的多态性检测中发现,有34对引物表现出多态性,共扩增出122个等位基因,各位点的等位基因数Na为2~7个,平均每个位点产生3.59个等位基因,观测杂合度Ho、期望杂合度He、多态信息含量PIC范围分别为0.000 ~ 0.600、0.078 ~0.771、0.106~0.718;Hardy-Weinberg平衡检测显示,13个位点偏离了平衡状态;用Nr和Swiss-Prot蛋白质数据库对含有多态性SSR的EST进行了基因注释,25个SSR位点来自注释基因序列.将34对泥蚶多态性SSR引物在格粗饰蚶中进行了通用性检测,结果有1 1对成功扩增,8对表现为多态,通用率为23.53%,这些通用引物可用于两种蚶的遗传多样性评价、系统进化分析、比较作图和基因发掘等研究.  相似文献

The objective of the present work was to develop species-specific microsatellite markers for P. scalare and to analyze the diversity and genetic structure of a wild population, from the Amazon River, and three commercial stocks (common, marble, and clown morphological variants), from farmers in Vieras-Minas Gerais. Through microsatellite-enriched genetic libraries, 11 microsatellite markers with adequate amplification patterns were characterized. Population genetic analysis identified eight polymorphic loci that generated 66 alleles ranging from two alleles (PSCA1B3) to nine (PSCA2H1). The polymorphic information content ranged from 0.031 to 0.827. High genetic differentiation was observed between the wild population and the stocks, and moderate differentiation between the three stocks. Deviation in the Hardy-Weinberg equilibrium was observed in one locus in the wild population, in five loci in the common morphological variant, in two in the marble, and in two in the clown morphological variant. Bayesian analysis of genetic structure revealed the existence of two clusters, one represented by the natural population and the other by the stocks. The developed microsatellite markers serve as a tool for the analysis of diversity and genetic structure and conservation studies of P. scalare.

Reproductive success of released animals becomes a growing interest in the context of stock enhancement program and conservation biology. In the case of the Ezo abalone (Pacific abalone; Haliotis discus hannai), limited information is available about the extent of interbreeding between natural and released hatchery populations. This study aimed to develop microsatellite DNA markers from Ezo abalone to address the issue. We developed 38 microsatellite markers that worked well in three full-sib families established from wild caught abalone. Of these, nine markers, with which alleles could be scored easily and accurately, were applied to two population samples derived from coastal areas where intensive releases of hatchery-produced abalone seedlings have been carried out. Multilocus estimates of genetic population differentiation calculated based on the nine markers (FST analogue θ and RST analogue ρ) rejected a null hypothesis of genetic homogeneity between the populations (θ=0.048, P<0.00001; ρ=0.029, P=0.0051). Although both populations conformed to Hardy Weinberg's equilibrium at almost all of the nine loci, the results of simulation analysis for variance of relatedness coefficient provided evidence of nonrandom mating in each population, possibly caused by the cumulative effect of stocking on the genetic make-up. The population analysis presented here is preliminary; but we consider that the microsatellite markers are potentially an efficient means to examine the reproductive contribution of released abalone to natural resources.  相似文献

为了建立黄鳍棘鲷微卫星亲子鉴定技术,利用荧光引物和自动测序技术检测了自主开发的12对微卫星分子标记在505尾黄鳍棘鲷个体中的遗传多态性,并构建了亲子鉴定技术。结果显示,该研究中筛选的12个微卫星标记共检测到119个等位基因,平均等位基因数(N_a)为9.91,平均观测杂合度(H_o)为0.651,平均期望杂合度(H_e)为0.661,平均多态性信息含量(PIC)为0.621,具有丰富的多态性。此外,运用Cervus 3.0软件对已知系谱信息的112尾黄鳍棘鲷亲本和393尾子代个体进行模拟分析,结果显示,当双亲未知且置信度为95%时,12个标记的累积排除概率达99.58%;当微卫星标记数量为8时,累积排除概率达到99.1%。因此确定AL49、AL37、AL01、AL20、AL14、AL18、AL15和AL51共8个多态性较高的微卫星标记为黄鳍棘鲷微卫星亲子鉴定的核心体系。在双亲性别未知的情况下,其双亲的累积排除率为99.1%。根据黄鳍棘鲷子代的实际基因分型数据,实际鉴定率为89.31%。该研究构建的微卫星标记组合能为黄鳍棘鲷不同家系混养后的亲子鉴定、种群选育和分子辅助家系管理提供科学的技术手段。  相似文献

利用微卫星DNA分子标记对广东湛江乌石(ZJ)、广西北海山口(BH)与越南锦普(JP)3个光裸星虫(Sipunculus nudus)野生群体的遗传多样性进行分析。结果表明,20个微卫星座位中有3个位点(Snu08、Snu10、Snu14)呈现单态,17个多态微卫星座位中的等位基因数介于2~9之间,多态信息含量(PIC)介于0.097~0.797之间。每个群体中有5个位点显著偏离了Hardy-Weinberg平衡,在位点Snu18上3个群体均偏离Hardy-Weinberg平衡。3个光裸星虫野生群体均表现出中等程度的遗传多样性水平,锦普群体遗传多样性水平高于北海和湛江群体。群体间F-统计量分析表明,群体遗传分化处于低等分化水平,差异不显著(Fst=0.019~0.049,P>0.05);基于DA遗传距离构建的NJ和UPGMA聚类树均显示,地理位置相邻的群体聚在一起。研究表明,3个光裸星虫地理群体均具有中等的遗传多样性,杂合度均偏低,群体间存在较大的基因流,杂合子明显缺失;群体间已产生遗传分化,但分化水平还较低。  相似文献

Solea senegalensis is a fish species in an early stage of domestication. A commercial hatchery in southern Spain experienced a dramatic failure in spawning and embryo survival, after incorporation into the broodstock of fish reared in their hatchery. To assess the impact of management on the stock quality, a population genetics study was made of adult broodstock present at this hatchery in 2000. The broodstock was composed of a group of fish of wild origin and two more groups from F1 progeny. A set of eight microsatellite loci was used to compare genetic variability among the three groups and to establish the relationships between pairs of individuals within each group.Individuals from F1 origin showed a substantial reduction in genetic variability when compared to those from the wild. There was a greater than 50% decrease in the number of alleles per locus corrected by sample size and 16% and 26% reductions in He values in the two F1 groups, respectively. The degree of genetic relationship between individuals, evaluated by the relatedness estimator, rxy, and by graphical approaches, revealed a high proportion, almost 75%, of full-sibs and half-sibs in both F1 groups, which might account for the reduction in overall variability. The high proportion of siblings also suggests a reproductive pattern in this stock in which few individuals breed successfully. This mating behaviour, along with factors inherent to selection and management systems, might be responsible for the negative effects on the reproductive capacity of the stock.  相似文献

利用60对微卫星分子标记分别对罗氏沼虾3个养殖群体进行遗传多样性分析,通过比较样本量及标记量对遗传多样性指标的影响,探讨最适宜的样本量及标记量。结果显示,样本量和标记量的大小均对遗传多样性指数有较大的影响,其中样本量与平均等位基因数和平均有效等位基因数呈高度正相关,与杂合度和Nei氏遗传多样性指数分别呈中度、高度相关,样本量为10~30时,遗传参数变化差异显著,样本量大于30,变化差异不显著;标记量与遗传参数也存在不同程度的相关性,标记量为5~25时,各遗传多样性指数变化有明显差异,标记量大于25时,各遗传参数变化较小,多态信息含量不同的标记直接影响到群体遗传参数。性别对群体遗传多样性指标无显著性影响。研究表明,应尽可能选择多态信息含量高的微卫星分子标记开展罗氏沼虾群体遗传多样性分析,可不考虑样本性别,样本量不宜小于30,标记量不宜少于25。  相似文献

Genetic population structure of artificially isolated white-spotted charr (Salvelinus leucomaenis) populations before and after translocation were examined using polymorphic microsatellite loci. We released 20 migrant females of below-dam populations in the Kame and Hitozuminai rivers, Japan, into small above-dam sites of each river, and also collected juveniles (0+ fish) of the pre-translocation and the post-translocation cohorts for DNA analyses. Reduced genetic diversity, expressed as the number of alleles and expected heterozygosity, was observed in all above-dam populations before translocation relative to below-dam populations. Highly significant genetic differentiation was also found between above- and below-dam populations. After translocation, on the other hand, genetic diversity of above-dam populations was restored to the levels of below-dam populations. Differences in gene frequencies between above- and below-dam populations were not detected after translocation. These results, in turn, suggest that the translocations in our survey had an immediate effect on the restoration of genetic diversity, and that the translocated individuals strongly introgressed into above-dam populations.  相似文献

为明确不同选育群体中间球海胆的遗传多样性和遗传结构,利用SSR-seq技术和15个微卫星位点,对1个家系选育群体(FP)、1个群体选育群体(IP)和1个未经选育的普通养殖群体(CP)的遗传多样性及遗传结构进行了分析。结果显示,15个微卫星位点共检测出112个等位基因,FP、IP、CP 3个群体的平均观测等位基因数(Na)分别为5.077、5.133和6.133个,平均有效等位基因(Ne)分别为2.816、2.873和3.638个,平均观测杂合度(Ho)分别为0.522、0.441和0.501,平均期望杂合度(He)分别为0.595、0.599和0.667,平均多态性信息含量(PIC)分别为0.546、0.543和0.623。家系选育群体(FP) He与Ho的差值(0.073)低于IP (0.158)和CP (0.166),平均固定指数(F)(0.115)低于IP (0.248)和CP (0.246)。3个群体间遗传分化系数(Fst)介...  相似文献
